Eight Previously Unreported Species of Fungi Identified in Mt. Manggyeong, Korea

  • A total of 118 fungal specimens were collected from Mt. Manggyeong, Chungnam province, Korea between July and October 2014. All specimens were identified to the species level using a combination of morphological characteristics and DNA sequencing. Out of 118 specimens, eight species, Amanita manginiana, A. pilosella, Calvatia holothurioides, Cantharellus tabernensis, Dicephalospora rufocornea, Gymnopus menehune, Marasmius brunneospermus, and Russula cerolens, have not been previously reported in Korea.

  • In November 2021, sponge gourd (Luffa aegyptiaca) plants with typical downy mildew symptom were observed in Korea. Based on morphological characteristics of sporangiophores and sporangia, combined with molecular phylogenetic analysis of the internal transcribed spacer rDNA and cox2 mtDNA regions, the causal pathogen was identified as Pseudoperonospora cubensis (Oomycota). This is the first report of downy mildew caused by P. cubensis on L. aegyptiaca in Korea. Given that P. cubensis exhibits high disease incidence and severity on many Cucurbitaceae crops, it could pose a significant threat to sponge gourd cultivation.

  • Numerous morphological studies on N. samarangae have been well conducted over the last ten years. In this context, we have attemtped to do molecular phylogenetic analysis by using metallothionein (MT) gene from N. samarangae. To this end, we cloned the full length cDNA of MT from cDNA library of N. samarangae. The complete cDNA sequences were obtained from the expressed sequence tag (EST) sequencing project of N. samarangae, The coding region of 195 bp gives an amino acid sequence of 65 residues including methionine. There are 5' (61 bp) and 3' (48 bp) untranslated region at both ends of the Ns-MT cDNA sequence. The combined results from BLAST analyses, multiple sequence alignment and molecular phylogenetic study of Ns-MT cDNA indicate that N. samarangae has similarity to land snails such as Helix pomatia, Helix aspersa and Arianta arbustorum.

Distribution Status of Hybrid Types in Large Liver Flukes, Fasciola Species (Digenea: Fasciolidae), from Ruminants and Humans in Vietnam

  • The aim of this study is to delineate 'admixed hybrid' and 'introgressive' Fasciola genotypes present in the Fasciola population in Vietnam. Adult liver flukes collected from ruminants in 18 Provinces were morphologically sorted out by naked eyes for small (S), medium (M) and large (L) body shapes; and human samples (n=14) from patients. Nuclear ribosomal (rDNA) ITS1 and ITS2, and mitochondrial (mtDNA) nad1 markers were used for determination of their genetic status. Total 4,725 worm samples of ruminants were tentatively classified by their size: 6% (n=284) small (S)-, 13% (n=614) medium (M)-, and 81% (n=3,827) large (L)-forms. All the representative (n=120, as 40 each group) and 14 human specimens, possessed maternal mtDNA of only F. gigantica and none of F. hepatica. Paternally, all (100%) of the L-(n=40) and 77.5% (n=31) of the M-flukes had single F. gigantica rDNA indicating 'pure' F. gigantica. A majority (90%, n=36) of the S- and 15% (n=6) of the M-worms had single F. hepatica rDNA, indicating their introgressive; the rest (10%, n=4) of the S- and 7.5% (n=3) of the M-flukes had mixture of both F. gigantica and F. hepatica rDNAs, confirming their admixed hybrid genetic status. Fourteen human samples revealed 9 (64%) of pure F. gigantica, 3 (22%) of introgressive and 2 (14%) of admixed hybrid Fasciola spp. By the present study, it was confirmed that the small worms, which are morphologically identical with F. hepatica, are admixed and/or introgressive hybrids of Fasciola spp., and able to be the pathogens of human fascioliasis.

Genetic Study of the Subfamily Salmoninae Based upon Mitochondrial DNA Control Region Sequences (미토콘드리아 DNA control region의 염기분석에 의한 연어아과 어류의 유전학적 연구)

  • The complete sequences of mtDNA control regions of six salmonines were determined: 1089 bp in lenok (Brachymystax lenok); 999 bp in cherry salmon (Oncorhynchus masou masou) and Ishikawa's cherry salmon (O. masou ishikauiae); 1002 bp in chum salmon (O. keta), and 1003 bp in rainbow trout (O. mykiss) and an albino mutant of rainbow trout. The estimated interspecific sequence divergences from PCR/direct sequencing data ranged from 5.42% to 16.49%. The organization of this region is similar to that of other vertebrates. A 81 bp tandemly repeated sequence, associated with length variation was observed in the 3' end of the salmonids control region in this study. In addition, The phylogenetic tree based on the control region sequences supported that cherry salmon was closer to chum salmon than to rainbow trout, while lenok was most distantly related species among six salmonines.

  • Except for a cosmopolitan and major pest of apples, Tetranychus urticae Koch, Tetranychus mites in Korea such as T. viennensis Zaher, T. kanzawai Kishida, and T. truncams Ehara have been considered as quarantine pests by Canada and United States. Even though these mites are not feeders on apples, they are suspected to attach accidentally on apple h i t s in autumn as females enter the diapause. The characters used to identify Tetranychus mites have been confined to the shape of aedeagus in adult male. To develope a fast and accurate alternative identification protocol applied to hibernating female mites on apples, their mitochondrial DNA (mtDNA) were examined to find out any polymorphisms to discriminate each species from the other ones. Three pairs of primers for polymerase chain reaction (PCR) were used to amplify cytochrome oxidase subunit I (CO-I) coding region in mitochondrial DNA5 of four species of Tetranycus mites. The longest amplified product was estimated its size as about 680 bp. Digestion with restriction enzymes, AluI, Ddel, and Sau3A, showed length polymorphisms, which will he useful as diagnostic markers to identify Tetranychus mites. Schematic restriction maps in amplified region were shown for each species.

Phylogenetic Analysis using mtDNA D-loop Sequences in Korean Native Ducks (mtDNA D-loop 염기서열 변이를 이용한 토종오리의 계통 분류)

  • Recently, the consumption of duck meat has been gradually increased in Korea. However, most of the duck breeds in Korea were imported from overseas. Based on the large demands for the breeding stocks of native ducks, a new project for the commercial use of the Korean native ducks has been launched. For the initial investigation of the relationships between Korean native duck (KND) with other duck breeds, the sequences from D-loop control region in mitochondrial DNA (mtDNA) was used. The results from phylogenetic analysis indicated that both KND and White Commercial Duck (WCD) breeds were classified well with wild duck breeds. However, mallard duck was not discriminated well with KND. The haplotype analysis indicated that KND and WCD have eight different haplotypes with eleven SNPs. Three haplotypes (haplotype 1, 3, 4) were shared both in KND and WCD. On the other hand, haplotype 1 was appeared only KND and haplotype 5, 6, 7, 8 were identified only in WCD population. With further verifications, the results presented here can be used for the conservation and commercialization of the Korean native ducks.

  • The nucleotide sequence of two hypervariable regions of the D-loop and the frequency of the 9-bp repeat in the region V of mitochondrial DNA (mtDNA) were investigated in the Korean population. Alignment of these sequences with the published reference revealed a unique pattern of base substitution and deletion compared with those of other races. The deletion and addition frequency of the 9-bp repeat in the region V was also distinct.

  • Metallothioneins (MT) are a group of low-molecular weight, cysteine-rich, intracellular proteins that are encoded by a family of genes containing at least 10 functional isoforms in human. The expression and induction of these proteins is associated with protection against DNA damage, oxidative stress, and apoptosis. Many studies have shown increased expression of MT in various human tumors, whereas MT is down-regulated in certain tumors such as hepatocellular carcinoma and liver adenocarcinoma. Hence, the expression of MT is not universal to all human tumors but may depend on the differentiation status and proliferative index of tumors, along with other tissue factors and gene mutations. Using Northern blot analysis, we found that laminin induced expression of MT-1 in HSG and PC12 cells, which can be differentiated by laminin, but had no effect on MB-231, MDA-435, and PC-3 cells, which cannot be differentiated by laminin. In addition, we analyzed the expression level of the MT-1 gene in five prostate cancer cell lines possessing different metastatic potential. The expression of MT-1 in normal and less malignant cells (RWPE-1 and WPE1-NA22) was high and up-regulated by laminin, whereas the expression of MT-1 in WPE1-NB14, WPE1-NB11, and WPE1-NB26 cells (malignant) was extremely low and not elevated by laminin. These results suggest that the MT-1 gene is involved in laminin-mediated differentiation and affects the metastatic potential of tumor cells.

  • 한국재래오골계는 천연기념물로 등록이 되어 있어 세계의 중요한 유전자원 중 하나이다. 현재 한국에서 사육되어 있는 오골계의 유전적 특성을 규명하기 위하여 미토콘드리아 DNA의 변이를 이용하여 계통 분석을 실시하였다. 총 31 마리의 한국재래오골계가 이 분석에 이용되었으며 10개의 haplotypes이 관찰되었다. NJ 방법으로 만들어진 계통도 분석을 통하여 이미 닭에서 알려진 A부터 C의 lineage를 포함하는 것으로 보아 한국 재래오골계는 아직도 높은 유전적 다형성을 유지하고 있음을 알 수 있었다. 이 연구 결과는 한국 재래오골계의 육종 및 보존 계획을 세우는데 유용하게 이용될 수 있을 것으로 사료된다.
