Effect of Water Extract of Korean White and Red Ginseng on Lipolytic Action of Toxohormone-L from Cancerous Ascites Fluid (고려인삼의 수용성 엑기스 성분이 암독소 호르몬-L의 체지방 분해작용에 미치는 영향)

  • This study was carried out to investigate the effect of water extracts from 4-, 5- and 6-year old red white ginseng roots on the inhibition of lipolysis in fat cells induced by toxohormone-L which has been known as lipolytic and anorexigenic factors. Toxohormone-L was obtained by partial purification of the ascites fluid from mice which had been inoculated with sarcoma-180. Each water extract of ginseng was effective in vitro against the inhibition of lipolysis induced by the toxohormone-L at the concentrations over 10~100 $\mu\textrm{g}$/ml. At the concentration of 1,000 $\mu\textrm{g}$/ml, the inhibition ratio of lipolysis by the water extracts of 4-, 5- and 6-year old white ginseng roots were 56.3, 59.7 and 59.4%, and those of red ginseng roots were 78.6, 79.1 and 82.5%, respectively, indicating that the 6-year old red ginseng was the most effective in the inhibition of the lipolysis. The total inhibitory activity per gram of ginseng sample was higher in the 4-year and 5-year old white ginseng roots at the concentrations of 100 $\mu\textrm{g}$/ml and 1,000 $\mu\textrm{g}$/ml, respectively, while for the red ginseng it was higher in the 6-year old ginseng than other ages regardless of the reaction concentration. Only once injection of the ginseng extracts(300 mg/ml in saline solution) with 10 ${mu}ell$/g of weight to the mice bearing sarcoma-180 was not effective against the extension of their life spans. On the contrary, the life spans of the mice were rather shortened by the injection of 5-year old white ginseng and 6-year old red ginseng, as compared to that of control.

Effect of Preheating Condition of Raw Ginseng on the Yield and Physical Property of Korean Red Ginseng Extract (수삼의 열처리 조건에 의한 홍삼 엑스의 수율 및 물리성 변화)

  • In order to enhance the yield and physical property of Korean red ginseng extract, preheating stage was added to the inception of red ginseng manufacturing process and its effect was investigated. Preheating of raw ginseng at $70^{\circ}C$ for 2 hour followed by steaming for 2 hours increased the yield of water and 60% alcohol extract most effectively. Those yields were the highest at the same condition as above except preheating time shortened to 1 hour at $70^{\circ}C$. Steaming time had little effect on the yield of water and 60% alcohol extract. The content of starch in red ginseng was reduced effectively by preheating of raw ginseng at $70^{\circ}C$. The brown color intensity of red ginseng increased in proportion to preheating temperature in the range of $50-70^{\circ}C$. However, there was no increase in the color intensity at $80^{\circ}C$.

The Effect of Korean Red Ginseng Water-extract on Aging-related Changes in the Xenobiotic Metabolizing Enzyme System in the Liver of Rats (홍삼의 물추출물이 노화에 따른 흰쥐 간의 이물질대사 효소계에 미치는 영향)

  • The present study was designed to investigate aging-related effects on the activities of xenobiotic metabolizing enzymes of rat liver by dietary supplementation of Korean red ginseng water-extract. Rat did not show any discernible signs of the rejection symptoms, and blood GOT and GPT levels were not influenced by ginseng water extracts, Cytochrome 450 levels and NADPH cytochrome P45O reductase, p-450 dependent ethoxycoumarin O-deethylase, and benzphetamine N-demethylase activities were decreased with aging, however, these phase I system enzymes activities in the ginseng group of24 months were well maintained compared with normal group. But, Levels of cytochrome bs and NADH-cytochrome b$_5$ reductase activities were also decreased with aging and were not found a clear difference between two groups. Glutathione-s-transferase activity, phase II enzyme system, in liver cytosols was also decreased in old ages, but the degree of decrease was higher in normal group than in giuseng supplemented group. These results indicate that long-term supplementation of red ginseng water extracts from weaning to 24 months do not show any side effects to rats, and retard age-related deteriorations of xenobiotic metabolizing enzymes activities in old ages.

Quality Characteristics of White pan bread with Led Ginseng powder (홍삼 분말을 첨가한 식빵의 품질특성)

  • This study investigated properties of bread utilizing extracts of ginseng powder. Ginseng powder bread was baked. Addition of 1~3% of ginseng powder extract to wheat flour was carried out. Rheological properties, dough pH, dough volume, bread volume, water absorption capacity, baking loss, bread color, bread texture, and sensory evaluation were tested to examine properties of bread baked with extracts of ginseng powder. The results are as follows. The dough pH decreased gradually with increasing ginseng powder extract concentration. The bread volume, baking loss, and bread weight increased with increasing use of ginseng powder extract, and springiness and cohesiveness increased as red ginseng powder additive concentration increased. Lightness of the L decreased while a and b increased. In the sensory evaluation, ginseng powder with 1% ginseng powder extract was evaluated as the best for taste, texture, flavor, and overall acceptability.

  • The purpose of this study is to investigate whether the intracellular hydrogen peroxide productions of mouse macrophage RAW 264.7 are modulated by Red Ginseng-Ejung-tang water extract (ER) and White Ginseng-Ejung-tang water extract (EG). Red Ginseng-Ejung-tang were composed of Red Ginseng, Atractylodes rhizome white, Zingiberis Rhizoma Siccus, and Glycyrrhizae Radix. White Ginseng-Ejung-tang were composed of White Ginseng, Atractylodes rhizome white, Zingiberis Rhizoma Siccus, and Glycyrrhizae Radix. The intracellular hydrogen peroxide productions were measured by dihydrorhodamine 123 assay with spectrofluorometer (excitation 485 nm; emission 535 nm). For 4, 20, 24, 44, 48, 68, and 72 h incubation, ER significantly increased hydrogen peroxide productions of RAW 264.7 at the concentration of 25, 50, 100, and $200{\mu}g/mL$ (P <0.05). EG for 4, 20, 24, 44, and 48 h incubation significantly increased hydrogen peroxide productions of RAW 264.7 at the concentration of 25, 50, 100, and $200{\mu}g/mL$ (P <0.05). For 68 and 72 h incubation, EG at the concentration of 50, 100, and $200{\mu}g/mL$ significantly increased hydrogen peroxide productions in RAW 264.7 (P <0.05). These results suggest that ER and EG have the immune-enhancing properties related with their increasing effects on the intracellular hydrogen peroxide production of macrophage.

Moisture Adsorption Preventative Effect of Fermented Red Ginseng Extract Spherical Granules by Using Hydrophobic Compounds (홍삼의 지용성 성분을 이용한 발효홍삼 농축액 알갱이의 흡습방지 효과)

  • For prevention of moisture adsorption, the fermented red ginseng extract spherical granules were coated by using hydrophobic fractions. The hydrophobic parts were extracted from red ginseng such as the perfect soluble part in 90% ethanol (PSE) and non-soluble part in hot water (NSHW). The optimum operating conditions for making fermented red ginseng extract spherical granules coated with PSE were a feeding rate (FR) of 0.49 mL/min, atomization air pressure (AP) of 2.14 bar, and product temperature (PT) of $48.96^{\circ}C$., whereas conditions for granules coated with NSHW were a FR of 0.61 mL/min, AP of 2.75 bar, and PT of $46.30^{\circ}C$. The solubility of coated fermented red ginseng extract spherical granule was lower than that of not coated. The fermented red ginseng extract spherical granules coated with NSHW showed more preventative effects for moisture adsorption than those coated with PSE, although there were no differences in solubility and fluidity. In the sensory evaluation, granules coated with hydrophobic fractions extracted from red ginseng were shown to be bitterer and less soluble than those not coated spherical granules. In conclusion, fermented red ginseng extract spherical granules coated with hydrophobic extracts from red ginseng showed a preventative effect against moisture.

Panax ginseng Improves Senile Testicular Function in Rats

  • We reported previously that the administration of Korean red ginseng water extract (KRG-WE) protected the guinea pig testis against damage induced by 2,3,7,8-tetrachlorodibenzo-p-dioxin (a potent endocrine disruptor). We also found that crude saponin from ginseng was the active ingredient responsible for this protection. Here, we examined the biological role of KRG-WE in an animal model of age-induced dysfunction of spermatogenesis. Twenty-four male Sprague-Dawley (six 2-month-old and eighteen 12-month-old) rats were used. The young and old control groups received only vehicle. The ginseng saponin (GS)- and KRG-WE-treated groups received GS (40 mg/kg body weight/day) and KRG-WE (200 mg/kg body weight/day), respectively, for 4 months. The number of cells, Sertoli cell index, Johnsen's score, and sex hormone levels decreased significantly with age. However, the administration of KRG-WE and GS markedly improved the number of germ cells, seminiferous tubular size, and Johnsen's score in the old rats. Ginseng produced a distinct testicular histological improvement in old rats. KRG-WE and GS elevated testosterone levels, while attenuating the aberrant increase in follicle stimulating hormone and luteinizing hormone levels. Sperm kinematics evaluated by a computer-assisted sperm analyzer demonstrated improvement in the percentage of motile sperm, progressive sperm motility, and curvilinear velocity associated with sperm quality, supporting the beneficial role of red ginseng in senile spermatogenesis. Overall, the total water extract had a more potent effect than the corresponding saponin fraction. In conclusion, Korean red ginseng rejuvenated age-induced testicular dysfunction. Additionally, the total water extract was more potent than the corresponding saponin fraction.

An Experimental Study on the Effect of Immunopotential and the Anticancer Effect of Red Ginseng Extract (한국 홍삼의 면역활성 및 항암효과에 관한 실험적 연구)

  • To evaluate the anticarcinogenic effect and its mechanism of red ginseng, the mice were treated with red ginseng and received subcutaneous Bl6 melanoma cell line injection on the back. Tumor incidence was same (100%) both in water and red ginseng-treated groups, but tumor production was delayed in red ginseng-treated group. Survival time was somewhat longer in red ginseng-treated group. The histopathological findings were similar in both groups, but lymphocytic infiltration around the tumor and melanin production in the tumor cells were prominent in the red ginseng-treated group. Flow cytometric analysis on T lymphocytes and natural killer cells revealed increased $T_H$/$T_S$ ratio and increased NK cells in red ginseng-treated group. These results suggest that the anticarcinogenic effect of red ginseng may be exerted by the increased cell-mediated immunity and natural killer cell activity.

  • Browning intensity is a major factor to estimate the quality of red ginseng or red ginseng products. The Maillard type of browning reaction proceeds nonenzymatically during extraction and concentration of red ginseng. The present studies were carried out to investigate the effects of amino acids and sugars on the browning reaction during extraction and concentration of red ginseng. Red ginseng was pulverized to 115 mesh and then tenfold (v/w) of water was added to the powder to make the substrate of red ginseng. Solution (0.1 M) of fourteen amino acids and of folly silgars were added to the substrates of red ginseng powder and these were then extracted and concentrated to examine their browning intensities. Amino acids were more effective than sligars in acrelerating the browning reaction. Acceleration of the browning reaction in the concentrate was in the order of arginine> histidine>glycine>alanine>lysine phenyl alanine>aspartic acid>lelicine>threonine>gllitamic acid>tyrosine>valine>istleucine>methionine for amino acids, and was glucose>frlictose >silcrose, maltose for sugars.

Ginsenoside Composition and Quality Characteristics of Red Ginseng Extracts prepared with Different Extracting Methods (추출방법에 따른 홍삼추출액의 사포닌 조성과 품질특성)

  • This study was conducted to compare the contents of ginsenoside according the water extract conditions of red ginseng. In method A, red ginseng extract was prepared at $75^{\circ}C$ for 18 hours by 1 time extraction, and method B, the preparation was done at $85^{\circ}C$ for 18 hours by 1 time extraction. In method C, the primary extract prepared at $75^{\circ}C$ for 9 hours was blended with the secondary extract prepared by re-extracting the red ginseng residue obtained after the primary extraction, at $85^{\circ}C$ for 9 hours. Method D was the same procedure as method C but the extraction temperature for the primary extraction was $85^{\circ}C$ and that for the secondary extraction was $95^{\circ}C$. The contents of total and $Rb_1$, $Rg_1$ and $Rg_3$ ginsenoside were highest in Method C. The content of prosapogenin (ginsenoside $Rg_2$, $Rg_3$, $Rb_1$ and $Rb_2$) was highest in Method B. There was no consistent tendency in Brix, pH, Hue value and absorbance among extraction methods.
