The Optimization of Mevinolin Production by Medium Composition of Penicillium citrinum (Penicillium citrinum 의 배지 조성에 의한 Mevinolin 생산 최적화 연구)

  • Theses studies were made on the mevinolin production from Penicillium citrinum Thom (KCTC 6990) Culture conditions pH temperature carbon sources nitrogen sources mineral sources surfactants and glucose concentration were optimized. The results of glucose concentration and maximum mevinolin production according to incubating time in the flask nearly disappeared after 5 days and appeared after 7 days respectively. temperature and pH conditions of maximum mevinolin production were $24^{\circ}C$ and 3.7 pH respectively. The results of maximum mevinolin production according to the kind of nutrients were as follows. Glucose of carbon sources were 3.5 mg/L. Peptone of nitrogen sources were 3.5 mg/L TEX>$K_2HP0_4$ of mineral sources was 3.8 mg/L Tween 20 of surfactants were 4.5 mg/L Maximum mevinolin productioni of glucose con-centration was 4.0mg/L of glucose 100 g/L In the batch culture Maximum mevinolin concentration was 10.3 mg/L after 8 days. maximum mevinolin specific production rate 0.016 mg/g-hr. These results need to be studied more than ever about temperature pH 야ㅕㅡ and treatment of by-product oil in the batch culture and must do the fad batch from now to increase mevinolin productivity.

Effect of Moss (Hypnum jutlandicum) Extracts and Mixing Treatments with Preservatives on Vase Life of Rosa hybrida 'Enjoy' (이끼 추출물과 절화 보존제 혼용처리가 절화장미 'Enjoy'의 수명에 미치는 영향)

  • This study was performed to develop a natural preservative which had the most successful effect on prolonging the vase life of cut flowers Rosa hybrida 'Enjoy'. To determine the effect of sucrose on vase life of cut roses, R. hybrida 'Enjoy', 0, 1, 3 or 5% of sucrose was mixed with Hypnum jutlandicum extract. The result showed that in the case of adding sucrose to extracts from H. jutlandicum, vase life of cut flowers was shortened, along with the cut flowers losing chlorophyll, becoming decolorized and finally showing wilting phenomenon. However, using hydroxy quinoline sulfate (HQS) and extracts from H. jutlandicum together exhibited an improved effect of prolonging vase life, especially when adding 100 $mg{\cdot}L^{-1}$ HQS, which had the effect of prolonging the vase life of R. hybrida 'Enjoy' the most. Regarding pH of extracts, 2 $mg{\cdot}L^{-1}$ of extracts from H. jutlandicum show originally pH 3.8. This was compared with adjusted extract solutions as pH 3.0, 4.0 or 5.0. The results showed that the solution of pH 3.8 had the most successful effect on prolonging the vase life of R. hybrida 'Enjoy'. Given all the results, H. jutlandicum extract (2 $mg{\cdot}L^{-1}$) and HQS (100 $mg{\cdot}L^{-1}$) was the most effective in prolonging the vase life of cut R. hybrida 'Enjoy'. Moss is expected to be worth developing as a natural preservative since it is easy to get and causes no damage to the environment.

The Experimental Study of Distribution Characteristics of Lift-force Acting under Pier Deck (잔교상판(棧橋床板)에 작용(作用)하는 양압력(揚壓力) 분포특성(分布特性)에 관한 실험적(實驗的) 연구(硏究))

  • This study describes the characteristics of distribution of lift-force acting under pier deck through physical experiment. The shape of peak wave pressure was sharp when compressed air existed but was not sharp without that. Values of lift-force was different between edge point and center point in the same block. Distribution of lift-force was expressed differently owing to dimensionless of deck length (l/L), wave steepness (H/L), clearance height per wave height (D/H). The dimensionless factor of D/H affected on the lift-force the clearance between still water surface and decks. This decided the maximum of lift-force. In the case of the same values of D/H, the lift-force are changed by the wave steepness (H/L). Because (D/H) become smaller as the wave steepness (H/L) is increased the height of decks must be decided with the condition which don't have the clearance with $D_{max}$ for the stable design of deck of pier. Effect of reducing lift force was greater in the on-shore than the off-shore according to compressed air existence. This researches points out that design of deck should retain compressed air in order to reduce wave lift force.

  • Proton magnetic resonance spectra of five glycine-containing dipeptides glycyl-L-valine, L-valyl-glycine, glycyl-DL-alanine, glycyl-DL-serine and glycyl-L-aspartic acid in $D_2O$ were investigated as a function of pH at room temperature. From the analysis of the spectra, it was found that the chemical shift of the $C_{\alpha}H,\;C_{\beta}H\;and\;C_rH$protons varies with pH as a one-step titration curve, and that the spin-spin coupling constant remains almost unchanged. Two distinct values of the chemical shift for $C_{\alpha}H,\;C_{\beta}H\;or\;C_rH$protons of constituent amino acids in dipeptide solutions indicate the existence of two magnetically non-equivalent sites in solution. From this study, the structures of the five dipeptides have been confirmed by proton magnetic resonance spectra and it has been suggested that the structural change, conformation and sequence determination can be explored for oligopeptides by an analysis of proton magnetic resonance spectra.

Lactobacillus acidophilus Inhibits the Helicobacter pylori Adherence

  • Lactobacillus acidophilus is known to have an inhibitory activity on growth of Helicobacter pylori and this activity has been attributed to lactic acid and antibacterial agents produced by Lactobacillus. Since every Lactobacilli produces lactic acid, an another factor must exist for L. acidophilus to inhibit H. pylori growth. In this work, the inhibitory activity of L. acidophilus on H. pylori adherence was studied. An immunoabsorbent assay using TLC plate was developed and used for screening the inhibitory activity of various Lactobacilli on H. pylori adherence. Glycolipid, the attachment site for H. pylori, was isolated from blood type O red blood cells and spotted on a TLC plate. The H. pylori adherence increased linearly with increasing amounts of glycolipid spotted on the TLC plate. Various L. acidophilus strains, but not L. casei, appeared to inhibit H. pylori adherence to glycolipid, and the adherence decreased linearly as the concentration of the Lactobacillus increased. The results show that the inhibitory activity of L. acidophilus on H. pylori adherence is an another factor for L. acidophilus to inhibit H. pylori growth.

Steady Boundary Layer Flow under the Influence of Progressive Finite Amplitude Wave (진행성 유한진폭파로 인한 정상성 경계층류)

  • The problem of the formation of steady stream of flat bottom boundary is revisited by applying a progressive finite amplitude wave as an external flow. A solution for the boundary layer is found by expanding the boundary equation into double Fourier series. A vertical profile of the stream is obtained as a function of the ratio, h/L, where h and L are the water depth and the wave length. For the best applicable range of the external wave, it is shown that the boundary stream is independent of the fluid viscosity, but a function of the wave parameters and the water depth. The stream velocity of the steady boundary layer flow is proportional to the wave phase velocity and the square of the ratio, H/h, where His the wave height. The magnitude of the velocity is insignificant when h/L is greater than 1/5.

  • To evaluate the performance of a bipolar packed bed cell (BPBC) filled with granular aluminium, the experiments were carried out in two groups as batch and continuous processes. In a batch process, removal efficiency of total phosphate (T-P) was 88% in case of electrolytic treatment of phosphate solution, T-P 10 mg/L at 6 V during 3 h by BPBC filled with granular aluminium. In a continuous process, residual T-P concentration was about 2 mg/L in case of electrolytic treatment of phosphate solution, 10 mg/L at 6 V, HRT 3 h by BPBC filled with granular aluminium. Break-through point was observed after running for 120 h at hydraulic retention time (HRT) of 3 h.

  • Serum levels of human placental lactogen have been measured by hemagglutination-inhibition reaction in 67 normal pregnant state and in 15 postpartum 24 hour state, HAIR is less sensitive and reliable method than radioimmunoassay, but simple, rapid, less expensive and fairly accurate, so it is more helpful in screening of large antenatal population with or without high risk complications. 1) Sensitivity of HPL-HAIR test kit was $0.1{\mu}g$/ml of H.P.L. serum level and had no cross reaction to HCG or male serum or non-pregenant female or newborn infant, 2) H.P.L. value was around $2{\mu}g$/ml until 24th week of pregnancy and rose to $6{\sim}8$ ${\mu}g$/ml continuously until about 36th week of pregnancy and then slightly decreased or stationary. 3) H.P.L. value in postpartum 24 hour state was undetectable. 4) There was poor correlation between maternal serum H.P.L. value at term and baby weight.

  • This experiment was carried out to investigate the effect of red ginseng extract on the growth of Lactobacillus sp. (L acidophilus, L casei, L salivarius), Escherichia coli and Listeria monocytogenes in pH controled medium by $\beta-Glycerol\;PO_4$ buffer. The growth of Lactobacillus sp. was show a similar pattern in control and MRS broth with red ginseng extract $1.0\%$ but was remarkably show inhibiting in MRS broth with over $2.0\%$ red ginseng extracts. The growth of E coli was inhibited in Trypticase soy broth with $1.0\%$ red ginseng extracts. Also the growth of L monocytogenes was inhibited in Trypticase soy broth with $5.0\%$ red ginseng extract The growth of L acidophilus KCTC3150, L casei KCTC3189, L salivarius ssp. salivarius CNU27, and E coli KCTC1039, L monocytogenes KCTC3443 were remarkably inhibited in pH non-control medium and pH control medium with $10\%$ red ginseng extract These results was suggested to effect of inhibition of microorganisms growth not pH decrease by organic acid but another components in red ginseng extract.
