Introduction and Expression of Foreign Genes in Rice Cells by Particle Bombardment

  • For establishing a transformation system of rice, an efficient introduction of foreign genes into embryogenic cell suspension by particle bombardment was conducted. The particle inflow gun based on the acceleration of DNA-coated tungsten particles using pressurized helium was constructed for delivery of DNA into rice cells. Several bombardment parameters were optimized using the transient expression of GUS gene. The conditions that gave the highest GUS gene expression of about 1000 blue spots per g fresh weight of bombarded cells include treatment of the cells with 0.5 M osmotic pressure, and use of the 410 kPa helium, 110 mm target distance, 13 mm syringe filter holder and 5 $\mu$L DNA/tungsten mixtures. It was also confirmed that rice actin promoter-intron construct gave the highest expression of all promoter-sequences studied. Eight weeks after the bombardment, stably transformed calluses were obtained on the selection medium containing 100 mg/L G418 and showed the strong activity in in situ GUS assay.

Isolation of Hepatoprotective Phenylpropanoid from Lactuca indica

  • In continuing our search for biologically active compounds from Korean Compositae medicinal plants, we investigated the constituents of the aerial parts of Lactuca indica L. and isolated a phenylpropanoid derivative from its MeOH extract. The chemical structure was characterized by spectroscopic methods, including 1D and 2D NMR to be di-E-caffeoyl-meso-tartaric acid (1). Compound 1 was isolated for the first time from this plant. In this paper, we suggest that the NMR assignment at C-2 of (+)-taraxafolin-B should be corrected. In the human HBV-transfected liver cell line HepG2.2.15, the compound 1 effectively reduced HBV DNA level in the release of mature HBV particles from HepG2.2.15 cultivation.

Effect of Organic Materials in Water Treatment by Hybrid Module of Multi-channel Ceramic Microfiltration and Activated Carbon Adsorption

  • We investigated the effect of organic materials on membrane fouling in advanced drinking water treatment by a hybrid module packed with granular activated carbon (GAC) outside multi-channel ceramic microfiltration membrane. Synthetic water was prepared with humic acid and kaolin to simulate natural water resouces consisting of natural organic matter and inorganic particles. Kaolin concentration was fixed at 30 mg/L and humic acid was changed as 2~10 mg/L to inspect the effect of organic matters. Periodic back-flushing using permeate water was performed for 10 sec per filtration of 10 min. As a result, both resistance of membrane fouling (Rf) and permeate flux (J) were influenced highly by concentration of humic acid. It proved that NOM like humic acid could be an important factor on membrane fouling in drinking water treatment. Turbidity and UV254 absorbance were removed up to above 97.4% and 59.2% respectively.

Isolation and characterization of bacteriophage infecting Lactobacillus plantarum KCCM 12116

  • Bacteriophages (phages) are known determinants of kimchi microbial ecology. Lactobacillus plantarum is related to kimchi over-acidification during the late stages of kimchi fermentation. A phage infecting Lac. plantarum was isolated from kimchi and characterized. The phage population for kimchi in a market was 2.3 log particles/mL, which corresponded to 32% of the bacterial population on a log scale. The isolated phage was designated as ΦLP12116. ΦLP12116 which belonged to the Siphoviridae family and has a very narrow host range, infecting only Lac. plantarum. The phage was stable at a lactic acid concentration of 1.0% and pH 4.0 at 4℃, indicating that it could survive in kimchi. In the kimchi extract broth treated by the phage, the growth of Lac. plantarum KCCM 12116 was inhibited by 2.2 log CFU/mL compared to the growth in non-phage-treated broth. Therefore, this study suggests that the growth of Lac. plantarum, which is known as an acid-producing strain during late fermentation in kimchi, may be controlled using the phage.

Antibacterial properties of composite resins incorporating silver and zinc oxide nanoparticles on Streptococcus mutans and Lactobacillus

  • Objectives: Recurrent caries was partly ascribed to lack of antibacterial properties in composite resin. Silver and zinc nanoparticles are considered to be broad-spectrum antibacterial agents. The aim of the present study was to evaluate the antibacterial properties of composite resins containing 1% silver and zinc-oxide nanoparticles on Streptococcus mutans and Lactobacillus. Materials and Methods: Ninety discoid tablets containing 0%, 1% nano-silver and 1% nano zinc-oxide particles were prepared from flowable composite resin (n = 30). The antibacterial properties of composite resin discs were evaluated by direct contact test. Diluted solutions of Streptococcus mutans (PTCC 1683) and Lactobacillus (PTCC 1643) were prepared. 0.01 mL of each bacterial species was separately placed on the discs. The discs were transferred to liquid culture media and were incubated at $37^{\circ}C$ for 8 hr. 0.01 mL of each solution was cultured on blood agar and the colonies were counted. Data was analyzed with Kruskall-Wallis and Mann-Whitney U tests. Results: Composites containing nano zinc-oxide particles or silver nanoparticles exhibited higher antibacterial activity against Streptococcus mutans and Lactobacillus compared to the control group (p < 0.05). The effect of zinc-oxide on Streptococcus mutans was significantly higher than that of silver (p < 0.05). There were no significant differences in the antibacterial activity against Lactobacillus between composites containing silver nanoparticles and those containing zinc-oxide nanoparticles. Conclusions: Composite resins containing silver or zinc-oxide nanoparticles exhibited antibacterial activity against Streptococcus mutans and Lactobacillus.

A study on influence of precipitation condition on rounding of AUC particles (AUC 침전조건이 둥근 AUC 입자 제조에 미치는 영향)

  • Mechanisms and conditions for rounding of AUC particles were examined during AUC precipitation. Rounding of AUC particle was possible only by external circulation using pump, not by internal circulation using agitator. The rate of AUC rounding $(dn_p/dt)$ was proporational to operation conditions such as magma density $(M_t:g-U/{\iota}l)$, turn over ratio $(T_o)$ and impeller tip velocity of pump (U); $ dn_p/dt{\propto}M_t{\cdot}T_o{\cdot}U^2$. The validity of this relationship was qualitatively confirmed by comparing the expermental results. Two rounding mechanisms were suggested. One is crack formation mechanism and the other etch-pit formation mechanism on the surface of AUC particle. It was found that the crack formation is more dominant at the initial stage and the etch-pit formation at the final stage of the AUC precipitation.

Estimation of damage area on membrane surface by application of fluorescent particles as a surrogate (형광입자를 이용한 분리막 표면 검측과 손상 면적 추정 오차에 대한 연구)

  • In this study, a novel method was proposed to test the integrity of water treatment system specifically equipped with membrane filtration process. We applied the silica particles coated with a fluorescent agent (rhodamine B isothiocyanate) as a surrogate to detect a membrane process integrity and evaluate the reliability of effluent quality in the system. Additionally, a series of experiments was conducted to evaluate the sensitivity of the method through the laboratory scale experiment. The laboratory scale experiments showed that the feasibility of application of proposed method to detect a breach or damaged part on the membrane surface. However, the sensitivity on predicting the area of a breach was significantly influenced by the testing conditions such as a concentration of surrogate, filtration flux, and detection time. The lowest error of predicting the area of breach was 3.5% at the testing condition of surrogate concentration of 80 mg/L injected with flux of $20L/m^2/hr$ for 10 minutes of detection time for the breach having the actual area of $7.069mm^2$. However, the error of estimation was increased at the small breach with area less than $0.785mm^2$. A future study will be conducted to estimate a damaged area with more accuracy and precision.

  • The preservation of biological activity of protein drugs in formulation is still a major challenge for successful drug delivery. Lipase was encapsulated in poly (D,L-lactide- co-glycolide) PLGA nano-particles using a w/o/w solvent evaporation technique. The lipase-containing PLGA/poly (vinyl alcohol) (PVA) nanoparticles were characterized with regard to morphology, size, size distribution, lipase-loading efficiency, in vitro lipase release, and stability of lipase activity. The size of nanoparticles increased as polymer concentration was increased. The size of particles was not significantly affected by the PVA concentration; on the other hand, the particle size distribution was the narrowest when 4% of PVA was used. In optimum conditions, we possessed nanoparticles that characterized 72.5% of encapsulation efficiency, $198.3{\pm}13.8 nm$ size diameter. During the initial burst phase, the in vitro release rate was very fast, reaching 83% within 12 days. Until days 6, enzyme activity increased as the amount of lipase released was increased.

Analysis and Mechanical Behavior of Coating Layer in Metallic Glass Matrix Composite (비정질 기지 복합재 코팅층의 미세조직 분석 및 기계적 거동)

  • For surface modification, bulk metallic glass coatings were fabricated using metallic glass powder and a mixture of a self-fluxing alloy or/and hard metal alloys with a heat-resisting property using a high velocity oxy-fuel coating thermal spraying process. Microstructural analyses and mechanical tests were carried out using X-ray diffraction, a scanning electron microscope, an atomic force microscope, a three-dimensional optical profiler, and nanoindenation. As a result, the monolithic metallic glass coating was found to consist of solid particle and lamellae regions that included many pores. Second phase-reinforced composite coatings with a self-fluxing alloy or/and hard metal alloy additives were employed with in-situ $Cr_2Ni_3$ precipitate or/and ex-situ WC particles in an amorphous matrix. The mechanical behaviors of the solid particles and lamella regions showed large hardness and elastic modulus differences. The mechanical properties of the particle regions in the metallic glass composite coatings were superior to those of the lamellae regions in the monolithic metallic glass coatings, but indicated similar trends in matrix region of all the coating layers.

A Study on CMP Characteristics According to Shape of Colloidal Silica Particles (콜로이달 실리카 입자 형상에 따른 CMP 특성에 관한 연구)

  • Slurry used for polishing semiconductors processed by exchange, pressurization, and multi-step feeding has been studied to investigate the effect of the size and shape of slurry particles on the oxide CMP removal rate. First, spherical silica sol was prepared by the ion exchange method. The spherical silica particle was used as a seed to grow non-spherical silica sol in accordance with the multi-step feeding of silicic acid by the ion exchange and pressurization methods. The oxide removal rate of both non-spherical silica sol and commercially available slurry were compared with increasing average particle size in the oxide CMP. The more alkaline the pH level of the non-spherical silica sol, the higher was the removal rate and non-uniformity.
