Aim of the study: Scaffolds are crucial to tissue engineering/regeneration. Biodegradable polymer/ceramic composite scaffolds can overcome the limitations of conventional ceramic bone substitutes such as brittleness and difficulty in shaping. In this study, poly(L-lactide)/hydroxyapatite(PLLA/HA) composite scaffolds were fabricated for in vivo bone tissue engineering. Material & methods: In this study, PLLA/HA composite microspheres were prepared by double emulsion-solvent evaporation method, and were evaluated in vivo bone tissue engineering. Bone marrow mesenchymal stem cell from rat iliac crest was differentiated to osteoblast by adding osteogenic medium, and was mixed with PLLA/HA composite scaffold in fibrin gel and was injected immediately into rat cranial bone critical size defect(CSD:8mm in diameter). At 1. 2, 4, 8 weeks after implantation, histological analysis by H-E staining, histomorphometric analysis and radiolographic analysis were done. Results: BMP-2 loaded PLLA/HA composite scaffolds in fibrin gel delivered with osteoblasts differentiated from bone marrow mesenchymal stem cells showed rapid and much more bone regeneration in rat cranial bone defects than control group. Conclusion: This results suggest the feasibility and usefulness of this type of scaffold in bone tissue engineering.
Application of membranes for guided tissue regeneration(GTR) have been confined to the subgingival barrier functions; however, many studies have provided evidence that some drugs, including tetracycline, initially can promote the growth of periodontal ligament or alveolar bone in peridontal therapy. Osseous regeneration in periodontal defects is increased by local administration of tetracycline due to its anti-collagenolytic effect, which enhances bone-forming ability via osteoblast cell chemotaxis and reduced bone resorption. The aim of this study was to evaluate effects of tetracycline loaded poly-L-lactide(PLLA) barrier membranes for guided bone regenerative potential. Tetracycline was incorporated into the PLLA membrane with the ratio 10% to PLLA by weight. Ability to guided bone regeneration of the membranes were tested by measuring new bone in the tibial defects($7{\times}10{\times}5\;mm^3$) of the beagle dog for 4,5, and 6 weeks. In control, drug-unloaded PLLA membranes were used in same size of defect. In histologic finding of the defect area, a few inflammatory cells were observed in both groups. These membrane were not perforated by connective tissue and maintained their mechanical integrity for the barrier function for 4-6 weeks. New bone formation was greater in defects covered by tetracycline-loaded membrane than in defects covered by drug- unloaded membranes. In bone regeneration guiding potential test, tetracycline-loaded membrane was more effective than drug- unloaded membranes(p<0.05). These results suggest that tetracycline-loaded PLLA membranes potentially enhance guided bone regenerative efficacy and might be a useful barrier for GTR in periodontal treatment.
Biodegradable and elastic poly(L-lactide-co-$\varepsilon$-caprolactone) (PLCL) was electrospun to prepare nanofibers, and N-isopropylacrylamide (NIPAAm) was then grafted onto their surfaces under aqueous conditions using $^{60}Co-{\gamma}$ irradiation. The graft yield increased with increasing irradiation dose from 5 to 10 kGy and the nanofibers showed a greater graft yield compared with the firms. SEM confirmed that the PLCL nanofibers maintained an interconnected pore structure after grafting with NIPAAm. However, overdoses of irradiation led to the excessive formation of homopolymer gels on the surface of thc PLCL nanofibers. The equilibrium swelling and deswelling ratio of the PNIPAAm-g-PLCL nanofibers (prepared with 10 kGy) was the highest among the samples, which was consistent with the graft yield results. The phase-separation characteristics of PNIPAAm in aqueous conditions conferred a unique temperature-responsive swelling behavior of PNIPAAm-g-PLCL nanofibers, showing the ability to absorb a large amount of water at < $32^{\circ}C$, and abrupt collapse when the temperature was increased to $40^{\circ}C$. In accordance with the temperature-dependent changes in swelling behavior, the release rate of indomethacin and FITC-BSA loaded in PNIPAAm-g-PLCL nanofibers by a diffusion-mediated process was regulated by the change in temperature. Both model drugs demonstrated greater release rate at $40^{\circ}C$ relative to that at $25^{\circ}C$. This approach of the temperature-controlled release of drugs from PNIPAAm-g-PLCL nanofibers using gamma-ray irradiation may be used to design drugs and protein delivery carriers in various biomedical applications.
Vascular tissue engineering has been accessed to mimic the natural composition of the blood vessel containing intima, media, and adventitia layers. We fabricated mechanically expanded PLLA/PLCL nanofibers using electrospinning and UTM. The pore size of the meshes was increased the gelatin immobilized AAc-PLLA/PLCL nanofibers ($203.30{\pm}49.62microns$) than PLLA/PLCL nanofibers ($59.99{\pm}8.66microns$) after mechanical expansion. To increase the cell adhesion and proliferation, we introduced carboxyl group, and gelatin was conjugated on them. The properties of the PLLA/PLCL nanofibers were analyzed with SEM, ATR-FTIR, TBO staining, and water contact angle measurement, general cell responses on the PLLA/PLCL nanofibers such as adhesion, proliferation, and infiltration were also investigated using smooth muscle cell (SMC). During the SMC culture, the initial viability of the cells was significantly increased on the gelatin immobilized AAc-PLLA/PLCL nanofibers, and infiltration of the cells was also enhanced on them. Therefore, gelatin immobilized AAc-PLLA/PLCL nanofibers and mechanically expanded meshes may be a good tool for vascular tissue engineering application.
In tissue engineering application, a fibrous structure of scaffolds has been issued as an alternative system to regulate cell survival and tissue regeneration, and electrospinning technique has been popularly used to generate fibrous meshes or sheets mimicking the structure of native extracellular matrix (ECM). However, recent strategy in the scaffold development is expanded to provide the structural property as well as a biological property of native ECM, a variety of surface modification techniques have been used to introduce biological property. In this study, we developed biomimetic poly(L-lactide-co-${\varepsilon}$-caprolactone) (PLCL) nano- and micro-fibrous scaffolds as a unique platform with structural and biological properties with native ECM using electrospinning method and gamma-ray irradiation. Surface morphology of the scaffolds was observed by scanning electron microscopy, and alteration of surface property was evaluated with toluidine blue O staining, water contact angle measurement and ATR-FTIR analysis.

In order to endow with new bioactive functionality from small intestine submucosa (SIS) powder as natural source to poly (L-lactide) (PLA) and poly (lactide-co-glycolide) (PLGA) synthetic biodegradable polymer, porous SIS/PLA and SIS/PLGA as natural/synthetic composite scaffolds were prepared by means of the solvent casting/salt leaching methods for the possibility of the application of tissue engineered bone and cartilage. A uniform distribution of good interconnected pores from the surface to core region was observed the pore size of 40~500 ${\mu}{\textrm}{m}$ independent with SIS amount using the solvent casting/salt leaching method. Porosities, specific pore areas as well as pore size distribution also were almost same. After the fabrication of SIS/PLA hybrid scaffolds, the wetting properties was greatly enhanced resulting in more uniform cell seeding and distribution. Five groups as PGA non-woven mesh without glutaraldehyde (GA) treatment, PLA scaffold without or with GA treatment, and SIS/PLA (Code No.3 ; 1 : 12 of salt content, (0.4 : 1 of SIS content, and 144 ${\mu}{\textrm}{m}$ of median pore size) without or with GA treatment were implanted into the back of nude mouse to observe the effect of SIS on the induction of cells proliferation by hematoxylin and eosin, and von Kossa staining for 8 weeks. It was observed that the effect of SIS/PLA scaffolds with GA treatment on bone induction are stronger than PLA scaffolds, that is to say, in the order of PLA/SIS scaffolds with GA treatment > PLA/SIS scaffolds without GA treatment > PGA nonwoven > PLA scaffolds only with GA treatment = PLA scaffolds only without GA treatment for the osteoinduction activity. The possible explanations are (1) many kinds of secreted, circulating, and extracellular matrix-bound growth factors from SIS to significantly affect critical processes of tissue development and differentiation, (2) the exposure of SIS to GA resulted in significantly calcification, and (3) peri-implant fibrosis due to covalent bonding between collagen molecule by crosslinking reaction. In conclusion, it seems that SIS plays an important role for bone induction in SIS/PLA scaffolds for the application of tissue engineering area.
The mixture density data for poly(lactide-co-glycolide) [PLGA] with supercritical $CO_2$, CHF$_3$ and CHClF$_2$ were obtained in the temperature range of 27 to 10$0^{\circ}C$ and at pressures as high as 3000 bar (PLGA$_{x}$, Where the molar concentration of glycolide in the backbone, x, range from 0 to 50 mol%). The PLA-$CO_2$, PLA-CHF$_3$, and PLA-CHClF$_2$ systems dissolve in the pressure less than 1430 below 700, and below 100 bar, respectively. The mixture density shows from 1.084 to 1.334 g/cm$^3$ at temperatures from 27 to 93$^{\circ}C$. The PLGA$_{15}$ -$CO_2$ mixture dissolves at pressures of below 1900 bar and the mixture density is in the range of 1.158 to 1.247 g/cm$^3$ at temperatures between 37 and 92$^{\circ}C$. The solubilities of the PLGA$_{25}$ for $CO_2$, CHF$_3$, and CHClF$_2$ are shown to pressure as high as 2390, 1470, and 118 bar, respectively, and the mixture density exhibits iron 1.154 to 1.535 g/cm$^3$ at temperatures from 29 to 81$^{\circ}C$. The PLGA$_{50}$-$CO_2$ system does not dissolve at 24$0^{\circ}C$ and 3000 bar while the PLGA$_{50}$-CHCIF$_2$ does easily at 5$0^{\circ}C$ and 100 bar. The mixture density for the PLGA-CHClF$_2$ system increases even at low pressures as the glycolide molar concentration increases.es.es.
Cell attachment and proliferation on the polymer films of triblock copolymer(ester-ether)s comprising po1y (L-1actide) (PLLA) and poly (oxyethylene-co-oxypropylene)(PN) were investigated using 3T3 fibroblasts. It was found that on the tissue culture polystyrene(TCPS) and the PLLA control film the cells could spread well while on the copolymer films the cells showed a rounded morphology without spreading and proliferated weakly. Especially, little cells proliferated on the films of copolymer having a LN composition of 20 wt%. While the water absorption of the copolymer films increased with increasing PN content, the contact angle against water of copolymer films immersed in aqueous medium was almost identical, being slightly lower than that of the PLLA film. These properties were compatible with the results of cell attachment. The in vitro hydrolysis of the films of triblock and multiblock type copolymers was faster with increasing PN content. The increased hydrolyzability, the flexibility and the decreased cell attachment suggested that these copolymers may have high potential as biodegradable materials for medical use.
The A-B-A type block copoly(ester-ether)s consisting of poly(L-lactic acid) (PLLA)(A) and poly(oxyethylene-co-oxypropylene)(B) were prepared to improve the mechanical properties and hydrolyzability of PLLA. The block copolymers showed an improved flexibility due to the incorporation of the soft segments. Then, the same copolymer has an improved anti-thrombogenicity probably due to the specific microphase separation structure in the surface. The AFM of the film of the block copolymer revealed that the surface was quite flat in comparison with that of PLLA. Therefore, the flatness of the surface may be related with the increased anti-thrombogenicity of the copolymer film.

Porous poly(L-lactic acid)(PLLA) scaffold membranes were prepared via. phase separation process. Chloroform, dichloromethane and 1,4-dioxane were used as solvent and, ethyl alcohol was used as non-solvent. Morphologies, mechanical properties and mass transfer characteristics of the scaffold membranes were investigated through SEM, stress-strain test and glucose diffusion test. The scaffold membranes obtained from the casting solutions with chloroform and with dichloromethane showed similar morphologies. They showed sponge-like porous structure with the pore size in the range of $3-10{\mu}m$ and, their porosities were in 50-80% range. Using 1,4-dioxane as solvent, nano-fibrous scaffold membranes with porosities over 80% were fabricated. When the polymer content in the solution with 1,4-dioxane was lowered to 4%, highly porous, macroporous and nano-fibrous scaffold membranes were obtained. The size of the macropore was tens of the microns and the porosity was around 90%. These results indicate that the solvent has significant effect on the scaffold membrane structure and, that scaffold membranes with various structures can be fabricated through phase separation method by choosing solvent and by controlling polymer concentration in the casting solution.
