Production of D-sorbitol and L-sorbose from Jerusalem artichoke by Zymomonas mobilis and Gluconobacter sMboxpydans (Zymomonas mobilis와 Gluconobacter suboxydans를 이용한 돼지감자로부터 D-sorbitol 및 L-sorbose 생성에 관한 연구)

  • The use of Jerusalem artichoke containing $\beta$-1, 2-fructose oligomer for the production of D-sorbitol and L-sorbose has been studied. The employment of inulinase(0.398%, v/v) for the hydrolysis of 40% (v/w) Jerusalem artichoke juice resulted in 36.7g/1 of glucose and 85.3g/1 of fructose at $50^{\circ}C$. These sugars were utilized as substrates for D-sorbitol and L-sorbose production. Coimmobilization of inulinase and permeabilized cells of Zymomonas mobilis in the mixture of chitin (5%, w/e) and x-carrageenan(4%, w/v) resulted in the production of 30.2g/1 of D-sorbitol by using inulin as a substrate. The process of L-sorbose production from D-sorbitol by Gluconobacter suboxydans was optimized with respect to the substrate concentration, level of dissolved oxygen and glucosic and concentration. Gluconlc acid produced by Zymomonas mobilis from glucose was found to inhibit Gluconobacter suboxtans in conversion of D-sorbitol to L-sorbose. In view of removing such inhibitory effect by gluconic acid, mutants were selected by the NTG (N-methyl-N'-N'-nitro-N-nitrosoguanidlne) treated method. Mutants selected by NTG mutagenesis showed no inhibitory effects of gluconic acrid against L-sorbone production when its concentration increased up to 100g/1. A mutant produced 40.1g/l of L-sorbose in the medium containing 100g/l D-sorbitol and 100g/l-gluconic acid. This result is consider able when compared with L-sorbose concentration (21.7g/1) obtained from the fermentation with wild type strain of Gluconobacter suboxnians.

Cellular DNA Repair of Oxidative Deoxyribose Damage by Mammalian Long-Patch Base Excision Repair

  • 2-Deoxyribonolactone (dL) arises as a major DNA damage induced by a variety of agents, involving free radical attack and oxidation of C1'-deoxyribose in DNA. We investigated whether dL lesions can be repaired in mammalian cells and the mechanisms underlying the role of DNA polymerase $\beta$ in processing of dL lesions. Pol $\beta$ appeared to be trapped by dL residues, resulting in stable DNA-protein cross-links. However, repair DNA synthesis at site-specific dL sites occurred effectively in cell-free extracts, but predominantly accompanied by long-patch base excision repair (BER) pathway. Reconstitution of long-patch BER demonstrated that FEN1 was capable of removing the displaced flap DNA containing a 5'-dL residue. Cellular repair of dL lesions was largely dependent on the DNA polymerase activity of Pol $\beta$. Our observations reveal repair mechanisms of dL and define how mammalian cells prevent cytotoxic effects of oxidative DNA lesions that may threaten the genetic integrity of DNA.

  • When the reaction rate constant k of phospholipase D on liposome was measured by the ANS fluorometry, k of phospholipase D on DMPC liposome which was made of L-$\alpha$-PI, cholesterol and L-$\alpha$-PS decreased than that of phospholipase D on DMPC liposome with cholesterol or with PI and cholesterol. Optimal $Ca^{2+}$ concentration, the most important factor on effect of phospholipase D, also decreased to 1mM, as compared with 10mM and 60mM respectively when cholesterol and PI were added, and cholesterol only was added. The change of cholesterol Mol% had a great influence on k value of phospholipase D. But in case of addition of L-$\alpha$-PS to cholesterol, the influence was relatively diminished.

We show that the index $ind(^cD) of ^cD$ divides $[Z(\overline{D}) : Z(\overline {^cD})] \cdot ind(D), where Z(\overline{D})$ is the center of the residue division ring $\overline{D}$.

Gamakamide C and D as Two New Analogues of Bitter-Tasting Cyclic Peptide with Hydantoin Structure from Oyster Crassostrea gigas

  • Two new bitter-tasting cyclic peptides comprising six amino acids, namely gamakamide C and D, were isolated from cultured oysters Crassostrea gigas. Dimethylaminoazobenzene sulfonyl-amino acid analysis detected Val and Leu in gamakamide C and Ile and Leu in gamakamide D. The molecular formula of gamakamide C was determined as $C_{43}H_{60}N_{7}O_8S$ by high-resolution fast atom bombardment mass spectroscopy (HR FAB-MS) ($[M+H]^+m/z822.4200{\Delta}-2.4mmu$), and that of gamakamide D was determined as $C_{43}H_{62}N_7O_8S$ by HR FAB-MS ($[M+H]^+m/z836.4379{\Delta}-2.0mmu$). Comparison of amino acid analyses and fragment ions by MS/MS among gamakamide C, D, and E (known), the structures of gamakamide C and D were confirmed $as-{\small{L}}-Val-{\small{L}}-Met(SO)-{\small{L}}-NMe-Phe-{\small{L}}-Leu-{\small{D}}-Lys-{\small{L}}-Phe-$ and $-{\small{L}}-Ile-{\small{L}}-Met(SO)-{\small{L}}-NMe-Phe-{\small{L}}-Leu-{\small{D}}-Lys-{\small{L}}-Phe-$, respectively.

  • The radio signal in GNSS was intentionally designed with two frequencies in order to combat the dispersion error caused by trans-ionospheric propagation. By measuring the path delay independently at the two, widely spaced GPS frequencies, L1 & L2, the TEC along the path from satellite to receiver can be measured directly. The issue with dual frequency measurement of the ionosphere is the calibration of L1/L2 interfrequency biases. L1/L2 interfrequency biases are generated because physical electric signal paths of L1 and L2 circuits are different from each other for both satellites and receiver. Conventionally L1/L2 interfrequency bias is estimated and broadcasted by 2D ionospheric model. In this paper, we estimated IFB (interfrequency bias) by 2D & 3D ionospheric models including real time filter methods and compared the result of those and concluded the merit of 3D tomography model to recover the problem of 2D thin shell model. We confirmed our conclusion by experimental data.

  • The design of missile require extremely small warheads that must be highly efficient and lethal. The penetration characteristics of each penetrator and the total number of penetrators on the warhead are obvious key factors that influence warhead lethality. The design of the penetrator shape and size are directly related to the space and weight of the warhead. The design of the penetrator L/D was directly related to the space and weight of the warhead. L and D are the length and the diameter of the projectile, respectively. The AUTODYN-3D code was used to study the effect of penetrator penetration. The objective of numerical analysis was to determine the penetration characteristics of penetrator produced by hypervelocity impacts under different initial conditions such as initial velocity, obliquity angle and L/D of penetrator. The residual velocity and residual mass were decreased with increasing initial impact velocity under $L/D{\leq}4$.

Concentration of Serum and Urine Creatinine in Children and Adolescents

  • Creatinine is a general marker as a screening test for renal disease. This study was conducted to provide basic data about pediatric concentration for serum and urine creatinine. The data from the 2011 Korean National Health and Nutrition Examination Survey was used. Analysis was done on 488 Children and Adolescents (boys 278, girls 210) aged 10 to 19, the Jaffe reaction method was used to gather the data. The highest serum creatinine levels were found in boys aged 19 to 20 years, the mean being 0.97 mg/dL (min 0.81 mg/dL, max 1.14 mg/dL). The levels showed increase over those in the 10 year old group. The highest urine creatinine levels were found aged 19 to 20 years, and the mean was 222.68 mg/dL (min 133 mg/dL, max 324 mg/dL). In the case of girls, the highest serum creatinine levels were found with those aged between 18 and 19, the mean being 0.71 mg/dL (min 0.49 mg/dL, max 0.84 mg/dL). The levels showed increase over the 11 to 18 year old group. The highest urine creatinine levels were found aged 14 to 15 years, and the mean was 218.44 mg/dL (min 131 mg/dL, max 321 mg/dL). The mean difference in serum creatinine in all age groups was statistically significant except for those aged 10 to 14, that of urine creatinine in all age groups was not significant statistically except for those aged 12 to 13, 17 to 18. Therefore, it is suggested that reference values for children and adolescents should be divided into different groups according to gender, further studies are needed using complementing data of the pediatrics.

C'est un homme qui a deux patries, deux racines : une roumaine, une autre fran?aise. Sa vie est a ?t? marqu?e par ses publications des l'adolescence. Ils font l'objet de ce livre-portrait d'Eugen Ionescu, po?te et critique, ?crivain de langue roumaine.
