Phaffia rhodozyma 세포파쇄액으로부터 항산화제 Astaxanthin의 미셀 형성을 통한 가용화 및 추출 (Solubillzation and Extraction Of Antioxidant Astaxanthin by Micelle Formation from Phaffia rhodozyma Cell Homogenate)

  • Astaxanthin (3,3'-dihydroxy-${\beta}$, ${\beta}$-carotene-4-4'-dione), a natural pigment of pink to red color, is widely distributed in nature particularly in the skin layer of salmonoids and the crust of shrimp, lobster, etc. Recently, it was produced from the yeast culture of Phaffia rhodozyma. Because of its high thermal stability and antioxidant functionality, its applications can be extended into food, cosmetics, and pharmaceutical ingredient beyond the traditional feed additive. Because of its very high lipophilicity, astaxanthin has been extracted traditionally by strong organic solvents such as chloroform, petroleum ether, acetone, etc. In this study, we developed a surfactant-based solubillization system for astaxanthin, and used it to extract astaxanthin from disrupted yeast cells. Among Tween 20, Triton X-100 and SDS, Tween 20 was identified as the most suitable surfactant in terms of extraction capacity and safety. The ethylene oxide group of Tween 20 was identified as the most significant factor to increase the HLB value that determined the extraction capacity. The effects of micelle formation condition, such as the molar ratio of astaxanthin and Tween 20, pH, and ionic strength were also investigated. pH and ionic strength showed no significant effects. The optimal molar ratio between astaxanthin and Tween 20 was 1 : 12. Antioxidant activity of astaxanthin was higher than ${\beta}$-carotene and ${\alpha}$-tocopherol. Astaxanthin in the crude extract from the yeast cell was more resistant to air and/or light degradation than pure astaxanthin, probably because of the presence of other carotenoids and lipids.

Callus induction and high-efficiency plant regeneration via somatic embryogenesis in Papaver nudicaule L., an ornamental medicinal plant

  • We describe culture conditions for a high-efficiency in vitro regeneration system of Papaver nudicaule through somatic embryogenesis and secondary somatic embryogenesis. The embryogenic callus induction rate was highest when petiole explants were cultured on Murashige and Skoog (MS) medium containing 1.0 mg $1^{-1}$ ${\alpha}$-naphthaleneacetic acid (NAA) and 0.1 mg $1^{-1}$ 6-benzyladenine (BA) (36.7%). When transferred to plant growth regulator (PGR)-free medium, 430 somatic embryos formed asynchronously from 90 mg of embryogenic callus in each 100-ml flask. Early-stage somatic embryos were transferred to MS medium containing 1.0 mg $1^{-1}$ BA and 1.0 mg $1^{-1}$ NAA to germinate at high frequency (97.6%). One-third-strength MS medium with 1.0% sucrose and 1.0 mg $1^{-1}$ $GA_3$ had the highest frequency of plantlet conversion from somatic embryos (91.2%). Over 90% of regenerated plantlets were successfully acclimated in the greenhouse. Secondary somatic embryos were frequently induced directly when the excised hypocotyls of the primary somatic embryos were cultured on MS medium without PGRs. Sucrose concentration significantly affected the induction of secondary embryos. The highest induction rate (89.5) and number of secondary somatic embryos per explant (9.3) were obtained by 1% sucrose. Most secondary embryos (87.2-94.3%) developed into the cotyledonary stage on induction medium. All cotyledonary secondary embryos were converted into plantlets both in liquid and on semisolid 1/3-strength MS medium with 1.0% sucrose.

북방전복 종묘생산을 위한 농축 규조류 Caloneis schroederi와 Rhaphoneis sp.의 효과 (Effect of the concentrated-diatom of Caloneis schroederi and Rhaphoneis sp. for seedling production of Haliotis discus hannai)

  • Even though seedling production of Haliotis discus hannai has fully developed, the culture of benthic diatom as a live food for larvae is still a barrier to solve in commercial hatchery. The farmer depends on mixed microalgae which are non-selectively attached on the plate by flowing of natural seawater. The adequate diatom on the plate for the larvae in terms of quality and quantity is always significant bottleneck in the hatchery. In this study, two benthic diatom species, Caloneis schroederi and Rhaphoneis sp. were separately cultured in mass and inoculated to four tons' settlement tank of the larvae. And the larvae and the spats were cultured for nineteen and nine weeks, respectively. The result on seedling production of H. discus hannai with this method was compared to that of the farmer's traditional method as a control. With regard to variation of species composition of benthic diatom on the plate, C. schroederi and Rhaphoneis sp. were dominant for first three weeks after inoculation. But the diverse diatoms mainly, Navicula, Amphora, Cylindrotheca, Licmophora, Pleurosigma began to attache on the plate from the 4th week. The larvae attached 2.5 times more in C. schroederi tank than in the control tank. The final total biomass of the seeds in Rhaphoneis sp. tank was 3.2 times more than that of the control tank. The retared-spats in the seedling production also showed significantly higher growth and survival in the spat fed Rhaphoneis sp. or C. schroederi than those in the control group. We suggest that C. schroederi is proper for settlement of the larvae and Rhaphoneis sp. is appropriate for the growth of the larvae and spats in the commercial hatchery of H. discus hannai.

Current strategies using 3D organoids to establish in vitro maternal-embryonic interaction

  • Importance: The creation of robust maternal-embryonic interactions and implantation models is important for comprehending the early stages of embryonic development and reproductive disorders. Traditional two-dimensional (2D) cell culture systems often fail to accurately mimic the highly complex in vivo conditions. The employment of three-dimensional (3D) organoids has emerged as a promising strategy to overcome these limitations in recent years. The advancements in the field of organoid technology have opened new avenues for studying the physiology and diseases affecting female reproductive tract. Observations: This review summarizes the current strategies and advancements in the field of 3D organoids to establish maternal-embryonic interaction and implantation models for use in research and personalized medicine in assisted reproductive technology. The concepts of endometrial organoids, menstrual blood flow organoids, placental trophoblast organoids, stem cell-derived blastoids, and in vitro-generated embryo models are discussed in detail. We show the incorportaion of organoid systems and microfluidic technology to enhance tissue performance and precise management of the cellular surroundings. Conclusions and Relevance: This review provides insights into the future direction of modeling maternal-embryonic interaction research and its combination with other powerful technologies to interfere with this dialogue either by promoting or hindering it for improving fertility or methods for contraception, respectively. The merging of organoid systems with microfluidics facilitates the creation of sophisticated and functional organoid models, enhancing insights into organ development, disease mechanisms, and personalized medical investigations.

강릉지방의 오징어 식해 개발에 관한 연구 -1. 숙성온도 및 기간에 따른 성분변화- (The Development of Squid(Todarodes pacificus) Sik-hae in Kang-Nung District -1. The Effects of Fermentation Temperatures and Periods on the Properties of Squid Sik-hae-)

  • 본 연구는 강릉지역에 향토식품으로만 명맥을 유지하고 있는 오징어 식해를 제조하여 숙성온도 및 숙성기간을 달리하였을 경우의 여러가지 화학적인 변화를 실험하였으며 그 결과를 요약하면 다음과 같다. 숙성기간동안 일반성분 및 염도의 변화는 거의 없었으며, 단지 조단백은 숙성기간이 증가함에 따라 다소 감소하는 경향을 나타내었다. pH는 $5^{\circ}C$를 제외하고는 숙성기간이 증가함에 따라 감소하였으며 특히 $15^{\circ}C$$20^{\circ}C$ 숙성에서는 숙성초기부터 급격하게 감소되었으며, 젓산 생성은 pH 감소와 반비례하여 증가하였다. 총 arginine량은 숙성초기에는 다소 증가하는 경향을 보였으나 전반적으로 변화가 거의 없었다. 아미노태 질소량은 숙성 15일까지는 급격하게 증가하다가 그 후로는 서서히 증가하였다. TMAO 량은 $5^{\circ}C$$10^{\circ}C$에서는 서서히 감소하였으며 $15^{\circ}C$$20^{\circ}C$에서는 숙성초기에 급격하게 감소하였다. TMA량은 숙성초기에는 $20^{\circ}C$를 제외한 숙성온도에서는 서서히 증가하다가 20일 이후 감소하는 경향을 보였다. 핵산관련물질은 숙성 초기에만 AMP가 존재하였고 Inosine 및 hypoxanthine 이 핵산관련물질의 대부분을 차지하였다. 이상의 결과를 고려할 때 식해숙성 조건은 15일 정도가 최적이며 저온($5^{\circ}C$ 이하)에서는 조금 더 숙성기간이 길어진다고 생각된다.

공옥진 1인 창무극에서 무(舞)의 한국적 정서와 예술적 가치 분석 (In Gong Ok-jin's solo performance of Changmugeuk Analysis of the Korean sentiments and artistic values of the choreography)

  • 아직 학계에서는 공옥진의 춤 인생에 관해 춤의 예술적 가치에 대한 연구는 매우 부진하다. 그녀의 인생은 타고난 예인의 집안에서 나고 자란 소질과 풍부한 감성, 각설이패시절과 유랑예인집단에서의 걸립 공연 당시의 애환, 창무극 활동기에 겪었던 <병신춤>의 위기와 극복, <동물춤>으로의 전환, 그리고 투병생활을 하며 망가진 몸을 극복하며 <환자춤>이라는 공연을 하기까지 삶은 그리 관대하지 않았다. 하지만 그녀의 삶의 질곡은 늘 예술적 발로가 되었고, 예술은 그녀가 살아가기 위한 위안이자 큰 원동력으로 작용되었음을 알 수 있다. 예로부터 악가무(樂歌舞) 일체 형식이란 우리민족 고유 예술의 특성에 있어 무(舞)는 음악과 노래의 비중에 비해 다소 부차적인 몸짓 예술로서 존재한 것만은 사실이다. 공옥진의 창무극에서 해학적인 재담과 판소리에 이어 독창적인 춤사위는 몸짓 행위하나하나에 무엇보다 집중하고 있다. 무엇을 전달하고자 그리도 원초적인 애를 쓰는지, 말보다 앞선 제스처와 끊임없이 이어지는 연기에는 무엇보다 춤이라는 의미를 각인시키며 춤의 의미를 다시한번 생각하게 한다. 특히 공옥진의 <병신춤>이라 부르는 춤은 역사상 유례가 없는 것으로, 무엇이 한국인의 정서에 부합되어 몸짓언어로 발화되며 예술적 가치를 지니는지에 대해서는 우리의 원초적인 정서를 공감하지 않고서는 이해하기 어렵다. 따라서 본 연구는 공옥진 1인 창무극에서 무(舞)의 독창적인 형태를 고찰해보고 한국적 소재와 전통의 계승방식에 있어서 우리전통춤의 독자적인 정서가치를 나름대로 정리하는 수준에서 연구해 본 것이다.

재래감귤 팔삭의 과피 추출물이 LPS로 활성화 된 RAW264.7 대식세포에서 염증매개물질 억제에 미치는 효과 (Inhibition of LPS-induced Inflammatory Biomarkers by Fraction of Citrus hassaku pericarp through Suppression of NF-${\kappa}B$ Activation in RAW264.7 Cells)

  • Objectives : Citrus is the fruit that is readily available around us. Therefore, we investigated the anti-inflammatory effects of fraction isolated from the Citrus hassaku pericarp in RAW264.7 macrophage cells. Methods : The effects of fraction from Citrus hassaku pericarp on cell viability on RAW264.7 cells were measured by the MTT assay. The mRNA levels of iNOS and COX-2, its protein level by fraction of Citrus hassaku pericarp treatment in RAW264.7 macrophage cells were investigated by RT-PCR and immunoblots. Nitrite accumulation in the culture was measured colorimetrically by the Griess reaction using a Griess reagent. The amount of IL-6 and TNF-${\alpha}$ production was determined using an enzyme-linked immunosorbent assay (ELISA) kit. Results : The results indicated that the fraction of Citrus hassaku pericarp concentration highly suppressed lipopolysaccharide (LPS)-induced nitric oxide (NO) and IL-6 productions without a cytotoxic effect on RAW264.7 cells. fraction of Citrus hassaku pericarp inhibited the expressions of LPS-induced iNOS and COX-2 protein and their mRNA in a dose-dependent manner. Particularly, fraction of Citrus hassaku pericarp suppressed the level of nuclear factor-${\kappa}B$ (NF-${\kappa}B$) activity, which was linked with the suppression of LPS-induced phosphorylation of p65 at serine 276 and p65 translocation into nuclei, but not MAPK signaling. In addition, treatment with fraction of Citrus hassaku pericarp inhibited the production of IL-6 and TNF-${\alpha}$ in LPS-stimulated RAW264.7 cells. Conclusion : Our results indicate that fraction of Citrus hassaku pericarp potentially inhibits the biomarkers related to inflammation through the blocking of NF-${\kappa}B$ p65 activation, and it may be a potential therapeutic candidate for the treatment of inflammatory diseases.

국내에서 분리된 황국균을 활용한 된장 제조 및 특성 분석 (Manufacturing and Quality Characteristics of the Doenjang made with Aspergillus oryzae Strains Isolated in Korea)

  • 국내 전통발효식품으로부터 protease 활성이 높은 Aspergillus oryzae SCF-6, SCF-37, 그리고 JJSF-1 균주를 선발하여 된장을 각각 제조하고 60일간 숙성하면서 품질을 비교 평가하였다. 60일 숙성 동안 수분 염도 및 pH는 시료 간에 큰 차이가 없었다. 된장의 숙성지표가 되는 아미노산성 질소의 함량은 숙성 경과에 따라 증가하여 60일 숙성 후 SCF-6을 활용하여 제조한 된장이 971.6 mg%로 다른 시료보다 높았으며, 유리아미노산 함량도 상대적으로 높은 8,064.9 mg%이었다. SCF-6과 37로 제조한 된장은 60일 숙성 후 GABA 함량이 각각 61.3과 53.7 mg%로 측정되었다. 색도는 숙성 경과에 따라 명도와 황색도는 점차 감소한 반면 적색도는 점차 증가하였다. 아플라톡신은 모든 된장에서 불검출 되었고, 바이오제닉 아민 중 histamine 함량은 2.55-5.60 mg/kg 그리고 tyramine은 3.70-5.87 mg/kg으로 매우 낮게 측정되었다. 본 결과로 A. oryzae SCF-6은 장류 제조의 종균으로 적합한 것으로 판단된다.

미생물분석법을 이용하여 한국인이 즐겨 섭취하는 일부 해조류 및 어패류와 그 가공식품의 비타민 $B_{12}$ 함량 분석 (Vitamin $B_{12}$ Content Using Modified Microbioassay in Some Korean Popular Seaweeds, Fish, Shellfish and Its Products)

  • There is a limitation to estimate vitamin $B_{12}$ intake due to a lack of data on vitamin $B_{12}$ content in many Korean foods. In this study, vitamin $B_{12}$ content was determined in some seaweeds, fish, and shellfish and their product that are consumed in Korea using a modified microbioassay with Lactobacillus delbruecki ATCC 7830. Dried laver and dried seasoned and toasted laver contained very high levels of vitamin $B_{12}$ (66.8 and $55.2-71.3\;{\mu}g$/100 g, respectively. Sea lettuce and seaweed fulvescene also contained high vitamin $B_{12}$ content of 5.47-9.41 and $6.46-7.20\;{\mu}g$/100 g, respectively, whereas sea mustard and sea tangle contained low levels of vitamin $B_{12}$; vitamin $B_{12}$ was not detected in seaweed fusifome. Pacific saury, trout, sea-bass, or squid contained 12.01, 2.00, 0.49 and $2.33\;{\mu}g$ vitamin $B_{12}$/100 g, respectively. Ochellatus octopus, and naked sand lance contained 0.72-1.43 and $3.68\;{\mu}g$ vitamin $B_{12}$/100 g, respectively. Dried Alaska pollack con-tained $0.19-2.64\;{\mu}g$ vitamin $B_{12}$/100 g. Shellfish such as little neck clam and small ark shellfish contained high levels of vitamin $B_{12}$ of $30.5-40.5\;{\mu}g$/100 g, and mussel and abalone contained 17.71 and $7.82\;{\mu}g$/100 g, respectively. Of unique Korean traditional fermented seafood products, salt-fermented products of squid ($2.91\;{\mu}g$/100 g), clams ($34.31\;{\mu}g$/100 g), Ala-ska pollack roe ($9.98-12.02\;{\mu}g$/100 g), hairtail guts ($4.58\;{\mu}g$/100 g) or small shrimp ($0.58-1.55\;{\mu}g$/100 g), and fish sauce from anchovies ($1.52-1.78\;{\mu}g$/100 mL), sand eel ($0.22-0.24\;{\mu}g$/100 mL) or small shrimp ($0.19-0.78\;{\mu}g$/100 mL) were analyzed. A few commercial brands of flying fish roe ($0.73-1.73\;{\mu}g$/100 g), canned tuna ($0.40\;{\mu}g$/100 g), and fried fish paste ($0.25-0.69\;{\mu}g$/100 g) were also analyzed. In conclusion, vitamin $B_{12}$ content in these foods, chosen considering the Korean food culture, should contribute to improve the present vitamin $B_{12}$ food database. It may be helpful to estimate vitamin $B_{12}$ intake more correctly than before, and provide additional information for dietary education related to vitamin $B_{12}$ and meal management.

전통발효식품 유래 Enterococcus faecium JK29에 의한 γ-aminobutyric acid의 생산 최적화 (Optimization of γ-Aminobutyric Acid Production by Enterococcus faecium JK29 Isolated from a Traditional Fermented Foods)

  • ${\gamma}$-Aminobutyric acid(GABA)를 생산하는 희귀 젖산균을 분리하기 위하여 전통발효식품으로부터 총 147개의 젖산균을 확보한 후 1% 글루탐산 나트륨(L-monosodium glutamate, MSG)를 사용하여 GABA를 생산하는 23개의 균주를 1차 분리하였다. 2차 분리를 위하여 글루탐산 탈탄산효소(glutamate decarboxylase)와 16S rRNA 유전자의 염기서열 분석을 통해 기본 MRS 배지에서 48시간 배양 후 1.56 mM의 GABA를 생산하는 Enterococcus faecium JK29를 최종 분리하였다. E. faecium JK29에 의한 GABA의 생산을 향상시키기 위하여 배양 조건을 최적화하였으며 그 결과 0.5% 자당(sucrose), 2% 효모 추출물(yeast extract), 0.5% 글루탐산 나트륨이 포함된 최적화 MRS 배지를 개발하였다. 최적화 MRS 배지를 활용하여 $30^{\circ}C$, pH 7.5에서 48시간 배양을 한 결과 E. faecium JK29이 14.86 mM의 GABA를 생산하는 것을 확인하였다.
