Heterophil Phagocytic Activity Stimulated by Lactobacillus salivarius L61 and L55 Supplementation in Broilers with Salmonella Infection

  • Newborn chicks are susceptible to Salmonella enterica serovar Enteritidis (SE). The objective of this study was to evaluate the effect of Lactobacillus probiotic isolated from chicken feces on heterophil phagocytosis in broiler chicks. A total of 150 newborn broiler chicks were divided into 5 groups (30 chicks per group) as follows: group 1 (normal control), given feed and water only, group 2 (positive control) given feed, water and SE infection, group 3 (L61 treated) given feed, water, SE infection followed by Lactobacillus salivarius L61 treatment, group 4 (L55 treated) given feed, water, SE infection followed by L. salivarius L55 treatment, and group 5 given feed, water, SE infection followed by L. salivarius L61 + L55 combination treatment. After SE infection, L. salivarius treatment lasted for 7 days. The results showed that L. salivarius L61 and L. salivarius L55 treatment, either alone or combination of both, increased the survival rate after SE infection, and upregulated heterophil phagocytosis and phagocytic index (PI). Conversely, chick groups treated with Lactobacillus showed lower SE recovery rate from cecal tonsils than that of the positive control group. The PI values of the chicken group with SE infection, followed by the combination of L. salivarius L61 and L. salivarius L55 were the highest as compared to either positive control or normal control group. Two Lactobacillus strains supplementation group showed significantly (p<0.05) higher PI value at 48 h than 24 h after treatment.

Differential Cytokine Regulatory Effect of Three Lactobacillus Strains Isolated from Fermented Foods

  • Lactic acid bacteria (LAB) isolated from fermented foods have potential as a treatment for immune-related disorders and the use of LAB has been increasing worldwide. In this study, the differential cytokine regulatory effect was examined with three isolates of lactobacilli strains; namely, Lactobacillus plantarum K55-5 isolated from dairy product, and L. sakei K101 and L. plantarum K8 previously isolated from kimchi (a Korean traditional fermented vegetable). Production of cytokines such as IL-10, IL-12, IFN-γ, and TNF-α was significantly increased in L. sakei K101- and L. plantarum K55-5-treated splenocytes as compared with controls. The oral administration of L. sakei K101 and L. plantarum K55-5 increased cytokine production in the immunosuppressed mouse splenocytes and blood. NK cell cytotoxic activity was also increased in L. sakei K101- and L. plantarum K55-5-fed mice. On the other hand, L. plantarum K8 did not affect cytokine induction in all the experiments performed in this study. The cytokine-inducing effect of L. plantarum K55-5 was significantly increased by lysates of heat-killed bacteria as compared with live, heat-killed, or supernatant of cell lysates. TNF-α production by lipoteichoic acids (LTAs) isolated from the three isolates of lactobacilli was compared, and it was found that K55-5 LTA had a highest cytokine-inducing ability, which was mediated by TLR2-mediated NF-κB and ERK activation. Taken together, our study suggests that L. plantarum K55-5 and L. sakei K101 can be used for the treatment of immunosuppressed disorders.

고농도 균체 배양에 의한 알콜 발효에서의 균체농도와 발효특성치 사이의 함수관계 규명 -균체농도와 비성장속도, 균체 수율, 알콜 생산성 사이의 함수관계- (A Study on the Functional Relationship between Biomass Concentration and Fermentation Characteristics in the Culture of High density Cell)

  • 회전식 진탕 배양기를 사용한 회분식 발효실험을 통하여 효모 S. cerevisiae ATCC 24858의 균체농도 변화가 비성장 속도, 균체수율, 에탄올수율, 에탄올 생산성 등에 미치는 영향을 규명하여 보았다. 균체의 비성장속도는 균체농도가 증가함에 따라 감소하여 균체농도가 55g/L에 이르게 되면 $Ohr^{-1}$에 도달하여 균체성장이 멈추게 되었다. 에탄올수율 $Y_{p/s}$는 균체농도 변화에 관계없이 일정한 값 0.43을 나타내었으나 균체수율$Y_{x/s}$는 균체농도 증가에 EK라 감소 경향을 나타내어 균체농도55g/L에서는 그 값이 0을 나타내었다. 에탄올 생산성은 균체농도 증가에 따라 선형적인 함수관계로 증가하는 경향을 나타내어 초기 당농도 170g/L, 초기 균체농도55g/L인 경우에는 30g/L까지 상승하였다. 또한 본 연구의 실험 결과를 Luong식에 적용하여 군체성장이 멈추는 에탄올의 농도가 95g/L임을 알 수 있었다.

The Expression and the Subcellular Localization of Regulatory Subunits of Class IA Phosphoinositide 3-Kinase in L6 Skeletal Muscle Cell

  • PI3-kinase activity through p85, the regulatory subunit of class IA PI3-kinase, is indispensable for the growth, differentiation, and survival of skeletal muscle cells, but little is known about the function of other regulatory subunits such as p55 and p50. We examined the subcellular localization and the expression of the regulatory subunits of class IA PI3-kinase in L6 myoblasts. Both p55 and p50 as well as p85 were expressed in L6 myoblasts. Whereas p85 was localized at both cytosolic and nuclear tractions, p55 and p50 were localized at only the nuclear traction. During the differentiation of L6 myoblasts, the protein concentrations of both p55 and p50 were decreased but that of p85 was not significantly changed. Menadione-induced oxidative stress induced the translocation of p85 from cytosol to nucleus and the increase of p55 expression. These results suggest that the regulatory subunits of class IA PI3-kinase play an important role in L6 myoblasts.

Complete genome and two plasmids sequences of Lactiplantibacillus plantarum L55 for probiotic potentials

  • In this study, we report the complete genome sequence of Lactiplantibacillus plantarum L55, a probiotic strain of lactic acid bacteria isolated from kimchi. The genome consists of one circular chromosome (2,077,416 base pair [bp]) with a guanine cytosine (GC) content of 44.5%, and two circular plasmid sequences (54,267 and 19,592 bp, respectively). We also conducted a comprehensive analysis of the genome, which identified the presence of functional genes, genomic islands, and antibiotic-resistance genes. The genome sequence data presented in this study provide insights into the genetic basis of L. plantarum L55, which could be beneficial for the future development of probiotic applications.

Association of Paraoxonase-1(Q192R and L55M) Gene Polymorphisms and Activity with Colorectal Cancer and Effect of Surgical Intervention

  • Background: Colorectal cancer (CRC) is a leading cause of cancer-related death. Oxidative DNA damage may contribute to cancer risk and the antioxidant paraoxonase is one endogenous free radical scavenger in the human body which could therefore exert an influeence. Purpose: Aim of this study was to determine the role of serum arylesterase (ARE) and paraoxonase 1(PON1) activities in CRC patients and to find any association between (PON1) Q192R and L55M gene polymorphisms in CRC patients. Also the serum ARE and PON1 activities in CRC patients will be investigated before and after surgery Materials and Methods: This study involved a total of 50 patients with newly diagnosed CRC and 80 healthy controls. PON1 and ARE activities were determined using an enzymatic spectrophotometric method. PON1 Q192R and L55M gene polymorphisms were determined using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) based restriction fragment analysis. The restriction enzyme AlwI was used to examine the Q192R polymorphism and Hsp92II for the L55M polymorphism. Results: Significant differences in the PON1 Q192R polymorphism were found between patients and controls. The Q allele was more frequent in the patient group than in controls, while the R allele was more frequent in the controls. Significant differences were found in the L55M polymorphism. Additionally, there were significant differences in L and M allele frequencies (p=0.001). The serum activities of PON1 and ARE were low in QQ and MM genotype. Conclusions: serum PON1 and ARE activities were significantly lower in CRC patients compared to healthy subjects. The R allele may protect against colorectal cancer.

갓김치에서 분리된 유산균의 활성산소종에 대한 저항성과 항산화 활성 (Resistance to Reactive Oxygen Species and Antioxidant Activities of Some Strains of Lactic Acid Bacteria from the Mustard Leaf Kimchi)

  • 갓김치로부터 분리된 유산균 5종(Lactobacillus acidophilus GK20, Lactobacillus brevis GK55, Lactobacillus paracasei GK74, Lactobacillus plantarum GK81, 및 Leuconostoc mesenteroides GK104)의 활성산소종에 대한 저항성 및 항산화 활성을 조사하였다. Hydrogen peroxide (0.5 mM)와 반응 5시간후 L. acidophilus GK20, L. brevis GK55, L. paracasei GK74 및 L. plantarum GK81들은 50% 이상의 생존율을 보였으며, superoxide anions에 의한 저해율은 L. acidophilus GK20와 L. paracasei GK74가 가장 낮았고, hydroxyl radical과의 반응에서는 L. paracasei GK74와 L. plantarum GK81이 가장 안정하였다. 한편, L. plantarum GK81는 실험 균주 중 가장 높은 DPPH 소거능($70.8{\pm}10.1%$)을 나타내었는데, 세포(IC)보다는 세포추출물(ICFE)의 활성이 더 높았다. 또한 L. plantarum GK81의 ICFE는 다른 균주들에 비해 높은 superoxide radical 소거능을 보였고, 이 균주의 IC와 ICFE에 의한 환원력은 BHA와 vitamin C보다 유의적으로 더 높게 나타났으며, L.paracasei GK74 IC의 환원력도 vitamin C와 비슷한 수준으로 나타났다. L. brevis GK55, L. paracasei GK74 및 L.mesenteroides GK104 IC의 $Fe^{2+}$-chelating 활성은 30% 이상이었고, L. plantarum GK81의 ICFE는 45% 이상의 활성을 나타내었다. 따라서 실험 균주 중 L. plantarum GK81은 다른 균들에 비해 ROS에 대해 비교적 안정하고, 항산화 활성도 가장 높은 것으로 확인되었다.

김치에서 분리한 Lactobacillus plantarum LHB55의 항균성과 요구르트 제조 적합성 연구 (A Study on the Yogurt Manufacture Suitability and Antimicrobial Activity of Lactobacillus plantarum LHB55 Isolated from Kimchi)

  • 이 연구는 김치에서 분리한 항균활성이 우수한 요구르트 제조용 유산균 스타터를 개발하기 위함이다. 분리한 103개의 산 생성 균주를 PCR로 screening하여 72개의 유산균을 분리하였다. 분리균의 배양액을 paper disk method를 사용하여 병원성 미생물(E. coli, S. enteritidis, S. aureus)에 대한 항균활성을 측정하였고, 활성이 강한 균주를 선별하여 API 50CHL과 16S rRNA sequencing 방법으로 균을 동정하였다. 균은 L. plantarum으로 확인되어 L. plantarum LHB55로 명명하였다. E. coli에 대해 높은 항균효과를 나타내는 L. plantarum LHB55를 사용하여 제조한 요구르트의 미생물학적, 이화학적 특성과 관능검사 결과 대조구와의 유의적인 차이는 거의 없었다. 그 결과 김치에서 분리한 L. plantarum LHB55의 항균활성이 우수한 요구르트 제조 스타터 균주로서 사용 가능함을 확인하였다. 항균성 등 인간에게 유리하게 작용하는 스타터 균주와 발효유제품 개발은 계속적인 연구가 필요할 것으로 생각된다.

저농도의 Ethanol에 의한 Listeria monocytogenes의 증식억제 (Inhibition of Listeria monocytogenes by Low Concentrations of Ethanol)

  • 저농도의 ethanol(3-7%, v/v)을 tryptic soy broth (TSB)에 첨가하여 L. monocytogenes의 증식과 생존에 미치는 효과를 이 세균의 최적온도(35$^{\circ}C$)와 저온(-20, 5$^{\circ}C$) 및 고온(45, 50, 55$^{\circ}C$)에서 검토하였다. 35$^{\circ}C$에서의 L. monocytogenes의 증식은 ethanol농도의 증가와 더불어 저해되었으며, 5% ethanol의 존재하에서는 긴 유도기를 거친 후에 증식이 시작되었으나 ethanol 7%에서는 생균수가 계속 감소하였다. 3-7%의 ethanol을 함유한 TSB에 $10^{5}$-$10^{6}$ cells/$m\ell$의 L. monocytogenes를 접종하여 저온(5$^{\circ}C$, -2$0^{\circ}C$)에 저장하였을 때 5$^{\circ}C$에서 세균은 5% 이하의 ethanol 첨가시에는 증식하였으나 -2$0^{\circ}C$에서는 저장초기에 생균수가 빠르게 감소한 후 감소속도는 느리게 진행되었다. 냉장과 동결저장에서 3%의 ethanol첨가로서 control의 90% 이상의 세균이 제거되었다. $10^{6}$-$10^{7}$ cells/$m\ell$의 L. monocytogenes를 접종하여 고온(45, 50, 55$^{\circ}C$)에 저장한 경우, L. monocytogenes의 생균수는 45$^{\circ}C$에서는 느리게 감소하였으며 3% 이하의 ethanol 첨가에 의한 항균효과는 뚜렷한 차이가 없었다. 5$0^{\circ}C$와 55$^{\circ}C$에서 생균수가 빠르게 감소하였는데 특히 55$^{\circ}C$에서는 3, 5, 7%의 ethanol첨가로서 세균의 사멸속도는 각각 control의 1.5배, 3배, 5배 정도 증가하였다.다.
