2014년 제주도 양식 넙치, Paralichthys olivaceus에 대한 VHS 및 RSIVD 모니터링 (Monitoring of VHS and RSIVD in cultured Paralichthys olivaceus of Jeju in 2014)

  • The outbreak of viral diseases caused by viral haemorrhagic septicaemia virus (VHSV) and red seabream iridovirus (RSIV) have been reported in cultured olive flounder, Paralichthys olivaceus. VHSV has been a serious viral disease that infects the olive flounders in South Korea. Clinical signs of VHSV infection are skin darkening, abdominal distension and haemorrhages. Outbreaks of fish iridoviral disease was first reported from red seabream, Pagrus major farms in Japan. Recently, iridovirus infection have occurred frequently from olive flounder farms in South Korea. In this study, disease surveillance was performed to monitor the prevalence of VHSV and RSIV in olive flounder in 2014. The samples were collected from 60 different olive flounder farms in Jeju from April, May, September, November and December in 2014. RT-PCR (VHSV) or PCR (RSIV) results showed that VHSV were detected in 5 farms, but RSIV has not been detected in any farms. The migration of olive flounder was restricted for the quarantine in 5 farms of VHS outbreak. The nucleocapsid protein (N) gene and glycoprotein (G) gene sequences of the 5 Korean VHSV isolates were successfully amplified and sequenced. Phylogenetic analysis was performed using the VHSV sequences reported here together comparison with the nucleotide sequences available from the GenBank database. Phylogenetic analysis indicated that most of Korea VHSV belong to the genotype IVa and closely related to the strains from Japan and China.

  • Han In-jin (Artemisia iwayomogi Kitamura) and Cham Dang-gwi (Angelica gigas Nakai) exhibit antibacterial, antiparasitic, antifungal, and antiviral properties in vitro. In this study, mixture of the extracts of these two medicinal plants was absorbed on pellets. Thereafter, these pellets were fed to olive flounder Paralichthys olivaceus for 12 weeks at laboratory (1st experiment) and 24 weeks at field test (2nd experiment), and the immune activity and disease resistance properties of the extracts were examined. It was observed that lysozyme activities of plasma, spleen, and kidney improved after 12 weeks. Furthermore, when the olive flounders were artificially infected with bacterial pathogens, their cumulative mortality decreased in the group that was fed the extracts for 12 weeks compared to that in control group, and the relative percent survival also improved. This study concluded that mixture of Han In-jin and Cham Dang-gwi extracts provides disease resistance in vivo.

  • Triploid fish have attracted the attention of fish farmers owing to of their larger cell size compared to that of diploid fish and their ability to utilize the energy required for sexual maturation for growth instead. However, the effect of increased growth in triploid fish has shown varying results. Therefore, this study aimed to compare growth between diploid and triploid starry flounders and diploid and triploid hybrid juveniles (starry flounder, Platichthys stellatus ♀×stone flounder, Kareius bicoloratus ♂) based on breeding water temperature to investigate the effect of increased growth in triploid fishes. The experiment was performed by categorizing the water temperature as high temperature (EXP. 1, 14.5-23.5℃) and low temperature (EXP. 2, 6.4-13.3℃) based on the optimal water temperature for starry flounder breeding. In EXP. 1, the growth of starry flounder triploids and hybrid triploids was significantly higher than that of diploids. However, in EXP. 2 the weight gain and specific growth rate of the starry flounder triploid were higher, and the growth of starry flounder was higher than that of the hybrids.

Expression of Vimentin Intermediate Filament for Vascular Development in Olive Flounder (Paralichthys olivaceus)

  • Cardiovascular system is the primary organ to develop and reach a functional state, which underscores the essential role of the vasculature in the developing embryo. The vasculature is a highly specialized organ that functions in a number of key physiological works including the carrying of oxygen and nutrients to tissues. It is closely involved in the formation of heart, and hence it is essential for survival during the hatching period. The expression of genes involved during vascular development in the olive flounder (Paralichthys olivaceus) in the days after hatching is not fully understood. Therefore, we examined the expression patterns of genes activated during the development of flounder. Microscopic observations showed that formation of blood vessels is related to the expression of the vimentin gene. Also, the temporal expression patterns of this vimentin-like gene in the developmental stages and in the normal tissues of olive flounder. The purpose of this study was to examine the expression patterns of vimentin in normal tissues of the olive flounder and during the development of the vascular system in newly hatched olive flounders and HIF-1 plays a vital role in the formation of blood vessels during development. Vimentin expression was strong at the beginning of the development of blood vessels, and was present throughout all developmental stages. Our findings have important implications with respect to the roles of vimentin and HIF-1 in the development and evolution of the first blood vessels in olive flounder. Further studies are required to elucidate the vimentin-mediated hypoxic response signal transduction and to decipher the functional role of vimentin in developmental stages.

Algorithm to determine injection position in olive flounder, Paralichthys olivaceus

  • This study aimed to develop an algorithm to determine the optimal injection position in olive flounder, Paralichthys olivaceus when a vaccine is injected into the fish by using a vision-based automatic equipment measruing the total length (TL), width and weight of the fish. Over a 5-month period, 500 olive flounders were examined to analyze the relation of the fish size and to the shape and location of its abdomen, using radiography, a ruler plate and scale. There were significant correlations between the TL and the shape and location of the abdomen. The abdomen was located 0.232TL - 2.7221 mm ($R^2$=0.8787) from the end of the mouth of the fish. The height and width of the abdomen in the fish were 0.1292TL + 1.8768 mm ($R^2$=0.7935) and 0.183TL-5.9791 mm ($R^2$=0.8641), respectively. The injection point in the abdominal region avoiding organs was determined by calculating the center of gravity of the abdomen volume. This can be expressed as g (0.2759TL - 2.0965, 0.1295TL - 4.2325) on the basis of TL line coordinates. The injection point by the expressed coordinates had errors for the x and y axes as 12.15 mm and 8.28 mm, respectively. These were enough to use the algorithm to injection for the equipment. This automated method to determine the position of any part in the fish can also be used for other purposes, for example, intramuscular injection or auto-tagging of fish.

다양한 농도의 에드워드 백신액에 대한 넙치, Paralichthys olivaceus의 침지 투여 효과 (Effects of immersion vaccination in different concentration of edwardsiellosis vaccine on olive flounder, Paralichthys olivaceus)

  • 본 연구는 양식 넙치, Paralichthys olivaceus에서 희석 백신의 장시간 처리 효과를 조사하였다. 넙치에 상업용 에드워드 백신 (5㎎/ℓ)을 $10^0$, $10^{-1}$, $10^{-2}$ and $10^{-3}$로 희석하여 2분 (short immersion, SI)과 24시간 (prolonged immersion, PI) 침지한 다음 시험어의 비특이적 면역계 (혈청 라이소자임, 살균능)와 특이적 면역계 (응집항체가)의 활성과 Edwardsiella tarda 공격에 대한 방어력의 변화를 조사하였다. 그 결과 에드와드 백신을 $10^{-1}$$10^{-2}$로 희석하여 장시간 침지 투여 시험구가 고농도 백신의 단시간 침지 시험구에 비하여 혈청 라이소자임 활성과 보체의 살균능이 높았으며 에드워드균 감염에 대한 방어력도 높았다. 따라서 희석 백신을 장시간 침지 처리하는 방법은 백신 효과를 높일 수 있을 것으로 기대된다.

양식 환경이 강도다리, Platichthys stellatus의 혈액 생화학적 성상 및 lysozyme 활성에 미치는 영향 (Effects of aquaculture conditions on blood chemistry property and lysozyme activity of starry flounder, Platichthys stellatus)

  • 어병세균인 Edwardsiella tarda, Vibrio ichthyoenteri, Streptococcus iniae의 강도다리에 대한 병원성, 염분농도가 어병세균의 성장 및 강도다리의 lysozyme 활성과 사육수온, 밀도가 강도다리에 미치는 영향을 조사하였다.연구 결과, E. tarda에 대해서는 병원성이 높았으나, V. ichthyoenteri와 S. iniae에 대해서는 항병력이 높았다.염분농도에 따른 균의 증식속도 조사 결과, E. tarda와 S. iniae는 저염분에서 증식속도가 빨랐으나, V. ichthyoenteri는 저염분에서 낮았으나, 유의적인 차이는 보이지 않았다. 저염분 사육에서 강도다리의 lysozyme 활성은 일반해수구에 비하여 다소 낮게 나타났으나 유의적인 차이는 보이지 않았다.사육수온, 밀도가 강도다리에 미치는 영향을 조사한 결과, 수온은 26℃ 이하, 밀도는 수조저면적의 100% 이하가 강도다리의 생리학적 성상 및 lysozyme 활성이 높게 나타났다.

양식넙치의 Ceroid중 치료효과에 대하여 (The treatment of Ceroidosis in Cultured flounder, Paralichthys Olivaceces)

  • 1. 과산화물가가 90.4mEq/kg인 산화사료를 180일간 투여하여 Ceroid증이 발생한 어류를 1991년 10월 1일부터 11월 4일까지 Ceroid증의 치료실험을 행하였다. 2. 치료하기 위하여 정상사료(POV : 15.5mEq/kg)에 glutathione을 첨가하지 않은 실험군, 10, 20, 30, 40mg/kg/day을 첨가한 실험군 그리고 산화사료(POV : 90.4mEq/kg)만 투여한 대조군에서의 성장을 비교한 결과 대조군 및 무첨가한 실험군에서는 거의 성장이 이루어지지 않았으며, glutathione을 첨가한 모든 실험군에 있어서는 실험군별 차이없이 비슷하게 나타났다. 3. 병리조직학적으로는 glutathione 10, 30mg/kg 첨가한 실험군에서는 투여 7일째부터 ceroid 침착량 및 손상된 조직의 회복상태를 나타나기 시작하여 투여 21일 이후부터는 거의 정상적인 조직상을 나타내었다. glutathione 10mg/kg과 30mg/kg 첨가한 실험군별 조직회복상태는 10mg/kg/day 첨가한 실험군 보다 30mg/kg/day 첨가한 실험군에서가 치료효과가 양호한 것으로 나타났다.

양식넙치의 Ceroid 증 발생에 대하여 (The Development Ceroidosis in Cultured Flounder, Paralichthys olivaceus)

  • 1. 1991년 4월 1일부터 9월 27일까지 넙치의 Ceroid증 발생실험을 행하였다. 2. Ceroid증을 발생시키기 위하여 과산화물가가 90.4mEq/kg, 36.9mEq/kg, 15.5mEq/kg, 15.2mEq/kg인 산화사료를 투여한 실험군과 대조군의 성장은 과산화물가가 90.4mEq/kg인 산화사료를 첨가한 실험군에서는 3개월째 부터, 과산화물가가 36.9mEq/kg인 산화사료를 첨가한 실험군에서는 4개월째 부터 과산화 물가가 15.5mEq/kg, 15.2mEq/kg 및 대조군보다 둔화하였다. 3. 병리조직학적으로는 대조군은 정상적인 조직상을 나타냈으며 과산화물가가 높은 실험군일수록 간장, 비장과 신장조직에 Ceroid 침착량 및 조직변화가 심하였고 낮은 실험군일수록 변화가 적었으나 근육과 장조직에서는 과산화물가의 함량에 관계없이 별다른 이상을 발견할 수 없었다.

In vitro에서 Photobacterium damselae subsp. damselae와 Vibrio 속 세균의 병원성 비교 (Comparative study of Photobacterium damselae subsp. damselae and Vibrios on pathogenicity in vitro)

  • 넙치, Paralichthys olivaceus 병어에서 분리된 Photobacterium damselae subsp. damselae와 Vibrio spp.(V. anguillarum, V. splendidus, V. harveyi and V. ordalii)의 병원성을 in vitro에서 조사하였다. Extracellular products (ECPs)의 성상은 효소 활성, 양과 넙치 적혈구에 대한 용혈능과 cell-line에 대한 세포 독성을 조사하였다. 그리고 세균의 internalization에서 signal transduction pathways를 signal transduction inhibitor를 이용하여 조사하였다.P. damselae는 phospholipase 활성, 넙치 적혈구에 대한 용혈능, 세포독성이 높게 나타났으며, 분리된 vibrios 균주에 비하여 internalization에서 다양한 경로를 나타내었다. 따라서 이러한 활성은 P. damselae의 병원성과 관련이 있을 것으로 생각된다.
