An Integrative Review of Health-related Quality of Life in Patients with Advanced Heart Failure (진행성 심부전 환자의 건강관련 삶의 질에 대한 통합적 고찰)

  • Purpose: Even though advanced heart failure (HF) severely affects the patient's health-related quality of life (HRQoL), there is little information regarding this issue. This review is aimed to describe the relevant clinical characteristics of patient with advanced HF and identify factors influencing HRQoL in these patients. Methods: Empirical articles were searched from electronic databases issued from January 2000 to June 2018 with using the key terms 'heart failure' and 'quality of life'. There were a total of 22 articles that met the inclusion criteria and were analyzed for this study. Results: First, nine studies among 22 studies clearly stated that their participants were samples of patients with advanced HF. Most reviewed studies showed the New York Heart Association (NYHA) class as the criteria for identifying advanced HF. Second, the level of HRQoL varied depending on the measurement tools utilized by the researchers. Third, the NYHA class, gender, and symptoms were mainly associated with HRQoL in patients with advanced HF. Also, nurse- or physician-led intervention, exercise, spiritual-focused intervention, and palliative care improved the HRQoL of the patients with advanced HF. Conclusion: This study found that the clear application of criteria for advanced HF and the development of advanced HF-specific HRQoL measurement was needed. Prospective studies should be considered for identifying differences in the levels and factors influencing HRQoL in patients with early stage or advanced HF to design patient-centered care.

Evaluation of Bacteriological Safety for the Shellfish growing Sea waters in Seocheon Area, Korea (충남 서천 패류생산해역에서의 세균학적 위생안전성 평가)

  • A Sanitary survey in an Important shellfish-growing area of Biin Bay in Seocheon-gun Korea, was conducted to evaluate bay conditions and compliance with the bacteriological criteria for areas designated for the production of shellfish for export. Seawater samples were collected monthly at 55 sampling stations established in the survey area from January 2006 to December 2008. Bacteriological water quality did not change in response to a small rainfall (10.0 mm), but it increased abruptly in response to rainfall to 62.5 mm The most probable number (MPN) value for total coliforms and fecal coliforms in 1, 980 seawater samples ranged from <1.8 to >1, 600 MPN/100mL and from <1.8 to 330 MPN/100mL, respectively. The geometric mean and estimated $90^{th}$ percentile value for total coliforms ranged from 2.0 to 10.3 MPN/100mL and from 7.0 to 42.6 MPN/100mL respectively. The geometric mean and estimated percentile value for fecal coliforms ranged from 1.8 to 4.0 MPN/100mL and from 1.9 to 18.3 MPN/100mL, respectively. Accordingly, the bacteriological water quality of Biin Bay met the National Shellfish Sanitation Program (NSSP) and Korea Shel1fish Sanitation Program(KSSP) criteria for areas designated for shellfish production for export in Korea.

Comparison of Synthetic Lysine Sources on Growth Performance, Nutrient Digestibility and Nitrogen Retention in Weaning Pigs

  • We compared the effects of supplementing $L-lysine{\cdot}SO_4$ to L-lysine HCl on growth performance, nutrient digestibility and nitrogen retention in weaning pigs. A total of 96 crossbred pigs, weaned at $21{\pm}3$ days of age and with an average initial body weight (BW) $6.23{\pm}0.01kg$, were given one of 4 treatments, which translated into 6 replicates of 4 pigs in each pen. The animals were randomly assigned to four dietary treatments according to a randomized completely block design (RCBD) as follows: 1) control-no synthetic lysine, lysine deficient (0.80% total lysine); 2) L-C (= 0.2% L-lysine HCl); 3) K-L-S (= 0.332% $L-lysine{\cdot}SO_4$, A company); 4) C-L-S (= 0.332% $L-lysine{\cdot}SO_4$, B company). Diets were formulated with corn, soy bean meal, and corn gluten meal as the major ingredients, and all nutrients except the lysine met or exceeded NRC requirements (1998). The lysine content of supplemented synthetic lysine was the same in all treatment groups except the control. No clinical health problems associated with the dietary treatments were observed. During the entire experimental period, body weight, average daily gain (ADG) and feed efficiency (G:F ratio) increased (p<0.01) in pigs fed the experimental diets supplemented with L-lysine??HCl or $L-lysine{\cdot}SO_4$ produced by A company, irrespective of the two synthetic lysine sources. Although the supplementation of $L-lysine{\cdot}SO_4$ produced by B company tended to improve the ADG and G:F ratio, significant differences were not seen among all treatments and tended to be lower than the L-C (L-lysine HCl) and K-L-S ($L-lysine{\cdot}SO_4$ groups using the product from A company). The digestibility of crude protein (CP) was increased by the supplementation of synthetic lysine (p<0.05), irrespective of the L-lysine source (L-C, K-L-S, C-L-S). The results of this study showed that ADG, G:F ratio, and CP digestibility improved when $L-lysine{\cdot}SO_4$ or L-lysine HCl was supplemented into the weaning pigs' diet. There was a clear difference in efficacy between the two $lysine{\cdot}SO_4$ products based upon the growth performance of weaning pigs. Consequently, the bioavailability of $lysine{\cdot}SO_4$ products should be evaluated before supplementation of synthetic lysine in swine diets.

The relationship between catechol-O-methyltransferase gene polymorphism and coronary artery abnormality in Kawasaki disease (가와사키병의 관상동맥 이상과 catechol-O-methyltransferase 유전자의 단일염기다형성)

  • Purpose : Many gene polymorphisms are associated with coronary artery abnormalities in Kawasaki disease. Catechol-O- methyltransferase (COMT) plays an important role in the metabolism of catecholamines, catechol estrogen, and catechol drugs. Polymorphisms of the COMT gene are reported to be associated with myocardial infarction and coronary artery abnormalities. The aim of this study was to evaluate the relationship between COMT gene polymorphisms and coronary artery abnormalities in Kawasaki disease patients. Methods : One hundred and one Korean children with Kawasaki disease and 306 healthy Korean control subjects were enrolled in this study. The polymorphisms of the COMT gene were analyzed by direct sequencing. Results : There were no differences in the genotype and allelic frequency of the rs4680 and rs769224 polymorphic sites between Kawasaki disease and control subjects. Further, no significant difference was found in the rs4680 polymorphism between patients with coronary artery abnormalities and patients without coronary artery abnormalities (codominant P=0.32, dominant P=0.74, recessive P=0.13). However, the distribution of the rs769224 polymorphism was significantly different between patients with coronary artery abnormalities and patients without coronary artery abnormalities (codominant P= 0.0077, dominant P=0.0021, recessive P=0.16). Conclusion : Our results indicate that the polymorphisms of the rs769224 gene might be related to the development of coronary artery abnormalities in Kawasaki disease.

Improvement of L-Lysine Productivity by Using Cell Fusion and Immobilized System (세포융합과 고정화 시스템을 이용한 L-Lysine의 생산성 향상)

  • This studies were designed to improve the productivity of L-lysine by protoplast fusion and immobilized system of fusants using strains of Brevibacterium flavum ATCC 21528, Brevibacterium lactofermentum ATCC 21086 and Corynebacterium glutamicum 820. Mutants were isolated with concentration method of $300{\mu}g/ml$ penicillin-G after treatment of $250{\mu}g/ml$ N-methyl-N-nitro-N-nitrosoguanidine. B. flavum $37-2(Hos^-,\;Kan^r,\;AEC^r)$, B. lactofermentum $6-2(Ile^-,\;Val^-,\;Str^r,\;AEC^r)$ and C. glutamicum 57-5$(Met^-,\;Thr^-,\;Rif^r,\;AEC^r)$ were isolated from mutants. Protoplasts were induced by being incubated with $500{\mu}g/ml$ lysozyme of lysis solution for 6 hr and the ratio of protoplast formation and regeneration were ranging from 97-99% and 33-37%, respectively. Fusion frequencies of fusants of BBFL 21, BCFG 37 and BCLG 59 were shown in the range from $1.25{\times}10^{-6}\;to\;5.83{\times}10^{-7}$ under the optimum conditions. The fusant BBFL 21 showed the highest productivity of $411.1\;ng/ml{\cdot}hr$ L-lysine in the lysine productivity broth at $30^{\circ}C$ for 72hr. In the immobilization systems, fusant BBFL 21 was employed in various polymer matrices such as sodium alginate, polyacrylamide, agar and ${\alpha}-carrageena$. The immobilization of sodium alginate showed the highest productivity of $413\;ng/ml{\cdot}hr$ L-lysine in the batch system. Continuous fermentation of immobilization system by using tube fermentor was produced the highest productivity $416.7\;ng/ml{\cdot}hr $ L-lysine under optimum condition.

Water Quality Monitoring for Hazard Analysis in Aquaculture Farm of Rainbow Trout (송어양식장의 위해요소 관리를 위한 수질 모니터링)

  • Water quality has been considered to be one of sanitation standard operating procedures (SSOP) for hazard analysis critical control points (HACCP) application in aquaculture farms. This study was conducted to evaluate a hazard caused by water used in aquaculture farm of rainbow trout. The water quality was analyzed to investigate both physiochemical and bacteriological level in water samples collected from aquaculture farm of rainbow trout, Oncorhynchus mykiss. No significant difference were observed on water temperature and pH from season to season. However, the levels of dissolved oxygen were decreased as the outside temperature was increased, even if the levels were adequate for aquaculture. Also, other physiochemical analysis including biochemical oxygen demand (BOD), chemical oxygen demand (COD) and suspended solid (SS) revealed that the waters for aquaculture analyzed in this study was suitable for rainbow trout aquaculture. The bacterial analyses were also revealed that the waters for aquaculture were met to both coliform group (<18 MPN/100mL) and viable cell count (<100 CFU/mL). However, some of waste waters from aquaculture farms showed higher levels of BOD and COD than those of waste water standard (<2 ppm), suggesting that regular cleaning of fish tank and precipitation tank is needed.

Purification and characterzation of the $\alpha$-L-Arabinofuranosidase from Escherichia coli Cells Harboring the Recombinant Plasmid pKMG11 (재조합 균주 Escherichia coli가 생산하는 Bacillus stearothermophilus $\alpha$-L-Arabinofuranosidase의 정제 및 특성)

  • $\alpha $-Arabinofuranosidase was produced by E. coli HB101 haboring the recombinant plasmid pKMG11 which contained the arfI gene of Bacillus stearothermophilus. The maximum production of the enzyme was observed when E. coli HB101 cells were grown at 37$\circ$C for 20 hours in the medium containing 0.5% arabinose, 1.0% tryptone, 0.5% yeast extract, and 1% NaCl. The $\ALPHA $-arabinofuranosidase produced was purified to homogeneity using a combination of 20-50% ammonium sulfate precipitation, DEAE-Sepharose CL-6B ion exchange column chromatography and Sepharose 6B-100 gel filtration. The purified enzyme was most active at 55$\circ$C and pH 6.5. The K$_{m}$ and V$_{max}$ values of the enzyme on $\rho $-nitrophenyl-$\alpha $-arabinofuranoside was determined to be 2.99 mM and 0.43 $\mu $mole/min (319.74 $\mu $mole/min/mg), respectively. The pI value was 4.5. The molecular weight of the native protein was estimated to be 289 kDa. The SDS-polyacrylamide gel clectrophoresis analysis suggested that the functional protein was a trimer of the 108 kDa identical subunits. The N-terminal amino acid sequence of the a-arabinofuranosidase was identified as X-Ser-Thr-Ala-Pro-Arg( \ulcorner )-Ala-Thr-Met-Val-Ile-Asp-X-Ala-Phe.

Chirality Conversion of Dipeptides in the Schiff Bases of Binol Aldehydes with Multiple Hydrogen Bond Donors

  • Novel binol aldehydes derivatized at 2' hydroxy position with both uryl and acetamide groups (2), and diuryl groups (3) have been synthesized. Both were designed for streospecific binding and chirality conversion of general dipeptides with support of multiple hydrogen bonding donor sites in the receptors. The receptors, 2 and 3, converted the chirality of N-terminal amino acids of peptides such as Ala-Gly, Met-Gly, Leu-Gly and His-Gly with stereoselectivity on D-form over L-form. The stereoselectivity ratios were in the range of 5-11, somewhat higher than those of the binol receptor with mono uryl group (1). The DFT calculation at the B3LYP/6-31G$^*$//MPWB1K/6-31G$^*$ level revealed that 3-D-Ala-Gly was 2.2 kcal/mol more stable than 3-L-Ala-Gly. The considerable steric hindrance between the methyl group of the alanine and the imine CH moiety of the receptor seems to be the main contributing factor for the thermodynamic preference.

Fusion between Protopldsts of Ganoderma applanatum and Oidia of Lyophyllum ulmarium (잔나비걸상버섯 원형질체(原形質體)와 만가닥버섯 분열자(分裂子)의 융합(融合))

  • The fusion between protoplasts of Ganoderma applanatum and oidia of Lyophyllum ulmarium (Hypsizigus marmoreus) was induced with polyethylene glycol and $CaCl_2$. When transferred to Ganoderma complete medium plates, fusants showed mixed morphologies both parents. During three times subcultivation the fusants were changed similar to those of L. ulmarium type. All fusants produced oidia, clamp connections and basidiocarps similar to those of L. ulmarium. Isozyme pattern of esterase of interorder fusants showed both parental and non-parental bands. Each individual fusant did not showed both parental and non-parental bands. Each individual fusant did not show any differences in mycelial growth rate, colony morphology, esterase band pattern and basidiocarp.

Effects of Aerobic Granular Sludge Separator on the Stability of Aerobic Granular Sludge (AGS) (호기성 그래뉼 슬러지 선별 분리기가 호기성 그래뉼 슬러지의 안정성에 미치는 영향)

  • In this study, the effect on the stability of Aerobic Granular Sludge (AGS) caused by an AGS separator was investigated. The AGS separator was a hydrocyclone. The main factors of the AGS separator were filter pore size (0.125~0.600 mm), conical-to-cylindrical ratio (1.5~3.0), and operating time (1~20 min). The AGS/mixed liquor suspended solid (MLSS) ratio gradually increased to 0.500 mm (AGS/MLSS: 84.3±3.0%). AGS was best separated at the conical-to-cylindrical ratio of 2.5 (AGS/MLSS: 84.7±3.3%). As the operating time increased, the AGS separation performance also tended to increase. The shortest AGS separator run time, but the highest AGS separation performance was 10 min (87.0±2.5%). AGS stability was evaluated by operating the selected AGS separator and sequencing batch reactor. The average removal efficiencies of TOC, TCODCr, SS, TN, and TP were 95.7%, 96.9%, 93.0%, 89.0%, and 96.2%, respectively, which met the effluent standards in Korea. In addition, the AGS/MLSS ratio tended to remain constant, and the sludge volume index demonstrated a tendency to decrease from 140 mL/g to 70 mL/g. During the operation, the particles of AGS in optical microscope observations gradually increased.
