해양미생물 Psychrobacter aquimaris LBH-10가 생산하는 산성 carboxymethylcellulase의 특성에 대한 연구 (Characterization of Acidic Carboxymethylcellulase Produced by a Marine Microorganism, Psychrobacter aquimaris LBH-10)

  • Carboxymethylcellulose (CMC)를 분해하는 미생물을 해수에서 분리하였으며, 16S rDNA의 염기서열을 분석하여 동정한 결과, Psychrobacter aquinaris로 학인 되어 P. aquinaris LBH-10로 명명하였다. 이 균주는 CMC, 셀로바이오스, 커드란, 여과지, p-nitrophenyl-$\beta$-D-glucopyranoside (pNPG), 풀루란 및 자일란을 분해하였으나, avicel 및 섬유소는 분해하지 못하였다. P. aquinaris LBH-10이 생산하는 carboxymethylcellulase (CMCase)의 최적 반응 온도는 $50^{\circ}C$이었으며, $20^{\circ}C$에서 $50^{\circ}C$의 온도 범위에서 24시간이 경과한 후에도 90% 이상의 활성을 유지하였다. 또한, 이 균주가 생산하는 CMCase의 최적 반응 pH는 3.5이었으며 pH 2.5에서 pH 7.0 사이의 산성 조건하에서 24시간이 경과한 후에도 70% 이상의 활성을 유지하였다. P. aquinaris LBH-10이 생산하는 CMCase의 최적 반응 pH는 지금까지 발견된 섬유소 분해효소 중에서 가장 낮은 pH로 판단된다. 제한된 농도의 $CoCl_2$, EDTA, 및 $PbCl_2$은 P. aquinaris LBH-10가 생산하는 CMCase의 활성을 증가시켰으나, $HgCl_2$, KCl, $MnCl_2$, $NiCl_2$, 및 $SrCl_2$는 이 균주가 생산하는 CMCase의 활성을 감소시켰다.

Synergistic efficacy of LBH and αB-crystallin through inhibiting transcriptional activities of p53 and p21

  • LBH is a transcription factor as a candidate gene for CHD associated with partial trisomy 2p syndrome. To identify potential LBH-interacting partners, a yeast two-hybrid screen using LBH as a bait was performed with a human heart cDNA library. One of the clones identified encodes ${\alpha}B$-crystallin. Co-immunoprecipitation and GST pull-down assays showed that LBH interacts with ${\alpha}B$-crystallin, which is further confirmed by mammalian two-hybrid assays. Co-localization analysis showed that in COS-7 cells, ${\alpha}B$-crystallin that is cytoplasmic alone, accumulates partialy in the nucleus when co-transfected with LBH. Transient transfection assays indicated that overexpression of LBH or ${\alpha}B$-crystallin reduced the transcriptional activities of p53 and p21, respectively, Overexpression of both ${\alpha}B$-crystallin and LBH together resulted in a stronger repression of the transcriptional activities of p21 and p53. These results showed that the interaction of LBH and ${\alpha}B$-crystallin may inhibit synergistically the transcriptional regulation of p53 and p21.

Whole genome sequencing of Luxi Black Head sheep for screening selection signatures associated with important traits

  • Objective: Luxi Black Head sheep (LBH) is the first crossbreed specialized for meat production and was developed by crossbreeding Black Head Dorper sheep (DP) and Small Tailed Han sheep (STH) in the farming areas of northern China. Research on the genomic variations and selection signatures of LBH caused by continuous artificial selection is of great significance for identifying the genetic mechanisms of important traits of sheep and for the continuous breeding of LBH. Methods: We explored the genetic relationships of LBH, DP, and several Mongolian sheep breeds by constructing phylogenetic tree, principal component analysis and linkage disequilibrium analysis. In addition, we analysed 29 whole genomes of sheep. The genome-wide selection signatures have been scanned with four methods: heterozygosity (HP), fixation index (FST), cross-population extended haplotype homozygosity (XP-EHH) and the nucleotide diversity (𝜃π) ratio. Results: The genetic relationships analysis showed that LBH appeared to be an independent cluster closer to DP. The candidate signatures of positive selection in sheep genome revealed candidate genes for developmental process (HoxA gene cluster, BCL2L11, TSHR), immunity (CXCL6, CXCL1, SKAP2, PTK6, MST1R), growth (PDGFD, FGF18, SRF, SOCS2), and reproduction (BCAS3, TRIM24, ASTL, FNDC3A). Moreover, two signalling pathways closely related to reproduction, the thyroid hormone signalling pathway and the oxytocin signalling pathway, were detected. Conclusion: The selective sweep analysis of LBH genome revealed candidate genes and signalling pathways associated with developmental process, immunity, growth, and reproduction. Our findings provide a valuable resource for sheep breeding and insight into the mechanisms of artificial selection.

Simultaneous Observation of FUV Aurora with Precipitating Electrons on STSAT-1

  • We present the results offar ultraviolet (FUV, 1350-1750 ${\AA}$) auroral observations made by the Far-ultraviolet IMaging Spectrograph (FIMS) instrument on the Korean microsatellite STSAT-1. The instrument was capable of resolving spatial structures of a few kilometers with the spectral resolution of 2-3 ${\AA}$. The observations were carried out simultaneously with the measurement of precipitating electrons using an electrostatic analyzer (ESA, 100 eV-20 keV) and a solid state telescope (SST, 170 keV-360 keV) on board the same satellite. With a careful mapping of the field lines, we were able to correlate the particle spectrum to the corresponding FUV spectrum of the footprints of the FIMS image that varied significantly in fine scales. We divided the FIMS spectral band into the LBH long (LBHL, 1640-1715 ${\AA}$) and LBH short (LBHS, 1380-1455 ${\AA}$) bands, and compared the electron energies with the intensities of LBHL and LBHS for the well-defined inverted-V structures. The result shows a strong correlation between the total LBH intensity and the energy flux measured by ESAwhile the peak energy itself does not correlate well with the LBH intensity. On the other hand, it was observed that the ratio of the LBHL intensity to that of LBHS increased significantly as the peak electron energy increased, primarily due to a smaller absorption by O2 at LBHL than at LBHS.

Analysis of FUV auroras by high energy electron precipitation

  • Our previous study showed the intensity of the long LBH (1600 - 1715 ${\AA}$) was enhanced very much compared to that of the short LBH (1400 - 1500 ${\AA}$) when the characteristic energy of the precipitating electrons increased from 1 keV to > 7 keV, in accordance with the theoretical models. In this presentation, we would like to present the results of our study for new modeling results about previous report and even higher energy electrons. We selected the events in which the fluxes both in the low energy (100 eV ~ 20 keV) and in the high energy (170 ~ 360 keV) were enhanced, and examined the auroral spectra for these events observed simultaneously by the imaging spectrograph on the same spacecraft. While the accurate characteristic energy could not be determined because of the gap in the energy range, our result showed the intensity ratio of the long LBH to the short LBH ranged from 1.2 to 2.0 in these events, in contrast to 1.0 or smaller for the events in which the highest enhancement was seen only in the low energy. Our study suggests that intense auroras might be accompanied by high energy electrons above 20 keV.

파이롯트 규모에서 미강을 이용한 해양미생물 Cellulophaga lytica LBH-14 유래의 cellobiase 생산 (Enhanced Production of Cellobiase by a Marine Bacterium, Cellulophaga lytica LBH-14, in Pilot-Scaled Bioreactor Using Rice Bran)

  • 본 연구의 목적은 통계학적 방법을 사용하여 해양미생물 Cellulophaga lytica LBH-14가 생산하는 cellobiase의 생산조건을 확립하는 것이었다. 이 균주의 생육에 최적인 미강, ammonium chloride 및 배지의 초기 pH는 100.0 g/l, 5.00 g/l 및 7.0이었으나, 이 균주가 생산하는 cellobiase의 생산에 최적인 조건은 각각 91.1 g/l, 9.02 g/l 및 6.6이었다. 이 균주의 생육에 최적인 $K_2HPO_4$, NaCl, $MgSO_4{\cdot}7H_2O$$(NH_4)_2SO_4$ 등과 같은 배지의 염농도는 각각 6.25, 0.62, 0.28 및 0.73 g/l이었으나, cellobiase 생산에 최적인 염들의 농도는 각각 4.46, 0.36, 0.27 및 0.73 g/l이었다. 또한, 균체의 생육 및 cellobiase의 생산에 최적인 온도는 각각 35 및 $25^{\circ}C$이었다. 플라스크 규모에서 최적화한 조건으로 파이롯트 규모의 생물배양기에서 cellobiase를 생산한 결과, 이 균주가 생산하는 cellobiase의 생산성은 92.3 U/ml이었으며, 이는 최적화하기 전에 비하여 5.4배 향상된 것 이었다. 본 연구를 통하여 쌀 도정공정의 부산물인 미강 및 ammonium chloride를 cellobiase를 생산하는 기질로 개발하였으며 해양 미생물을 사용하여 cellobiase의 생산기간을 7일에서 3일로 단축시켰다. 또한, 본 연구를 통하여 C. lytica LBH-14가 생산하는 cellobiase의 최적 생산조건은 이 균주가 생산하는 CMCase의 최적 생산조건과 다르다는 사실을 확인하였다.

Far ultraviolet observations of diffuse, monoenergetic, and broadband auroras

  • Discrete auroras, with unique shapes embedded in diffuse auroras, are generally associated with precipitating electrons that originate from the plasma sheet and are accelerated on the way as they travel to polar regions along the field lines. Two acceleration mechanisms have been proposed: quasi-static electric fields and dispersive Alfven waves, which are believed to yield monoenergetic peaks and broadband features in the particle spectra, respectively. Hence, it should be interesting to see how the two different mechanisms, through their characteristic spectra of the accelerated electrons, produce distinct auroral images and spectra, especially in the far ultraviolet (FUV) wavelengths as the long and short Lyman-Birge-Hopfield (LBH) bands exist as well as the strong absorption band of molecular oxygen in the FUV band. In fact, we have previously shown, using the simultaneous observations of precipitating electrons and the corresponding FUV spectra, that the discrete auroras associated with inverted-V events have a stronger relative intensity of the long LBH to the short LBH compared to diffuse auroras, especially when the peak energy is above a few keV. In this paper, we would like to focus on the differences in the FUV images and spectra between the two discrete auroras of the monoenergetic and broadband cases, again based on the study using the dataset of simultaneous observations of particles and FUV spectral images.

통계학적인 방법과 왕겨를 기질로 사용하여 해양에서 분리한 Bacillus licheniformis LBH-52 를 사용한 carboxymethylcellualse의 생산조건 최적화 (Statistical Optimization for Production of Carboxymethylcellulase from Rice Hulls by a Newly Isolated Marine Microorganism Bacillus licheniformis LBH-52 Using Response Surface Method)

  • 왕겨를 기질로 사용하여 carboxymethylcellualse (CMCase)를 생산하는 미생물을 해수에서 분리하였으며 16S rDNA의 염기서열을 분석하여 동정한 결과, Bacillus lichemiformis로 확인되었다. CMCase를 생산하기 위한 최적의 탄소원과 질소원은 왕겨와 암모니움 나이트레이트이었다. 통계학적인 방법인 response surface method (RSM)을 사용하여 CMCase를 생산하기 위한 조건을 최적화하였다. 통계학적인 분석 결과, 왕겨가 균체의 생육에 미치는 영향이 가장 높았으며, 왕겨와 배지의 초기 pH가 CMCase 생산에 미치는 영향이 높았다. Design Expert Software를 사용하여 결과를 분석한 결과, 균체의 생장에 최적인 조건은 48.7 g/l 왕겨, 1.8 g/l 암모니움 나이트레이트, 배지의 초기 pH 6.8 및 배양온도 35.7$^{\circ}C$이었으나, CMCase의 생산에 최적인 조건은 43.2 g/l 왕겨, 1.1 g/l 암모니움 나이트레이트, 배지의 초기 pH 6.8 및 배양온도 35.7$^{\circ}C$이었다. 최적화된 조건에서 왕겨를 기질로 사용하여 B. lincheniformis LBH-52가 생산하는 CMCase는 79.6 U/ml이었다. 본 연구를 통하여 왕겨와 암모니움 나이트레이트를 CMCase를 생산하는 기질로 개발하였으며, 해수에서 분리한 미생물을 사용하여 생산기간을 3일로 단축하였다.

연육 (surimi) 소재로서 어체중량이 다른 넙치의 품질 특성 비교 (Quality of Bastard Halibut with Different Weights as a Surimi Source)

  • This study investigated the physicochemical and enzymatic properties of unmarketable cultured bastard halibut (Paralichthys olivaceus) of different weights as a potential source of surimi and surimi gel. The proximate composition of cultured bastard halibut of different weights did not differ significantly at P<0.05 (light weight (LBH) 400~500 g, medium weight (MBH) 600~800 g, and heavy weight (HBH) > 1,000 g). Compared to Alaska pollock muscle, the bastard halibut muscle had a 4% higher crude protein content and 6% lower moisture content. The collagen content of LBH bastard halibut muscle was 1.58 g/100 g, which was lower than or no different from bastard halibut weighing different amounts. Regardless of fish weight or pH, the enzymatic activities of crude fish extracts ranged from 0.34~0.48 U/mg for casein and hemoglobin, 11.0~12.7 U/mg for LeuPNA, 5.4~6.1 U/mg for ArgPNA, 2.3~2.9 U/mg for SAAPFNA, and 0.1~0.2 U/mg for BAPNA. The yield of surimi gel from LBH was 24.4%, which was similar to that from MBH and lower than that from HBH. The surimi gel from LBH was similar to that from HBH, while weaker than that from MBH. The surimi gel from LBH gel was stronger than grade SA gel from commercial Alaska pollock.

Analyzing environmental factors influencing the gestation length and birth weight of Hanwoo cattle

  • Background: This study focused on reproductive traits in Hanwoo cattle, specifically the environmental factors affecting gestation length and birth weight. Methods: The records of 1,540 cows calved at the Hanwoo Research Institute from 2015 to 2023 were examined. This study analyzed two populations, line-breeding Hanwoo (LBH) and general Hanwoo (GH), with all cows undergoing estrus synchronization and artificial insemination. The R software was used to compare the differences between the two populations and analyze the environmental factors affecting each trait. Results: The results showed that the average gestation length for LBH was 283.28 ± 5.93 days, which was significantly shorter than that of the GH, which had an average of 285.63 ± 6.21 days (p < 0.001). The average birth weight of LBH calves was 25.10 ± 3.69 kg, significantly lighter than GH calves, which weighed 27.26 ± 4.11 kg on average (p < 0.001). Analysis of environmental factors revealed significant differences in the gestation length of LBH based on dam parity, year, and season of calving. However, no significant differences were observed based on calf sex. For LBH, birth weight showed significant differences based on dam parity, year of calving, and sex of the calf, but not the season of calving. In GH, gestation length varied with dam parity and calving season, but not with calving year or calf sex. The GH birth weight showed differences based on dam parity, year of calving, and calf sex, but not the season of calving. Conclusions: Reproductive traits in the Hanwoo cattle industry are economically vital but are heavily influenced by environmental factors due to their low heritability. An accurate evaluation of the genetic potential of these traits requires an analysis of the environmental factors affecting them. The results of this study serve as foundational data for predicting the potential for genetic improvement in the gestation length and birth weight of Hanwoo cattle.
