Studies on Dorsal Aspect Target Strengths of Rock Bream, Oplegnathus Fasciatus and Dusky Spinefoot, Siganus Fuscescens (돌돔과 독가시치의 등방향 반사강도에 관한 연구)

  • In order to obtain fundamental data for estimation of fisheries resource by echo sounder, we carried out the measuring of dorsal aspect Target strengths for rock bream and dusky spinefoot fishes that were caught much around the Jeju Island and in South Sea of Korea. The appropriate equations share the common form. TS=A+20 log L, where TS is the average dorsal aspect target strength in decibels, L is the fish total length in centimeters, and the coefficient A is determined by a least mean squares regression analysis. For rock bream, the result is TS=-72.97+20 log L and, for dusky spinefoot it is TS=-63.16+20 log L And, we have investigated the bearing range of maximum dorsal aspect target strength for all of rock bream and dusky spinefoot by the echo sounder with transducer of which frequency is 200kHz. They are $-12^\circ$-$-21^\circ$and $-1^\circ$--8 espectively, when the fishes is swimming down to the bottom. The maximum dorsal target strengths are -41.50dB at -18 or rock bream and -30.69dB at $-6^\circ$for dusky spinefoot.

Estimating Surface Orientation Using Statistical Model From Texture Gradient in Monocular Vision (단안의 무늬 그래디언트로 부터 통계학적 모델을 이용한 면 방향 추정)

  • To recover three dimensional information in Shape from Texture, the distorting effects of projection must be distinguished from properties of the texture on which the distortion acts. In this paper, we show an approximated maximum likelihood estimation method in which we find surface orientation of the visible surface (hemisphere) in gaussian sphere using local analysis of the texture. In addition, assuming that an orthogonal projection and a circle is an image formation system and a texel (texture element) respectively, we drive the surface orientation from the distribution of variation by means of orthogonal projection of a tangent direction which exists regularly in the arc length of a circle. We present the orientation parameters of textured surface with slant and tilt in gradient space, and also the surface normal of the resulted surface orientationas as needle map. This algorithm is applied to geographic contour (artificially generated chejudo) and synthetic texture.

  • This study was carried out to estimate drying schedule of Fagaceae four species grown in Kangwon-Do by oven-drying at $100^{\circ}C$ which aimed to elucidate the characteristics such as current moisture content, drying process, initial check, collapse and internal check during drying. The results were summerized as follows ; Current moisture content of each board showed a rapid drying curve with the hight initial moisture content of board and species with the high initial moisture content took long to do drying. Appearance of checks for Quercus variabilis were extremely severe and estimated schedule was $T_5-C_2$. The shrinkage rate of grain direction was in the order of tangential direction > radial direction > longitudial direction and the shrinkage rate of oven-drying at $100^{\circ}C$ was bigger than normal shrinkage rate.

Quantifying Uncertainty of Vitamin C Determination in Infant Formula by Indophenol Titration Method (인도페놀 적정법에 의한 성장기용조제식 중 비타민 C 함량분석의 측정불확도 산정)

  • Uncertainty involved during determination of vitamin C content in infant formula was quantified by indophenol titration method. Uncertainty sources in measurand, such as purity, weight, final volume of standard, volume of standard solution used for titration, sample weight, final volume of sample, extraction solution used for titration, titration of extraction solution and standard solution by indophenol solution were identified and used as parameters for combined standard uncertainty based on Guide to the expression of uncertainty in measurement (GUM) and Draft EURACHEM/CITAC Guide. Uncertainty parameters of each source in measurand were identified as resolution, reproducibility and stability of chemical balance, standard material purity, repeatability, reproducibility, end point of titration, 1 mL pipet, 5 mL autopipet, and 100 mL mass flask. Each uncertainty component was evaluated by types A and B and included to calculate combined uncertainty. Analytical test result for traceability under laboratorial conditions using Certified Reference Material (CRM) test was certified as $108.4{\pm}1.7mg/100g$, which was within CRM certification range of $114.6{\pm}6.6mg/100g$. Uncertainty test result of vitamin C content of 5 g sampling was $56.7{\pm}2.44mg/100g$. Uncertainty could be reduced by identification of uncertainty sources and components related with vitamin C determination by indophenol titration method and by decreasing uncertainty sources and components.

Optimization of Biopolymer Production from Alkali-Tolerant Bacillus sp. (알칼리 내성 Bacillus sp.의 생물고분자 생산조건의 최적화)

  • Cultural conditions for the biopolymer production by an alkali tolerant Bacillus sp. isolated from soil were investigated and determination of optimal conditions was carried out by response surface method. The maximal production of biopolymer was obtained after cultivation at $30^{\circ}C$ for 36hrs in the mixture of 8% soluble starch, 0.75% yeast extract, 0.1% $NaNO_3$, 0.05% $MgSO_4\;7H_2O$ and 1% $Na_2CO_3$ adjusted to pH 10. Under these conditions, about 44 g/l of biopolymer were produced. From the results of response surface analysis, optimal condition for the production of biopolymer were obtained at stationary point with 15.16 of C/N ratio, $34.62^{\circ}C$ of temperature and 9.50 of pH. On the basis of these conditions, it was estimated that 66.84 g/l of the biopolymer could be produced.

  • This experiment was carried out to investigate the lectin of soybean (Glycine max L.) seed. Purification was done by 50-80% ammonium sulfate precipitation, CM-cellulose and Sephadex G-100 column. The purity was ascertained by electrophoresis. The molecular weight of purified lectin was estimated as 132,000. It was composed of three subunits which molecular weight was 45,000. The lectin was identified as glycoprotein by Schiff's reagent staining and Dubois method. The lectin agglutinated erythrocytes of rabbit and human. The amounts of the lectin to agglutinate human erythrocytes differed among the blood types: The blood type A required the least amount, the next was B, O, and AB in order. The agglutination was specifically inhibited by 5${\mu}$g/ml of N -acetyl.-D-galactoseamine and 200${\mu}$g/ml of D-galactose. Other tested sugars could not inhibit the agglutination of the erythrocytes by the lectin.

Optimization of Electro-UV-Ultrasonic Complex Process for E. coli Disinfection using Box-Behnken Experiment (Box-Behnken법을 이용한 E. coli 소독에서 전기-UV-초음파 복합 공정의 최적화)

  • This experimental design and response surface methodology (RSM) have been applied to the investigation of the electro-UV-ultrasonic complex process for the disinfection of E. coli in the water. The disinfection reactions of electro-UV-ultrasonic process were mathematically described as a function of parameters power of electrolysis ($X_1$), UV ($X_2$), and ultrasonic process ($X_3$) being modeled by use of the Box-Behnken technique, which was used for fitting 2nd order response surface model. The application of RSM yielded the following regression equation, which is empirical relationship between the residual E. coli number (Ln CFU) in water and test variables in coded unit: residual E. coli number (Ln CFU) = 23.69 - 3.75 Electrolysis - 0.67 UV - 0.26 Ultrasonic - 0.16 Electrolysis UV + 0.05 Electrolysis Ultrasonic + 0.27 $Electrolysis^2$ + 0.14 $UV^2$ - 0.01 $Ultrasonic^2$). The model predictions agreed well with the experimentally observed result ($R^2$ = 0.983). Graphical 2D contour and 3D response surface plots were used to locate the optimum range. The estimated ridge of maximum response and optimal conditions for residual E. coli number (Ln CFU) using 'numerical optimization' of Design-Expert software were 1.47 Ln CFU/L and 6.94 W of electrolysis, 6.72 W of UV and 14.23 W of ultrasonic process. This study clearly showed that response surface methodology was one of the suitable methods to optimize the operating conditions and minimize the residual E. coli number of the complex disinfection.

Quantitative analysis of cholesterol in infant formula by isotope dilution liquid chromatography-tandem mass spectrometry (동위원소희석 액체크로마토그래피 질량분석법에 의한 분유 내 콜레스테롤의 정량)

  • An isotope dilution liquid chromatography tandem mass spectrometry was developed as a primary method for the quantitative analysis of cholesterol in infant formula. Cholesterol-d4 was used as an internal standard and spiked into the infant formula sample. In order to release cholesterol out of cholesteryl ester, which is cholesterol bound to fatty acids in infant formula, saponification was carried out. Saponification conditions were optimized with heating temperature, reaction time and the concentration of KOH. The optimum conditions were as follows; heating temperature was 70 ℃, reaction time was 180 min and the concentration of KOH was 0.8 mL of 8 M KOH for about 0.1 g infant formula sample. Extraction of cholesterol out of sample solution was carried out with hexane uisng liquid-liquid extraction. Chromatographic analysis was carried out using Phenomenex Kinetex C18 column. Mobile phase was 0.1% acetic acid in methanol/water (v/v, 99/1) and flow rate was 0.3 mL/min. Cholesterol and cholesterol-d4 were monitored at mass transfer m/z 369/259 and 373/263 respectively. Reproducibility of the method was evaluated to be 0.23% of the measurement result. The expanded uncertainty of the measurement result of cholesterol in infant formula was approximately 1.9% at a 95% confidence level. NIST standard reference material having certified values of cholesterol in infant formula, was analyzed in order to verify this method. The ID-LC/MS/MS results were well agreed with the certified values of NIST SRM within the uncertainty.

Evaluation of reference value of anti-Glutamic acid decarboxylase antibody for cerebrospinal fluid (뇌척수액에서 항 Glutamic acid decarboxylase 항체검사의 참고치 설정)

  • Purpose Anti-Glutamic acid decarboxylase antibody test (GAD Ab) has been used as a predictor of type 1 diabetes. GAD Ab has also been shown to be highly potent in cerebrospinal fluid (CSF) of patients with suspected diabetic peripheral neuropathy. Recently, it has been known that clinical significance of GAD Ab using CSF is useful for the neurological disorders. However, the reference value of anti-GAD Ab has been provided only for serum. In this experiment, we estimated the reference value of anti-GAD antibody for CSF in neurological patients. Materials and Methods A total of 211 neurological patients were enrolled. Serum and CSF were analyzed by radioimmunoassay (RIA) using commercial RIA anti-GAD Ab kit (RSR, London, United Kingdom). Normal saline was used as the normal CSF control because CSF is most similar to 0.9% normal saline. Results The mean value of normal CSF control was 1.97 U/mL, and two standard deviations (2SD) was 1.44 U/mL. Based on this data, the expected reference range of CSF could be estimated from 0.54 U/mL to 3.40 U/mL Conclusion The reference range of normal CSF control using normal saline obtained with Hoffmann's method. However, there will be a need to validate the CSF reference values using human normal CSF.

  • Background : In diagnosis or monitor of the airway obstruction in bronchial asthma, the measurement of $FEV_1$ in the standard method because of its reproducibility and accuracy. But the measurement of peak expiratory flow(PEF) by peak flow meter is much simpler and easier than that of $FEV_1$ especially in children. Yet there have been still no data of the predicted normal values of PEF measured by peak flow meter in Korean children. This study was conducted to provide equations to predict the normal value of PEF and correlation between PEF and $FEV_1$ in healthy children. Method : PEF was measured by MiniWright peak flow meter, and the forced expiratory volume and the maximum expiratory flow volume curves were measured by Microspiro HI 501(Chest Co.) in 346 healthy children(age:5-16 years, 194 boys and 152 girls) without any respiratory symptoms during 2 weeks before the study. The regression equations for various ventilatory parameters according to age and/or height, and the regression equations of $FEV_1$ by PEF were derived. Results : 1. The regression equation for PEF(L/min) was: $12.6{\times}$age(year)+$3.4{\times}$height(cm)-263($R^2=0.85$) in boys, and $6{\times}$age(year)+$3.9{\times}$height(cm)-293($R^2=0.82$) in girls. 2. The value of FEFmax(L/sec) derived from the maximum expiratory flow volume curves was multiplied by 60 to compare with PEF(L/min), and PEF was faster by 125 L/min in boys and 118 L/min in girls, respectively. 3. The regression equation for $FEV_1$(ml) by PEF(L/min) was:$7{\times}$PEF-550($R^2=0.82$) in boys, and $5.8{\times}$PEF-146 ($R^2=0.81$) in girls, respectively. Conclusion : This study provides regression equations predicting the normal values of PEF by age and/or height in children. And the equations for $FEV_1$, a gold standard of ventilatory function, was predicted by PEF. So, in taking care of children with airway obstruction, PEF measured by the peak flow meter can provide useful information.
