Effects of an Extract from the Roots of Platycodon Grandiflorum on the Levels of p53 and pRB in NCI-H460 Human Lung Carcinoma Cells (길경 수용액 추출물에 의한 NCI-H460 인체 폐암세포의 p53 및 pRB의 발현에 미치는 영향)

  • Platycodi Radix, the root of Platycodon grandiflorum A. DC (Campanulaceae), commonly known as Doraji in Korea (Chinese name, 'Jiegeng', and Japanese name, 'Kikyo') has been used as an expectorant in traditional Oriental medicine. Extracts from the roots of P. grandiflorum have been reported to have wide ranging health benefits. In Korea, Platycodi Radix is also used as a food and employed as a folk remedy for adult diseases, such as bronchitis, asthma and pulmonary tuberculosis, hyperlipidemia, diabetes, and inflammatory diseases, and as a sedative. Several studies on its chemical and immunopharmacological effects including immunostimulation and antitumor activity have been performed. However, the relevant molecular mechanisms are poorly understood. Platycodi Radix, the root of Platycodon grandiflorum A. However, the relevant molecular mechanisms are poorly understood. In the present study, we investigated the effects of an aqueous extract from the roots of P. grandiflorum AEPG) on the cell growth of human lung adenocarcinoma NCI-H460 cells in order to understand its anti-proliferative mechanism. AEPG treatment down-regulated the cyclin D1 expression in both transcriptional and translational levels without alteration of cyclin E. In AEPG-treated cells, the levels of cyclin-dependent kinase (C아) 6 mRNA and protein were significantly inhibited, but the levels of Cdk2 and Cdk4 were slightly inhibited by treatment of AEPG. AEPG treatment induced a marked accumulation of Cdk inhibitors, p16 and p27. However, AEPG treatment did not affect not only retinoblastoma protein (pRB) but also tumor suppressor p53 protein expression. The present results indicated that AEPG-induced inhibition of lung cancer cell proliferation is associated with the blockage of G1 phase progression through induction of Cdk inhibitors such as p16 and p27, and inhibition of cyclin D1 and Cdk6. AEPG exposure, as offered by this study, provides cluse for the mechanism of AEPG action. Taken together, these findings suggest that P. grandiflorum has strong potential for development as an agent for prevention and treatiment against human lung cancer.

Medical Achievements of Doctor-Lee, Seokgan and Interpretation of the first unveiled 「Daeyakbu」 (조선 중기 유의(儒醫) 이석간(李碩幹)의 가계와 의약사적 연구 - 새로 발견된 대약부(大藥賦)를 중심으로 -)

  • This study confirmed that a doctor named Lee, Seok-gan whose name has been widely known but whose real identity has remained unclear, was an active Confucian doctor in the 16th century. In addition, through the newly discovered "Daeyakbu" among his family line, writings, and relics that have been handed down in a family, this study looked into his medical philosophy and medicine culture. The author of "Ieseokgangyeongheombang"(Medical Book by Lee, Seok-gan(李石澗), Seok-gan is the same person as an active famous doctor Lee, Seok-gan(李碩幹, 1509-1574) in the 16th century. Such a fact can be confirmed through "Samuiilheombang", "Sauigyeongheombang" and the newly opened "Ieseokgangyeongheombang". Lee, Seok-gan was born in the 4th ruling year of king Jungjong (1509) and was active as a doctor until the 7th ruling year of king Seonjo(1547); his first name is Jungim with the pen name-Chodang, and he used a doctor name of 'Seokgan.' He was known as a divine doctor, and there have been left lots of anecdotes in relation with Lee, Seok-gan. Legend has it that Seokgan went to China to give treatment to the empress, and a heavenly peach pattern drinking cup and a house, which the emperor bestowed on Seokgan in return for his great services, still have remained up to the present. Usually, Seokgan interacted with Toegye Lee Hwang and his literary persons, and with his excellent medical skills, Seokgan once gave treatment to Toegye at the time of his death free of charge. His medical skills have been handed down in his family, and his descendant Lee, Ui-tae(around 1700) compiled a medical book titled "Gyeongheombangwhipyeon(經驗方彙編)". Out of Lee, Seok-gan's keepsakes which were donated to Sosu museums by his descendant family, 4 sorts of 'Gwabu'(writings of fruit trees) including "Daeyakbu" were discovered. It's rare to find a literary work left by a medical figure like this, so these discoveries have a deep meaning even from a medicine culture level. Particularly, "Daeyakbu" includes the typical "Uigukron". The "Uigukron", which develops its story by contrasting politics with medicine, has a unique writing style as one of the representative explanatory methods of scholars' position during the Joseon Dynasty; in addition, the distinctive feature of "Uigukron" is that it was created in the form of 'Gabu' other than a prose.

The Possibility of Geographical Range Change of 'Ho-Nam' as a Place Name (지명 '호남(湖南)'의 형성과 지리적 범위 변화 가능성)

  • This study looks into existing discussion about the entry time and the geographical criteria point of 'Ho-Nam' as a place name. And discusses the reason which the geographical criteria point is not clear with emphasis on the possibility of the geographical range change of place name. The result is as follows. Firstly, the regional criteria of 'Ho-Nam' is Kum-River if we consider direction and the role as a regional barrier among the existing offered criteria. Secondary, the criteria of Ho-Nam and Ho-Seo might be different each other. A place name of Ho-Nam had appeared before Chosun period as a geographical concept and changed to the cultural concept during Chosun period. And Ho-Seo was established in Chosun period. But 'Ho-Nam=Jeon-la Province' and 'Ho-Seo=Chung-cheong Province' is established during Chosun period as a cultural concept. Thirdly, the geographical range of Ho-Nam of Go-Ryeo period and Chosun period were different each other. Fourthly, the criteria of Ho-Nam may be the Han-River if we consider the entry time and geographical range change.

Supplementing punicalagin reduces oxidative stress markers and restores angiogenic balance in a rat model of pregnancy-induced hypertension

  • Pre-eclampsia (PE) is a pregnancy disorder that is characterised by severe hypertension and increased risks of foetal and maternal mortality. The aetiology of PE not completely understood; however, maternal nutrition and oxidative stress play important roles in the development of hypertension. The treatment options for PE are currently limited to anti-hypertensive drugs. Punicalagin, a polyphenol present in pomegranate juice, has a range of bioactive properties. The effects of supplementation with punicalagin on angiogenesis and oxidative stress in pregnant rats with induced hypertension were investigated. The pregnant rats were randomly divided into five experimental groups (n=12 per group). Hypertension was induced using an oral dose of NG-nitro-L-arginine methyl ester (L-NAME, 50 mg/kg/day) on days 14-19 of pregnancy. Punicalagin (25, 50 or 100 mg/kg) was given orally on days 14-21 of pregnancy. Punicalagin treatment at the tested doses significantly reduced diastolic, systolic, and mean arterial blood pressure in L-NAME treated rats from day 14. Punicalagin also restored angiogenic balance by increasing the expression of vascular endothelial growth factor and downregulating vascular endothelial growth factor receptor-1/fms-like tyrosine kinase-1. Punicalagin, significantly increased the placental nitric oxide levels as compared to PE group. The increased levels of oxidative stress in rats with PE were markedly decreased by treatment with punicalagin. Punicalagin at the tested doses markedly (p<0.05) enhanced the placental antioxidant capacity in L-NAME-treated rats. The raised catalase activity observed following L-NAME induction was significantly (p<0.05) and restored to normal activity levels in punicalagin treatment. Further, 100 mg dose of punicalagin exhibited higher protective effects as compared to lower doses of 25 and 50 mg. This study shows that supplementation with punicalagin decreased blood pressure and oxidative stress and restored angiogenic balance in pregnant rats with induced PE.

Effectiveness of the Internet Real-Name Verification Law: Evidence from Self-censorship and Intention to Detour the Regulation (인터넷실명제와 우회로의 선택: 인터넷 공론장 참여자들의 자기검열과 우회로 선택의향을 중심으로)

  • This paper investigates the effectiveness of Real-Name System on Internet, which has been applied to some Internet service provider bulletin boards in Korea. The goal of the Real-Name system on the Internet is controlling expression of slanderous or abusive language. The effectiveness of the regulation is dependent on the people's perceptions and attitude toward to the legislation. This empirical study examines the antecedents that affect the perceptions and attitude toward the regulation. Further, we investigate the relationships among variables such as internet public sphere participation, attitude to the regulations, and intention to choose detours. Results of regression analysis show that the younger and the more progressive individuals are, the more they oppose the implementation of the law. Also, it was found that people who have more political interest and participate more actively in public sphere via internet, tend to oppose the law. Structural equation models reveal that participation in online public sphere drives negative attitudes about the regulation. In addition, people with negative perceptions about the law tend to choose the detours such as 'cyber asylum' and 'service asylum'.

Correlation between Short Stature and Obese Degree - Intended for the Case of 236 Patient in the Name of Short Stature (저신장과 비만도의 상관성 분석 - 저신장을 주소로 내원한 환아 236명을 대상으로)

  • Objectives The purpose of this study was to investigate correlation between short stature and obese degree. And we also wanted to know the characteristics of patients visited clinic in the name of 'short stature'. Methods Height, body weight, BMI, fat mass, lean body mass, percent body fat were taken intended for 236 patients in the name of short stature. To all the patient questionnaire for growth clinic was drawn up, and ultrasound scan was taken through calcaneous of the right foot. The patients were classified to three groups - short, average, tall group - according to relative position of mid-parental height. It was analysed that the differences between groups in obese degree. Also was investigated correlation between position of short stature and obese degree, and between obese degree and bony maturity. Results & Conclusion 1. The average ages of patients in the name of 'short stature' were $12.69{\pm}3.93$ years old in boys, $10.66{\pm}3.67$ years old in girls. And it seemed to be just before second rapid maturing period. 2. The average BMI were $20.58{\pm}4.07kg/m^2$ in boys, $18.65{\pm}2.85kg/m^2$ in girls, and average percent body fat were $21.99{\pm}7.35%$ in boys, $26.01{\pm}6.35%$ in girls. 3. The numbers of obese children were 34(31.2%) in boys, 19(14.9%) in girls on the basis of BMI. And the numbers were 39(35.8%) in boys, 53(41.7%) in girls on the basis of percent body fat. There was a big difference in case of girls. 4. The numbers of AG(average group) were 48(44.0%) in boys, 60(47.2%) in girls, the numbers of SG(short group) were 35(32.1%) in boys, 31(24.4%) in girls, and numbers of TG(tall group) were 26(23.9%) in boys, 35(27.6%) in girls. 5. There were no significant differences among the groups in BMI, lean body mass, fat mass, percent body fat. Only significant difference in DI(disease index). 6. There were no significant correlation between PH(percent height) and BMI, leanbody mass, fat mass, percent body fat. Only significant correlation in DI(disease index). 7. There were no significant differences between DA(difference between bone age and chronorogical age) and BMI, leanbody mass, percent body fat. Only significant correlation in fat mass.

Folk Plants in the Inland of Northern Area in Gangwon-do (강원도 내륙 북부지역의 민속식물)

  • This study was carried out to classify the folk plants in inland of northern region of Gangwon-do. According to the survey results from 35 places of 9 counties and cities from April to October, 2010, the folk plants in inland of northern region of Gangwon-do consisted of a total 261 taxa; 73 families, 185 genera, 225 species, 2 subspecies, 30 varieties, and 4 form. Among the investigated 261 taxa, 3 Korean endemic, 11 rare plants and 9 naturalized plants were also included. The use by usage of 261 taxa was: 378 times; Edible, 53 times; Medicinal, 8 times; Dye, 7 times; Ornamental, 6 times; Fuel, 5 times; Spice, 3 times; Aroma, 2 times; Nectar, 41 times; Others, respectively, so the edible use is the highest. The most useful part was the leaf, followed by whole plant and root. The consistency comparison between the scientific name and the local name were the highest in the 20's and the lowest in 80's.

Interaction between Renin-Angiotensin and Endothelium-Derived Nitric Oxide Systems in Two-Kidney, One Clip Hypertensive Rats

  • The present study was aimed to investigate the role of endothelium-derived nitric oxide (NO) in the control of renin release and to examine if NO is implicated in the development of two-kidney, one clip (2K1C) hypertension. Male Sprague-Dawley rats $(150{\sim}200\; g)$ were constricted at the left renal artery. They were then supplemented with $N^{G}-nitro-L-arginine\;methyl\;ester\;(L-NAME,\; 5mg/100\;mL)$ or with L-arginine hydrochloride (400 mg/100 mL) in the drinking water. The control group was supplied with normal tap water. The sham-clipped rats were operated as in 2K1C rats except for that no clip was made. The kidneys were taken to examine in vitro release of renin at days 7 and 14 following clipping the renal artery. Northern blot analysis was also done to assess the expression of renin gene in the kidney. In sham-clipped rats, L-NAME caused a sustained increase of the blood pressure, whereas L-arginine was without effect. Neither L-NAME nor L-arginine-supplementation significantly affected the development of hypertension in 2K1C rats. Plasma renin concentration (PRC) measured on day 28 did not significantly differ among the L-NAME, L-arginine and control groups either in 2K1C or in sham-clipped rats. Renin contents (RRC) in the clipped kidney were increased, while those in the contralateral kidney were decreased. The release of renin in vitro from cortical slices was also enhanced in the clipped kidney, whereas it was attenuated in the contralateral. Comparing the RRC and in vitro release, the latter was more rapidly decreased than the former in the contralateral kidney. The renin mRNA levels in the contralateral kidney were almost at their nadir at days 7 and 14 in 2K1C rats. It is suggested that NO does not affect the development of 2K1C hypertension in which the renin-angiotensin system has been activated. The data also confirm that RRC and renin gene expression are increased in the clipped kidney and suppressed in the contralateral kidney in 2K1C rats.

Analysis of Confusion of Azalea Cultivars and Its Actual State of Use in Landscaping Businesses (조경업체에서의 철쭉재배품종의 혼선 및 이용실태 분석)

  • This study was conducted to examine the state of confusion about those azalea cultivars(hereinafter, Azalea Cultivars) that are used among practicians in landscaping businesses(designers, constructors, and producers). Which Azalea Cultivars would be delivered when they were ordered to producers using the names of Azalea Cultivars notified by the Public Procurement Service. In addition, the situations of azalea cultivar planting designs in the planting design drawings of three apartments completed recently and whether the Azalea Cultivars on the design drawings were the same as those Azalea Cultivars that were actually planted were examined. When Azalea Cultivars were ordered using the notified names, three Azalea Cultivars; Beni, Three, and Daewang were delivered for the name Yeongsanhong and two Azalea Cultivars; Zasanhong and Hancheol were delivered for the name Zasanhong. Yeongsanhong was perceived as a common name of Beni, Three, and Daewang by producers. Therefore, this caused confusion between producers and purchasers. In the case of Zasanhong, as the traits of Zasanhong are similar to those of the cultivar Hancheol, the producers or purchasers might have been unable to clearly distinguish between the cultivars and thus two cultivars might have been delivered without being distinguished from each other. The names of Azalea Cultivars used in planting design drawings were the names notified by the notifying agencies or comprehensive and unclear names such as azalea and Yeongsanhong. Completely different Azalea Cultivars from those written in planting design drawings were planted in actual construction sites in many cases. These situations should have occurred because designers did not write accurate cultivar names in planting design drawings and constructors could not accurately perceive the classification characteristics of individual Azalea Cultivars but planted those Azalea Cultivars with similar flowers colors or those that could be easily bought to fit the conditions of construction sites.

The Political Geography of Place Names : The Decisions of City Names in the Process of Administrative District Reorganization (지명의 정치지리학: 행정구역개편으로 인한 시 명칭 결정을 사례로)

  • Human being has used place names to differentiate one place from another. Place names are the products of collective human cognition in that a place name is chosen when it successfully represents the identity of a place. In addition, place names have been changed by the competition and struggle between social agencies and the political imperatives of hegemonic groups to impose their identity on the places. Recent geographic studies on place names have focused on the social and political processes behind the change of place names. In this vein, the purpose of this study is investigating the debates on the decision of city names in the process of the administrative reorganization in mid 1990s by the lens of political geography. Residents in cities and counties tried to justify their arguments by emphasizing historical backgrounds and popularity of their names. Additionally, economic power and potential were mobilized for the political resources to win over the battle over city names. The result, however, shows that the decision of newly consolidated cities' name was mainly made by the amount of political resources, such as population and number of seats in local assemblies. Several regions tried to use city names to negotiate with counterparts. In sum, the decision of place names is the product of political competitions, and the place name becomes the symbol of territorial identity. Place names have been in the center of disputes in local politics even after the name was decided, which suggest further studies on reasonable solution to mitigate the disputes to be expected when additional reorganization of administrative districts.
