Performance of Growing Period of Two-way Crossbreed Parent Stock for Producing of Laying-Type Korean Native Commercial Chickens (산란전용 토종 실용계를 생산하기 위한 2원교배 종계의 육성 능력 검정)

  • This work was carried out to investigate the performance of growing period of two-way crossed of Korean native chickens parental stocks. A total of four hundred eighty female 2-crossbred chicks were used in this study and they were from National Institute of Animal Science. Groups were four crossbreds (4 replications/crossbred, 30 birds/replication) as A) C strain ${\times}$ Y strain, B) C strain ${\times}$ L strain, C) C strain ${\times}$ G strain and D) C strain ${\times}$ W strain. Body weight of A crossbred was the highest at the age of 8 week (P<0.05) and that of D strain was the lowest for growing period (P<0.05). Body weights of A and B crossbreds were higher than those of C and D crossbreds at the 12 and 16 weeks (P<0.05). Weekly body weights of A and B crossbreds were higher than C and D crossbreds (P<0.05), and weekly body weight of B crossbred was higher compared to other crossbreds at 0~20 weeks old. Weekly feed intake of D crossbred was the lowest among all crossbreds at 0~12, 0~16 and 0~20 weeks old (P<0.05). Weekly feed conversion ratio of C crossbred was the highest among crossbreds (P<0.05). These results can give the basic information for growth related data in 2-way crossbreed Korean Native Chickens, which can be used for the parental stocks for the laying-type of Korean native commercial chickens.

A Cellulolytic and Xylanolytic Enzyme Complex from an Alkalothermoanaerobacterium, Tepidimicrobium xylanilyticum BT14

  • A cellulolytic and xylanolytic enzyme complex-producing alkalothermoanaerobacterium strain, Tepidimicrobium xylanilyticum BT14, is described. The cell was Grampositive, rod-shaped, and endospore-forming. Based on 16S rRNA gene analysis and various lines of biochemical and physiological properties, the strain BT14 is a new member of the genus Tepidimicrobium. The strain BT14 cells had the ability to bind to Avicel, xylan, and corn hull. The pH and temperature optima for growth were 9.0 and $60^{\circ}C$, respectively. The strain BT14 was able to use a variety of carbon sources. When the bacterium was grown on corn hulls under an anaerobic condition, a cellulolytic and xylanolytic enzyme complex was produced. Crude enzyme containing cellulase and xylanase of the strain BT14 was active in broad ranges of pH and temperature. The optimum conditions for cellulase and xylanase activities were pH 8.0 and 9.0 at $60^{\circ}C$, respectively. The crude enzyme had the ability to bind to Avicel and xylan. The analysis of native-PAGE and native-zymograms indicated the cellulosebinding protein showing both cellulase and xylanase activities, whereas SDS-PAGE zymograms showed 4 bands of cellulases and 5 bands of xylanases. Evidence of a cohesinlike amino acid sequence seemed to indicate that the protein complex shared a direct relationship with the cellulosome of Clostridium thermocellum. The crude enzyme from the strain BT14 showed effective degradation of plant biomass. When grown on corn hulls at pH 9.0 and $60^{\circ}C$ under anaerobic conditions, the strain BT14 produced ethanol and acetate as the main fermentation products.

Laying Performance of Korean Native Breeder Ducks for Producing Korean Native Commercial Ducks (토종 실용오리 생산을 위한 토종 종오리의 산란 능력)

  • This study was carried out to investigate the performance of laying period of A and B strains of Korean native ducks (KND). One hundred sixty eight ducks were used in this work and divided into A and B strains (6 replications/strain, 14 birds/replication). Ducks were fed with laying ducks' feedstuff (CP 15%, ME 2,900 kcal/kg) for the age of 20~80 wk old. There was no significant difference on body weight, feed intake and egg weight between A and B strains. Weekly body wt of KNDs was the lowest at the age of 40~48 wk old, and that of those was high at the age of 24~32 wk and 64~80 wk old (P<0.05). Weekly feed intake significantly decreased at the age of 36~48 wk and 68~72 wk old (P<0.05), but there was no significant difference at the age of 52~56 wk old. Weekly egg wt was the lowest at the age of 32~40 wk old. Egg production ratio of B strain was higher compared to that of A strain at the age of 24~28 wk, 60~64 wk and 20~80 wk old. Weekly egg production ratio was the highest at the age of 28~32 wk old, and was high maintained until the age of 48 wk old. However, weekly egg production ratio decreased from the age of 52 wk old to the age of 68 wk old. Number of egg of B strain (267.5) was higher than that of A strain (235.6) at the age of 20~80 wk old. There was no significant difference on feed conversion ratio between A and B strain at the age of 28~36 wk old, but feed conversion ratio of A strain was higher than that of B strain at the other weeks (P<0.05). These results provided the basic data on the record of laying period of Korean native ducks.

  • This study was carried out to investigate the performance of laying period of two-way crossbred of Korean native chickens. Treatments were four crossbreds (4 replications/crossbred, 30 birds/replication) as A) C strain ${\times}$ S strain, B) C strain ${\times}$ H strain, C) R strain ${\times}$ S strain, and D) L strain ${\times}$ H strain, respectively. There were no significant difference on age, egg weight, and body weight at first egg among crossbreds (P>0.05). Body weight of D crossbred was lower compared to other crossbreds at the age of 24, 32, 40 and 60 week (P<0.05). Feed intake of A crossbred was highest among crossbreds (P<0.05). Weekly egg weight of A crossbred was the highest among crossbreds all laying period except 48~52 and 60~64 weeks. Hen-house egg production of A and B crossbreds was higher than that of C and D crossbreds (P<0.05). Hen-day egg production decreased after at the age of 36 weeks. Weekly egg production of A and B crossbreds was higher compared to C and D crossbreds (P<0.05). These results suggested the basic data on the record of laying period of 2-crossbreed Korean Native Chickens.

Genetic Relationship between Populations and Analysis of Genetic Structure in the Korean Native Chicken and the Endemic Chicken Breeds (한국재래닭 및 토착화 품종간의 유연 관계 및 유전 특성 분석)

  • The purpose of this study was to assess the genetic variation and establish the relationship amongst breeds and strains using 7 chicken specific microsatellite markers. A total of 317 DNA samples from four Korean native chicken (KNC) strains (KR: Korean Native Red chicken strain, KY: Korean Native Yellow chicken strain, KL: Korean Native Black chicken strain, KO: Ogol chicken strain) and three introduced endemic chicken breeds (LE: Leghorn chicken breed, RI: Rhode Island Red chicken breed, CO: Cornish chicken breed). The size of microsatellite markers was decided using GeneMapper Software (v.4.0) after being analyzed using an ABI 3130 Genetic Analyzer. Frequencies of microsatellites markers were used to estimate heterozygosities and genetic distances. The lowest distance (0.074) was observed between the KY and KL breeds and the highest distance (0.779) between the KL and LE breeds. The KNC strains (KR, KY, KL) have comparatively near genetic distance each other. On the other side, each individual was not ramified to different groups and were spread evenly in phylogenetic dendrogram about all the KNC of each strain populations. But the endemic breed populations (LE, RI, CO) were ramified to different groups. The microsatellite polymorphism data were shown to be useful for assessing the genetic relationship between Korean native strains and other foreign breeds.

Population, Symbiotic Effectiveness, and Protein Profile Patterns of Indigenous Rhizobium leguminosarum biovar viciae to Korean Soils

  • Some symbiotic characteristics of native Korean Rhizobium leguminosarum biovar viciae were analysed to get some informations desirable for cultivation of hairy vetch (Vicia villosa Roth) using its symbiont in Korea. The size of indigenous populations of R. leguminosarum biovar viciae was higher in seven upland soils showing $1.7{\times}10^2{\sim}5.8{\times}10^4cells\;g{\cdot}soil^{-1}$, which appeared to be 10% and 37% higher for cultivated and uncultivated soils of hairy vetch, respectively, than seven paddy soils with $1.7{\times}10^2{\sim}1.7{\times}10^4cells\;g{\cdot}soil^{-1}$. In symbiotic potentials, however, the yields of hairy vetch treated with 10-fold-diluted ($10^{-1}$) inoculum and 1000-fold-diluted ($10^{-3}$) one was 11.2% and 8.8% more, respectively, in paddy than upland. Hairy vetch inoculated with either strain KHR 106 from Sacheon or strain KHR 120 from Yesan among native Korean R. leguminosarum biovar viciae isolates was of similar yield increment of 16% (p < 0.05) in upland soils with native R. leguminosarum biovar viciae of $5.8{\times}10cells\;g{\cdot}soil^{-1}$. In case of coinoculation of the two strains, however, the yields was not significantly increased. In especial, isolate KHR 106, KHR 120, and KHR 122 from Suwon, which has also good symbiotic effectiveness, showed different protein profile patterns each other. As a result, hairy vetch is possibly able to use atmospheric nitrogen through symbiotic relationship with diverse native R. leguminosarum biovar viciae in Korean arable lands. For safe and good production of hairy, however, the use of superior strains with high symbiotic effectiveness and competitiveness will be desirable.

Characterization of an Iron- and Manganese-containing Superoxide Dismutase from Methylobacillus Sp. Strain SK1 DSM 8269

  • A superoxide dismutase was purified 62-fold in seven steps to homogeneity from Methylobacillus sp. strain SK1, an obligate methanol-oxidizing bacterium, with a yield of 9.6%. The final specific activity was 4,831 units per milligram protein as determined by an assay based on a 50% decrease in the rate of cytochrome c reduction. The molecular weight of the native enzyme was estimated to be 44,000. Sodium dodecyl sulfate gel electrophoresis revealed two identical subunits of molecular weight 23,100. The isoelectric point of the purified enzyme was found to be 4.4. Maximum activity of the enzyme was measured at pH 8. The enzyme was stable at pH range from 6 to 8 and at high temperature. The enzyme showed an absorption peak at 280 nm with a shoulder at 292 nm. Hydrogen peroxide and sodium azide, but not sodium cyanide, was found to inhibit the purified enzyme. The enzyme activity in cell-free extracts prepared from cells grown in manganese-rich medium, however, was not inhibited by hydrogen peroxide but inhibited by sodium azide. The activity in cell extracts from cells grown in iron-rich medium was found to be highly sensitive to hydrogen peroxide and sodium azide. One mol of native enzyme was found to contain 1.1 g-atom of iron and 0.7 g-atom of manganese. The N-terminal amino acid sequence of the purified enzyme was Ala-Tyr-Thr-Leu-Pro-Pro-Leu-Asn-Tyr-Ala-Tyr. The superoxide dismutase of Methylobacillus sp. strain SK1 was found to have antigenic sites identical to those of Methylobacillus glycogenes enzyme. The enzyme, however, shared no antigenic sites with Mycobacterium sp. strain JC1, Methylovorus sp. strain SS1, Methylobacterium sp. strain SY1, and Methylosinus trichosproium enzymes.

Enhanced antibacterial activity of tilmicosin against Staphylococcus aureus small colony variants by chitosan oligosaccharide-sodium carboxymethyl cellulose composite nanogels

  • Background: The poor bioadhesion capacity of tilmicosin resulting in treatment failure for Staphylococcus aureus small colony variants (SASCVs) mastitis. Objectives: This study aimed to increase the bioadhesion capacity of tilmicosin for the SASCVs strain and improve the antibacterial effect of tilmicosin against cow mastitis caused by the SASCVs strain. Methods: Tilmicosin-loaded chitosan oligosaccharide (COS)-sodium carboxymethyl cellulose (CMC) composite nanogels were formulated by an electrostatic interaction between COS (positive charge) and CMC (negative charge) using sodium tripolyphosphate (TPP) (ionic crosslinkers). The formation mechanism, structural characteristics, bioadhesion, and antibacterial activity of tilmicosin composite nanogels were studied systematically. Results: The optimized formulation was comprised of 50 mg/mL (COS), 32 mg/mL (CMC), and 0.25 mg/mL (TPP). The size, encapsulation efficiency, loading capacity, polydispersity index, and zeta potential of the optimized tilmicosin composite nanogels were 357.4 ± 2.6 nm, 65.4 ± 0.4%, 21.9 ± 0.4%, 0.11 ± 0.01, and -37.1 ± 0.4 mV, respectively; the sedimentation rate was one. Scanning electron microscopy showed that tilmicosin might be incorporated in nano-sized crosslinked polymeric networks. Moreover, adhesive studies suggested that tilmicosin composite nanogels could enhance the bioadhesion capacity of tilmicosin for the SASCVs strain. The inhibition zone of native tilmicosin, tilmicosin standard, and tilmicosin composite nanogels were 2.13 ± 0.07, 3.35 ± 0.11, and 1.46 ± 0.04 cm, respectively. The minimum inhibitory concentration of native tilmicosin, tilmicosin standard, and tilmicosin composite nanogels against the SASCVs strain were 2, 1, and 1 ㎍/mL, respectively. The in vitro time-killing curves showed that the tilmicosin composite nanogels increased the antibacterial activity against the SASCVs strain. Conclusions: This study provides a potential strategy for developing tilmicosin composite nanogels to treat cow mastitis caused by the SASCVs strain.

Genetic Characterization of Two Putative Toxin-Antitoxin Systems on Cryptic Plasm ids from Bacillus thuringiensis Strain YBT-1520

  • A novel putative toxin-antitoxin segregational stability system named KyAB system was identified in a novel native plasmid pBMB8240 from Bacillus thuringiensis strain YBT-1520, based on sequences homology with other toxin-antitoxin systems, the lethal activity of the KyB putative toxin in Escherichia coli and the stabilizing effect of the kyAB system in Bacillus thuringiensis. Secondarily, the native plasmid pBMB9741 from the same strain was resequenced and the corrected plasmid was named as pBMB7635. Based on sequence homology with the tasAB system and the lethal activity of toxin protein in Escherichia coli, a tasAB-like putative toxin-antitoxin system was identified on pBMB7635.
