Molecular and Morphological Characteristics of a New Species Collected from an Insect (Cicindela transbaicalica) in Korea

  • To exploit insect-derived fungi, insects were collected from seven different regions in Korea, including Gyeongbuk, Goryeong, and several fungi were isolated from them. A fungal strain designated 21-64-D was isolated from riparian tiger beetle (Cicindela transbaicalica) and morphologically identified as a species belonging to the genus Oidiodendron. Phylogenetic analysis using the nucleotide sequences of internal transcribed spacer (ITS) regions and the partial sequence of the large subunit of the nuclear ribosomal RNA (LSU) gene revealed the distinct phylogenetic position of the isolate among recognized Oidiodendron species including its closest neighbors O. chlamydosporicum, O. citrinum, O. maius, and O. pilicola. The hyphal and conidial morphology of the strain, particularly club-shaped hyphae, clearly differentiated it from its close relatives. Results indicated that 21-64-D is a novel species in the genus Oidiodendron, for which the name Oidiodendron clavatum sp. nov. is proposed.

Isolation and Identification of Two Unreported Sordariomycetes Fungi in Korea: Pestalotiopsis clavata and Botryotrichum iranicum

  • Two fungal strains, designated KNUF-21-006 and KNUF-21-028, were isolated from soil samples collected from Gyeongbuk Province, Korea. The strain KNUF-21-006 was similar to other Pestalotiopsis species in terms of morphological characteristics, including whitish to pale brown mycelium, conidial shape, and size. The isolate had aerial hyphae that produced black fruiting bodies on the mycelium. The conidia were fusoid to ellipsoid, four-septate, and appendage-bearing. Phylogenetic analysis using the internal transcribed spacer (ITS) region, translation elongation factor 1-alpha (TEF), and β-tubulin (TUB) gene sequences confirmed that the closest relationship of the isolate at the species level was with Pestalotiopsis clavata. The strain KNUF-21-028 exhibits similar morphological characteristics to other Botryotrichum species, including white aerial mycelium with sulcate and irregular margins, conidial shape, and size. The conidia were globose, single, and hyaline. Upon molecular analysis-using the ITS region, large subunit (LSU) rRNA gene, and TUB gene sequences-the fungus was identified as Botryotrichum iranicum. This is the first record of these fungal species in Korea.

Unveiling the Diversity of Hydnum in the Republic of Korea with One New Species, Hydnum paucispinum

  • Hydnum is a genus of ectomycorrhizal fungi belonging to the Hydnaceae family. It is widely distributed across different regions of the world, including North America, Europe, and Asia; however, some of them showed disjunct distributions. In recent years, with the integration of molecular techniques, the taxonomy and classification of Hydnum have undergone several revisions and advancements. However, these changes have not yet been applied in the Republic of Korea. In this study, we conducted an integrated analysis combining the morphological and molecular analyses of 30 specimens collected over a period of approximately 10 years in the Republic of Korea. For molecular analysis, the sequence data of the internal transcribed spacer (ITS) region, the large subunit of nuclear ribosomal RNA gene (nrLSU), and a portion of translation elongation factor 1-a (TEF1) were employed as molecular markers. Through this study, we identified eight species that had previously not been reported to occur in the Republic of Korea, including one new species, Hydnum paucispinum. A taxonomic key and detailed descriptions of the eight Hydnum species are provided in this study.

Morphological and molecular characterization of root-lesion nematode Pratylenchus hippeastri from Korea

  • The root-lesion nematode Pratylenchus spp. is the most important plantparasitic nematode due to its worldwide distribution, wide host ranges, and migratory endoparasitic characteristics. One population of Pratylenchus collected from the giant pussy willow (Salix chaenomeloides Kimura) in the Andong area as part of a nematode survey in Korea was characterized morphologically and by molecular methods. The analysis of morphological measurements and morphometric characteristics, as well as DNA sequencing of the rRNA large subunit (LSU) D2/D3 expansion segments and the internal transcribed spacer (ITS) gene sequence, confirmed the identity of this population as P. hippeastri. This study is the first report of P. hippeastri associated with Salix chaenomeloides in Korea and worldwide. Further studies on distribution and pathogenicity in different P. hippeastri host crops, such as grapevines, strawberries, and apples, are necessary. The taxonomic keys to 16 Pratylenchus species in Korea are provided.

거제도 장목항에서 적조원인생물 Akashiwo sanguinea(Dinophyceae): 형태, 분자계통학적 특성 및 온도와 염분에 따른 성장 특성 (Bloom-forming dinoflagellate Akashiwo sanguinea(Dinophyceae) in Jangmok Harbour of Geoje Island, Korea: Morphology, phylogeny and effects of temperature and salinity on growth)

  • 거제도 장목항에서 분리한 Akashiwo sanguinea의 형태와 계통학적 특성을 명확히 하고, 여러 온도와 염분구배에 따른 성장조건을 파악하고자 하였다. A. sanguinea의 세포는 오각형이었고, 세포의 길이는 54.7~70.3 ㎛, 폭은 31.5~48.5 ㎛로 나타났다. 핵은 세포의 중심에 위치하였고, 엽록체는 황갈색으로 세포 전체에 퍼져있었다. 상추구는 알파벳 e 모양이었다. 계통분석 결과 장목항에서 분리한 본 배양주는 ribotype A에 포함되었다. 온도 및 염분구배에 따른 성장 실험은 5℃ 이하의 온도를 제외한 모든 온도구배에서 성장이 나타났다. 그리고, 최대성장속도는 온도 20℃, 염분 20 psu에서 0.50 day-1로 나타났고, 최대세포밀도는 온도 25℃, 염분 30 psu에서 1,372 cells mL-1였다. 이 결과는 A. sanguinea가 가을철에 한국 연안에서 최대 증식을 보일 수 있다는 것을 나타낸다.

남해안에서 분리한 유독 와편모조류 Gymnodinium catenatum Graham (Dinophyceae): 형태, 분자계통학적 특성 및 온도와 염분에 따른 성장 특성 (Toxic dinoflagellate Gymnodinium catenatum Graham(Dinophyceae) from the southern coast of Korea: morphology, phylogeny and effects of temperature and salinity on growth)

  • 남해안에서 분리한 Gymnodinium catenatum의 형태, 계통학적 분석 및 다양한 온도 및 염분에 반응하는 성장 조건을 파악하였다. G. catenatum의 세포는 세로로 길거나 세로와 가로의 길이가 유사한 오각형이었다. 세포의 길이는 $38.1{\sim}77.4{\mu}m$, 폭은 $26.1{\sim}40.8{\mu}m$로 나타났다. 핵은 세포의 중간에 위치하였고, 상추구는 말굽의 편자 모양이었고, 이는 국내외 배양주와 형태적으로 매우 유사한 결과 였다. 계통분석 결과도 염기유사도를 비교해 보았을 때, 기존에 보고된 배양주와 100% 일치하여 이 종은 단일 계통으로 판단되었다. 온도와 염분에 대한 성장실험에서 G. catenatum은 $15^{\circ}C$ 이상의 온도에서 염분 15 psu를 제외한 모든 염분구간에서 성장을 보였으며, 고온인 $30^{\circ}C$에서는 성장을 하지 않았다. 최대성장속도는 온도 $25^{\circ}C$, 염분 35 psu에서 $0.37day^{-1}$로 나타났고 최대세포밀도는 온도 $20^{\circ}C$, 염분 25에서 $1,073cells\;mL^{-1}$였다. 이 결과는 G. catenatum이 한국 남해안에서 여름철 및 가을철, 특히 평균 수온이 $20^{\circ}C$ 이상인 여름철에 최대 증식을 보일 수 있다는 것을 나타낸다.

Single-cell PCR을 이용하여 분석한 득량만 Chattonella 종 (Raphidophyceae)의 분자계통학적 특성 (Molecular Phylogeny of Chattonella (Raphidophyceae) Species from Deungnyang Bay, Korea Using Single-Cell PCR)

  • 최근 우리나라 연안에서 출현빈도가 점차 늘어나고 있는 침편모조류에 속하는 Chattonella는 대표적인 유해조류 중 하나로, 이들 종은 세포벽이 없어, 단순히 세포의 형태나 크기 등 광학현미경 관찰만으로는 정확하게 동정하는 것이 어렵다. 따라서 본 연구에서는 2017년 득량만에서 발생한 Chattonella 적조 시료를 대상으로 단일 세포를 분리하고, 이들 시료의 28s rDNA, rbcL, psaA 영역을 대상으로 single-cell PCR 기법을 이용하여 종 동정을 실시하였다. 현미경 관찰 결과 장축은 평균 $74.0{\pm}10.1{\mu}m$이고 단축은 평균 $33.1{\pm}3.6{\mu}m$로 일반적인 Chattonella의 형태적 특징을 보였다. 28s rDNA, rbcL, psaA 영역을 대상으로 한 염기서열 비교 결과에서는 세 영역 모두에서 하나의 종으로 명확히 구분되지는 않았다. 하지만 C. marina, C. marina var. antiqua, C. marina var. ovata 그룹과 99~100 % 높은 서열 유사성을 보였다.

국내에 서식하는 꽃에서 분리한 야생 효모 분포 및 종 다양성 (Distribution and Species Diversity of Wild Yeasts Isolated from Flowers in Korea)

  • 국내에 서식하는 꽃에서 다양한 토착 효모를 분리 및 확보하고 효모의 분포 특성과 종 다양성에 관해 연구하였다. 국내 25지역에서 꽃 9종 77개체를 채집한 후 효모를 순수분리하였다. 효모 502균주의 large-subunit (LSU) rDNA 염기서열을 분석 및 상동성 비교로 각 효모의 종을 동정하였다. 분리된 효모는 총 50종으로 동정 되었고, 모든 꽃 시료로부터 Aureobasidium pullulans, A. leucospermi, Filobasidium magnum이 분리되었다. 꽃에 우점한 효모 3종을 제외하고, 효모 균주 수가 적게 분리된 종은 지역과 분리원에 따른 유의성을 확인할 수는 없었다. 본 연구를 통해 꽃으로부터 확보한 국내 토착 효모 50종은 산업적 활용을 위한 자원으로써 역할을 기대할 수 있고, 추후 효모와 기주 간의 상호작용이나 효모다양성 연구의 기초자료로써 활용될 수 있을 것이다.

New Species of the Genus Metschnikowia Isolated from Flowers in Indonesia, Metschnikowia cibodasensis sp. nov.

  • A novel species, Metschnikowia cibodasensis, is proposed to accommodate eight strains (ID03-$0093^T$, ID03-0094, ID03-0095, ID03-0096, ID03-0097, ID03-0098, ID03-0099, and ID03-0109) isolated from flowers of Saurauia pendula, Berberis nepalensis, and Brunfelsia americana in Cibodas Botanical Garden, West Java, Indonesia. The type strain of M. cibodasensis is ID03-$0093^T$ (= NBRC $101693^T$ =UICC $Y-335^T$ = BTCC-$Y25^T$). The common features of M. cibodasensis are a spherical to ellipsoidopedunculate shaped ascus, which contains one or two needle-shaped ascospores, and lyse at maturity. Asci generally develop directly from vegetative cells but sometimes from chlamydospores. The neighbor-joining tree based on the D1/D2 domain of nuclear large subunit (nLSU) ribosomal DNA sequences strongly supports that M. cibodasensis (eight strains) and its closest teleomorphic species, M. reukaufii, are different species by a 100% bootstrap value. The type strain of M. cibodasensis, ID03-$0093^T$, differed from M. reukaufii NBRC $1679^T$ by six nt (five substitutions and one deletion) in their D1/D2 region of nLSU rDNA, and by 18 nt (five deletions, four insertions, and nine substitutions) in their internal transcribed spacer regions of rDNA, respectively. Four strains representative of M. cibodasensis (ID03-$0093^T$, ID03-0095, ID03-0096, and ID03-0099) showed a mol% G+C content of $44.05{\pm}0.25%$, whereas that of M. reukaufii NBRC $1679^T$ was 41.3%. The low value of DNA-DNA homology (5-16%) in four strains of M. cibodasensis and M. reukaufii NBRC $1679^T$ strongly supported that these strains represent a distinct species.

Production of a New Biosurfactant by a New Yeast Species Isolated from Prunus mume Sieb. et Zucc.

  • Biosurfactants reduce surface and interfacial tension due to their amphiphilic properties and are an eco-friendly alternative for chemical surfactants. In this study, a new yeast strain JAF-11 that produces a biosurfactant was selected using drop collapse method, and the properties of the extracts were investigated. The nucleotide sequences of the strain were compared with closely related strains and identified based on the D1/D2 domain of the large subunit ribosomal DNA (LSU) and internal transcribed spacer (ITS) regions. Neodothiora populina CPC 39399T, the closest species with strain JAF-11, showed a sequence similarity of 97.75% for LSU and 94.27% for ITS, respectively. The result suggests that the strain JAF-11 represents a distinct species that cannot be assigned to any existing genus or species in the family Dothideaceae. Strain JAF-11 produced a biosurfactant reducing the surface tension of water from 72 mN/m to 34.5 mN/m on the sixth day of culture and the result of measuring the critical micelle concentration (CMC) by extracting the crude biosurfactant was found to be 24 mg/l. The molecular weight 502 of the purified biosurfactant was confirmed by measuring the fast atom bombardment mass spectrum. The chemical structure was analyzed by measuring 1H nuclear magnetic resonance (NMR), 13C NMR, and two-dimensional NMRs of the compound. The molecular formula was C26H46O9, and it was composed of one octanoyl group and two hexanoyl groups to myo-inositol moiety. The new biosurfactant is the first report of a compound produced by a new yeast strain, JAF-11.
