Effects of Extraction Method on Physiological Activity and Antibiosis of Mulberry Powder (오디분말의 추출 방법에 따른 생리활성 및 항균효과에 미치는 영향)

  • The purpose of this study was to analyze the physiological activity and antibiosis of mulberry powder by extract method. The total phenol content of mulberry powder extracted with methanol was 82.9 mg/g, while that of powder extracted with water was 46.9 mg/g. Extractions with methanol were therefore more effective than with water. The total flavonoid contents of mulberry powder extracted with methanol was 13.0 mg/g, while that of powder extracted with water was 9.4 mg/g. Also, the nitrite-scavenging ability of mulberry powder was lower than ascorbic acid and BHT. The SOD-like activity of mulberry powder extracts, natural antioxidant, and artificial antioxidant at 5 mg/mL arranged in order of decreasing concentration were ascorbic acid (98.3%) > BHT (88.1%) > water extract (9.8%) > methanol extract (3.0%). And, the OH radical scavenging activities of mulberry powder extracts and natural antioxidant at 5 mg/mL in order of decreasing concentration was ascorbic acid (97.0%) > methanol extract (46.2%) > water extract (35.8%). The antimicrobial effects of mulberry powder extracted with methanol could be detected on Bacillus cereus ($10,000{\mu}g/disc$) and Staphylococcus aureus ($10,000{\mu}g/disc$).

Comparative bioavailability of synthetic vitamin C and Nutra-C (calcium ascorbate) in Korean healthy volunteers

  • The purpose of this study was to compare the relative bioavailability of synthetic Vitamin C and Nutra-C® (calcium ascorbate) using a randomized parallel pharmacokinetics study design. Under fasting conditions, 20 healthy volunteers were randomly allocated to receive a single oral dose (500 mg of ascorbic acid) of either synthetic Vitamin C or Nutra-C®. Fasting blood was collected pre-dose and 1, 2, 3, 4, 7 and 10 hr post-dose. The ascorbic acid content of human serum was determined using HPLC with ultraviolet detection. The fasting serum ascorbic acid concentrations of synthetic Vitamin C and Nutra-C® were 6.734 ± 2.09 ng/mL (n = 10) and 7.542 ± 2.96 ng/mL (n = 10), respectively. The bioavailability of Nutra-C® was significantly greater (128 %, p < 0.05) than that of the synthetic Vitamin C.

  • Paenibacillus sp. JB-13 producing the cyclodextrin glucan-otransferase(CGTase) [EC 2.4.1.19] that glucosylated ascorbic acid(AA) at the C-2 position was isolated form soil and the optimal conditions for the production of 2-O-$\alpha$-D- Glucopyranosl L-Ascorbic acid(AA-2G) with CGTase were investigated. CGTase produced AA-2G efficiently using dextrin as a substrate and AA as an aceptor. Several AA-2-oilgosaccharides(AA-2Gs) were also produced in this reaction mixture, and these were efficiently hydro-lyzed to AA-2G and glucose by the treatment with glucoamylase. The optimal temperature for AA-2G production was $37^{\circ}C$ and the optimal pH was around 6.5. CGTase also utilized $\alpha$-,$\beta$-,${\gamma}$-CDs, soluble starch, com statch, dia-static solution from rice and diastatic solution from malt as substrate, but not glucose. The reaction mixture for the maximal production of AA-2G was following; 15% total substrate concentration, 2,500 units/ml of CGTase and a mixing ration of 3:2(g of AA: g of dextrin). Under this condition, 56 mM of AA-2G ,which corresponded to 12.4% yield based on AA. was produced after incubation for 44 hrs at $37^{\circ}C$ and pH 6.5.

Effect of L-Ascorbic Acid Contents in Tissue on Collagen Synthesis in Guinea Pigs (Guinea pig의 조직중 L-Ascorbic acid함량이 콜라겐 생합성에 미치는 영향)

  • To clarify the requirement of L-ascorbic acid (AsA) in collagen synthesis, the incorporation of 1-$^{14}$ C-proline into the tissues of guinea pigs and the specific radioactivity ratio (proline/hydroxyproline) in collagen were investigated. Male guinea pigs maintained on the AsA-deficient diet were divided into three groups ; group A (AsA-deficient animals) : group B (control animals) supplemented with 5mg AsA/day ; group C (high dose animals) with 300mg AsA/day, and orally supplemented with or with-out AsA for 14 days. Collagen synthesis was estimated by measuring the incorporation of labeled pro-line into collagen in lung and dorsal skin, and the hydroxyproline contents in lung and skin. The AsA contents in the tissues were determined by high-peforrnance liquid chromatography (HPLC), and serum alkaline phosphatase activity was also measured. The serum alkaline phosphatase activity of AsA deficient group was very low as compared with those of AsA supplemented group. Incorporation of labelled proline into collagen and its specific radioactivity ratio in collagen increased with increasing levels of AsA in the tissues. There was a significantly positive relationship between the levels of AsA and hydroxyproline in the tissues.

Changes of Antioxidant Activities of Soft Tofu Manufactured with Red Ginseng Extract during Storage (홍삼 추출물 첨가 연두부의 저장 중 항산화 활성 변화)

  • To develop tofu with bioactivity, red ginseng extract was added and soft tofu was prepared without pressuring and forming steps. Then, the content of antioxidant components and antioxidative activities were measured using the prepared soft tofu during storage. The contents of polyphenols of the control and the sample added red ginseng extract were $605.25{\mu}g/mL$ and $598.51{\sim}681.65{\mu}g/mL$. The content of the polyphenols was proportional to the concentration of added red ginseng extract. The content of ascorbic acid of the control was 6.42 mg%, and the ascorbic acid contents of the sample added red ginseng extract were higher than that of the control. The contents of polyphenols and ascorbic acid of control were lower than those of the sample added red ginseng extract during the storage. The addition of red ginseng extract increased reducing power and ABTS radical cation decolorization.

Effect of Antioxidants on the Thermostability of Red Pigment in Prickly Pear (선인장열매 적색색소의 열안정성에 대한 항산화제의 효과)

  • The color stability of betacyanins and effects of antioxidants from Opuntia dillenii Haw were determined in the fruit juice at temperature up to $90^{\circ}C$. The absorption maxima of betacyanins occurred between 536 nm and 538 nm. When fruit juice was heated at $90^{\circ}C$ for various times, the red color gradually diminished and the absorption maxima slightly shifted toward uv region. The kinetic analysis of the data obtained indicated that the discoloration for betacyanins obeyed first order reaction pattern, when the thermal stability test was performed at $50{\sim}90^{\circ}C$. And the rate constant increased from $1.56{\times}10^{-3}/min\;to\;71.91{\times}10^{-3}/min$ with the half-life decreasing from 444.23 min to 9.64 min. The results also indicated that the thermal stability of pigment decreased with increasing temperature. The energy of activation was 10.94 kcal/mole for the pigment. N-propyl gallate, L-cysteine, and ascorbic acid were added to cactus fruit juice at concentrations of $0.01{\sim}0.3%$ at different temperatures. Npropyl gallate and L-cysteine had a little antioxidant effect on betacyanins stability at $50^{\circ}C\;and\;70^{\circ}C$, whereas ascorbic acid had a great antioxidant effect with the half-life value of 2 to 10 times to that of the control.

Some Vitamin Status in Healthy Elderly Koreans from Urban Households (도시 지역 노인의 일부 비타민 영양상태에 관한 연구)

  • Riboflavin, Ascorbin Acid & Vitamin D status of 74 elderly Korean (35 men and 39 women)from urban households in Incheon were evaluated by blood analysis. Mean EGR-AC value of men was 1.05 while that of women was 1.03. Marginal deficiency of riboflavin(EGR-AC 1.15-1.35) was shown in 33.3% of men and 20% of women. Average plasma ascorbic acid contents of subjects were very low and men had significantly lower amount than women(0.23mg/㎗ vs 0.44mg/㎗). The percentages of subjects who had plasma ascorbic acid less than 0.4mg/㎗ were 88.9% of men and 45% of women. Mena serum Calcium, Inorganic phosphate contents and mean serum ALP activity of men were 9.97mg/㎗, 3.28mg/㎗ and 61.85 Unit/L respectively and those of women were 9.78mg/㎗, 3.49mg/㎗ and 67.80Unit/L respectively. From these results, Vitamin D status of subjects was considered to be normal.

Antioxidant, Anti-inflammatory and Cytotoxicity on Human Lung Epithelial A549 Cells of Jerusalem artichoke (Helianthus tuberosus L.) Tuber

  • This study investigated in vitro antioxidant, anti-inflammatory and cytotoxicity on human lung epithelial A549 cells of different solvent extracts from Jerusalem artichoke (Helianthus tuberosus L.) tuber. The EtOH extract contained amounts of phenolics (22.20 tannic acid equivalent ㎎/ɡ) and exhibited the highest antioxidant activity and anti-inflammatory activity. Several methods were employed for measure the antioxidant activity: 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity (IC50 = 206.79 ㎍/㎖), reducing power activity (21.26 ascorbic acid equivalent ㎎/ɡ) and total antioxidant activity (19.05 ascorbic acid equivalent ㎎/ɡ). Meantime, the EtOH extract inhibited the NO production completely with a concentration of 800 ㎍/㎖. Besides, the H2O extract exhibited more potent effect on human lung epithelial A549 cells. This study suggested that Jerusalem artichoke tuber had antioxidant, anti-inflammatory and cytotoxicity on human lung epithelial A549 cells.

Antioxidant Activity and Anti-wrinkle Effects of Aceriphyllum rossii Leaf Ethanol Extract

  • We evaluated the antioxidant activity and anti-wrinkle effects of Aceriphyllum rossii leaf ethanol extract (ARLEE) in vitro using human dermal fibroblasts. The total polyphenol and flavonoid contents of ARLEE were 578.6 and 206.3 mg/g, respectively. At a concentration of $250{\mu}g/mL$, the electron-donating ability of ARLEE was 87.1%. In comparison with the vehicle, ARLEE treatment at $100{\mu}g/mL$ significantly increased type I procollagen synthesis (p < 0.01) by 50.7%. In vitro ARLEE treatment (10 mg/mL) inhibited collagenase and elastase activity by 97.1% and 99.2%, respectively. Compared with the control, ascorbic acid treatment at $100{\mu}g/mL$ significantly decreased matrix metalloproteinase (MMP)-1 protein expression (p < 0.01) by 37.0%. ARLEE treatment at $50{\mu}g/mL$ significantly decreased MMP-1 protein expression (p < 0.01) by 46.1%. Ascorbic acid and ARLEE treatments at $100{\mu}g/mL$ significantly decreased MMP-1 mRNA expression (p < 0.01) by 26.1% and 36.1%, respectively. From these results, we conclude that ARLEE has excellent antioxidant activity and even better anti-wrinkle effects than ascorbic acid in human dermal fibroblasts. These results suggest that ARLEE could be used in functional cosmetics for the prevention or alleviation of skin wrinkles induced by ultraviolet rays.

Effect of Curing Additives on Color Property of Smoked Duck Meat (염지제가 훈연오리의 육색 특성에 미치는 영향)

  • This study was conducted to investigate the effect of curing additives on color property of smoked duck meat. Curing process of samples was performed one of the following treatments: C, non-curing: T1, 2.43% salt: T2, 2.43% salt + 0.49% tripolyphosphate (TPP): T3, 2.43% salt + 0.49% TPP + 0.002% nitrite: T4, 4.76% duck seasoning: and T5, 1.47% salt + 0.24% TPP + 0.2% L-ascorbic acid. Instrumental meat color of both breast and thigh of smoked duck showed that the CIE $a^*$ value of the T4 was significantly (p<0.05) higher than that of the other treatments, whereas T5 had a significantly (p<0.05) higher CIE $b^*$ value than the other treatments. In results of nitroso pigment, T5 of smoked duck breast was significantly (p<0.05) higher value compared to other treatments, whereas T3 and T5 of smoked duck thigh had a significantly (p<0.05) higher value than other treatments. Heme pigment contents of control and T5 was significantly (p<0.05) higher value compared to other treatments in smoked duck breast. Meat color of T3 by sensory evaluation showed redder (p<0.05) than other treatments. These results suggested that using L-ascorbic acid is revealed to be pink color without nitrite or pigment when manufacturing of smoked duck meat.
