소수성 및 친수성 담체를 이용한 Trickling Bed Biofilter의 생물학적 수소생산 (The Fermentative Hydrogen Production in Trickling Bed Biofilter Filled with Hydrophilic-and Hydrophobic-Media)

  • Two mesophilic trickling bed bioreactors filled with two different types of media, hydrophilic- and hydrophobic-cubes, were designed and conducted for hydrogen production under the anaerobic fermentation of sucrose. Each bioreactor consisted of the column packed with polymeric cubes and inoculated with heat-treated sludge obtained from anaerobic digestion tank. A defined medium containing sucrose was fed by the different hydraulic retention time(HRT), and recycle rate. Hydrogen concentrations in gas-phase were constant, averaging 40% of biogas throughout the operation. Hydrogen production rate was increased till $10.5\;L{\cdot}h^{-1}{\cdot}L^{-1}$ of bioreactor when influent sucrose concentrations and recycle rates were varied. At the same time, the hydrogen production rate with hydrophobic media application was higher than its hydrophilic media application. No methane was detected when the reactor was under a normal operation. The major fermentation by-products in the liquid effluent of the both trickling biofilters were acetate, butyrate and lactate. In order to run in the long term operation of both reactor filled with hydrophilic and hydrophobic media, biofilm accumulation on hydrophilic media and biogas produced should be controlled through some process such as periodical backwashing or gas-purging. Four sample were collected from each reactor on the opposite hydrogen production rate, and their bacterial communities were compared by terminal restriction fragment length polymorphism (T-RFLP) analysis of PCR products generated using bacterial 16s rRNA gene primers (8f and 926r). It was expressed a marked difference in bacterial communities of both reactors. The trickling bed bioreactor with hydrophobic media demonstrates the feasibility of the process to produce hydrogen gas. A likely application of this reactor technology can be hydrogen gas recovery from pre-treatment of high carbohydrate-containing wastewaters.

  • Six cases of congenital heart disease were operated on by means of cardiopulmonary bypass between December, 1975 and April, 1976. Two cases of ventricular septal defects (VSD), two cases of VSD, associated with ruptured aneurysm of sinus Valsalva, two cases of atrial septal defects (ASD) and one case of pulmonic stenosis with patent ductus arteriosus were operated. Sarns roller pumps and Bentley Temptrol oxygenators were used for extracorporeal circulation. Pump oxygenator was primed with Ringer's lactate solution, 5% dextrose in water, mannitol, and ACD blood. Flow rate ranged from 2.0 to $2.4L/M^2/min$. Bicarbonate was added to the oxygenator with estimated amount as 15 mEq/L/hr. Venous catheters were introduced into superior and inferior vena cava, and oxygenated blood was returned to the body through aortic cannula inserted into ascending aorta. Moderate hypothermia ($30^{\circ}C$) was induced by core cooling. Aorta was cross clamped for 15 minutes and released for 3 minutes, and repeated clamping when necessary. Atrial and ventricular septal efects were closed by direct sutures. Aneurysms of sinus Valsalva ruptured into the right ventricle were repaired through right ventriculotomy by d:rect closure with Dacron patch reinforcement. Cardiopulmonary bypass time varied from 66 to 209 minutes, and aorta cross clamping time ranged from 13 to 56 minutes. Postoperative bleeding was minimal except one case who needed for evacuation of substernal hematoma. Intra- and postoperative urinary output was satisfactory. Acid-base balance, partial pressure of $O_2$, electrolytes, and hematological changes during intra- and post-perfusion period remained at the acceptable ranges. No mortality was experienced.

고빌리루빈혈증이 유도된 신생자돈에서 근적외석 발광기를 이용한 뇌 혈역학적 변화에 대한 연구 (Near-Infrared Spectroscopy for Monitoring Cerebral Hemodynamics in Hyperbilirubinemia-induced Newborn Piglets)

  • 목 적 : 본 연구는 고빌리루빈혈증이 유도된 신생자돈에서 비 침습적인 NIRS을 통한 뇌의 혈역학적 변화에 대해 알아보고자 하였다. 방 법 : 17 마리의 신생자돈을 대상으로 정상 대조군(CG, n=6), 고빌리루빈혈증군(BG, n=7마리), 7-NI를 투여한 고빌리루빈 혈증군(NG, n=4)으로 무작위 구분하였다. 고빌리루빈혈증의 유도는 40 mg/kg의 빌리루빈을 농축괴로 정주한 후 곧 이어 30mg/kg/hr로 4시간 동안 지속 정주하여 혈중 빌리루빈 농도를 20 mg/dL 이상으로 유지하였고 7-NI는 빌리루빈을 농축괴로 투여한 전과 후에 50 mg/kg을 복막투여 하였다. 모든 실험군은 실험 기간 동안 뇌의 혈역학적 변화를 위해 NIRS로 감시하였고 뇌 조직을 적출하여 생화학적인 변화를 관찰하였다. 결 과 : 동맥혈의 base excss, pH, 평균 동맥압은 BG군과 NG군에서 CG군에 비해 유의하게 감소하였다. BG군에서 유의하게 뇌 조직의 $Na^+$, $K^+$-ATPase activity, ATP, PCr은 유의하게 감소하고 conjugated dienes는 유의하게 증가하였으나 NG 군은 이런 이상소견이 유의하게 완화되었다(P<0.05). 뇌 혈역학적 검사상 [$HbO_2$], [HbT], 및 [HbD]는 BG군에서 CG군에 비해 유의하게 감소하였고(P<0.05) NG군은 CG군과 차이가 없었다. 실험 종료시 $ScO_2$는 세 군간에 유의한 차이가 없었다. 결 론 : 고빌리루빈혈증이 유도된 신생자돈에서 뇌의 혈역학적인 변화를 비침습적인 NIRS의 감시를 통해 유용하게 관찰할 수 있었다.

Involvement of Lysosome Membrane Permeabilization and Reactive Oxygen Species Production in the Necrosis Induced by Chlamydia muridarum Infection in L929 Cells

  • Chlamydiae, obligate intracellular bacteria, are associated with a variety of human diseases. The chlamydial life cycle undergoes a biphasic development: replicative reticulate bodies (RBs) phase and infectious elementary bodies (EBs) phase. At the end of the chlamydial intracellular life cycle, EBs have to be released to the surrounded cells. Therefore, the interactions between Chlamydiae and cell death pathways could greatly influence the outcomes of Chlamydia infection. However, the underlying molecular mechanisms remain elusive. Here, we investigated host cell death after Chlamydia infection in vitro, in L929 cells, and showed that Chlamydia infection induces cell necrosis, as detected by the propidium iodide (PI)-Annexin V double-staining flow-cytometric assay and Lactate dehydrogenase (LDH) release assay. The production of reactive oxygen species (ROS), an important factor in induction of necrosis, was increased after Chlamydia infection, and inhibition of ROS with specific pharmacological inhibitors, diphenylene iodonium (DPI) or butylated hydroxyanisole (BHA), led to significant suppression of necrosis. Interestingly, live-cell imaging revealed that Chlamydia infection induced lysosome membrane permeabilization (LMP). When an inhibitor upstream of LMP, CA-074-Me, was added to cells, the production of ROS was reduced with concomitant inhibition of necrosis. Taken together, our results indicate that Chlamydia infection elicits the production of ROS, which is dependent on LMP at least partially, followed by induction of host-cell necrosis. To our best knowledge, this is the first live-cell-imaging observation of LMP post Chlamydia infection and report on the link of LMP to ROS to necrosis during Chlamydia infection.

산화적 손상에 의해 유도된 신경세포독성에 대한 검정콩 껍질 추출물의 보호효과 (Protective Effects of Black Soybean Seed Coat Extracts against Oxidative Stress-induced Neurotoxicity)

  • 본 연구는 산화적 스트레스로부터 유도되는 신경세포 사멸을 보호할 수 있는 생리활성물질을 탐색하기 위하여 검정콩 껍질 추출물의 항산화 효과 및 PC12 신경세포 보호효과에 대하여 조사하였다. 다양한 용매에 의해 추출된 검정콩 껍질 추출물의 총 페놀화합물 함량을 측정한 결과 70% acetone 추출물이 다른 용매추출물에 비하여 가장 높은 총 페놀화합물 함량을 나타내었으며, 이를 기초로 70% acetone 추출물을 이용한 세포 내 항산화 활성도 높은 것으로 나타났다. 또한 PC12 신경세포 보호효과를 조사한 결과, 세포생존율과 세포막 손상 보호효과에서 70% acetone 추출물 처리구는 최대 $500{\mu}g/mL$까지 $H_2O_2$를 처리한 대조구에 비하여 농도 의존적으로 신경세포 보호효과가 나타남을 알 수 있었다. 동일 추출물을 활용한 in vivo 인지학습능력 평가에서도 대조구인 $A{\beta}$ ICV injection을 한 group과 비교하였을 때 전체적으로 추출물의 농도가 증가할수록 Y-maze test와 passive avoidance test에서 control구와 유사하거나 높은 인지 및 기억 능력 회복효과를 보여주었다. 본 연구결과를 종합해 보면 BSSCE는 항산화, 뇌 신경세포 보호효과 및 in vivo 인지 기억 회복능을 갖는 것으로 판단되며, 추후 검정콩 껍질에 존재하는 유효 생리활성물질을 확인함으로써 현대 사회의 가장 큰 질병 중 하나인 알츠하이머성 치매(AD)와 같은 퇴행성 뇌신경질환 예방에 유용한 소재가 될 것으로 판단된다.

Effects of Organic Acids Mix and Modified Atmosphere Packaging on the Storage Quality of Sliced Bacon

  • The effects of organic acids mix (0.4%) and modified atmosphere packaging (MAP) on the storage quality of sliced bacon were investigated. Pork bellies were treated with or without organic acids at the curing stage. The organic acids mix comprised 35% sodium acetate, 25% salt, 15% calcium lactate, 11% trisodium citrate, 7% ascorbate, and 7% citric acid. The cured pork bellies were smoked and packaged with 50% $CO_2$ + 50% $N_2$ (50% $CO_2$-MAP) and 100% $N_2$ (100% $N_2$-MAP), and stored at $5^{\circ}C$ for 14 d. The 50% $CO_2$-MAP showed a higher pH value (p<0.05) up to 10 d, a lower protein deterioration (p<0.05) as measured by volatile basic nitrogen (VBN) from 6 to 14 d, and a higher color value of lightness (CIE $L^*$) compared to 100% $N_2$-MAP. The development of lipid oxidation measured by thiobarbituric acid reactive substance (TBARS) values seemed to be effectively controlled throughout the storage period in both 50% $CO_2$-MAP and 100% $N_2$-MAP regardless of the application of organic acids. The 50% $CO_2$-MAP inhibited the growth of aerobic and anaerobic bacteria (p<0.05) both in non-added and bacon added with organic acids mix. The 50% $CO_2$-MAP alone seemed to be effective in delaying the growth of bacteria since the use of organic acids mix gave no additional effects. The addition of organic acids mix lowered the pH value (p<0.05), effectively retarded the protein deterioration (p<0.05), and showed a higher color value of lightness (CIE $L^*$) value (p<0.05) and lower color value of redness (CIE $a^*$) value (p<0.05). In conclusion, 50% $CO^2$-MAP showed better quality and self-life of sliced bacon during storage. However, the beneficial effect of organic acids mix was not noticed in the concentration used in this experiment.

보리수나무 열매, 잎 및 줄기 에탄올 추출물의 함유성분 분석과 간암 세포 증식억제 효과 (Component Analysis and Anti-Proliferative Effects of Ethanol Extracts of Fruits, Leaves, and Stems from Elaeagnus umbellata in HepG2 Cells)

  • 경남 합천 일대에서 재배된 보리수나무(Elaeagnus umbellata Thunb.)의 생리활성을 부위별(열매, 잎, 줄기)로 평가한 결과는 다음과 같다. 항산화 특성을 측정한 결과, 총폴리페놀 함량이 높게 나타난 줄기(829.6 mg/g) 에탄올 추출물에서 DPPH 라디칼 소거능($EC_{50}=54.04{\mu}g/mL$)이 가장 우수하였다. 유리 아미노산 분석에서는 잎(6,179.12 mg/100 g) 에탄올 추출물이 열매(378.88 mg/100 g)와 줄기(1,211.69 mg/100 g) 에탄올 추출물보다 유리 아미노산 함량이 높았으며, 열매의 proline, 잎의 asparagine 및 줄기의 GABA가 대표적인 유리 아미노산인 것을 확인하였다. LC-MS/MS를 이용한 주요 페놀 성분 분석에서는 잎 에탄올 추출물에서 gallic acid, 줄기 에탄올 추출물에서 catechin의 함량이 가장 높았으며, HepG2 cell과 Chang cell의 암세포 증식억제 효과를 측정한 결과 잎 에탄올 추출물이 HepG2에 특이적으로 높은 억제 활성이 나타났다. 향후 보리수나무를 국내 우수 자원으로 활용하기 위해서는 추가로 다양한 암세포에 대한 활성 차이, 부위별 활성 물질의 구명 및 작용 기전 등에 대한 계속된 연구가 수행되어야 할 것이다.

RAW 264.7 대식세포에서 NO, TNF-α, IL-6 생산에 관한 Squalene, Alkoxy Glycerol, Batyl, Chimyl의 효과 (Production of NO, TNF-α and IL-6 by Squalene, Alkoxy Glycerol, Batyl and Chimyl Solutions in RAW 264.7 Macrophage Cells)

  • 본 연구에서는 대식세포인 RAW 264.7 세포주에 SQ ($0.1{\%}$), AG($0.1{\%}$), Batyl(1 mg/mL), Chimyl(1 mg/mL) 용액 등을 단독 혹은 C. albicans와 함께 첨가하여 배양한 후LDH, NO, TNF-$\alpha$, IL-6 발현에 어떤 영향을 주는 지를 관찰하였다. 모든 실험군에서 실험에 사용한 농도는 LDH, NO생산에 영향을 주지 못하였다. 실험에 사용한 용액을 단독 처리한 실험의 경우 TNF-$\alpha$와 IL-6발현의 경우 6시간째에는 큰 차이를 나타내지는 못하였으나, 24시간째에는 발현에 영향을 주었다(p < 0.05). C. albicans을 함께 처리한 실험의 경우에는 C. albicans단독 처리군에 비해 실험 용액을 함께 처리한 군에서 C. albicans유도 TNF-$\alpha$와 IL-6발현을 감소시키는 효과를 관찰하였다(p < 0.05, p < 0.01). 본 연구를 통해 SQ, AG, Batyl, Chimyl 용액이 C. albicans 유도에 의해 감소된 면역반응을 증가시킬 수 있을 것으로 생각되어진다.

이식을 위한 가토심장의 장기 보존방법에 관한 비교 연구 (Comparative Study of Prolonged Preservation Methods in Rabbit Heart for Transplantation)

  • 심장이식 수술의 성공의 관건은 공여자와 수혜자의 적절한 조합과 이미 정립된 수술수기와 합께 적출된 심장의 효과적인 기능 보존에 달려있다. 심장 보존에 있어서 괄목할 만한 발전이 있었음에도 불구하고 최근의 가장 진보된 방법으로 심 허 혈 상태의 최대 허용기간은 약 4~6시간정도이다 저자는 이식을 위한 적출 심장의 보존 용액 이 갖추어야 할 적정 요건에 대한 자료를 축적하고자, 단순 저온 침적방법에 의거하여, H/S용액(I군)을 대조군으로 하여 저자가 고안한 CK용액(W군)을 기존의 MEC용액( ll군) 및 MUW용액(E군)과 비교 실험하였다. 적출된 가토의 심장을 실험 대상으로 하고 정압형 Langendorff실험모형을 사용하여, 20분간의 평형 상태, 4시간동안의 심장 저장 및 20분간의 재관류시기를 거치게 한 후, 심근 조직을 절제하여 냉동 보관하였다. 실험 계획에 따라 관관류량,좌심실압, 압력 미분치를 측정하였고, 동결 심근조직내의 효소치를 정량 분석하였다. 결론적으로, 저자의 비교 실험에서 MUW용액과 CK용액의 심근보호 능력이 우수하였고, MEC용액은 대조군에 비해 저조하였다. potassium의 농도가 MVW용액보다 낮고()4.2nM/L), 문헌상에서 \ulcorner\ulcorner보호에 유익한 것으로 보고된 각종의 substrate을 첨가하여 제작된 CK용액의 성적이 MUW용액 과 비슷한 수준으로 우수하였는데, 이는 심장보존용액의 전해질 농도의 조정과 첨가물질의 선택 및 조정에 있어서 지속적인 연구와 발전이 필요함을 시사한다고 할 수 있다.

Prognostic Factor Analysis of Overall Survival in Gastric Cancer from Two Phase III Studies of Second-line Ramucirumab (REGARD and RAINBOW) Using Pooled Patient Data

  • Purpose: To identify baseline prognostic factors for survival in patients with disease progression, during or after chemotherapy for the treatment of advanced gastric or gastroesophageal junction (GEJ) cancer. Materials and Methods: We pooled data from patients randomized between 2009 and 2012 in 2 phase III, global double-blind studies of ramucirumab for the treatment of advanced gastric or GEJ adenocarcinoma following disease progression on first-line platinum- and/or fluoropyrimidine-containing therapy (REGARD and RAINBOW). Forty-one key baseline clinical and laboratory factors common in both studies were examined. Model building started with covariate screening using univariate Cox models (significance level=0.05). A stepwise multivariable Cox model identified the final prognostic factors (entry+exit significance level=0.01). Cox models were stratified by treatment and geographic region. The process was repeated to identify baseline prognostic quality of life (QoL) parameters. Results: Of 1,020 randomized patients, 953 (93%) patients without any missing covariates were included in the analysis. We identified 12 independent prognostic factors of poor survival: 1) peritoneal metastases; 2) Eastern Cooperative Oncology Group (ECOG) performance score 1; 3) the presence of a primary tumor; 4) time to progression since prior therapy <6 months; 5) poor/unknown tumor differentiation; abnormally low blood levels of 6) albumin, 7) sodium, and/or 8) lymphocytes; and abnormally high blood levels of 9) neutrophils, 10) aspartate aminotransferase (AST), 11) alkaline phosphatase (ALP), and/or 12) lactate dehydrogenase (LDH). Factors were used to devise a 4-tier prognostic index (median overall survival [OS] by risk [months]: high=3.4, moderate=6.4, medium=9.9, and low=14.5; Harrell's C-index=0.66; 95% confidence interval [CI], 0.64-0.68). Addition of QoL to the model identified patient-reported appetite loss as an independent prognostic factor. Conclusions: The identified prognostic factors and the reported prognostic index may help clinical decision-making, patient stratification, and planning of future clinical studies.
