  • BACKGROUND: Arsenic (As)-contaminated groundwater used for long-term irrigation has emerged as a serious problem by adding As to soils. Phytoremediation potential of fiber crops viz., kenaf (Hibiscus cannabinus L.), mesta (Hibiscus sabdariffa L.), and jute (Corchorus capsularis L.) was studied to clean up As-contaminated soil.METHODS AND RESULTS: Varieties of three fiber crops were selected in this study. Seeds of kenaf, mesta, and jute varieties were germinated in As-contaminated soil. Uptake of As by shoot was significantly higher than that by root in the contaminated soil. In As-contaminated soil, kenaf and mesta varieties accumulated more As, than did jute varieties. In the plant parts above ground, mainly the shoots, the highest As absorption was recorded in kenaf cv. HC-3, followed by kenaf cv. HC-95. Kenaf varieties produced more biomass. In terms of higher plant biomass production, and As absorption, kenaf varieties showed considerable potential to remediate As-contaminated soil.CONCLUSION: The overall As absorption and phytoremediation potentiality of plant varieties were in the order of kenaf cv. HC-3 > kenaf cv. HC-95 > mesta cv. Samu-93 > jute cv. CVE-3 > jute cv. BJC-7370. All varieties of kenaf, mesta, and jute could be considered for an appropriate green plant-based remediation technology in As-contaminated soil.

  • The adsorption ability of wood-based activated carbon to adsorb methylene blue (MB) and crystal violet (CV) from aqueous solution has been investigated. Adsorption studies were carried out on the batch experiment at different initial MB and CV concentrations (MB=150 mg/L~400 mg/L, CV=50 mg/L~350 mg/L), contact time, and temperature. The results showed that the MB and CV adsorption process followed the pseudo-second-order kinetic and intraparticle diffusion was the rate-limiting step. Adsorption equilibrium data of the adsorption process fitted very well to both Langmuir and Freundlich model. The maximum adsorption capacity ($q_m$) by Langmuir constant was 416.7 mg/g for MB and 462.4 mg/g for CV. The thermodynamic parameters such as ${\Delta}H^{\circ}$, ${\Delta}S^{\circ}$ and ${\Delta}G^{\circ}$ were evaluated. The MB and CV adsorption process was found to be endothermic for the two dyes.

  • We studies seed set in the interspecific F1 /backcross hybrids of Lilium species. In the interspecfic hybrid of L. longiflorum cv. Gelria with L.${\times}$ fomolongi cv. Raizan,93% fruit set was obtained by stigmatic pollination in comparison to 53% from cut-style pollination. Accordingly, the number of seed set resulting from stigmatic and cut-style pollination was 147 and 53, respectively. Pollination o( both stigmatic and cut-style pollination resulted in 47% fruit set in the hybrid of L. longiflorum cv. Lorina with L.${\times}$ fomolongi cv. Raizan. However, stigmatic pollination formed 413 seeds, whereas only 24 seeds were obtained by cut-style pollination in this cross. The hybrid of L.${\times}$ fomolongi cv. Raizan with L. longiflorum cv. Come set 40% fruit with a total of 43 seeds by stigmatic pollination. However, no fruit set was observed in cut-style pollination in this hybrid. Backcrossing the F1 hybrid by cut-style pollination of L. longiflorum cv. Lorina ${\times}$ Asiatic hybrid cv. Chicago with the latter parent led to 53% fruit set, and 109 embryos were obtained. Likewise, backcrossing following cut-style pollination of L. longiflorum cv. Lorina ${\times}$ Asiatic hybrid cv. Corsia with the latter parent formed 67% fruits and 107 embryos. However, in the remaining interspecific hybrids, cut-style pollination set no fruit.

  • The possibility of employing biomass of Salix viminalis cv. Q683 as a resource of bio-energy was evaluated. The chemical analysis of S. viminalis cv. Q683 leaf biomass showed components such as, extractives (2.57%), lignin (39.06%), hemicellulose (21.61%), and cellulose (37.83%), whereas, its stem was composed of extractives (1.67%), lignin (23.54%), hemicellulose (33.64%), and cellulose (42.03%). The biomass of S. viminalis cv. Q683 was saccharified using two enzymes celluclast and viscozyme. The saccharification of S. viminalis cv. Q683 biomass was influenced by enzymes and their strengths. The optimal enzyme combination was found to be celluclast (59 FPU/g substrate) and viscozyme (24 FBG/g substrate). On saccharification the glucose from leaf and stem biomass was 7.5g/L and 11.7g/L, respectively after 72 hr of enzyme treatment. The biomass and enzyme-treated biomass served as the feedstock for ethanol production by fermentation. The ethanol production from stem and leaf biomass was 5.8 g/L and 2.2 g/L respectively, while the fermentation of the enzymatic hydrolysates yielded 5 g/L to 8 g/L bioethanol in 72 hours.

  • Effect of the antibiotic carbenicillin on callus and shoot formation from the leaf disc culture of tobacco (Nicotiana tabacum L. cv. BY-4) was examined. The number of shoot induced from the leaf explants was decreased as the amount of carbenicillin increased from 250 mg/L to 2,000 mg/L on MS medium containing 0.5 mg/L of BAP or kinetin. In addition, calli formation from the leaf explants was increased by the treatment of 250 mg/L ∼ 500 mg/L carbenicillin with or without 0.5 mg/L of 2,4-D or NAA. However, the fresh weight of 4-week-cultured explants was decreased with increasing carbenicillin from 250 mg/L to 2,000 mg/L on MS medium containing 0.5 mg/L of 2,4-D or NAA. These results indicate that carbenicillin has an auxin-like effect, such as promoting callus formation and inhibiting shoot induction. It leads to the conclusion that the auxin-like property should be taken into account for the production of transgenic plants via Agrobacterium-mediated transformation.
