Survey on mycoplasmal pneumonia of swine in Youngnam area and antimicrobial susceptibility of Mycoplasma hyopneumoniae isolated from Slaughter pigs (영남지방 도축돈의 Mycoplasma 폐렴조사 및 분리균에 대한 약제 감수성)

  • The present study was carried out to investigate the prevalence of mycoplasmal pneumonia of slaugter pigs in Youngnam area during the period from 1995 to 1997. The prevalence and pathomorphology of gross lung lesions were studied from 682 slaughter pigs in 8 swine herds. Gross lesions of pneumonia were recorded in the lungs of 442(64.8%), from 367 out of them(83.0%) were diagnosed as mycoplasmal pneumonia. Microbiological examination was performed with 197 lungs with gross lesions of mycoplasmal pneumonia of slaughter pigs from 8 different swine herds. M hyopneumoniae, P multocida, A pleuropneumoniae, Streptococcus spp, Corynebacterium spp, and H parasuis were detected in 24. 4%, 48.2%, 2.5%, 11.2%, 3.6%, and 1.0% of the pneumonic lungs, respectively. A total of 48 strains of M hyopneumoniae was investigated for their in vitro susceptibility to 8 antibiotics. Among the drugs tested, lincomycin, oxytetracycline, tiamulin and tylosin showed the high activity in minimal inhibitory concentration(MIC) of $0.04{\sim}5{\mu}g/ml$ while erythromycin showed low activity in MIC values($1.25{\sim}{\geq}40{\mu}g/ml$).

Epidemiological characteristics of classical swine fever outbreak at Jeonbuk area in 2003 (전북지역에서 발생한 돼지콜레라의 역학적 특성)

  • Classical swine fever (CSF) was confirmed in 19 herds in Jeunbuk provence (Iksan, Gimje, Wanju, Buan, and Jangsu) in Korea between March and May, 2003 and 10,263 pigs were slaughtered. Pigs contacted with CSF virus in primary outbreak farm show fever, reduced appetite, arched back and chill in company with sever respirative sign and then most infected farms also were observed to fever, reduced appetite, sudden death, and leukopenia (101 pigs). In order to detecting infectious pig with CSF virus, A total of 555 pigs were inspected in 65 herds and blood samples were collected and serological test (ELISA), antigen ELISA, and reverse transcription-polymerase chain reaction (RT-PCR) had been done. Positive rate were $74\%$ (410 pigs) in antibody ELISA, $2\%$ (11 pigs) in antigen ELISA and $33\%$ (182 pigs) in RT-PCR, respectively. As shown that the RT-PCR was useful than the ELISA for determining CSF virus in blood, meat, and other organs.

  • Porcine respiratory coronavirus (PRCV) is antigenically related to transmissible gastroenteritis virus (TGEV). Differential serological diagnosis between PRCV and TGEV infection is not possible with the classical sero-neutralization test. Infection with PRCV or TGEV induces antibodies which neutralize both viruses to the same titer. However, the enzyme-linked immunosorbent assay (ELISA) and polymerase chain reaction (PCR) can differentiate between PRCV and TGEV infection. This study was carried out to investigate the prevalence of PRCV infection of swine in Gyeongnam province. A total of 391 serum samples from 37 herds in Gyeongnam were examined for antibody to PRCV using blocking ELISA. All serum samples were collected from 130- to 150-day-old pigs between August and December 2006. By ELISA, 182 out of 391 sera tested (46.5%) and 29 out of 37 sample herds (78.4%) were positive against PRCV. Our data suggested that seropositive herds for PRCV are distributed diffusely throughout Gyeongnam. The PCR methods were established to diagnose PRCV spike protein (S) gene. PCR were conducted to identify the PRCV genome against 150 pigs in PRCV antibody positive herds.

Seroprevalence of Mycoplasma hyopneumoniae infection in Korean swine herds (우리나라 양돈장에서 Mycoplasma hyopneumoniae 유병율 조사)

  • Serum samples of 1,175 pigs from 148 Korean swine farms not using Mycoplasma hyopneumoniae (M. hyo) vaccines were collected for seroepidemiological study of M. hyo infection by indirect ELISA method. Informations of each farm were provided about province where the farm was located and season when blood samples were collected. Then, the selected farms were divided into farm units which had 5 serum samples according to production stages : sow, suckling piglet (<30 days old), nursery pig (30-70 days old), and growing pig (>70 days old). Seroprevalence of M. hyo infection according to production stages, province, and season was investigated by using ELISA-positve rate of the selected samples for each study. This study showed that 85.34% (78.94-91.78%, 95% CI) of farms were positive to M. hyo infection and 34.81% (32.09-37.53%, 95% CI) among pigs were sero-positive to M. hyo infection in Korean swine farms. In the study of seroprevalence by production stage, most farms had sows and growing pigs which were sero-positive to M. hyo infection (sow: 83.05%, growing pigs: 87.72%) and most pigs seemed to be naturally infected by M. hyo at 8-10 weeks of age. Also, M. hyo infection showed seasonal pattern that most pigs were infected in late fall to early winter. However, in the study of seroprevalence by province, there was no significant correlation between province and M. hyo sero-positive rate.

Risk assessment of the potential for a classical swine fever outbreak in Korea based on a herd immunity

  • The risk of an outbreak of classical swine fever (CSF) was evaluated in relation to the vaccination and seroprevalence of antibodies. Blood samples were collected from 60 piggeries throughout Korea and information regarding CSF vaccination habits was also obtained via in-person interviews with pig farmers. The results of the survey indicated that 51 out of 60 farms were regularly performing CSF vaccinations in reproductive herds. Farmers preferred to vaccinate their reproductive pigs before weaning (41 farms) than on (9 farms) or after weaning (1 farm). In growing herds, however, double vaccinations as recommended were implemented for only 40 farms (66.7%) and vaccine schedules were identified as being frequently ignored. Moreover, many farms (18/40) were found to vaccinate earlier or later than the recommended time. According to antibody seroprevalence levels, only 36 farms (60%) were considered to be safe regarding potential risk for a CSF outbreak. Among the remaining pig farms, 6 were at low-risk (10%), 12 were at medium-risk (20%), and another 6 were at high-risk (10%). Antibody levels were found to be consistent with vaccination status obtained from personal interview surveys. Our findings demonstrate the importance of vaccinations regarding the prevention of a CSF outbreak, suggesting that vaccinations must be promoted toward pig raisers and practitioners.

Detection of Lawsonia intracellularis in swine feces by polymerase chain reaction (돼지분변에서 PCR에 의한 Lawsonia intracellularis 검색)

  • Swine proliferative enteritis(SPE) caused by inかsoma intracellularis is a common enteric disaese of grower and finisher pig. Swine affected with SPE show variable clinical signs including diarrhea, weight loss, aberrant growth and death. The characteristic lesion of ileitis at necropsy is marked thickening of the last section of the small intestine. The inner lining of the thickened intestine proliferates almost like a cancer and curved rod bacteria(L intracellularis) are always seen inside the intestinal wall. Infected swine shed the organism in the feces. Isolation and growth of pure L intracellularis in vitro requires a suitable cell culture. This procedure is difficult and not a practical means of diagnosis, thus the polymerase chain reaction(PCR) test of feces can be used to determine whether a pig is shedding the infective organism. A sensitive assay based on amplification of a 319bp DffA fragment of the L intracellularis of Swine proliferative enteritis was attempted for the detection of the organism in the 62 feces of swine. L intracellularis was identified on three herds and detected in 6 fecal samples, representing a infection rate of 9.7%. The PCR was very sensitive and specific on the individual level. The PCR technique could be very useful for the diagnosis of this disease.

  • The outbreak of hepatosis dietetica from 2 different herds of pigs was diagnosed for the first time in Korea. The livers from 5 affected pigs showed yellow atrophy, necrotic foci and ecchymotic hemorrhages anatomically, and hemorrhagic necrotic hepatitis histopathologically.

Seroprevalence of swine influenza and porcine reproductive and respiratory syndrome in Korea

  • A total of 501 serum samples were selected from blood samples that were submitted to Department of Veterinary Pathology, Kangwon National University from all provinces in Korea from September 2001 to August 2002. Their sera were examined for antibodies to swine influenza virus subtype H1N1 (SlV H1N1) and porcine repro-ductive and respiratory syndrome virus (PRRSV) according to the age of pig, season, and herd size using enzyme-linked immunosorbent assay. The seroprevalence of SIV H1N1, PRRSV, and dual infection were 39.12%, 61.48%, and 25.95%, respectively. The seroprevalence of SIV H1N1 according to herd size was not significant differences (p>0.05). The results showed that the PRRSV infection spread widely in swine herds throughout the country.

Survey on the red internal organs gross lesions of slaughtered pigs in Jeonbuk (전북지역 도축돈 적내장 육안병변 조사)

  • Respiratory diseases are common in swine industry and have great economic importance. Respiratory disorders cause substantial losses to the swine industry. Losses associated with respiratory disease vary considerably between herds and seasons. In a survey was carry out to investigate the lesion of red internal organs in slaughtered pigs and provided assistant data which are useful for each farm. From november 2012 to december 2013, 1,680 pigs out of 84 farms were sampled in Jeonbuk province. Gross lesions such as swine enzootic pneumonia (SEP), pleuritis, pleuropneumonia, pericarditis, liver milk spot were examined for the pigs. Overall prevalence of SEP was 55.7%. According to season, the incidence occurred higher in fall than winter, spring, and summer. The mean SEP score was 0.91, the highest incidence occurred in fall. The prevalence of pleuropneumonia, pleuritis, pericarditis, and milk spot was 36.4%, 49.7%, 2.3%, and 8.8%, respectively. The positive rate of PRRS and PCV2 was 2.9% and 70.0% by PCR analysis.

Seroprevalence of porcine proliferative enteropathy before initiating vaccine marketing in Korea

  • Proliferative enteropathy caused by Lawsonia intracellularis is one of the most common enteric diseases in pigs. The objective of this study was to determine the prevalence of serum antibodies against L. intracellularis in the general swine population of Korea from 2005 to 2008. In total, 8,008 swine serum samples obtained from 1,001 herds were tested. The samples were analyzed with an immunoperoxidase monolayer assay to detect anti-L. intracellularis antibodies. The overall 4-year average true prevalence was 40.0% (CI: 39.4 - 40.6%) at the individual animal level and 71.9% (CI: 70.3-73.4%) at the herd level.
