Inheritance between Le Gene and Ti Gene in Soybean (Glycine max L.)

  • Lectin protein and Kunitz trypsin inhibitor (KTI) protein of mature soybean seed are a main antinutritional factor in soybean seed. The Le gene controls a lectin protein and Ti gene controls the KTI protein in soybean. Ti locus has been located on linkage group 9 in the classical linkage map of soybean. Position of Le locus on linkage map was not identified. Genetic relationship between Ti locus and Le locus could be useful in soybean breeding program for the genetic elimination of these factors. The objective of this study was to determine the independent inheritance or linkage between Ti locus and Le locus in soybean seed. Two $F_2$ populations were developed from three parents (Gaechuck#1, T102, and PI548415). The $F_1$ seeds from Gaechuck#1 (titiLeLe) ${\times}$ T102 (TiTilele) and Gaechuck#1 (titiLeLe) ${\times}$ PI548415 (TiTilele) were obtained. The lectin and KTI protein were analysed from $F_2$ seeds harvested from the $F_1$ plants to find independent assortment or linkage between Ti locus and Le locus. The segregation ratios of 3 : 1 for Le locus (129 Le_ : 44 lele) and Ti locus (132 Ti_ : 41 titi) and were observed. The segregation ratios of 9 : 3 : 3 : 1 (95 Le_Li_ : 34 Le_titi: 37 leleTi_ : 7 leletiti) between Le gene and Ti gene in $F_2$ seeds were observed. This data showed that Ti gene was inherited independently with the Le gene in soybean. These results will be helpful in breeding program for selecting the line with lacking both KTI and lectin protein in soybean.

Helicobacter pylori에 감염된 소아의 위점막에서 Lewis 항원의 발현 (Expression of lewis antigen in gastric mucosa of children with Helicobacter pylori infection)

  • 목 적 : Lewis b($Le^b$) 항원과 BabA 단백은 H. pylori가 위점막에 부착하는 것과 연관이 있는 것으로 알려져 있다. 소아에서 H pylori 감염에 대한 위점막의 면역 반응은 성인에서의 반응과 차이가 있을 것으로 추정되지만 Le 항원의 발현에 관한 연구는 매우 드물다. 이에 저자들은 국내 소아 H. pylori 양성 환자의 위점막에서 Le 항원의 발현 양상을 알아보기 위하여 본 연구를 시행하였다. 방 법 : 2002년 1월부터 2004년 1월까지 상부 위장관 증상 때문에 소아과를 방문하여 내시경 검사를 시행받은 소아 환자를 대상으로 하였다. 위전정부에서 최소한 2개 이상의 조직 검체를 생검하여 CLO 검사와 조직 특수 염색(Warthin-starry) 검사를 각각 시행하여 모두 양성인 경우에 H. pylori에 감염된 것으로 판정하였다. H. pylori 양성 35명과 H. pylori 음성 19명의 위점막 파라핀 조직에서 DNA를 추출하여 cagA, babA2 특이 시발체를 이용하여 PCR을 시행하였다. 위점막 생검 조직을 대상으로 $Le^a$, $Le^b$, $Le^x$$Le^y$ 항체를 사용하여 면역조직화학적 염색을 시행한 다음 각각의 Le 항원의 발현 정도를 평가하였다. 결 과 : H. pylori 양성 환자에서 $Le^a$ 항원은 60%(21명/35명), $Le^b$ 항원 97%(34명/35명), $Le^x$ 항원 88%, $Le^y$ 항원은 100%에서 발현이 확인되었다. H. pylori 음성 환아군에서는 $Le^a$, $Le^b$, $Le^x$$Le^y$ 항원이 각각 52%, 100%, 89%, 100%에서 발현되었다. H. pylori 양성 유무에 따른 $Le^b$ 항원 발현의 차이는 없었다. CagA 및 babA2 유전자는 H. pylori 양성 환자군 중 각각 65.7%(23명/35명), 25.6%(8명/35명)에서 확인되었다. Lewis 항원들의 발현에 따른 H. pylori 감염, cagA 양성 및 babA2 양성율의 차이는 없었다. 결 론 : 국내 소아의 위점막에서 높은 $Le^x$$Le^y$ 항원의 발현율을 확인할 수 있었으나 H. pylori 감염 및 babA2의 양성 여부와는 관련이 없었다. Lewis 항원 이외의 다른 점막 수용체들과 세균 독소들에 대한 지속적 연구가 필요할 것으로 생각된다.

Effects of a lipid-encapsulated zinc oxide supplement on growth performance and intestinal morphology and digestive enzyme activities in weanling pigs

  • This study compared the effects of varying lipid content and dietary concentration of a lipid-encapsulated (LE) ZnO product to those of native ZnO and thereby to find insights into optimal lipid coating and dosage of the Zn supplement. A total of 192 21-d-old weanling pigs were allotted to 48 pens, after which each six pens received a ZnO-free basal diet supplemented with 125 ppm ZnO (100 ppm Zn; BASAL), 2,500 ppm Zn as native ZnO (HIGH), or 100 or 200 ppm Zn as LE ZnO (LE-100 or LE-250) containing 8%, 10%, or 12% lipid [LE-8%, LE-10%, or LE-12%, respectively; $2{\times}3$ factorial arrangement within the LE-ZnO diets (LE-ALL)] for 14 d. Forty pigs were killed at the end for histological and biochemical examinations. None of ADG, ADFI, gain:feed, and fecal consistency score differed between the LE-ALL and either of the BASAL and HIGH groups. Hepatic and serum Zn concentrations were greater (p <0.05) in the HIGH vs. LE-ALL group, but did not differ between LE-ALL and BASAL, between LE-100 and -250, or among LE-8%, -10%, and -12% groups. Villus height (VH), crypt depth (CD), and the VH:CD ratio in the duodenum, jejunum, and ileum did not differ between the LE-ALL and either of the BASAL and HIGH groups, except for a greater CD in the duodenum in the LE-ALL vs. HIGH group. Additionally, VH and CD in the duodenum and VH:CD in the jejunum were greater in the LE-250 vs. LE-100 group. Specific activities of sucrase, maltase, and leucine aminopeptidase in these intestinal regions and those of amylase and trypsin in the pancreas were not influenced by the lipid content or dietary concentration of LE ZnO and also did not differ between the LE-ALL and either of the BASAL and HIGH groups, except for a greater pancreatic amylase activity in the former vs. HIGH group. In conclusion, the present results indicate that the LE ZnO, regardless of its lipid percentage or supplementation level examined in this study, has no significant effect on growth performance, fecal consistency, or digestive enzyme activities of weanling pigs under the experimental conditions.

Improved Understanding of LeTID of Single-crystalline Silicon Solar Cell with PERC

  • Light elevated temperature induced degradation (LeTID) was noted as an issue in multi-crystalline silicon solar cells (MSSC) by Ram speck in 2012. In contrast to light induced degradation (LID), which has been researched in silicon solar cells for a long time, research about both LeTID and the mechanism of LeTID has been limited. In addition, research about LeTID in single-crystalline silicon solar cells (SSSC) is even more limited. In order to improve understanding of LeTID in SSSC with a passivated emitter rear contact (PERC) structure, we fabricated four group samples with boron and oxygen factors and evaluated the solar cell characteristics, such as the cell efficiency, $V_{oc}$, $I_{sc}$, fill factor (FF), LID, and LeTID. The trends of LID of the four group samples were similar to the trend of LeTID as a function of boron and oxygen.

식물체(솔잎, 자초)의 에탄올 추출물이 유탁액의 지방산화에 미치는 영향 (Effect of Ethanol Extracts in Pinus densiflora, Lithospermum erythrorhizon on the Lipid Oxidation of Oil Emulsion)

  • This study was carried out to investigate the effects of ethanol extracts on lipid oxidation of oil emulsion. The results are as follows; The scavenging ability of plant extracts for hydroxyl radical was found, and plant extracts played an important role as a strong chelating agents to bind iron if Fe2+ ion exists in oil emulsion. Pinus densiflora(PD), Lithospermum erythrorhizon(LE) and PD+LE acted as strong chelating agents to bind iron to reduce lipid oxidation in oil emulsion. The content of Fe2+ ion in ethanol extracts from LE and PD+LE were significantly higher(p<0.05) than that of ethanol extracts from PD. The content of total iron has same tendency. The ascorbic acid content of PD(16.36ppm) was slightly higher than those of LE(13.08ppm). Electron donating ability of PD was significantly higher(p<0.05) than those of LE. However, the superoxide(SOD) like ability of LE showed a little higher than those of LE and PD+LE, which means the strong antioxidant activity of LE. The nitrite scavenging effects were dependent on pH value, however, they decreased as pH value increased. Especially, they almost didn't show the nitrite scavenging effect in pH 6.0. In conclusion, the PD and LE extracts may be used as natural antioxidant sources to reduce lipid oxidation in oil emulsion.

Effects of Loquat (Eriobotrya japonica Lindl.) Leaf Extract with or without Ascorbic Acid on the Quality Characteristics of Semi-Dried Restructured Jerky during Storage

  • Deterioration of jerky during storage is a major concern; this is usually combated with natural or synthetic antioxidants. This study aimed to evaluate the quality characteristics of semi-dried restructured jerky with and without loquat leaf extract (LE) powder and ascorbic acid (AA) during storage for 180 days. The jerkies were formulated with 0%, 0.15%, and 0.3% LE and/or 0.05% AA (Control, no antioxidant; AA, 0.05% AA; LE 0.15, 0.15% loquat LE; LE 0.15-AA, 0.15% loquat LE+0.05% AA; LE 0.3, 0.3% loquat LE; LE0.3-AA, 0.3% loquat LE+0.05% AA). LE is a phenolic compound, whose 1,1-diphenyl-2-picrylhydarzyl radical scavenging activity and metal chelating activity were found to be higher than AA. All antioxidant combinations having higher LE concentration and containing AA were effective in delaying protein and lipid oxidation compared to the control or AA. At the end of storage period, LE 0.15-AA and AA had higher CIE a* and lower shear force than the control. Therefore, the combination of 0.15% LE and 0.05% AA can result in reduced protein and lipid oxidation without any negative effect on the quality characteristics of semi-dried restructured jerky.

난류유동의 대 와동 모사(LES) (Large Eddy Simulaion of Turbulent Flows)

  • 이 글에서는 난류유동의 수치모사 기법으로서 최근 주목을 받고 있는 LES에 대해서 살펴보았다. LES의 개념은 공간에서의 여고에 근거를 두고 있으며 따라서 난류모델은 작은 스케일의 유동 구조에만 국한된다. LES의 이러한 장점은 DNS에 좀도 가까운 난류모델링의 가능성을 의미한다. 또한 그간 발전된 LES의 기법들에 대해 고찰해보았으며 LES는 DNS의 처선책으로서 난류유 동을 예측하기 위한 도구로서 그 전망은 매우 밝다.

무측지성 국화 형질전환 계통 영양번식 제2세대의 형태적 및 분자생물학적 특성 (Phenotypic and molecular characteristics of second clone (T0V2) plants of the LeLs-antisense gene-transgenic chrysanthemum line exhibiting non-branching)

  • 환경위해성평가 연구 대상인 형질전환 이벤트로서의 자격을 확인하고자 형질전환세대($T_0V_0$)에서와 같이 영양번식 제1세대($T_0V_1$)에서도 도입유전자 LeLs-antisense의 발현 특성인 무측지성을 유지한 국화 무측지성 형질전환계통 LeLs80의 영양번식 제2세대($T_0V_2$)의 형태적 및 분자 생물학적 특성을 조사하였다. LeLs80 계통의 $T_0V_2$에서도 LeLs-antisense 유전자의 발현 특성인 무측지성이 안정적으로 유지되는 것을 확인하였다. 또한, Southern 및 Northern 분석에 의해 LeLs-antisense 유전자가 3 copy 도입되었으며, LeLs-antisense 유전자의 전사체가 정상적으로 발현되는 것도 확인할 수 있었다. 또한 flanking T-DNA sequencing method를 이용하여 LeLs-antisense 유전자의 주변 염기서열 분석 통해 LeLs80 계통의 genome내 LeLs-antisense 유전자 주변에 186 ~ 464 bp의 pCAMBIA2300 T-DNA right border 부근으로 추정되는 염기서열이 확인되었고, pCAMBIA2300 전 염기서열과의 비교 분석한 결과, pCAMBIA2300 T-DNA left border와 right border내 선발마커 유전자 NPT II의 발현 promoter 부분과 LeLs-antisense 유전자 발현 terminator 일부 염기서열과 일치하였다.

Effects of Supplementation of Mixed Cassava (Manihot esculenta) and Legume (Phaseolus calcaratus) Fodder on the Rumen Degradability and Performance of Growing Cattle

  • Two experiments were conducted to assess the effect of replacing a conventional concentrate with mixed cassava (Manihot esculenta) foliage and legume (Phaseolus calcaratus) foliage. In Exp. 1, three rumen fistulated crossbred cows were used for in sacco rumen degradability studies. In vitro gas production was also studied. In Exp. 2, 11 crossbred F2 heifers (Red Sindhi$\times$Holstein Friesian), with initial live weight of $129{\pm}6kg$ and aged six months, were allocated in a Completely Randomized Design (CRD) to evaluate a mixture (ratio 3:1) of cassava and legume foliage (CA-LE feed) as a protein source compared to a traditional concentrate feed (Control) in diets based on fresh elephant grass (Pennisetum purpureum) and urea treated rice straw ad libitum. The Control feed was replaced by the CA-LE feed at levels of 0% (Control), 40% (CA-LE40), and 60% (CA-LE60) based on dry matter (DM). The in sacco degradation of CA-LE feed was higher than Control feed (p<0.05). After 48 h incubation the degradation of CA-LE feed and Control feed was 73% vs. 58% of DM and 83% vs. 65% of CP, respectively. The gas production of CA-LE feed was also significantly higher than of Control feed during the first 12 h of incubation. The results of the performance study (Exp. 2) showed that the level of CA-LE feed in the concentrate had no effect on total dry matter intake (p>0.05), but live weight gains (LWG) in CA-LE40 and CA-LE60 were significantly higher (551 and 609 g/d, respectively) than in the Control group (281 g/d). The intake of CP was higher (p<0.05) for the treatments CA-LE40 and CA-LE60 (556 and 590 g/d, respectively) compared to that of Control (458 g/d), while there was no significant difference in ME intake. The feed conversion ratio was 16.8, 9.0 and 7.9 kg DM/kg LWG in Control, CA-LE40 and CA-LE60, respectively. The feed cost of CA-LE40 and CA-LE60 corresponded to 43% and 35%, respectively, of the feed cost of Control feed. The best results were found when CA-LE feed replaced 60% of DM in Control feed and considerably decreased feed cost. It is concluded that feeding cassava foliage in combination with Phaseolus calcaratus legume as a protein supplement could be a potentially valuable strategy which leads to reduced feed costs and a more sustainable system in smallholder dairy production in Vietnam.
