The Induction of ROS-dependent Autophagy by Particulate Matter 2.5 and Hydrogen Peroxide in Human Lung Epithelial A549 Cells (미세먼지와 산화적 스트레스에 의한 인간 폐 상피 A549 세포에의 ROS 의존적 자가포식 유도)

  • Recently, interest in the harmful factors of particulate matter (PM), a major component of air pollution, has been increasing. In particular, PM2.5 with a diameter of less than 2.5 ㎛ is well known to induce oxidative stress accompanied by autophagy in human lung epithelial cells. However, studies on whether PM2.5 increases autophagy under oxidative stress and whether this process is reactive oxygen species (ROS)-dependent are insufficient. Therefore, in this study, we investigated whether PM2.5 promotes autophagy through the generation of ROS in human alveolar epithelial A594 cells. According to our results, cells co-treated with PM2.5 and hydrogen peroxide (H2O2) showed a lower cell viability than cells treated with each alone, which was associated with increased total and mitochondrial ROS production. The co-treatment of PM2.5 and H2O2 also increased autophagy induction, which was confirmed through Cyto-ID staining, and the expression of autophagy biomarker proteins increased. However, when ROS generation was artificially blocked by N-acetyl-L-cysteine pretreatment, the reduction in cell viability and induction of autophagy by PM2.5 and H2O2 co-treatment were markedly attenuated. Therefore, the present results suggest that PM2.5-induced ROS generation may play a critical role in autophagy induction in A549 cells.

Growth Characteristics of Tomatoes Grafted with Different Rootstocks Grown in Soil during Winter Season (대목 종류에 따른 저온기 토경재배에서의 토마토 생육 특성 분석)

  • Cultivation of tomatoes in Korea grown in soil covers 89% of the total area for tomato cultivation. Tomatoes grown in soil often encounter various environment stresses including not only salt stress and soil-borne diseases but also cold stress in the winter season. This study was conducted to comparatively analyze the performance of rootstocks with cold stress by measuring the growth, yield, and photosynthetic efficiency in tomatoes grown in soil. The rootstocks were used 'Powerguard', 'IT173773', and '20LM' for the domestic rootstock cultivars and 'B-blocking' for a control cultivar. The tomato cultivar 'Red250' was used as the scion and the non-grafted tomatoes. Stem diameter, flowering position, leaf length, and leaf width were investigated for the growth parameters. The stem diameter of the non-grafted tomatoes decreased by 15% compared to the grafted tomatoes at 80 days after transplanting when exposed to low temperatures of 9-14℃ for 14 days. The leaf length and width of the non-grafted tomatoes were the lowest with 42.4 cm and 41.8 cm at 80 days after transplanting. The total yield per plant was the highest in tomato plants grafted on 'Powerguard' with 1,615 g and lowest in non-grafted tomatoes with 1,299 g. As the result of measuring the chlorophyll fluorescence parameters, PIABS and DI0/RC, which mean the performance index and dissipated energy flux, 'Powerguard' was the highest with 3.73 in PIABS and the lowest with 0.34 in DI0/RC, whereas non-grafted tomatoes was the lowest with 2.62 in PIABS and the highest with 0.41 in DI0/RC at 80 days after transplanting. The stem diameter has positive correlation with PIABS, while it has negative correlation with DI0/RC. The results indicate that can be analyzed by chlorophyll fluorescence parameters can be used for analyzing the differences in the growth of tomato plants grafted on different rootstocks when exposed to cold stress.

Effects of Dry Heat Treatment on the Reduction of Main Food-Borne Bacteria on Alfalfa Seeds (건열처리를 이용한 알팔파의 주요 식중독균 저감화)

  • In this study, the conditions of dry heat treatment (21 days at 65℃, 16 days at 70℃, 10 days at 75℃, and 7 days at 80℃) were investigated to inactivate Bacillus cereus ATCC 12480, Listeria monocytogenes ATCC SSA81, Staphylococcus aureus ATCC 6538, Escherichia coli O157:H7 ATCC 43894, and Salmonella Typhimurium ATCC 14028 on alfalfa seeds, without affecting the rate of germination of seeds. Alfalfa seeds were inoculated at levels of 6-7 log CFU/g and treated with dry heat at 65℃, 70℃, 75℃, and 80℃; thereafter, the rate of seed germination was determined. The rate of germination was set at 70%, according to the market standards. The bacteria were inactivated when B. cereus was treated with dry heat for 21 days at 65℃, 18 days at 70℃, 14 days at 75℃, and 4 days at 80℃; L. monocytogenes was treated for 21 days at 65℃, 18 days at 70℃, 12 days at 75℃, and 7 days at 80℃; S. aureus was treated for 18 days at 65℃, 18 days at 70℃, 11 days at 75℃, and 4 days at 80℃; E. coli O157:H7 was treated for 21 days at 65℃, 18 days at 70℃, 12 days at 75℃, and 6 days at 80℃; and Sal. Typhimurium was treated for 24 days at 65℃, 22 days at 70℃, 14 days at 75℃, and 7 days at 80℃. For all bacteria, the D-value (R2 = 0.5656-0.7957) significantly decreased when the temperature increased from 65℃ to 80℃ (P<0.05). Since dry heat treatment of alfalfa seeds at 80℃ for 7 days affects their germination rate, dry heat treatment at 75℃ for 14 days is the most effective way to ensure their safety. This study suggests a potential method of bacterial inactivation using dry heat treatment to increase the microbiological safety of sprouts.

Studies on the Selection Efficiency for Clear Rice in the wx - carrier technique. (wx-Carrier Technique를 이용한 맑은 쌀의 선발에 관한 연구)

  • The main objective of this experiment was to investigate the selection efficiency for clear rice by the application of the wx-carrier technique in rice. Twelve semi-dwarf waxy lines were bred through backcrosses to the two recurrent semi-dwarf parents, IR667 and IR1317, utilizing six different waxy cultivars as waxy donor parent. These waxy lines were crossed to three clear non-waxy varieties, IR24, Suweon 287 and Suweon294. Their F$_3$ seeds were separated into waxy and non-waxy and the clearness of non-waxy grains were counted. The results are summarized as follows: l. Mean clearness per plant for twelve waxy lines ranged from 3.7% to 78.9% at the 35 days after heading. It was higher in the lines which utilized IR 1317 as recurrent parent and it was lower in the lines which utilized IR667 as recurrent parent. Mean clearness per plant for recurrent parent, IRl317 and IR667 were 47.6% and 5.6% respectively. The clear non-waxy parents; Suweon287, IR24 and Suweon294 showed 83.7% 80.5% and 73.5% clearness respectively. 2. Mean clearness of F$_3$ seeds of the crosses between different waxy parents and IR24, Suweon 294 and Suweon 287 ranged 16.7-73.9%, 21.9-42.9% and 10.6-26.9%, respectively. IR24 crosses showed the highest mean clearness. 3. Highly positive correlation was found between the clearness of F$_3$ seeds of Suweon 287 crosses and those of Suweon294. 4. Significant differences were observed in mean clearness of F$_3$ seeds of the crosses between the different waxy lines, which was bred through the same number of backcrosses to the same recurrent parent, and a clear non-waxy parent. Crosses of IR1317 recurrent parent showed higher mean clearness than those of IR667 recurrent parent. 5. In some crosses, clearness was higher in homo-non-waxy than in hetero-non-waxy, but, in other crosses it was higher in hetero-non-waxy. Thus no distinct pattern in the segregation of clearness was observed along the homo-non-waxy or hetero-non-waxy. 6. From the results it was concluded that, the selection efficiency for the clear rice can be improved by choosing the proper waxy parent, as well as proper recurrent parent under the wx-carrier technique.

Biological Yielding Potential of Rice in Association with Climatic Factors in Yeongnam Region (영남지역 기상과 수도의 한계생산력 해석)

  • Meteorological year variations for rice crop from 1973 to 1984 were compared by using air temperature and sunshine hour for nursery period, cooling index for reproductive stage and meteorological yield productivity index for ripening period. The most optimum transplanting date and heading date for crop yield based on real transplanting date-grain yield relationship or heading date-grain yield relationship, meteorological yield productivity index and actual results showed good agreement each other. Around May 26 for transplanting and August 10 for heading were the most optimum date in Indica/Japonica hybrid cultivars while these were about June 8 and August 23 for Japonica cultivars, respectively. On the other hand, theoretical late limiting heading date for safe ripening were August 20 for Indica/Japonica hybrid cultivars and August 30 for Japonica cultivars, respectively, for both methods, cumulative temperature method during ripening with 80% believable frequency and meteorological yield productive index method having 1000(kg/10a) yielding potential. Based on the yield forecast trial, the highest values of photosynthetic efficiency, 2.5%, and crop growth rate, 23g/㎡/day, were recorded during 30 days before rice heading. Considering the photosynthetic efficiency and solar radiation, the potential crop growth rate was more or less 30g/㎡/day and the biological grain yielding potential in a existing cultural practices was approximately 900-1000(kg/10a) in Milyang weather condition. To increase further yielding potential, either photosynthetic efficiency or harvest index or both should be improved by manipulating appropriate canopy architecture, plant spacing, fertilizer, chemical, etc.

Effects of Planting Date on Agronomic Characteristics and Varietal Differences in Sesame Varieties (파종기 이동에 따른 참깨 품종들의 유용형질 변화 및 품종간 차이)

  • This study was conducted to obtain basic informations for breeding and improving cultural practices of sesame (Sesamum indicum L.) through investigation of several agronomic characters of 82 major varieties plants in April 20, May 15 and June 20. Stem diameter and plant height were largest in early planting(April 20). On the contrary, they were smallest in late planting(June 15). Therefore, the later planting, the poorer the vegetative growth on the basis of stem diameter and plant height. Number of capsules per plant and number of grains per capsule were 88 and 54, respectively, in the early planting, while they were decreased to 25 capsules and 40 grains in late planting. The percentage of ripeness and 1000 grain weight 78 and 2.3g in the April 20planting while they were 58% and 2.1g in June 20 planting indicating the later planting, the lower the ripeness. Grain yield per m1 in April 20 and June 20 planting was 112 and 18g respectively. Consequently, grain yield significantly decreased as planting delayed. On the basis of the vegetative and reproductive growth, varieties could be classified into early, conventional, late and indetermediate planting adapted groups. The major yield components were highly and postively correlated with accmulated sunshine hours and temperatures confirming that grain yield was closely related with sunshine and temperature. Among the varieties tested, Gurye and Local 135 had more than 150 capsules. Haenam and Woogang had more than 75 grains per capsule. Since grain yield oflate planted sesame were significantly different among the varieties, breeding of sesame for after barly cropping would be more effective under late cultural condition of after barly.

Studies on the Flowering and Maturity in Sesame 1. Flowering Habit by Different Plant Types (참깨 개화, 등숙에 관한 연구 -제 1 보 참깨 초형에 따른 개화특성에 관한 연구-)

  • This experiment was performed to investigate the flowering habit of sesame (Sesamum indicum L.). Sesame varieties tested could be classified into 8 different plant types by their morphological traits such as capsule shape, capsule setting habit and branching types among sesame gene pool of Crop Experiment Station, ORD. The first flower was appeared at the lowest node on main stem. Flowers were appeared progressively toward the tip of the main stem and also toward the tips of branches. The interval of flowering for a node was about one day, but 3 to 8 days for the flowers on the tips. Side flowers started at 4 to 5 nodes lower than those of center flower at the same day. Flowers were beared 2 by 1 node on the middle part of flower setting node (7-9) in mono capsule setting habit in spite of its normal is 1 by 1 node on the other nodes. Flowers were beared opposite direction on each node of stem and flowering toward the tip of main stem composed of cross shape between nodes and spiral, reverse of clockwise direction. We called this habit as cross spiral flowering order and cross spiral phyllotaxis. The first flower on branches was appeared when center flower on the 5th node of main stem began to flower. The branches produced at higher nodes on main stem showed larger flowering periods and more number of flowers than that at lower parts. BTB (Branch, Tricapsule, Bicarpels, 4 Loculi) type showed three capsule setting habits and same flowering period both on main stem and branches while BTQ (Branch, Tricapsule, Quadricarpels, 8 Loculi) type showed three capsule setting habit on main stem and mono-capsule setting habit on branches. In BTQ type, the period of flowering was much shorter on branches than on main stem. Branching type was considered more promising than non branching type for the breeding of early maturing high yielding variety because branching type has the advantage of bearing a lot of flowers in comparatively short flowering period.

Feed Value of Pearl Millet [Pennisetum americanum(L.) Leeke] Grown as a New Forage Crop (진주조의 사료적 가치)

  • Pearl millet has been detected as a promising new forage crop of excellent quality and productivity since 1985. Its green fodder yields were 10.7 to 12.8 tons per hectare in average of 26 accessions in Suwon, 1985. The yield level was much higher than those of Italian and pro so millets and com. Com was better than Italian and proso millets, and proso millet was better than Italian millet for a green fodder crop. Suwon 1 pearl millet hybrid was the best of the 13 hybrids examined in Suwon, 1986 being 149 tons per hectare of three times cut green fodder yields. The pearl millet hybrid was higher in green fodder yield than com and sorghum/sudan grass hybrids. Leaf area index was 32.4 for the three times cut pearl millet, while 5.8 for the one time cut corn, and 20.8 for the three times cut sorghum/sudan grass. Crude protein content was 16.3 percent for pearl millet being six to five percent higher than corn and sorghum/sudan grass, 11.8 percent for the one time cut at maturity and 16.1 percent for four times cut being higher than corn and sorghum/sudan grass. Crude fat content was 3. percent for pear 1 millet grain being some what higher than corn and sorghum/sudan grass and 1.3-1.4 percent for green fodder crop. Crude fiber content in grain was 1.9 percent for pearl millet 2.6 percent for corn, and 4.3 percent for sorghum/sudan grass. Crude fiber content in pearl millet plant was 24.4 to 26.8 percent, Crude ash content was 2.4 percent in grain and 10.8 to 11.6 percent in the plants of pearl millet hybrid. In vitro digestibility of grain was 93.7 percent for pearl millet, 95.4 percent for corn, and 55.8 percent for sorghum/sudan grass. The digestibility of whole plant was 57.6 to 63.4 percent for pearl millet, 46.3 percent for corn, and 47.3 to 57.6 percent for sorghum/sudan grass. Heavier nitrogen fertilizer applications increased green fodder yields, protein content and digestibility, but reduced fat and ash content of pearl millet inbred line T 186.

Varietal Difference and Environmental Variation in Protein Content and/or Amino Acid Composition of Rice Seed (쌀의 단백질함량과 아미노산 조성의 품종간 차이와 환경변이)

  • Varietal difference of protein content in forty eight Korean recommended rice cultivars and environmental Variation in protein content of milled rice harvested at six sites of the middle and/or southern plain and four locations of mid-mountainous and/or alpine area in 1989 were investigated. Also, the composition of amino acid in milled rice was compared among three rice varieties: a high-protein japonica rice, Nongbaek, a high-protein Tongil-type rice, Yongjubyeo, and a low-protein japonica rice, Hwaseongbyeo. Korean recommended rice varieties showed 7.93% of average protein content with varietal variation from 5.5% to 10.2% for milled rice harvested in 1988, and 9.17% of mean protein content with the variation from 6.3% to 12.0% for milled rice harvested in 1989. Tongil-type rice was about 1% higher in protein content of milled rice than japonica. The low-protein japonica rice, Hwaseongbyeo exhibited lower content of essential amino acids per g of rice flour sample than the high-protein japoinica, Nongbaek and/or Tongil-type rice, Yongjubyeo, but the relative content of essential amino acids per 16.8g of nitrogen in milled rice of the former was not so different with those of the latters. Among amino acids the content of glutamic acid was highest and among essential amino acids the content of leucine was highest while methionine was lowest. The protein content of milled rice was negatively correlated with days from seeding to heading, K/Mg ratio, alkali digestion value(l-7) and amylose content, but it was positively correlated with translucency and magnesium content of milled rice. The protein content of milled rice harvested in the southern plain paddy field was about 1% higher compared with those harvested in the Middle plain. Also, the protein content of milled rice harvested in the southern mid-mountainous and alpine area was about 0.8% higher compared with those harvested in the resemble altitude area of the middle-northern part of Korea. The contribution of environmental variation to total in plain area was about 28.1% while that in mid-mountainous and alpine area was about 56.4%.

Relationship between Planthoppers (Nilaparvata lugens and Sogatella furcifera) and Rice Diseases (멸구류(類)(벼멸구 및 흰등멸구)와 수도병해(水稻病害)의 복합발생피해(複合發生被害)에 관(關)한 연구(硏究))

  • The locational preference of the brown planthopper (BPH) Nilaparvata lugens ($St{\aa}l$) and the whitebacked plant hopper (WBPH) Sogatella furcifera(Horvath) was studied on rice cultivars IR22 and IR36 as an integral part of subsequent research on insect-fungal pathogen relationships. The BPH was observed to stay consistently on the basal portion while the WBPH showed a general preference for the upper portion regardless of varieties, rice growth stages and insect population density levels. The habitat preference of both species (BPH and WBPH) was found not to be affected by the presence of the other species when both species are present on the same host plant Five rice cultivars with different reactions to BPH biotype 2 were used in the study on BPH-Rhizoctonia solani relationship: IR22 and TN1 (susceptible); Triveni and ASD7 (moderately resistant); and IR42 (resistant). Test plants were inoculated with R. solani (Kuhn) $3{\sim}4$days after insect infestation. Sheath blight disease severity/incidence was significantly higher in the treatment where BPH+R. solani were together than in the treatment with only the pathogen. Symptom expression of the disease in the BPH-pathogen combination was faster and mycelial growth was more profuse inducing the formation of more infection structures. Regardless of varietal reaction to BPH biotype 2, the degree of hopperburn was significantly higher in the combination of the two pests as compared with that of BPH alone. There could be a synergistic relationship between the insect pest and the pathogen indicated by a positive interaction between the two species.
