Dipeptide의 陽性子 磁氣共鳴硏究 (Proton Magnetic Resonance Studies of Dipeptides)

  • Glycine이 포함된 dipeptide인 glycyl-L-valine, L-valyl-glycine, glycyl-DL-alanine, glycyl-DL-serine 및 glycyl-L-aspartic acid의 중수용액속에서의 양성자 자기공명스펙트럼이 실온에서는 pH의 함수로 연구되었다. 이들 스펙트럼의 분석으로부터 $C_{\alpha}H,\;C_{\beta}H\;및\;C_rH$양성자의 chemical shift는 pH에 따라 일단계의 적정곡선으로 변하고 있으며 spin-spin coupling constant는 거의 변하지 않고 있음을 알 수 있다. Dipeptide 용액속에 있는 구성 아미노산의 $C_{\alpha}H\;C_{\beta}H\;및\;C_rH$양성자의 chemical shift가 각각 두개의 다른 값들로 나타나는 것은 이들 용액속에 두개의 자기적으로 서로 다른 site들이 들어있음을 말해준다. 이 연구에서는 다섯개 dipeptide의 구조를 양성자 자기공명스펙트럼에 의해 확인하였으며 결과는 앞으로 oligopeptide에서의 구조 변화, conformation, 구성 아미노산의 순서 결정등을 양성자 자기공명스펙트럼의 분석에 의해 알아낼 수 있음을 암시하고 있다.

유전자 조작한 Klebsiella pneumoniae로부터 L-트립토판 생산을 위한 발효배지 조건 (Media Optimization for L-tryptophan Production by Genetically Engineered Klebsiella pneumoniae)

  • The optimum medium composition for the production of L-tryptophan with Klebsiella pnuemoniae pheA tyrA trpE trpR/pSC 101-trp$^{+}$ and the effect of precusors in the optimum medium were studied. The specific growth rate in the optimum medium was almost the same as that in the basal medium, the former showing 1.01 and the latter 1.07 hr $$^{-1}$, but the produced tryptophan was increased 45% in the optimum medium. The maximum amount of produced tryptophan was 159 mg/l within 14 hours. Tryptophan production was ceased by casamino acid addition over 4 g/l in medium, but cell maSS increased with its addition. Indole and anthranilate as precusors had toxic effect on growth and tryptophan production at experimented concentration range (over 20 mg/l), but L-serine had good effect on tryptophan production, resulting in 175 mg/l tryptophan within 14 hours.

  • Vivrio mimicus (ATCC 33568)의 최적 배양조건하에서 배양액으로부터 collagenase를 황산암모늄 염석과 DEAE-Sephadex A-50 이온코환크로마토그래피에 의해 분리. 정제하였다. SDS-PAGE 전기영동분석법 및 겔여과법으로 정제된 collagenase의 분자량은 42 kDa 이였다. 기질인 불용성 콜라겐(Type I)에 대한 collagenase의 최적 pH 및 온도는 각각 7.75 및 28$\circ$였다. 금속착물제와 serine protease 저해제는 collagenase의 활성을 저해하였지만 L-cysteine과 histidine은 효소의 활성을 저해하지 않았다. collagenase의 아미노산 조성은 glycine 및 alanine의 아미노산 잔기가 많이 함유되어 있었다. 불용성 (Type I) 콜라겐에 대한 collagenase의 속도상수인 K$_{m}$ 값 및 R$_{cat}$/K$_{m}$값은 각각 2.86 mg/ml 및 972.28 U/mg-protein 이었다.

  • Properties of a protease purified from Neungee[Sarcodon aspratus(Berk, ) S. Ito] have been investigated. The enzyme displays a glycosylated serine protease. The enzyme is able to hydrolyze alanine glycine methionine glutamine and cysteine of N-CBZ and N-t-BOC-L-amino acid derivatibes relatively strongly but splits valine proline and isoleucine derivatives with low affinity which means the enzyme has the broad substrate spectrum toward the amino acids. Interestingly the enzyme was inhibited by bromelain inhibitor. That is the active site environ-ment of the enzyme is believed to be similar to that of bromelain However peptide mapping studies show that the two enzymes have distinct different cleavage sites.

Purification and Characterization of a Collagenolytic Protease from the Filefish, Novoden modestrus

  • A serine collagenolytic protease was purified from the internal organs of filefish Novoden modestrus, by ammonium sulfate, ion-exchange chromatography on a DEAE-Sephadex A-50, ion-exchange rechromatography on a DEAE-Sephadex A-50, and gel filtration on a Sephadex G-150 column. The molecular mass of the filefish serine collagenase was estimated to be 27.0 kDa by gel filtration and SDS-PAGE. The purified collagenase was optimally active at pH 7.0-8.0 and $55^{\circ}C$. The purified enzyme was rich in Ala, Ser, Leu, and Ile, but poor in Trp, Pro, Tyr, and Met. In addition, the purified collagenolytic enzyme was strongly inhibited by N-P-toluenesulfonyl-L-lysine chloromethyl ketone (TLCK), diisopropylfluorophosphate (DFP), and soybean trypsin inhibitor.

  • Geranyllinalool, a polyisoprenoid compound, was found to block the early biosynthetic pathway of sphingolipids in LLC-PKl cells. Sphinganine, an intermediate in sphingolipid biosynthetic pathway, was abruptly accumulated in LLC-PKl cells at $2{\;}{\mu}M$ of fumonisin B1(FB1), a specific inhibitor of sphinganine N-acyltransferase, for 24 hr. Geranyllinalool lowered the $B_1(FB_1)$, a specific inhibitor of sphinganine N-acyltransferase, for 24 hr. Geranyllinalool lowered th FB1 and $50{\;}\mu$M geranyllinalool. l-Cy-closerine, an inhibitor of serine-palmitoyl transferase, was used as a positive control to evaluate the inhibitory effect of geranyllinalool. These results suggest that geranyllinalool may inhibit the serine-palmitoyl transferase, the first enzyme in de novo sphingolipid biosynthesis, resulting in the altered regulation of sphingolipid metabolism.

마늘의 alcohol 침지 중 휘발성 향기성분과 침출유리아미노산 함량 (Elution Profiles of Volatile Compounds and Free Amino Acids during Alcohol Soaking of Garlic(Allum sativum L.))

  • 마늘주의 산업화에 앞서 기초 자료로써 활용하고자, 마늘을 소주 등의 술에 침지하였을 때 마늘의 휘발성 기능성분과 유리아미노산이 술로 침출되는 정도 및 형태에 관하여 조사하고자, 생마늘을 20% 주정에 5주간 침지하고 매주 휘발성 향기성분과 유리아미노산들이 침출되는 양적 및 형태적 변화를 조사하였다. 마늘의 주요 유리 아미노산은 L-aspartic acid, L-glutamic acid, L-arginine, L-alanine, L- proline, L-asparagine 및 L-serine이었으며, 유리 아미노산들 모두 침지 기간의경과에 따라 침출량이 증가하였다. 또한, L-threonine, L-proline, L-valine 및 L-leucine 등의 중성아미노산들과 방향족아미노산인 tyrosine과 phenylalanine은 3주 후부터 80% 이상 침출된 반면에, 산성, 염기성 및 함황아미노산들은 5주 후에도 80% 미만이었다. 생마늘의 휘발성 향기성분은 diallyl trisulfide, diallyl disulfide, methyl allyl disulfide, 2-vinyl-4H-1,3-dithiin, 3-vinyl-3,4-dihydro-1,2-dithiin, 3,5-diethyl-1,24-trithiolane, isobutyl isothiocyanate 및 diallyl sulfide 등의 황화합물들이 주요 성분들이었다. 생마늘로부터 주정으로 침출된 휘발성 향기성분은 allyl alcohol, diallyl disulfide, 3,5-diethyl-1,2,4-trithiolane, diallyl trisulfide, 3,4-dimethoxyfuran의 5종이었으며, 침지기간의 경과에 따라 3,5-diethyl-1,2,4-trithiolane는 계속 침출량이 증가하였지만 그 외 4종은 감소하였다. 침 지기간 중 furfural, 5-methylfurfural, 5-hydroxymethylfurfural, dimethyl pyrazine, furfuryl alcohol, 3-hydroxy-2-bytanone과 2,3-dihydro-3,5-dihydroxy-6-methyl-4H-pyran-4-one이 새로이 생성되었으며, 침지기간의 경과에 따라 침출주에 대부분 축적되는 양상을 보였다.

  • The nutritional significance of essential amino acids, as well as non-essential amino acids, is well documented in poultry production with regards to growth performance and protein accretion. However, the function of amino acids in the stress response is still unclear. L-Pipecolic acid, a L-lysine metabolite in the brain, induced a hypnotic and sedative effect acting via the ${\gamma}$- aminobutyric acid receptors. L-Arginine also induced a sedative effect via its metabolism to L-ornithine. In addition, three-carbon nonessential amino acids like L-alanine, L-serine and L-cysteine also induced sedative effects. These facts suggest that the requirement for amino acids in both essential and non-essential types may require reconsideration to add the concept of stress amelioration in the future.

Threonine의 생물공학적 생산 (Biotechnology for the Production of Threonine Production)

  • Various methods are available for the production of L-threonine. The microbial production of L-threonine has been achieved by breeding L-threonine analog-resistant auxotrophic mutants of various bacteria. The enzymatic production of L-threonine has been demonstrated by use of threonine metabolic enzymes such as threonine deaminase, threonine aldolase, or threonine dehydrogenase complex. Threonine synthesis from glycine and ethanol seems to be catalyzed by the enzymes Methanol dehydrogenase(MDH) and Serine hydroxymethyltransferase(SHMT), which was also found to catalyze the aldol condensation of glycine with acetaldehyde. The improved production of L-threonine has been achieved by amplifying the genes for the L-threonine biosynthetic enzymes using recombinant DNA techniques.
