인삼열매 발효추출물을 함유한 제품의 품질안정성 (Quality Stability of Products Containing Fermented Ginseng Berry Extracts)

  • 인삼열매는 진세노사이드 함량이 인삼뿌리와 다르게 나타나며, 항 고혈당효과, 항암활성 등의 다양한 약리적 효능이 있다고 보고되고 있다. 본 연구에서는 인삼열매를 수확하여 Rb1 및 Rg1 등의 진세노사이드 함량이 풍부한 4회 증포 인삼열매와 Rg3를 포함한 총 진세노사이드 함량이 풍부한 7회 증포 인삼열매를 혼합하여 열수추출물과 에탄올 추출물을 얻었고, 이를 L. plantarum으로 발효시킨 추출물을 이용하여 액상 스틱을 제조한 후 10, 25, $35^{\circ}C$에서 4개월 동안 저장하면서 진세노사이드 함량 변화 및 이화학적 특성을 조사하였다. 인삼열매 추출물을 함유하는 액상스틱을 $35^{\circ}C$에서 4개월 동안 저장한 후에 pH는 4.81에서 3.81로 저하되었으나, 산도와 고형분 함량은 변화하지 않았다. DPPH 소거능은 $10^{\circ}C$$25^{\circ}C$에서 4개월 동안 저장했을 때 큰 변화를 나타내지 않았으나 $35^{\circ}C$에서 4개월 저장했을 때 크게 감소하였다. L값(명도)와 b값(황색도)는 저장기간 동안 감소하였으나, a값(적색도)는 변화하지 않았다. Rg1, Rb1, F2, Rg3(S), Rg3(R), Rg5 등 6종류의 총 진세노사이드 함량은 10, 25, $35^{\circ}C$에서 4개월 동안 저장하는 동안에 큰 변화가 없었으며, 일반세균과 대장균군도 검출되지 않았다.

  • Panax ginseng C. A. Meyer is well known as traditional herbal medicine, and ginseng berries are known to exhibit potential immune-enhancing functions. However, little is known about the in vivo immunomodulatory activity of Korean ginseng berries. In this study, crude Korean ginseng berries polysaccharides (GBP) were isolated and their immunomodulatory activities were investigated using cyclophosphamide (CY)-induced immunosuppressive BALB/c mice. In CY-treated mice, oral administration of GBP (50-500 mg/kg BW) remarkably increased their spleen sizes and spleen indices and activated NK cell activities. GBP also resulted in the proliferation of splenic lymphocytes (coordinating with ConA: plant mitogen which is known to stimulate T-cell or LPS: endotoxin which binds receptor complex in B cells to promote the secretion of pro-inflammatory cytokines) in a dose-dependent manner. In addition, GBP significantly stimulated mRNA expression levels of immune-associated genes including interleukin-1β (IL-1β), IL-2, IL-4, IL-6, tumor necrosis factor-α (TNF-α), interferon-γ (IFN-γ), toll-like receptor 4 (TLR-4), and cyclooxygenase-2 (COX-2) in CY-treated mice. These results indicate that GBP is involved in immune effects against CY-induced immunosuppression. Thus, GBP could be developed as an immunomodulation agent for medicinal or functional food application.

고려인삼에 대한 제초제 2, 4-D의 반응연구 (Study on the Response of Korean Ginseng (Panax ginseng C.A. Meyer) to the Herbicide 2,4-D Application)

  • Various rates of 2,4-D were sprayed on 2 and 3 year old ginseng plants as foliar spray to define the critical concentration. No apparent plant injury was noticable for those ginseng plants when application concentration of 2,4-D doubled the recommended dosage (70 ml/10a). Neither abnormal foliar change occurred nor any inhibition in leaf and stem growth was resulted for the plants treated with 2,4-D concentrated two times of the recommended dosage. When the rates of 2,4-D application were increased greater than this level, injury ratings increased linearly with the rates of 2,4-D application and plant you was inhibited. Ethylene gas was not produced from the ginseng plant treated with 2 times concentrated 2,4-D, however the ginseng plants produced 0.03 to 0.09 ppm ethylene gas when the rate of application were increased 3 and 4 times, respectively. On the other hand the soybean treated with the recommended amount of 21-D produced ethylene gas of 10-20 times higher compared with ginseng plants and died. Photosynthesis ability of the ginseng leaf was significantly decreased by 2,4-D foliar application but it was recovered 4 weeks after 2,4-D foliar treatment. The herbicide 2,4-D was applicated to 2,3 and 4 years old ginseng plants as foliar spray with the rates of 0.5, 1.0, 1.5 and 2.0 times of the recommended dosage to define the effects of 2,4-D on the plant growth and root yield of the ginseng. There were no significant differences in the leaf and stem growth between untreated and 2,4-D treated plant. Berry maturing of 3 and 4 year old ginseng was not influenced by 2,4-D. The root weight of 4 years old ginseng plant was not reduced by application of 2,4-D concenrated 2 times of the recommended dosage, Application time of the herbicide 2,4-D had no effects on the leaf or stem growth of 2,3 and 4 year old year old ginseng plants. When the ginseng seedling was treated with 2,4-D, detrimental phenomena as stem bending and docoration of seedling leaf margin occurred, but stem bending was recovered in a few day s. Unraveling dynamic metabolomes underlying different maturation stages of berries harvested from Panax ginseng

  • Background: Ginseng berries (GBs) show temporal metabolic variations among different maturation stages, determining their organoleptic and functional properties. Methods: We analyzed metabolic variations concomitant to five different maturation stages of GBs including immature green (IG), mature green (MG), partially red (PR), fully red (FR), and overmature red (OR) using mass spectrometry (MS)-based metabolomic profiling and multivariate analyses. Results: The partial least squares discriminant analysis score plot based on gas chromatography-MS datasets highlighted metabolic disparity between preharvest (IG and MG) and harvest/postharvest (PR, FR, and OR) GB extracts along PLS1 (34.9%) with MG distinctly segregated across PLS2 (18.2%). Forty-three significantly discriminant primary metabolites were identified encompassing five developmental stages (variable importance in projection > 1.0, p < 0.05). Among them, most amino acids, organic acids, 5-C sugars, ethanolamines, purines, and palmitic acid were detected in preharvest GB extracts, whereas 6-C sugars, phenolic acid, and oleamide levels were distinctly higher during later maturation stages. Similarly, the partial least squares discriminant analysis based on liquid chromatography-MS datasets displayed preharvest and harvest/postharvest stages clustered across PLS1 (11.1 %); however, MG and PR were separated from IG, FR, and OR along PLS2 (5.6 %). Overall, 24 secondary metabolites were observed significantly discriminant (variable importance in projection > 1.0, p < 0.05), with most displaying higher relative abundance during preharvest stages excluding ginsenosides Rg1 and Re. Furthermore, we observed strong positive correlations between total flavonoid and phenolic metabolite contents in GB extracts and antioxidant activity. Conclusion: Comprehending the dynamic metabolic variations associated with GB maturation stages rationalize their optimal harvest time per se the related agroeconomic traits.

Ginsenoside Re Enriched Fraction (GS-F3K1) from Ginseng Berries Ameliorates Ethanol-Induced Erectile Dysfunction via Nitric Oxide-cGMP Pathway

  • Erectile dysfunction (ED) is a highly prevalent disorder that affects millions of men and considered to be an early symptom of atherosclerosis and a precursor of various systemic vascular disorders. The aim of the present study was to prepare ginsenoside Re enriched fraction (GS-F3K1, ginsenoside Re 10%, w/w) from ginseng berries flesh and to investigate the enhanced activities of GS-F3K1 on alcohol-induced ED. GS-F3K1 was prepared by the continuous liquid and solid separating centrifugation and circulatory ultrafiltration from ginseng berries flesh. GS-F3K1 was administered for 5 weeks in ethanol-induced ED rat by oral administration of 20% ethanol. To investigate the effects of GS-F3K1 on ED model, the levels of nitrite expression, cyclic guanosine monophosphate (cGMP) and erectile response of the penile corpus cavernosum of rat were measured. The erectile response of the corpus cavernosum was restored after GS-F3K1 administration, to a level similar to the normal group. The level of nitrite and cGMP expression in the corpus cavernosum of GS-F3K1-administered male rats was increased significantly compared to positive control group. GS-F3K1 from ginseng berries should effectively restore ethanol-induced ED in male rats and could be developed as a new functional food for the elderly men.

Protective effect of ginsenoside Re on acute gastric mucosal lesion induced by compound 48/80

  • The protective effect of ginsenoside Re, isolated from ginseng berry, against acute gastric mucosal lesions was examined in rats with a single intraperitoneal injection of compound 48/80 (C48/80). Ginsenoside Re (20 mg/kg or 100 mg/kg) was orally administered 0.5 h prior to C48/80 treatment. Ginsenoside Re dose-dependently prevented gastric mucosal lesion development 3 h after C48/80 treatment. Increases in the activities of myeloperoxidase (MPO; an index of neutrophil infiltration) and xanthine oxidase (XO) and the content of thiobarbituric acid reactive substances (TBARS; an index of lipid peroxidation) and decreases in the contents of hexosamine (a marker of gastric mucus) and adherent mucus, which occurred in gastric mucosal tissues after C48/80 treatment, were significantly attenuated by ginsenoside Re. The elevation of Bax expression and the decrease in Bcl2 expression after C48/80 treatment were also attenuated by ginsenoside Re. Ginsenoside Re significantly attenuated all these changes 3 h after C48/80 treatment. These results indicate that orally administered ginsenoside Re protects against C48/80-induced acute gastric mucosal lesions in rats, possibly through its stimulatory action on gastric mucus synthesis and secretion, its inhibitory action on neutrophil infiltration, and enhanced lipid peroxidation in the gastric mucosal tissue.

제초제 2, 4-D에 대한 고려인삼의 반응 I. 2, 4-D의 농도가 인삼의 생육 및 근수량이 미치는 영향 (Response of Korean ginseng (Panax ginseng C. A. Meyer) to 2, 4-D I . Effects of 2, 4-D concentrations on Growth and Root Yield)

  • 제초제 2, 4-D의 처리가 인삼의 생육 및 근수량에 미치는 영향을 구명하기 위하여 2, 4-D 제초제량 (유제 :70ml/100$\ell$/10a, 수화제 : 250g/100$\ell$/10a)의 0.5, 1.0, 2.0 배액을 2년, 3년 및 4년생 인삼에 대하여 출아후 40일에 각각 엽면처리아여 경엽의 상육과 장과의 착생 및 근수량의 변이를 조사하고 무처리대조구와 비교 분석하였던 바 그 결과를 요약하면 다음과 같다. 1. 2년, 3년 및 4년생 인삼에 2, 4-D를 일반제초약량의 2배로 경엽산포하여도 지상부의 전체적인 생육상은 무처리구와 차이가 없었으며 특별한 약해현장도 나타나지 않았다. 2. 2년, 3년 및 4년생 인삼에 있어 무처리구와 2, 4-D 유제 및 수화제 처리 농도간에 엽장, 여폭, 경장 및 경직경 등 유의차가 인정되지 않았다. 3. 3년생 및 4년생 인삼의 장과착생수는 2, 4-D 유제를 일반체초제의 2배로 처리한 구에서도 무처리구와의 유의차는 인정되지 않았으며 외관상의 형태에도 전혀 이상이 없었다. 4. 4년생 인삼에 있어 무처리구와 2, 4-D 유제 및 수화제의 처리농도간에 근장, 근식경, 지근수 및 근주의 차이는 인정되지 않았다. 5. 묘삼에 2, 4-D 유제 및 수화제의 일반제초제량을 경엽처리할 경우 줄기가 연화되어 구부러졌으며 특히 수화제 처리시는 잎의 끝이 백화고사되는 약해현장을 나타내었고 구부러진 줄기는 2~3일 후 다시 상향으로 회복되기는 하였으나 이상신장된 결과를 보였다.

인삼줄기버섯파리(신칭) Phytosciara procera에 의한 피해 실태 보고 (A Report on the Damage Caused by Phytosciara procera, Ginseng Stem Fungus Gnat)

  • 최근 2005년 파주 6년생 포지를 시작으로 철원, 영월, 괴산 등지에서 정체불명의 해충이 인삼 줄기 속을 파고들어 피해를 주는 현상이 빈번하여, 해당 해충을 채집하여 동정한 결과 국내 미기록종인 Phytosciara procera(신칭: 인삼줄기버섯파리)인 것으로 확인되었다. 이 해충은 이번 연구를 통해 처음으로 인삼 해충으로 기록되는 종이며, 검정날개버섯파리과 (Sciaridae) 곤충 중에서 인삼 해충이 확인된 것도 세계 최초이다. 인삼줄기버섯파리에 의한 피해로 추정되는 증상은 1993년 경기도 용인의 6년생 인삼 포지에서 처음으로 확인되었다. 당시 피해주율이 25%에 이르고 경작자가 막대한 경제적 손실을 입었지만, 명확한 동정 결과를 얻지 못하였고 피해 실태도 정확히 밝히지 못했었다. 인삼줄기버섯파리는 인삼의 화경, 줄기 부분에서 산란하며, 유충이 줄기 내부를 파고 섭식하여 고사시키는 해충인데, 인삼줄기버섯파리가 만든 상처는 세균의 2차 감염 통로로 이용될 수 있기 때문에 결과적으로 줄기 속무름병의 발생에도 영향을 미칠 것으로 판단된다. 인삼줄기 버섯파리 성충이 많이 발생하는 시기는 두 차례로 7월 하순부터 8월 상순 사이와 8월 하순부터 9월 상순 사이로 조사되었다. 인삼의 화경제거시 열매를 잔존시켜 건전한 형태로 남겨두거나 피해줄기를 조기에 제거하는 등의 경종적 방제를 이용하면, 인삼줄기버섯파리에 의한 피해가 확산되는 것을 방지할 수 있을 것으로 기대되지만, 아직 이 해충에 대한 생리, 생태 연구가 미흡한 만큼 기초 조사와 방제대책 수립을 위한 연구가 지속적으로 이루어져야 할 것으로 판단된다.

Melanin Biosynthesis Inhibition Effects of Ginsenoside Rb2 Isolated from Panax ginseng Berry

  • Ginsenoside Rb2 (Gin-Rb2) was purified from the fruit extract of Panax ginseng. Its chemical structure was measured by spectroscopic analysis, including HR-FAB-MS, 1H-NMR, and IR spectroscopy. Gin-Rb2 decreased potent melanogenesis in melan-a cells, with 23.4% at 80 μM without cytotoxicity. Gin-Rb2 also decreased tyrosinase and MITF protein expression in melan-a cells. Furthermore, Gin-Rb2 presented inhibition of the body pigmentation in the zebrafish in vivo system and reduced melanin contents and tyrosinase activity. These results show that Gin-Rb2 isolated from P. ginseng may be an effective skin-whitening agent via the in vitro and in vivo systems.
