This study was conducted to investigate the effect of microbial liquid manure(MLM) on the growth and yield characteristics of Altari radish (Raphanus sativus L.). MLM 1,000mg$.$L$^{-1}$ MLM 2,000mg$.$L$^{-1}$ and MLM 1,000mg L$^{-1}$ + microbial fermentation compost(MFC) 600kg/10a were treated. MLM was sprayed on leaves at 10, 20, and 40 days after planting. Leaf length and width of radish in all microbial fertilizer treatments were more vigorous than that of control significantly. Number of leaves was great in order of 1,000 mg$.$L$^{-1}$ MLM+600kg/10a MFC 〉2,000mg$.$L$^{-1}$ MLM〉1.000mg$.$L$^{-1}$ MLM. SPAD reading value was increased with treatments of 2,000mg$.$L$^{-1}$ MLM and 1,000mg$.$L$^{-1}$ MLM+600kg/10a MFC. Root length and weight of all the microbial fertilizer treatments tended to increase compared with control, and 1,000mg$.$L$^{-1}$ MLM+600kg/10a MFC treatment was the most effective.
A plant regeneration system was developed through shoot organogenesis from in vitro leaf explants of Chrysanthemum morifolium 'Ohblang'. The effects of different concentrations of plant growth regulators and AgNO3 on efficient shoot regeneration and inhibition of browning were evaluated in chrysanthemum. The explants were cultured on MS shoot induction medium supplemented with 12 combination treatments of 6-benzyladenine (BA) 0.5, 1.0 and 2.0 mg/L, and α-naphthaleneacetic acid (NAA) 0.2, 0.5, 1.0 and 2.0 mg/L in darkness for 6 weeks and cultured under a 16/8 h photoperiod for 6 weeks. The highest shoot regeneration was obtained from the explants cultured on the medium with 1.0 mg/L BA and 1.0 mg/L NAA. Based on this result, AgNO3 was added to a shoot induction medium containing MS salts, vitamins, 1.0 mg/L BA, 1.0 mg/L NAA, 30 g/L sucrose, and 6 g/L agar to reduce browning of chrysanthemum leaf explants. In the control treatment without AgNO3, leaf explants turned brown at the cut edge; however, browning was not observed in AgNO3 treatments. Shoot organogenesis was higher at low concentrations of AgNO3 and decreased with an increase in AgNO3 concentration. The explants cultured on shoot induction medium (MS salts, vitamins, 1.0 mg/L BA, 1.0 mg/L NAA) with 1 mg/L of AgNO3 produced the highest shoot regeneration with 2.6 shoots per explants and a browning index of 0.7. When the regenerated shoots were detached from the explants and cultured on MS medium, the shoots were elongated and rooted successfully.
Two lysis-defective but DNA synthesis non-defective temperature-sensitive (ts) mutants of mycobacteriophage L1, L1G23ts23 and L1G25ts889 were found to be defective also in phage-specific RNA synthesis in the late period of their growth at 42$^{\circ}C$each to the extent of 50% of that at 32$^{\circ}C$The double mutant, L1G23ts23G25ts889 showed the ts defect in phage RNA synthesis that was nearly additive of those shown individually by the two single-mutant parents. Both G23 and G25 were shown to start functioning sometimes between 30 and 45 min after infection but the former gene might be dispensable after 45 min, while the latter was not. Northern analysis also shows that at 42$^{\circ}C$>, L1G23ts23 affects RNA synthesis more strongly than L1G25ts889 from L1 DNA segments that serve as the template for late gene transcription. Among the 21 virion and 12 non-virion late proteins synthesized by L1, L1G23ts23 is defective in the synthesis of at least 9 virion and all of non-virion proteins at 42$^{\circ}C$>. In contrast, L1G25ts889 is completely defective in synthesis of all the 33 late proteins. Possible roles of G23 and G25 in the positive regulation of transcription of different sets of late genes of L1 have been discussed.
We have established a high-frequency plant regeneration system via organogenesis from mature seed of Korean lawngrass(Zoysia japonica Steud.). The effects of 2,4-dichiorophenoxy acetic acid (2,4-D), 6-furfuryl amino purine (kinetin), $\alpha$-naphthaiene acetic acid (NAA), N6-benzyl amino pu-rine (BAP), and casein hydrolysate (CR) on cailus induction and multiple shoot formation on ex-posure to light were evaluated. Callus produced on the Murashige and Skoog (MS) medium containing 2,4-D and kinetin had high organogenesis potency. A single addition of 1.0 mg $L-^1$ 2,4-D significantly induced callus. Also, 1.0 mg L-$^1$ 2,4-D, with the addition of 0.1 mg $L-^1$ kinetin highly enlianced callus induction. The trend of cailus induction was also found on mediurn containing 0.1 mg $L-^1$ BAP with 1.0 mg $L-^1$2,4-D, and 1 g $L-^1$ CR with the addition of 1.0 mg $L-^1$ 2,4-D. However, NAA was no effective on callus formation. The growth of root was significantly high in the presence of 0.1 mg $L-^1$ kinetin compared to other concentrations. Over 2 mg $L-^1$ kinetin highly lengthened roots. Fresh weight of plantlet was highest on medium containing 0.1 mg $L-^1$ 2,4-D. Also, on medium containing 0.1 mg $L-^1$ BAP, fresh weight of piantlet was highly enhanced. BAP was significantly effective on multiple shoot formation, particularly when 2.0 mg $L-^1$ was added with 0.1 mg $L-^1$ 2,4-D. Callus induction and multiple shoot formation were achieved on MS basal medium containing 1.0 g $L-^1$ CH.
This experiment was carried out to clarify the effect of pyroligneous liquor and coconut water on plantlet multiplication and in vitro flowering of Dendrobium moniliforme. Plantlet growth and multiplication was good in 1.0 ml/L pyroligneous liquor treatment which was added to the basal media of 3 g/L hyponex and 4 g/L peptone ($H_{3} P_{4}$) containing 0.1 mg/L NAA and 1.0 mg/L kinetin. In the treatment of 30 ml/L coconut water showed good results on plantlet growth and multiplication. In vitro flowering showed highest rate in the treatment of 1.0 ml/L pyroligneous liquor and 30 ml/L coconut water which were added to the basal media of $H_{3} P_{4}$ containing 0.1 mg/L NAA and 1.0 mg/L kinetin.
This experiment was to investigate the Leptospiral antibody in the pigs with the serological test in two areas of Kangwondo from March to April. 1989. Twelve different living antigen (L. icterohaemorrhagiae, L. pomona, L. hardjo, L. australis, L. canicola, L. autumnalis, L. grippotyphosa, L. tarassovi, L. pyrogenes, L. bataviae, L. lai and L. ballum) of Leptospira interrogans was used for the serological test in the pigs. The blood sample of 280 pigs collected from the slaughter houses were detected by microscopic-agg-lutination test. The results were as follows, 1. Among the serum sample of 280 heads of the pigs, 6 heads of the swine (2.14%) were positive. 2. Among the positive sample of 6 heads, 3 heads of the swine (1.1%) showed the antibody of L. ict-erohaemorrhagiae, 2 heads of the swine (0.7%) showed the antibody of L. canicola, and 1 head of the swine (0.4%) showed the antibody of L. pomona. 3. The positive rate of Leptospira interrogans in the swine of Kangwondo appeared to be very low.
This experiment was to investigate the Leptospiral antibody in the pigs with the serological test by the microscopic-agglutination-test (MAT) from November 1991. to Januaury 1992. Antigen(living antigen) was used L. icterohemorrhagiae, L. pomona, L. canicola, L. hardjo, L. ballum, L. australis, L. autumnalis, L. grippotyphosa, L. tarassovi, L. pyrogenes, L. bataviae, L. hebdomadis for the serolgical test in the pigs. The result obtained are summarized as follows of the total 202 serum samples examined, 1. Among the serum samples of 202 heads, 19 heads of the pigs(9.4%) were positive. 2. Among the positive samples of 19 heads, The detected were L. icterohemorhagiae 10 heads(5.0%), L. pomona 3 heads(1.5%), L. canicola 6 heads(3.0%). 3. Antibody titers of positive sera were ranging from 1:100 to 1 : 400. Serotiters appeared to be very low, 4. The seroprevalence of Leptospira in Chechon was higher than that other districts (5. 4% -5.8%), but the lower than Chung-nam, Kyonggi(13.7-15.9%)
This study was carried out to find the effects of plant growth regulators on callus formation, rooting and shooting from cotyledon explant in oriental me]on. Various combinations of 0.1 mg/L auxins (IAA, NAA) and 0.5, 1.0. 1.5, 2.0 mg/L cytokinins (BA, kinetin, zeatin) were treated to the MS basal medium, respectively. Callus was induced mort effectively as 2,437.0 mg (FW)/explant in MS medium supplemented with 0.1 mg/L NAA and 2.0 mg/L BA, but that was non-embryogenic callus as colored yellow white and broke easily. Root was induced most effectively at a frequency of 98.0% in MS medium supplemented with 0.1 mg/L NAA and 0-5 mg/L kinetin. Shoots formed on cut part of vein at a frequency of 98.0% in MS medium supplemented with 0.1 mg/L IAA and 2.0 mg/L BA, that were multiple shoots. in case of its concentration, BA and lower concentration of IAA and NAA (0.01 and 0.05 mg/L). respectively. shooting ratio was not increased. The result of treatment with BA 0-5 mg/L and IAA 0.1 mg/L, callus induced at a week, and shoot start to form multiple shoots about 3 weeks after inoculation. After 2 times subculture as 2 weeks intervals, divided shoots rooted and developed into intact plantlets at 10 weeks and then that grown normally on pots after acclimatization.
In order to examine efficient L-threo-2,3-Dihydroxyphenylserine (L-threo-DOPS) synthesis process using whole cell biocatalyst, a thermostable L-threonine aldolase (L-TA), which cloned from Streptomyces coelicolor A3(2) and improved for stability, was expressed in a Corynebacterium glutamicum R strain. The constructed Corynebacterium expression vector, pCG-H44(1) successfully expressed L-TA in C. glutamicum R strain, but showed very low expression level. In order to improve the expression level, the expression vector named pCG-H44(2) was reconstructed by eliminating 1 nucleotide between SD sequence and start codon of L-TA. The pCG-H44(2) vector plasmid was able to overexpress L-TA approximately 3.2 times higher than pCG-H44(1) in C. glutamicum R strain (CGH-2). When the whole cell of CGH-2 was examined in a repeated batch system, L-threo-DOPS was successfully synthesized with a yield of 4.0 mg/ml and maintain synthesis rate constantly after 30 repeated batch reactions for 130 h.
The purpose of this study were analysed fatigue character of lumbar extension Muscle during repeated trunk flexion-extension motion we used EMG multi-telemeter (WEB 5000, Nihon Koden, Japan), Medex lumbar extension machine(Ocala, USA). We evaluated 20 persons on chronic low back pain group and 20 persons on control group. We analysed the quantitative variables IMF, MF, decreased ratios of MF to investigate fatigue. As a result, the following conclusion was drawn : 1. Each lumbar IMF of CLBP was L5> L3> L1 in the order of their size. CON group was similar quantitative and same order. But the relationship of two groups were not significantly. 2 The IMF within lumbar position(L1-L3, L1-L5, L3-L5) of two groups was significantly all positions(p<.05). 3. The MF shifting at lumbar position of two groups during repeated trunk flexion-extension motion was decrease shifting pattern with increase repeating motion, especially CLBP group was suddenly dropping than CON at all lumbar positions(L1, L3, L5) on early stage motion(34 set). 4. The lumbar fatigue of CLBP was higher than CON during repeated trunk flexion-extention motion, so CLBP supposed more tired than CON. And it was significantly within two groups at lumbar positions(L1, L3 : p<.05, L5 : P<.01).
