HPV Infection and Cervical Abnormalities in HIV Positive Women in Different Regions of Brazil, a Middle-Income Country

  • Human papillomavirus is a virus that is distributed worldwide, and persistent infection with high-risk genotypes (HR-HPV) is considered the most important factor for the development of squamous cell cervical carcinoma (SCC). However, by itself, it is not sufficient, and other factors may contribute to the onset and progression of lesions. For example, infection with other sexually transmitted diseases such as human immunodeficiency virus (HIV) may be a factor. Previous studies have shown the relationship between HPV infection and SCC development among HIV-infected women in many regions of the world, with great emphasis on low- and middle-income countries (LMICs). Brazil is considered a LMIC and has great disparities across different regions. The purpose of this review was to highlight the current knowledge about HPV infection and cervical abnormalities in HIV+ women in Brazil because this country is an ideal setting to evaluate HIV impact on SCC development and serves as model of LMICs and low-resource settings.

High prevalence of avian hematozoan parasite infection in wild owls in Chungbuk province of Korea (mid-South Korea)

  • Wild owls are widespread in Korea, even being common in urban areas due to the presence of hills or small mountains in and around the cities. This results in high levels of contact between owls and domestic animals. This study investigated the prevalence of avian hematozoan parasite infection in 2 common owl species in Chungbuk province of Republic of Korea for 3-year period: the Eurasian eagle owl (Bubo bubo) and the Brown hawk owl (Ninox scutulata). Peripheral blood smears taken from 56 wild owls were examined by microscopic examination and confirmed by molecular technique. Twenty (36%) of these samples tested positive for the Haemoproteus infection. The infection rate was higher in Brown hawk owls (50%) than in Eurasian eagle owls (33%). These results indicate that the wild owls may act as a reservoir for Haemoproteus infection in susceptible domestic birds.

Cytomegalovirus Infection among Pregnant Women in Beijing: Seroepidemiological Survey and Intrauterine Transmissions

  • Primary cytomegalovirus (CMV) infection during pregnancy can cause congenital defects. Available data for CMV infection during pregnancy in north China are inadequate. The aim of this study was to evaluate the epidemiology of maternal CMV infection and explore the incidence of congenital infection. In this prospective study, serum CMV IgG and IgM antibodies were measured in 2,887 pregnant women using ELISA, and the IgG avidity test was performed on all IgM-positive subjects. The seroprevalence of anti-CMV IgG was 94.70%, and of anti-CMV IgM was 1.28%. CMV IgG prevalence increased significantly with age (p < 0.01). Women living in downtown areas showed higher IgG prevalence than those residing in urban areas (p = 0.023). CMV-IgM seroprevalence was highest in autumn (p = 0.021). There was no difference in IgM seroprevalence by age, socioeconomic status, geographical area, or gravida. The rate of primary CMV infection was 0.45% (13/2,887) at the first trimester. The seroconversion rate during pregnancy was 0.76% (22/2,887). One woman underwent seroconversion during pregnancy and gave birth to an infant with asymptomatic CMV infection. Congenital CMV infection was diagnosed in five of the 14 infants from 14 mothers with active infection, for a vertical transmission rate of 35.71% (5/14). Three infants were asymptomatic, whereas two infants presented symptomatic infection with hearing deficits. Although CMV IgG prevalence is relatively high in north China, significant attention to primary CMV infection during pregnancy is still needed.

PCR을 이용한 경북 동부지역 소의 러시아범안열원충 감염률 조사 (Prevalence of Theileria sergenti infection in cattle of eastern areas in Gyeongbuk province by PCR)

  • This study was performed to determine the prevalence of Theileria sergenti (T. sergenti) in cattle reared in eastern areas of Gyeongbuk province by PCR. Among 443 samples collected from 42 cattle farms, 96 (21.7%) samples from 13 (31.0%) farms were positive for T. sergenti. By regions, 87 (26.6%) out of 327 cattle in Gyeongju, 8 (9.5%) out of 84 cattle in Pohang, and 1 (3.1%) out of 32 cattle in Ulleung were positive for T. sergenti. T. sergenti infection in dairy cattle (27.9%) was significantly higher than that in Korean cattle (9.4%, P<0.0001). Accordingly, Korean cattle were more resistant to T. sergenti infection. Prevalence of T. sergenti in cattle was increased with age (P<0.0001). The infection rate in cows (23.3%) was significantly higher than that in bulls (5.0%, P<0.01). Seasonally, prevalence of T. sergenti in cattle was highest in autumn (32.7%, P<0.01). Prevalence of T. sergenti in grazing cattle (66.7%) was significantly higher than that in non-grazing cattle (15.8%, P<0.0001). Since prevalence of T. sergenti infection is high in cattle reared in eastern areas of Gyeongbuk province, survey on other hemoparasites and appropriate control programs need to be established in this region.

Investigation of Immune Biomarkers Using Subcutaneous Model of M. tuberculosis Infection in BALB/c Mice: A Preliminary Report

  • Evaluation and screening of vaccines against tuberculosis depends on development of proper cost effective disease models along with identification of different immune markers that can be used as surrogate endpoints of protection in preclinical and clinical studies. The objective of the present study was therefore evaluation of subcutaneous model of M.tuberculosis infection along with investigation of different immune biomarkers of tuberculosis infection in BALB/c mice. Groups of mice were infected subcutaneously with two different doses : high ($2{\times}10^6CFU$) and low doses ($2{\times}10^2CFU$) of M.tuberculosis and immune markers including humoral and cellular markers were evaluated 30 days post M.tuberculosis infections. Based on results, we found that high dose of subcutaneous infection produced chronic disease with significant (p<0.001) production of immune markers of infection like $IFN{\gamma}$, heat shock antigens (65, 71) and antibody titres against panel of M.tuberculosis antigens (ESAT-6, CFP-10, Ag85B, 45kDa, GroES, Hsp-16) all of which correlated with high bacterial burden in lungs and spleen. To conclude high dose of subcutaneous infection produces chronic TB infection in mice and can be used as convenient alternative to aerosol models in resource limited settings. Moreover assessment of immune markers namely mycobacterial antigens and antibodies can provide us valuable insights on modulation of immune response post infection. However further investigations along with optimization of study protocols are needed to justify the outcome of present study and establish such markers as surrogate endpoints of vaccine protection in preclinical and clinical studies in future.

Transcription Factor PU.1 Inhibits Aspergillus fumigatus Infection via Surfactant Protein-D

  • Aspergillosis is a life-threatening disease in individuals with compromised immune systems. Fungal invasion is a highly critical process during host cellular infection. Several papers have reported that transcription factors are responsible for the infection process. To investigate what transcription factors are involved in the process in an effort to inhibit fungal infection into cells, I checked the surfactant protein family and PU.1 transcription factor levels in A549 cells infected with A. fumigatus conidia. PU.1 and surfactant protein-D levels were reduced in cells infected with fungal conidia. I then observed an increase in surfactant protein-D on PU.1-overexpressed cells. Infection of A. fumigatus conidia was decreased in PU.1-overexpressed cells, whereas the suppression of PU.1 did not lead to any changes in cases of A. fumigatus conidia infection. These results indicate that PU.1 inhibits the infection of A. fumigatus conidia via the expression of surfactant protein-D, suggesting that PU.1 is a key transcription factor for protection against A. fumigatus invasion.

치과기공사의 감염에 대한 인식과 관리 실태 연구 (A study on the perception and management of dental technician's infection prevention)

  • Purpose: The purpose of this study was to investigate dental technicians' awareness of infections and to enhance their interest and commitment towards infection prevention. Methods: A self-reported questionnaire survey was conducted among dental technicians. Total of 195 responses were used for analysis. A cross-tabulation analysis was used to compare dental technicians' awareness of infections according to their levels of education on infection prevention (α=0.05). Results: Dental technicians were educated on infection prevention (40.5%). Most participants received infection prevention education from school (29.8%). Dental laboratories had an infection control guideline (34.9%). Dental technicians were not aware of infection risks at their workplaces or believed that they were not at risk of infections at their workplaces (59.3%). Conclusion: Dental technicians must be consistently educated on infection prevention and control through systematic education at school and wider promotion through media. Research is needed to economically and efficiently improve equipment used by dental technicians and to protect them from infectious diseases. A budget for infection prevention must also be determined. Institutional measures such as providing support for dental technicians at the policy level and developing a basic infection prevention manual are necessary. Solving these issues not only protects dental technicians from infections but also allows them to provide high-quality medical services.

Distribution of Oncogenic Human Papillomavirus Genotypes at High Grade Cervical Lesions above CIN 2 Grade with Histological Diagnosis

  • High risk human papillomavirus (HR-HPV) is major risk factor for uterine cervical cancer. There are approximately 15 types of HR-HPV. Liquid based cytology samples (116 samples) with high grade cervical lesions belonging to cervical intraepithelial neoplasia (CIN) 2, CIN 3, carcinoma in situ (CIS) and squamous cell carcinoma (SCC) were used after histologic confirmation. HR-HPV genotype assay was conducted using DNA chips. The HR-HPV infection rate was 81.9% with SCC samples showing the highest HR-HPV infection rate of 31%. CIN 3, CIS and CIN 2 showed infection rates of 25%, 16.4% and 9.5%, respectively. According to age with HR HPV infection rate, the 30~39 years-old group showed the highest infection rate by 92.3%. According to distribution with HR HPV genotyping, HPV 16 showed the highest infection rate by 42.3% whereas HPV 33 and HPV 58 showed infection rates of 11.7% and 10.8%, respectively. HPV 18 which is the second most common infected HPV genotype in the world showed 3.6%. Of the three most common oncogenic HR-HPV genotypes in CIN 2, we detected HPV 16, 35, 58; CIN 3 was HPV 16, 33, 58; CIS was HPV 16, 58, 33 (35/52); and SCC was HPV 16, 33, and 18 (31/52/58). Among the HPV 18, CIN 2, CIN 3, CIS and SCC showed 0.9%, 0.9%, 0% and 1.8%, respectively. The most often used preventive vaccines for cervical cancers use HPV 16 and HPV 18 as targets. However, results derived from this study suggest that a preventive vaccine against HPV 16 and HPV 18 would not be optimal for populations in this study.

Nucleotide-binding oligomerization domain 1 is dispensable for host immune responses against pulmonary infection of Acinetobacter baumannii in mice

  • Nucleotide-binding domain 1 (Nod1) is a cytosolic receptor that is responsible for the recognition of a bacterial peptidoglycan motif containing meso-diaminophimelic acid. In this study, we sought to identify the role of Nod1 in host defense in vivo against pulmonary infection by multidrug resistant Acinetobacter baumannii. Wildtype (WT) and Nod1-deficient mice were intranasally infected with $3{\times}10^7CFU$ of A. baumannii and sacrificed at 1 and 3 days post-infection (dpi). Bacterial CFUs, cytokines production, histopathology, and mouse ${\beta}$-defensins (mBD) in the lungs of infected mice were evaluated. The production of cytokines in response to A. baumannii was also measured in WT and Nod1-deficient macrophages. The bacterial clearance in the lungs was not affected by Nod1 deficiency. Levels of IL-6, $TNF-{\alpha}$, and $IL-1{\beta}$ in the lung homogenates were comparable at days 1 and 3 between WT and Nod1-deficient mice, except the $TNF-{\alpha}$ level at day 3, which was higher in Nod1-deficient mice. There was no significant difference in lung pathology and expression of mBDs (mBD1, 2, 3, and 4) between WT and Nod1-deficient mice infected with A. baumannii. The production of IL-6, $TNF-{\alpha}$, and NO by macrophages in response to A. baumannii was also comparable in WT and Nod1-deficient mice. Our results indicated that Nod1 does not play an important role in host immune responses against A. baumannii infection.

실험용 흰쥐의 내부기생충 감염실태조사 (Prevalence of internal parasites in the laboratory rats)

  • A survey on the prevalence of internal parasites in the laboratory rats in the Chonnam and Kyunggi areas was carried out. A total of 208 rats was selected for necropsy and the parasites were collected from the organs and the fecal samples. The infection rate of the internal parasites was high as 63.9% and eight species of parasites were identified. Among the species identified A tetraptera(21.6%) and H diminuta(20.7%) appeared with relatively higher infection rates than any other species identified. A significant difference in numbers of parasites was recognized between the regions as 8 species in Chonnam and 4 species in Kyunggi. The laboratory rats reared in stainless steel cage showed the higher infection rate than those in polycarbonate cage, with some exception in H spumosa and H nana. In this survey T crassicauda was firstly in the laboratory rats in Korea.
