This research aims to develop a multiple channel electrochemical DNA chip using micro- fabrication technology. At first, we fabricated a high integrated type DNA chip array by lithography technology. Several probe DNAs consisting of thiol group at their 5-end were immobilized on the gold electrodes. Then target DNAs were hybridized by an electrical force. Redox peak of cyclic-voltammogram showed a difference between target DNA and mismatched DNA in the anodic peak current. Therefore. it is able to detect a various genes electrochemically after immobilization of a various probe DNA and hybridization of label-free DNA on the electrodes simultaneously. It suggested that this DNA chip could recognize the sequence specific genes.
Detection of exon 19 deletion mutation in the epidermal growth factor receptor (EGFR) gene, which results in increased and sustained phosphorylation of EGFR, is important for diagnosis and treatment guidelines in non-small-cell lung cancer. Here, we have developed a simple and convenient detection system using the interaction between G-quadruplex and fluorophore thioflavin T (ThT) for discriminating EGFR exon 19 deletion mutant DNA from wild type without a label and quencher. In the presence of exon 19 deletion mutant DNA, the probe DNAs annealed to the target sequences were transformed into G-quadruplex structure. Subsequent intercalation of ThT into the G-quadruplex resulted in a light-up fluorescence signal, which reflects the amount of mutant DNA. Due to stark differences in fluorescence intensity between mutant and wild-type DNA, we suggest that the induced G-quadruplex structure in the probe DNA can report the presence of cancer-causing deletion mutant DNAs with high sensitivity.
현재까지 연구 개발된 바이오칩 및 센서의 경우에는 생체분자 상호작용의 분석을 수행하기 위해서 효소나 형광 물질 등과 같은 표지물질을 생체분자에 주입할 필요성이 있었다. 이러한 표지작업은 단백질 등과 같이 고차구조를 형성하는 생체분자에 있어서 그 분자인식능을 저하시키는 문제가 발생하게 된다. 그리고 표지작업은 일련의 조작이 필요하기 때문에 조작의 복잡성을 띄게 되고, 간편성을 저해하는 문제가 발생하게 되며, 분석 결과를 얻기 위해서는 장시간을 필요로 하게 된다. 또한, 생체분자 상호작용의 분석에 적용되는 측정장치도 대형화하게 되어 온사이트 모니터링에 이용하기 어렵다. 이러한 문제점들을 해결하기 위해서 비표지로 생체분자의 상호작용 분석이 가능한 SPR 광학특성, QCM 및 전기화학법 등을 이용한 비표지 바이오칩 및 센서가 개발되었다. 하지만 표지 바이오칩 및 센서와 마찬가지로 장치의 대형화 및 복잡화, 간편성 및 감도 등에 문제가 있었다. 따라서 지금까지 개발되어진 표지 및 비표지 바이오칩의 문제들을 해결하기 위해서 나노구조에서만 발현되는 새로운 광학특성인 LSPR을 기반으로 하는 새로운 형태의 코어-쉘 구조 나노입자 바이오칩이 제작되었다. 코어-쉘 나노입자 바이오칩의 표면에 수직방향으로부터 입사광을 조사하고 바이오칩 표면으로부터 반사된 반사광을 검출기로 검출하여 흡수 스펙트럼을 소형의 분광기로 해석함으로서 코어-쉘 나노입자 바이오칩 기반 비표지 광학 바이오센서를 완성하였다. 또한 단백질 항원-항체 반응에 대한 비표지 검출 및 정량특성을 평가한 결과, 감도, 간편성, 유연성, 폭넓은 응용성 등에 양호한 특성을 확인할 수 있었다. 이상에서 살펴본 바와 같이, 코어-쉘 나노입자 바이오칩 기반 비표지 광학 바이오센서는 생체분자 상호작용의 분석에 많이 이용되고 있는 단백질, DNA, 세포 등의 생체분자에 유연하게 대처할 수 있을 것으로 생각되어지며, 그 밖에 의료, 식품분석, 환경 및 공정 모니터링 등 분야에 폭넓게 이용될 것으로 기대되고 있다. 또한 본 코어-쉘 나노입자 바이오칩 기반 비표지 광학 바이오센서는 소형으로 저렴한 분광기를 이용하여 측정을 실시하고 있기 때문에 온사이트 모니터링에의 적용도 가능할 것으로 생각된다.
청과물의 품질 영양표시 모델을 개발하기 위하여 경북의성군 옥자두를 대상으로 품질 및 영양적 성분을 분석하고 품질 영양표시 모델을 확립하였다. 일반성분은 수분 92%, 조단백 0.74%, 조지방 0.64%, 조섬유 1.65%, 조회분 0.32%, 무질소물 4.29%로 확인되었으며, 열량은 32kcal/100g 으로 계산되었다. 당도는 $10.33{\pm}0.85Brix^{\circ}$로, 산도는 $1.08{\pm}0.12%$로 나타났으며, 자두의 비타민 C 함량은 $7.037{\pm}0.317mg%$로 나타났다. 무기질은 황, 칼륨, 칼슘, 나트륨, 마그네슘 등의 순으로 함량이 높았고, 주요 유리당으로는 과당, 포도당, 자당로 나타났다. 유기산으로는 호박산, 사과산, 구연산이 확인되었다. 총 페놀화합물의 함량은 과육과 과피 간에 현저하게 차이를 보였으며 전과로는 $75.55{\pm}0.73mg%$의 함량을 보였다. 품질 표시에는 당과 산 함량, 영양 정보 표시에는 열량, 미네랄, 비타민 C, 총 페놀 성분이 표기된 자두의 품질 영양 표시 모델을 개발하여 제시하였다.
Strong interactions between electromagnetic radiation and electrons at metallic interfaces or in metallic nanostructures lead to resonant oscillations called surface plasmon resonance with fascinating properties: light confinement in subwavelength dimensions and enhancement of optical near fields, just to name a few [1,2]. By utilizing the properties enabled by geometry dependent localization of surface plasmons, metal photonics or plasmonics offers a promise of enabling novel photonic components and systems for integrated photonics or sensing applications [3-5]. The versatility of the nanoplasmonic platform is described in this talk on three folds: our findings on an enhanced ultracompact photodetector based on nanoridge plasmonics for photonic integrated circuit applications [3], a colorimetric sensing of miRNA based on a nanoplasmonic core-satellite assembly for label-free and on-chip sensing applications [4], and a controlled fabrication of plasmonic nanostructures on a flexible substrate based on a transfer printing process for ultra-sensitive and noise free flexible bio-sensing applications [5]. For integrated photonics, nanoplasmonics offers interesting opportunities providing the material and dimensional compatibility with ultra-small silicon electronics and the integrative functionality using hybrid photonic and electronic nanostructures. For sensing applications, remarkable changes in scattering colors stemming from a plasmonic coupling effect of gold nanoplasmonic particles have been utilized to demonstrate a detection of microRNAs at the femtomolar level with selectivity. As top-down or bottom-up fabrication of such nanoscale structures is limited to more conventional substrates, we have approached the controlled fabrication of highly ordered nanostructures using a transfer printing of pre-functionalized nanodisks on flexible substrates for more enabling applications of nanoplasmonics.
Transfer printing is a method to combine printing and dyeing technology by the use of sublimation. It is an environmentally-friendly printing method that saves costs, reduces the production processes by the omission of the washing process, and saves time by maintaining quality. Due to the development of transfer printing, a high value added printing technology is available now but color fastness to sublimation of the printing products is still low since there are few dyes that have an affinity to the fabrics and the application technology is still inadequate. Specially, in case of high concentration black dyes, eco-label type black dyes, which is a substitution for general dispersal dyes, have been developed while general dispersal black dyes are still used, creating issues such as color differences on the surface and back side of the fabrics and contamination by friction after transfer printing. There are also some restricted substances such as allergens. To address these issues, high concentration black dyes and application technology that are environmentally-friendly and that have over 16 K/S through the use of single dyes with excellent color fastness, fixation ability, and similar melting temperature were developed for this study.
The purpose of this thesis is to suggest a Practical Plan for ethical consumption by reviewing the category of Vegan Fashion and investigating its Social Value of vegan fashion. This will be achieved through investigating the papers and official home pages of 13 selected Vegan Fashion brands. It was found that in terms of use of materials such as leather, fur and organic fibers the brands can be divided into three sections: fur-free, cruelty-free and perfect vegan. A Practical Plan is suggested based on the aspects of production, consumption, distribution and education. Firstly, the provider should be required to understand vegan materials deeply, it is also desirable for them to get vegan certifications. Secondly, the seller should also understand about vegan materials, and be able to explain this to consumers. The education from the seller is vital and the meaning of logos and associated contents used by the label should be clearly explained to consumers. Thirdly, the association of consumers, and fashion brands should cooperate to enhance the level of general understanding in society further, this should influence new laws, that address ethical issues regarding the use of fur in fashion. Environmental problem of the future might be reduced if the stakeholders in Vegan Fashion are cooperatively and actively trying to educate the general population and make Vegan Fashion popular and ethical consumption popular.

Purpose: To evaluate effects of metformin on clinical outcome of non-diabetic patients with stage IV NSCLC. Materials and Methods: A prospective, randomized, open-label, controlled pilot study was conducted on patients with stage IV NSCLC with an Eastern Cooperative Oncology Group Performance Status (ECOG PS) of 0-2, excluding patients with diabetes and lactic acidosis. Thirty chemo-$na\ddot{i}ve$, non-diabetic patients with stage IV NSCLC were enrolled. Fifteen patients received intravenous gemcitabine/cisplatin regimen alone (arm B) while fifteen patients received the same regimen plus daily oral metformin 500mg (arm A). The effect of metformin on chemotherapy-response rates, survival, and adverse events in these patients was evaluated. Results: Objective response rate (ORR) and median overall survival (OS) in arms A and B were 46.7% versus 13.3% respectively, p=0.109 and 12 months versus 6.5 months, respectively, p=0.119. Median progression free survival (PFS) in arms A and B was 5.5 months versus 5 months, p=0.062. No significant increase in toxicity was observed in arm A versus arm B. Percentage of patients who experienced nausea was significantly lower in arm A versus arm B, at 26.7% versus 66.7% respectively, p=0.028. Conclusions: Metformin administration reduced occurrence of chemotherapy induced-nausea. Non-statistically significant improvements in the ORR or OS were observed. Metformin had no effect on PFS.
Surface enhanced Raman scattering (SERS) is considered as one of promising medical and diagnostic technologies. The SERS effect is caused by the localized surface plasmon resonance (LSPR) from metal nanoparticles with narrow hot spots. The mechanism of LSPR, development of nanostructure fabrication, and corresponding researches are discussed. The flexible, label-free, low-cost, and highly-sensitive Au/ZnONRs/G is introduced. The Au/ZnONRs/G detects and distinguishes cataract, age-related macular degeneration, and diabetic macular edema from aqueous humor. Comprehension of SERS provides further improvement in bio sensing technology including early diagnosis and prolonged life expectancy.realize highly stretchable electrodes.
In this study, DNA-templated copper nanoclusters (DNA-CuNCs) were used to detect exonuclease III (Exo III) activity, which is important for the diagnosis and therapy of several diseases. The results of this study showed that Exo III was affected by the concentrations of magnesium ions and sodium ions, and its optimal conditions for cleavage were $5mM\;Mg^{2+}$ and less than $25mM\;Na^+$. With a blunt-end DNA, more than 98% of DNA was digested by Exo III. As expected, with two or four cytosines in the terminal position of a 4-base overhanging DNA such as 5'-GGCC-3' and 5'-CCCC-3', there was little cleavage by Exo III compared with a blunt-end DNA.
