Physiological Properties of Sarcodon aspratus Extracts by Ethanol Concentration (능이버섯 추출물의 생리활성 효과)

  • Physiological properties of Sarcodon aspratus extracts were investigated. Yields of water, 30, 60, and 90% ethanolic extracts were 52.10, 46.90, 41.50, and 32.20%, respectively. The 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity of 60% ethanolic extract was 40.40% at a concentration of 4 mg/mL, which was highest of all the extracts. The polyphenol content of 60% ethanolic extract was the highest at 16.23 mg/g at a concentration of 1 mg/mL. Superoxide anion radical scavenging activities were 56.30~77.05% at concentrations of 1, 2, and 4 mg/mL, which were better than the ascorbic acid activity (56.19% at 10 mg/mL) (P<0.05). Nitrite-scavenging ability was the most effective in 30% ethanolic extract. A pH level of 3.0 was the most effective for all extracts. Angiotensin-converting enzyme inhibitory activities of Sarcodon aspratus extracts were from 52.67 to 89.52%.

Quality Control on the BOD, COD, TN, TP of Water Quality Pollutant (수질오염물질 BOD, COD, TN, TP의 정도관리(I))

  • Quality control was carried out the three analytes, BOD, COD, TN, TP of high and low concentration for thirteen laboratories. Evaluation values of Quality control was determined from average value which were made by six times test, repeatedly and periodically. This value had not only error ratio of within 5% from prepared sample value but also high precision of standard deviation, 0.44 (BOD, low concentration), 2.15 (BOD, high concentration), 0.12 (COD, low concentration), 1.63 (COD, high concentration), 0.35 (TN, low concentration), 1.99 (TN, high concentration), 0.05 (TP, low concentration), 0.14 (TP, high concentration). Upper (Lower) Warning Limit (ULWL) and Upper (Lower) Acceptance Limit (ULAL) values made from quality control chart. Three (5.30, 9.70, 5.30 mg/L) and five laboratories were over ULAL value in low and high BOD concentration, respectively. Two (41.00, 30.60 mg/L) and four laboratories were over ULAL value in low and high COD concentration, respectively. while, other eleven laboratories appeared reliable data. One laboratory (0.70 mg/L) was over ULAL at low TN concentration, while other eleven laboratories had reliable values. In case of high TN concentration, eleven laboratories were over ULAL value, it should be focuced for improvement of reliability about measurement and analysis of TN. Four (1.14, 0.45, 0.64, 1.49mg/L) and seven laboratories were over ULAL in low and high TP concentration, respectively.

  • This experiment was carried out to investigate the effect of hypoglycemic yogurt as a clinical test with person both who have diabetes and who don't have diabetes. Diabetes is a disease caused by unused and accumulated glucoses produced via the foods digestion due to the lack of insulin or lower tissue reaction to insulin. Patients with diabetes or complications of diabetes are on the rise annually. For the therapy and prevention of diabetes, a yogurt made from functional materials such as insulin-resistant FK-23 lactic acid bacteria, Pinitol, edible fiber which delays glucose-absorption, extract powder of glucose-absorption-inhibitory white bean and low-caloric Erythritol and banaba extract is effective for reduce or restrain the blood glucose levels. The blood glucose levels after a meal of non-diabetic persons were lower after drinking a hypoglycemic yogurt than they after drinking general yogurt. The blood glucose levels after a meal of 10 diabetic patients were checked after drinking the yoghurt. As a result, 19 mg/dL, 30 mg/dL, 31 mg/dL of blood glucose levels decreased in 30 min, 60 min and 120 min respectively after drinking the yogurt. Blood glucose inhibition rates of 80 % of the tested patients also decreased. The blood glucose level of the diabetic patients having drunk the yogurt for 30 days consistently decreased into 59 mg/dL in 120 min after a meal.

The Characteristics of Tetrachloroethylene (PCE) Degradation by Pseudomonas putida BJ10 (Pseudomonas putida BJ10의 Tetrachloroethylene (PCE) 분해 특성)

  • In this study, biological PCE degradation by using a BTEX degrading bacterium, named BJ10, under aerobic conditions in the presence of toluene was examined. According to morphological, physiological characteristics, 16S rDNA sequencing and fatty acid analysis, BJ10 was classified as Pseudomonas putida. As a result of biological PCE degradation at low PCE concentrations (5 mg/L), PCE removal efficiency by P. putida BJ10 was 52.8% for 10 days, and PCE removal rate was 5.9 nmol/hr (toluene concentration 50 mg/L, initial cell density 1.0 g (wet weight)/L, temperature 30, pH 7 and DO $3.0{\sim}4.2\;mg/L$. At high PCE concentration (100 mg/L), PCE removal efficiency by P. putida BJ10 was 20.3% for 10 days, and PCE removal rate was 46.0 nmol/hr under the same conditions. The effects of various toluene concentration (5, 25, 50, 100, 200 mg/L) on PCE degradation were examined under the same incubation conditions. The highest PCE removal efficiency of PCE was 57.0% in the initial PCE concentration of 10 mg/L in the presence of 200 mg/L toluene for 10 days. Furthermore, the additional injection of 5.5 mg/L PCE (total 7.6 mg/L) made 63.0% degradation for 8 days in the presence of 50 mg/L toluene under the same conditions. Its removal rate was 13.5 nmol/hr, which was better than the initial removal rate (8.1 nmol/hr).

  • A microbial BOD sensor for the continuous estimation of BOD was been developed by immobilizing Trichosporon cutaneum, which was immobilized between a dialysis and a gas-permeable membrane, on an oxygen electrode. The optimum pH and temperature for BOD measurement using this sensor were pH 7.0 and $32{\sim}33^{\circ}C$, respectively. The best result was obtained at 2~3ml/min flow rate in 0.1M phosphate buffer solution. A linear relationship was observed between ${\Delta}DO$ and the concentration of standard GGA solution below 60mg/l(90ppm $BOD_5$). The reproducibility was found to be within 3% for the standard solution containing glucose 30mg/l and glutamic acid 30mg/l. The output DO value of this sensor was almost constant for 30 dalys. The response time and the recovery time were about 5 and 10 min, respectively. This sensor was employed for the BOD measurement of waste pollutants and was compared with $BOD_5$ method.

Lipid and Citric Acid Production by Wild Yeasts Grown in Glycerol

  • In this study, crude glycerol was used as a carbon source in the cultivation of wild yeasts, aiming at the production of microbial lipids and citric acid. Forty yeasts of different sources were tested concerning their growth in crude and commercial glycerol. Four yeasts (Lindnera saturnus UFLA CES-Y677, Yarrowia lipolytica UFLA CM-Y9.4, Rhodotorula glutinis NCYC 2439, and Cryptococcus curvatus NCYC 476) were then selected owing to their ability to grow in pure ($OD_{600}$ 2.133, 1.633, 2.055, and 2.049, respectively) and crude ($OD_{600}$ 2.354, 1.753, 2.316, and 2.281, respectively) glycerol (10%, 20%, and 30%). Y. lipolytica UFLA CM-Y9.4 was selected for its ability to maintain cell viability in concentrations of 30% of crude glycerol, and high glycerol intake (18.907 g/l). This yeast was submitted to lipid production in 30 g/l of crude glycerol, and therefore obtained 63.4% of microbial lipids. In the fatty acid profile, there was a predominance of stearic (C18:0) and palmitic (C16:0) acids in the concentrations of 87.64% and 74.67%, respectively. We also performed optimization of the parameters for the production of citric acid, which yielded a production of 0.19 g/l of citric acid in optimum conditions (38.4 g/l of crude glycerol, agitation of 184 rpm, and temperature of $30^{\circ}C$). Yarrowia lipolytica UFLA CM-Y9.4 presented good lipid production when in the concentration of 30 g/l of glycerol. These data may be used for production in large quantities for the application of industrial biodiesel.

A Study on the Combined Treatment of Municipal Solid Waste Landfill Leachate (도시폐기물매립지침출수의 병합처리에 관한 연구)

  • An experimental research was conducted in order to study the combined treatment o of municipal landfill leachate and municipal sewage. The landfill leachate was that of Nanjido landfill site, and the municipal sewage was that of Chungnang municipal sewage treatment plant in Seoul. Several sets of bench~scale sequencing batch reactor(SBR) were used as e experimental apparatus. Specially investigated items in this experiment were the removal efficiency of substrate and the influence of treatment time. The experiment lasted for about 2 years. The result are as follows ; 1. The characteristics of leachate were pH 7.5~8.2, BOD 80~336mg/L, COD 908~1,460mg/L, NH3-N 1,409~2,330mg/L, T~P 2.7~7.lmg/L, Cl~3,540~4,085mg/L, a and heavy metals are a very small amount. And the characteristics of sewage were pH 6.9~7.3, BOD 78.4~129.3mg/L, COD 121.2~305.0mg/L, T~N 14.9~36.4mg/L, T-P 2.3~8.9mg/L. 2. The treatability of leachate alone was not treat well. So for the good treatment of leachate, it was necessary to deal with the pretreatment before bi이ogical treatment and a combined treatment of municipal sewage. 3. The various contents of the leachate were 5%, 10%, 30%, and 50%, and the removal efficiency of COD was 86.0%, 82.8%, 60.6%, and 31.7%. The maximum content of the leachate which could be sucessfully treated by SBR in the combined treatment was 10% of that of sewage. And the removal efficiency of COD increased n notably, as its treatment time increased. 4. The various contents of the electrolytic treated leachate were 5%, 10%, 30%, and 50%, and the removal efficiency of COD was 89.9%, 86.1%, 79.2%, and 69.8%. The maximum content of the leachate which could be sucessfully treated by SBR in the combined treatment was 30 % of that of sewage. And the removal efficiency of C COD increased notably, as its treatment time increased.

Induction and in vitro proliferation of adventitious roots in Dendropanax morbifera (황칠나무(Dendropanax morbifera)의 부정근 유도 및 기내증식조건)

  • Dendropanax morbifera (Araliaceae) is an endemic species in Korea and distributed in the southern part of Korea. The roots and stems of this plant have been used for folk medicine for the treatment of migraine headache, dysmenorrheal, and remove wind dampness and for Vanishes production. Production of adventitious roots in D. morbifera by in vitro cultures could be used as alternatives materials. Leaf, stem, and root segments from D. morbifera seedling were cultured on Murashige and Skoog (MS) medium supplemented with 3.0 mg/L IBA and 30 g/L sucrose. After 4 weeks of culture, the highest induction of adventitious roots was obtained from the leaf segment. Frequency of adventitious root formation on medium with various kinds of auxins (IAA, NAA, 2,4-D, and IBA) and various concentrations of IBA (0, 0.1, 0.5, 1.0, 3.0, and 5.0 mg/L) was tested. The maximum induction of adventitious root was obtained on medium with 1.0 mg/L IBA. In liquid culture, growth of root was best 1/2MS medium supplemented with 1.0 mg/L IBA and 30 g/L sucrose. Adventitious roots were cultured in 5 L bioreactor containing 1/2 MS medium supplemented with 1.0 mg/L IBA and 30 g/L sucrose and mass-production of adventitious roots was successfully achieved. This study demonstrated for the first time to produce adventitious roots in D. morbifera.

Corrosion Behavior of Inconel X-750 for Carbon Anode Oxide Reduction Application

  • The corrosion behavior of the Inconel X-750 alloy was investigated for its potential application under a Cl2-O2 mixed gas flow in an Ar atmosphere. The corrosion rate was found to be negligible at temperatures up to 400℃ under a flow rate of 30 mL·min-1 Cl2 + 170 mL·min-1 Ar, whereas an exponential increase was observed in the corrosion rate at temperatures greater than 500℃. The suppression of the corrosion reaction due to the presence of O2 was verified experimentally at flow rates of 30 mL·min-1 Cl2 (4.96 g·m-2·h-1), 20 mL·min-1 Cl2 + 10 mL·min-1 O2 (2.02 g·m-2 ·h-1), and 10 mL·min-1 Cl2 + 20 mL·min-1 O2 (1.34 g·m-2·h-1) under a constant Ar flow rate of 170 mL·min-1 at 600℃ for 8 h. The surface morphology analysis results revealed that porous surfaces with tunnel-type holes were produced under the Cl2-O2 mixed-gas condition. Furthermore, the effects of the Cl2 flow rate on the corrosion rate were investigated, indicating that its impact was negligible within the range of 5-30 mL·min-1 Cl2 at 600℃.

Medicinal Plant Lemon Grass (Cymbopogon Citratus) Growth under Salinity and Sodicity

  • Salinity with sodic condition disturbs germination, retards emergence, and slow down seedling development of Lemon Grass (Cymbopogon citratus).Lemongrass is a perennial grass plant widely distributed worldwide and most especially in tropical and subtropical countries. This research experiment was designed to evaluate the influences of (4 dSm-1+ 13.5 (mmol L-1)-1/2, 5 dSm-1+ 25 (mmol L-1)1/2, 5 dSm-1+ 30 (mmol L-1)1/2, 10 dSm-1+ 25 (mmol L-1)1/2 and 10 dSm-1+ 30 (mmol L-1)1/2) on biomass produce of lemon grass against salt tolerance. The uppermost biomass yield (45.53 gpot-1) was produced by 4 dSm-1+ 13.5 (mmol L-1)1/2 treatment. The increase in the intensity of salts reduced the growth of lemon grass. Lower biomass yield (79.33 gpot-1) was gained at 10 dSm-1+ 30 (mmol L-1)1/2. 5 dSm-1+ 25 (mmol L-1)1/2 treatment performed enhanced outcome i.e. the least reduction % over control (5.87). Salinity- sodicity showed serious effect on the growth reduction from 5.87% to33.60%. This reduction gap was affected by the negative effect of salinity and sodicity on Linseed growth. Salinity- sodicity showed severe impact on the growth reduction from 5.87% to33.60%. Based on the findings, lemon Grass (Cymbopogon citratus).was capable to grow up the maximum at 4 dSm-1+ 13.5 (mmol L-1)1/2 treatment.
