Fortification of γ-aminobutyric acid and bioactive compounds in whey by co-fermentation using Bacillus subtilis and Lactobacillus plantarum (유청을 이용한 Bacillus subtilis와 Lactobacillus plantarum의 혼합발효를 통한 γ-aminobutyric acid와 생리활성물질 강화)

  • Biologically active substances including gamma-aminobutryric acid (GABA) were added into whey by co fermentation using Bacillus subtilis HA and Lactobacillus plantarum EJ2014. The first fermentation using B. subtilis HA with 5% monosodium glutamate (MSG) and 2% glucose enhanced the production of poly-${\gamma}$-glutamic acid (PGA), resulting in higher consistency of $4.09Pas^n$ as well as whey protein peptides. After the second fermentation using L. plantarum EJ2014, the remaining MSG (3.40%) as a precursor was completely converted to 2.21% GABA. Furthermore, the lactose content in whey decreased from 6.73 to 3.68% after co-fermentation, and the tyrosine content increased from 20.47 to 38.24%. Peptides derived of whey proteins were confirmed by SDS-PAGE. Viable cell counts of B. subtilis and L. plantarum were 5.83 log CFU/mL and 9.20 log CFU/mL, respectively. Thus, co-fermentation of whey could produce the novel food ingredient fortified with biologically active compounds including GABA, ${\gamma}$-PGA, peptides, and probiotics.

L-Lysine Production by Amplification of the ddh Gene in a Lysine-producing Brevibacterium lactofementum. (Brevibacterium lactofermentum에서 ddh 유전자의 증폭에 의한 L-Lysine의 생산)

  • The ddh gene encoding meso-DAP-dehydrogenase (DDH) involved in the dehydrogenase pathway is essential for high-level lysine production in Brevibacterium lactofermentum. To investigate the effect of the ddh gene amplification on lysine production by B. lactofementum, we constructed two E. coli -B. lactofermentum shuttle vector, pEB1 and pEB2. The recombinant plasmids, pRK1 and pRK2, carrying the ddh gene were introduced into B. lactofermentum by electroporation. The specific activity of DDH by amplification of the ddh gene was increased 7-fold, and also L-Lysine production of B. lactofermentum strains harboring recombinant plasmids were 18∼20% higher than that of the control.

A Design of Dual-band Microstrip Antenna Loading Inverted-L-shaped Parasitic Elements Vertically at Radiation Apertures for GPS Applications (방사개구면에 역 L형 기생소자를 세운 GPS용 이중대역 마이크로스트립 안테나 설계)

  • In this paper, we present novel dual-band microstrip antennas using inverted-L-shaped parasitic elements vertically at radiation apertures for GPS L1(1.575 GHz) and L2(1.227 GHz) bands. For making dual band which has large interval, the inverted-L-shaped parasitic element was loaded at the radiation aperture of a half-wavelength patch antenna(GPS L1) in opposite direction of the feeding point for receiving the low frequency(GPS L2). The low frequency occurs by perturbation and coupling between the patch and parasitic. Next, due to use circular polarizations at the GPS applications, two inverted-L-shaped parasitic elements were loaded at radiation apertures of each polarizations and the feeding point was moved at diagonal part of the patch. The dimensions of the designed circularly polarized antenna were $88.5{\times}79{\times}10.4mm^3$ ($0.36{\lambda}L{\times}0.32{\lambda}L{\times}0.04{\lambda}L$, ${\lambda}L$ is the free-space wavelength at 1.227 GHz). Measured -10 dB bandwidths were 116.3 MHz(7.4%) and 64.3 MHz(5.2%) at GPS L1 and L2 bands, respectively. All of these cover the respective required system bandwidths. The measured 3 dB axial ratio bandwidths were 11.7 MHz(0.74%) and 14 MHz(1.14%), respectively. Within each of the designed bands, broadside radiation patterns were observed.

Isolation, Identification, and Fermentation Characteristics of Bacillus sp. with High Protease Activity from Traditional Cheonggukjang (전통 청국장으로부터 protease 분비능이 우수한 Bacillus sp. 균주의 분리 동정 및 발효 특성)

  • Twenty one strains strongly producing protease were isolated from Korean traditional Cheonggukjang. Eight strains selected by sensory evaluation on Cheonggukjang prepared with isolated strains were identified with based on biochemical properties a and 16S rDNA sequencing. Identified strains were Bacillus subtilis MB4, and Bacillus amyloliquefaciens A1, A2, B1, MC1, SB2, SC1, and SD1. Protease activities, important strain selection factor, were higher in Cheongjukjang prepared with B. subtilis MB4 (179.6 Unit) and B. amyloliquefaciens SB2 (201.9 Unit) than commercial traditional Cheonggukjang (97.9 Unit). Sensory evaluation revealed Cheonggukjang prepared with B. subtilis MB4 had flavor very similar to commercial traditional Cheonggukjang. Cheonggukjang prepared with B. suhtilis MB4 (0.0006 Pa s) and commercial traditional Cheonggukjang (0.0002 Pa s) revealed lower viscosities than those of Cheonggukjang prepared with B. amyloliquefaciens SB2, MC1, B1, A1, SD1, A2, and SC1 (0.006 to 0.008 Pa s at 1001/s. Results show Cheonggukjang could be prepared using single strain of B. subtilis MB4, maintaining high protease activity and very similar sensory and viscosity qualities with those of commercial traditional Cheonggukjang.

  • The addition products of glutathione to ${\beta}$ -nitrostyrene derivatives were synthesized. ${\beta}$ -Nitrostyrene (1a), p-methyl-${\beta}$-nitrostyrene (1b), 3,4,5-trimethoxy-${\beta}$-nitrostyrene (1c), o-, m- and p-chloro-${\beta}$-nitrostyrene (1e, 1f, 1g) and o-, m- and p-methoxy-${\beta}$-nitrostyrene (1h, 1i, 1j) undergo addition reactions with glutathione to form S-(2-nitro-1-phenylethyl)-L-glutathione (5a), S-[2-nitro-1-(p-methyl)phenylethyl]-L-glutatione (5b), S-[2-nitro-1-(3', 4', 5'-trimethoxy)phenylethyl]-L-glutathione (5c), S-[2-nitro-1-(o-chloro)phenylethyl]-L-glutathione (5e), S-[2-nitro-1-(m-choro)phenylethyl]-L-glutathione (5f), S-[2-nitro-1-(p-chloro)phenylethyl]-L-glutathione (5g), S-[2-nitro-x-(o-methoxy)-phenylethyl]-L-glutathion e(5h), S-[2-nitro-x-(m-methoxy)phenylethyl]-L-glutathion e (5i), and S-[2-nitro-1-(p-methoxy)phenylethy])-L-glutathione (5j), respectively. The structure of adducts were identified by UV and IR-spectra, molecular weight measurement, and elemental analysis.

Effect of Jasmonic Acid on the Root Growth and the Eleutheroside Accumulation in the Adventitious Root Culture of Eleutherococcus senticosus (가시오갈피나무 부정근(不定根) 배양(培養)에서 부정근(不定根)의 생장(生長)과 Eleutheroside유(類)의 생산(生産)에 미치는 Jasmonic acid처리(處理)의 영향)

  • This study was carried out to investigate the dose-dependent effect of jasmonic acid on both the adventitious root growth and the accumulation of various eleutherosides in the Eleutherococcus senticosus bioreactor culture. The highest biomass production (4.6 g DW/L) was observed in the absence of jasmonic acid and the root growth was significantly decreased by increasing the jasmonic acid concentration. However, jasmonic acid stimulated the production of both eleutheroside B, E and $E_1$. The highest level of eleutheroside B and E ($476.3{\mu}g/g\;DW$ and $676.0{\mu}g/g\;DW$) was obtained by 0.5 mg/L jasmonic acid treatment, while eleutheroside $E_1$ was accumulated at the highest level by 0.01 mg/L jasmonic acid treatment. The highest content of total eleutheroside was $2468.6{\mu}g/L$ when 0.01 mg/L jasmonic acid was applied. In addition, when the adventitious roots were cultured with 0.01 mg/L jasmonic acid, the highest levels of eleutheroside B, E and $E_1$ were observed at the 6th, 8th and 4th day of culture, respectively.

Classification of Listeria monocytogenes Isolates from Korean Domestic Foods Using Random Amplification of Polymorphic DNA and Serotyping Analysis (Random Amplification of Polymorphic DNA와 혈청학적 분석을 이용한 국내식품에서 분리한 Listeria monocytogenes의 분류)

  • Molecular subtyping of Listeria monocytogenes, including type strains and isolates from Korean foods, were performed using random amplification of polymorphic DNA (RAPD). Each Listeria species showed specific RAPD band patterns, and L. monocytogenes serotypes and isolates were divided into two clusters. RAPD results showed that L. monocytogenes isolates from Korean foods were divided into two groups. Group I contained L. monocytogenes serotypes 1/2b and 4b, whereas Group II contained serotypes 1/2a and 1/2c. These results suggested RAPD as possible subtyping methods for Listeria species. Also, RAPD Results showed significant correlation between molecular subtyping and serotyping of L. monocytogenes, and classified two different groups of L. monocytogenes isolated from Korean foods.

  • PD-L1 is expressed in a variety of antigen-presenting cells and provides T cell tolerance via ligation with its receptor PD-1 and B7-1 on T cells. Stimulation with lipopolysaccharide (LPS) can increase the level of PD-L1 expression in B cells and macrophages, which suggests that this molecule plays a role in the immunosuppression observed in severe sepsis. The aim of this study was to identify which of the downstream pathways of TLR4 are involved in the up-regulation of PD-L1 by LPS in macrophages. Flow cytometry was used to examine the expression of PD-L1 in RAW 264.7 macrophages stimulated with LPS. The following chemical inhibitors were used to evaluate the role of each pathway: LY294002 for PI3K/Akt, SB202190 for p38 MAPK, and U0126 for MEK. LPS induced the expression of PD-L1 in a time- and dose-dependent manner. Transfection of siRNA for TLR4 suppressed the induction of PD-L1. Pretreatment with LY294002 and SB202190 decreased the level of PD-L1 expression but U0126 did not. Overall, the PI3K/Akt and p38 MAPK pathways are involved in the up-regulation of PD-L1 expression in RAW 264.7 macrophages stimulated with LPS.

Design and Fabrication of the One-Chip MMIC Mixer using a Newly Proposed Bias Circuit for L-band (새로운 바이어스 회로를 적용한 L-band용 One-Chip MMIC 믹서의 설계 및 제작)

  • In this paper, the study of a design and fabrication of the receiver MMIC mixer for L-band application is described. The mixer is composed of active LO and RF balun to integrate on a chip and applied a newly proposed bias circuit to compensate the process variations of active devices. The conversion gain of the mixer is -14 dB, IIP3 is approximately 4 dBm and port-to-port isolation is over 25 dB. The newly proposed bias circuit is composed of a few FETs and resistors, and can compensate the variation of the threshold voltage by the process variations, temperature changes and etc. The designed chip size is $1.4\;mm{\times}1.4\;mm$.

Bioconversion of Linoleic Acid to Conjugated Linoleic Acid by Bifidobacterium breve

  • The bioconversion of linoleic acid (LA) to conjugated linoleic acid (CLA) was investigated to examine LA-adaptation of Bifidobacterium breve KCTC 3461 to additions of 1 to 5 mg/mL of LA overtime. To induce LA-adaptation, B. breve KCTC 3461 was treated with LA, according to three schemes. For LA-adapted B. breve the maximum concentration of CLA, $300\~350{\mu}g/mL$, was obtained in cys-MRS medium containing 1 mg/mL of LA. The CLA production significantly increased with increasing LA concentration, from 1 to 4 mg/mL, but the conversion of LA to CLA gradually decreased. The CLA production capability of B. breve, and its tolerance, improved significantly with LA-adaptation. The addition of LA (1 mg/mL) into the culture broth after 24 h of cultivation in a 100-mL media bottle was most effective at promoting CLA production. In a 2.5-L stirred-tank bioreactor, the observed conversion and productivity of $56.6\%\;and\;35.4{\mu}gmL^{-1}h^{-1}$, respectively, by LA-adapted B. breve were approximately 6.6 and 9.8 times higher than those of LA-unadapted B. breve.
