The Effects of Korean Safflower(Carthamus tinctorious L.) Seed Powder Supplementation Diet on Bone Metabolism Indices in Rats during the Recovery of Rib Fracture (한국산 홍화씨분말 보충식이의 급여가 골절된 흰쥐의 골대사지표에 미치는 영향)

  • This study was carried out to investigate the effects of Korean safflower(Carthamus. tinctorious L.) seed powder supplementation on bone metabolism during the recovery of rib-fracture induced by surgical operation in rats. Male Sprague-Dawley rats of 10 weeks old, weighing 370$\pm$5g, were divided into two groups after arrival : the control group(C group, AIN-76 semifurified diet) and safflower seed group(S group, AIN-76 semifurified diet+10% safflower seed powder). They were fed experimental diets for 12 days before the rib fracture operation and for 30 more days after the operation. A number 9 rib was fractured surgically and sham-operation was also performed. After the rib-fracture operation, rats were sacrificed on the 8th, 11th, 16th, 21st, and 30th day, respectively. The body weights were decreased on the 8th day after operation in both groups but recovered gradually thereafter. The fractured sections of the S group were more strongly adhered and more rapidly repaired than those of C group. Effects of safflower seed powder supplementation on indices of bone metabolism during the repairing of rib-fracture can be summarized as following. There were no differences between C and S group. In the levels of PTH and calcitonin, which regulate the concentration of plasma calcium. However, PTH level on the 8th day was significantly higher than that on the 16th day in control group. There were also no differences between groups or within groups in the levels of calcitonin. Osteocalcin levels, one of the bone-formation indices, were similar in two groups. Activities of total-ALP in plasma were highest at the 8th day in both groups and decreased thereafter, except on the 30th day in control group. The activities of bone-specific ALP that is directly related to bone calcification, showed higher levels in the S group than in the C group throughout the experimental period. They were significantly increased in the S group on the 11th, 16th, and 21st days. In comparisons of plasma calcium and phosphate levels, Ca levels were tended to be lower in the S group than in the C group and P levels showed the opposite trend. It seems that the fractured ribs were more rapidly repaired in rats supplemented with safflower seed powder than in control rats, possibly due to stimulation of bone calcification from increased activity of bone-specific ALP.   • Much attention has been given to the role of safflower (Carthamus tinctorius L.) seed in preventing and treating osteroporosis recently. Although numerous studies were done on effects of safflower oil, no attention was given to the role of dietary fiber of safflower seeds. This study was aimed at investigating physiological significance of defatted safflower seed as a source of dietary fiber. Sprague Dawley female rats were assigned one of 5 groups: ovariectomized control (Ovx-control) group and ovariectomized rats 15% (Ovx-ss15) and 30% defatted safflower seed (Ovx-SS30), sham-operated (Sham) group and a normal group. Cholesterol was supplemented to all diets at 0.5% except the normal diet. Ovariectomized rats were pair-fed isocalorically to the Sham group. Ovariectomy caused heavier body weight, but feeding 30% safflower seed brought back to the level of Sham group. Activities of disaccharidases of jejunal mucosa were significantly lowered in Ovx-control group compared to those of Sham, and supplementation of safflower seed tended to increase the activities. Fecal weight of Ovx-SS15 and Ovx-SS30 were almost twice as those of Ovx-control Sham. Propionic acid and butyric acid concentrations per g of feces of Ovx-SS15 and daily excretion of these fatty acids were significantly increased as compared to those of Sham and Ovx-control. In concentration defatted safflower seed supplementation significantly increased fecal bulk and short chain fatty acid production in large intestine of rats.

  • A study was carried out to investigate the effects of safflower seed powder on the improvement of lipid metabolism in high fat and high cholesterol fed rats. Male Sprague Dawley rats of 10 weeks old, weighing 325$\pm$5g, were divided into two groups; the control group(C group, high fat(10% lard) and high cholesterol(1% cholesterol)) and safflower seed group(S group, 10% safflower seed powder), they were fed experimental diets for 6 weeks. Food intake, body weight gains and organ weight had little differences between the groups. Concentration of lipoprotein in serum was remarkably lower in S group than in C group. Serum cholesterol levels were significantly lower in S group(72.94$\pm$4.08 mg/dl) than in C group(89.41$\pm$4.19mg/dl). The level of serum HDL cholesterol was higher in S group than in C group. The level of serum LDL C was significantly lower in S group than in C group. The ratio of HDL cholesterol to total cholesterol were higher in the S group than in the C group, too. The value of atherogenic index(AI) was determined to be low in S group. The content of liver triglyceride and cholesterol in the S group was lower than that of C group. ACAT activities which involves in cholesterol esterification in liver, was not significantly different between two groups.

  • The therapeutic effect of safflower seed powder on experimental osteoporosis in the rat induced by ovariectomy was evaluated. Thirty Sprague-Dawley rats were ovariectomized at the age of 12 weeks. Seven weeks postovariectomy, rats were divided into two groups: control and safflower seed powder treated group. Five animals from each group were sacrificed at the following time points: 1, 3, and 5 weeks. Scanning electron microscopic observation and morphometric analysis of the tibiae epiphysis showed that the administration of safflower seed powder significantly prevented reduction of cortical bone width and bone volume compared with the control group. In conclusion, safflower seed powder contains something that prevent bone loss due to estrogen deficiency, and was effective in preventing the osteoporotic decrease of bone mass.

Effect of Addition of Various Mesh Sifted Powders from Safflower Seed on Quality Characteristic of Yangeng (입도별 홍화씨 분말 첨가가 양갱의 품질에 미치는 영향)

  • Safflower Yangengs were prepared with composite dried powder of small red bean(Phaseolus radiatus L.) containing various ratios of safflower(Carthamus tinctorius L.) seed powder sifted through 20, 35, 45 and 60 mesh size and kinds of mixed water, their cooking characteristics were evaluated. Water content and water activity of cooked products were increased as the content and sieve mesh number of safflower seed powder increasing from 5%, 20 mesh to 20%, 60mesh, respectively. Color values of yangeng were increased in green tea extract mixed water. Rheological properties of yangengs were measured by compression test with texture analyzer, as results, hardness and fracturability increased that were shown in high content and high mesh number sifted safflower seed powder, but adhesiveness and springiness decreased, respectively. From the sensory evaluation test for yangeng, sensory scores were good scores in more mesh number sifted powder addition, especially overall acceptance, texture and fracturability. The 45mesh and 15% powder added yangeng was noted as having high sensory scores and preferable acceptability in sensory evaluation.

  • To clarify the efficient application method of sulfur in safflower, we investigated the growth and seed yield as affected by ammonium sulfate (AS) and sulfur powder. AS was applied to the soil with four levels of 0, 4, 8, and 12 kg/10a and was applied by foliar application with 2 kg/10a as sulfur content compared with sulfur powder 20 kg/10a. By the application of sulfur fertilizer, plant height, stem diameter, and weight of stem and leaves tended to be greater than control. AS was more efficient than sulfur powder in growth of safflower. Sulfur application showed positive effect on yield components and seed yield was increased by $4-10\%$ compared to control. In application effects, AS and foliar application were more efficient than sulfur powder and soil application, respectively.

Effects of Safflower Seeds on the Serum Levels of Insulin-like Growth Factors, Insulin-like Growth Factor Binding Protein-3 and BALP in Osteoporosis Induced-ovariectomized Rats (흰쥐의 난소제거로 유발한 골다공증에 대한 홍화씨의 IGFs, IGF binding protein-3 그리고 BALP에 대한 혈청내 효과)

  • This study was carried out to investigate the effects of the Korean Safflower (Carthamus inctorius L) seed powder on serum level of hormones and trabecula area during the recovery from osteoporosis induced ovariectomized rats. Four month-old rats were ovariectomized (OVX), remained untreated for 8 weeks, and were subsequently administered safflower seed (0.03 g/kg) every other day 30 for days. We examined the effects of treated safflower seed every 10 days on ovariectomy-related changes in Insulin-like Growth Factors, Insulin-like Growth Factor binding protein-3 (IGFBP-3), Estrogen, Bone-specific alkaline phosphotase, Calcium, and Phospotase in the serum, and also histomorphology of the proximal fibula metaphysis and femur/body weight rate. Ten and 20 days after safflower seed treatment in OVX rats, serum levels of IGF-I, -II and IGFBP-3 were not different from the Sham and OVX groups. In 30 days, serum levels of IGF-I,-II and IGFBP-3 were higher after safflower seed treatment in OVX rats as compared to the other two groups (p<0.05). Bone alkaline phosphatase levels were increased through safflower seed treatment in OVX rats compared to the other two groups in 30 days. There were no differences between OVX and safflower seed treated OVX rats in serum levels of estrogen and femur/body weight rate, but estrogen levels for the sham group were higher than for the other two groups. The safflower seed is increased to serum levels of IGFs, IGFBP-3 and BALP of osteoporosis induced by ovariectomized rats. Thus, we conclude that the safflower seed is a possible role for improvement of osteoporosis induced-ovariectomized rats.

Effects of Safflower Seed Powder on the Quality Characteristics of Kimchi (홍화씨 분말 첨가가 김치의 품질에 미치는 영향)

  • This study was carried out to investigate the effect of safflower powder on the characteristics of kimchi. Quality indexes were pH, titratable acidity, reducing sugar content, microbial counts, and sensory evaluation. Safflower powder enhanced the decrease in pH and increase in titratable acidity during fermentation at l0$^{\circ}C$, and the changes were more conspicuous until 15 days. Stabilized levels in pH was attained, but steady increase in titratable acidity was shown after that time. Microbial loads of total and lactic acid bacteria showed a faster stationary phase for kimchi samples with safflower powder than control. Control was better scores in sensory evaluation, but there were no significant differences in aroma and taste except sample with 3% safflower powder.

Physicochemical Characteristics of Ground Pork with Safflower Seed Powder as an Animal Fat Replacer (동물성지방 대체제로서 홍화씨(Carthamus tinctorius L.)를 첨가한 분쇄돈육의 이화학적 품질특성)

  • This study was carried out to investigate the effect of the addition of safflower seed (Carthamus tinctorius L.) on the physicochemical properties of ground pork as an animal fat replacer. Three types of ground pork were evaluated: 20% pork fat added (control), 10% pork fat and 10% safflower seed powder added (10% SS), and 20% safflower seed powder added (20% SS). The moisture, protein, and ash contents were highest in 20% SS, and the fat content was highest in the control (p<0.05). The cooking yield, moisture retention, fat retention, and water-holding capacity were highest in 20% SS, and the control showed a reduction in the diameter (p<0.05). The external and internal L-, a-, and b-values of the control were higher than those of the 10% SS and the 20% SS (p<0.05). The cholesterol content of the control, the 10% SS, and the 20% SS was 50.85, 21.77, and 17.91 mg/100 g, respectively, and that of the 20% SS was lowest among the samples (p<0.05). The linoleic acid content of the control, the 10% SS, and the 20% SS was 28.68%, 41.04%, and 54.26%, respectively. The total unsaturated fatty acid content of the control, the 10% SS, and the 20% SS was 50.53%, 55.76%, and 64.93%, respectively. The linoleic acid and the total unsaturated fatty acid content were highest in the 20% SS (p<0.05). There was no significant difference in amino acid composition.

Sensory and Mechanical Characteristics of Yukwa Added Safflower Seed Powder (홍화종실분말 첨가 유과의 관능적.기계적 특성)

  • The quality of Yukwa added with safflower seed powder (SSP) was investigated using chemical, sensory and mechanical properties, The result of chemical properties showed that degree of expansion of Yukwa were decreased as the amount of SSP increased, while fat absorption increased. Yukwa with 1% SSP was the best in appearance, taste and overall preference of acceptability. Lightness decreased by the addition of SSP, while redness and yellowness increased. Texture measurement showed that hardness was the lowest in the Yukwa with 5% SSP, and cohesiveness was the highest in the Yukwa with 1% SSP. Hardness and brittleness of mechanical properties were positively correlated with the acceptability. The degree of expansion was positively correlated with cohesiveness and springiness. Fat absorption and moisture content were positively correlated with gumminess in the mechanical properties. Yukwa with 1% SSP showed a good overall preference.
