Cytokine Production in Canine PBMC after Bartonella Henselae Infection (Bartonella Henselae 감염 후의 개 말초혈액단핵구에서의 사이토카인 양상)

  • Bartonella henselae is the causative agent of cat scratch disease. Although cats are the main zoonotic reservoirs of Bartonella spp., unusual cases of cat scratch disease caused by a domestic dog scratch have been recently reported. For the in vivo B. henselae infection, eight dogs were inoculated intradermally with $2{\times}10^8CFU$ of B. henselae Houston-1 suspended in 1 ml of phosphate buffered saline on day 0 and subsequent injections of the same amount given intradermally on days 21, 28, 36, 58 and 64. After in vivo canine B. henselae infection was confirmed by nested PCR, the IFN-$\gamma$ levels of the culture supernatant of PBMC stimulated with B. henselae was significantly higher in the B. henselae-PCR positive group than the B. henselae-PCR negative group. Our results showed that the canine immune responses against B. henselae were different from those of cats. Th1 activation by B. henselae stimulation was characterized in dog peripheral blood mononuclear cells, whereas Th2 activation was reported in B. henselae-infected cats.

Hemodynamic Evaluation of Acute Mitral Valve Insufficiency Model induced by Chordae Tendinae Rupture in Normal Dogs (개에서 건삭파열로 유발한 급성 이첨판 폐쇄부전 모델의 혈류역학적 평가)

  • The study was to observe hemodynamic alterations of cardiac function to design a model of canine mitral valve insufficiency (MVI) based on chordae tendinae rupture (CTR). Ten healthy beagles with normal heart function were used in this study. To measure hemodynamics, the patient monitor was equipped for invasive blood pressure and a Swan-Ganz catheter. Hemodynamic alterations were checked promptly during CTR procedures. MVI model was made by transection of the chordae tendinae with small arthroscopy hook knife through $5^{th}$ intercostal open chest. Color Doppler at the level of the mitral valve showed high-velocity regurgitant flow immediately after CTR at intraoperative echocardiography. In hemodynamic measurements, pulmonary capillary wedge pressure (PCWP) was significantly increased, while mean arterial pressure (MAP), venous pressure (VP), pulmonary arterial pressure (PAP), cardiac output (CO) and cardiac index (CI) were significantly decreased after CTR. It was known that the left atrium was overloaded by regurgitant volume from the left ventricle. In conclusion, the MVI model induced by CTR technique in this study should be used as suitable one for the effective research of canine mitral valve disease. Further study should be needed to measure the chronic alternation of mitral valve in the model.

THE HISTOPATHOLOGICAL STUDY ON THE RESPONSE OF THE REMAINING PULP TISSUES TO THE ADHESIVE RESIN AFTER PULPOTOMY (치수절단 후 접착레진 도포에 대한 잔존치수조직 반응에 관한 연구)

  • This study was performed to observe the histopathological response to the bonding resin directly applied on the remaining pulp tissues. 40 teeth from 3 adult dogs were pulpotomized with a sterile round bur and sharp excarvater. In the control group, $Ca(OH)_2$ powder was applied on the pulp tissue and the cavities were sealed with IRM cement. In the experimental group 1, Superbond C&B was applied on the remaining pulp and the cavities conditioned with 10-3 solution were filled with the mixture of the MMA liquid, PMMA powder and Catalyst. Multi-purpose adhesive was used on the remaining pulp tissue in the experimental group 2 and Z-100 was filled in the cavities. In the experimental group 3, Clearfil photobond applied and directly photo-cured on the pulp tissue, then the cavities were treated with CA agent (10% citric acid and 20% $CaCl_2$ aqueous solution) for 20 seconds, washed and applied with Clearfil photobond then filled with Protect liner. The experimental animals were sacrified at the 1st, 2nd, and 4th week. The specimens were routinely processed and stained with H-E for light microscopic observation. The results were as followed : 1. In the experimental group 1, the number and characteristics of the dentin bridge formation case was similar to those in the control group and less cases were observed in the experimental group 2 and 3 than experimental group 3. The inflammatory response in experimental group 1 was less than that in the control group at 1st week but there had been little difference at between 2nd and 4th week. 2. The number of the dentin bridge in experimental group 2 was less than that in control group and experimental group 1. The inflammatory response of the experimental group 1 was similar to that of experimental group 1 but less than that of the control group. A number of bleeding and vascular congestion were observed. The least inflammatory response was seen in the experimental group 2 among all groups. 3. In the experimental group 3, one case of the dentin bridge formation was observed and that was the same as that in the experimental group 2 but smaller than that of the control and experimental group 1. The inflammatory response of the experimental group 3 was least at the 1st week and most at the 4th week in the all group.

Efficacy of Twice a Week Selamectin Application as a Treatment for Mixed Canine Scabies and Ear Mite Infestation in a Dog (개에서 피부개선충(Canine Scabies)과 귀진드기(Ear Mite) 혼합감염에 대한 주 2회 selamectin 적용 일례)

  • A 6-month-old, male, American cocker spaniel was presented with severe pruritus, alopecia and head shaking. In physical examination generalized erythema, papules, scales and crust were observed. Otic discharge was dark-brown colored in both ear canals. After extensive dermatologic diagnostic evaluations, the infestation of scabies, ear mites, Malassezia spp, cocci were proved. Therefore, he was treated with selamectin 6 ㎎/㎏ which was applied to the skin in a single spot twice a week for 2 weeks in order to investigate the effect of selamectin on canine scabies and ear mites by this treatment protocol. When he was rechecked at 14th day after fourth being administered, scabies and ear mites wasn't seen. The dermatological signs, pruritus, papules were remarkably decreased and only slight scales were observed. After treatment of scabies and ear mites, the clavamox and koto-conazole was administered for cocci and Malassezia spp, respectively, for 1 week and topical therapy was initiated. with sebolytic and chlorhexidine shampoo once a week for 3 weeks. Two months later skin lesions were normal dermatologically and the hair was fully recovered. Despite of the expensiveness the selamectin is very effective in the dogs that infected by scabies and ear mites as this treatment protocol.

The Efficacy of Needle-Acupuncture at Nei Guan (PC06) and Xin Shu (BL15) on Canine Tachycardia (개 빈맥에 대한 내관(PCO6) 및 심수(BLl5) 자침의 효과)

  • This study examined the efficacy of needle-acupuncture (AP) at Nei Guan (PC06) and Xin Shu (BL15) on canine tachycardia. A total of 18 beagle dogs were divided into three groups: a control (6 heads), PC06 (6 heads) and PC06 (6 heads) group. Tachycardia was induced by an intramuscular injection of glycopyrrolate. No treatment was given to the control group. Needle-AP at PC06 and BL15 were applied simultaneously at the same time as the glycopyrrolate injection for 20 minutes in PC06 and BL15 groups, respectively. The changes in R-R intervals and respiration rates were determined. The changes in the R-R intervals in the PC06 and BL15 groups were significantly higher than the control group. The R-R intervals of the BL15 group were significantly higher than in the PC06 group. The changes in the respiration rates in the PC06 group were similar to those of the control group. However, the respiration rates in the BL15 group were significantly lower than the control and PC06 groups. In conclusion, needle-AP at PC06 and BL15 is effective in assisting in the recovery from induced canine tachycardia. However, needle-AP at BL15 is more effective than that at PC06.

A STUDY ABOUT EARLY OSTEOCONDUCTIVITY OF POROUS ALLOPLASTIC CARBONAPATITE AND ANORGANIC BOVINE XENOGRAFT IN CANINE MAIXLLIARY AUGMENTATION MODEL (탄산아파타이트로 된 인공골과 소뼈에서 유래한 무기질 골의 초기 골전도에 대한 연구)

  • Introduction: Although several types of calcium-phosphate coumpound have been frequently applied to osseous defects at maxillofacial area for many years, there is a controversy about its efficiency on bone conductivity comprared to xenograft bone substitute. Alloplastic carbonapatite has been introduced to improve disadvantages of hydroxyapatite and to mimic natural bone containing carbon elements. However, a preclinical study about its efficiency of osteoconductivity has not been reported. This study was performed to evaluate the early osteoconductive potential of synthetic carbonapatite with multiple pores relative to anorganic bovine xenograft. Materials and methods: Total 5 beagle dogs were used for maxillary augmentation model. The control (anorganic bovine xenograft) and experimental groups (synthetic carbonapatite) were randomly distributed in the mouth split design. After bone graft, all animals were sacrificed 4 weeks after surgery. Histological specimens with Masson Trichrome staining were made and histomorphometrically analysed with image analyser. The statistical analysis was performed using paired t-test. Results: In both groups, all animals had no complications. The experimental group showed relatively much new bone formation around and along the bone substitutes, whereas it was clearly reduced in the control group. The ratios of new bone area to total area, to material area and to the residual area excluding materials were higher in the experimental group ($0.13{\pm}0.03,\;0.40{\pm}0.13,\;0.20{\pm}0.06$ respectively) than in the control group ($0.01{\pm}0.01,\;0.03{\pm}0.02,\;0.03{\pm}0.03$, respectively). And the differences between both groups were statistically significant (p<0.001, <0.01, <0.01, respectively), while the ratio of material area to total area in two groups was not significant. Conclusion: Carbonapatite showed a high osteoconductivity in the early stage of bone healing compared to bovine derived anorganic bone substitute. This study suggests that this bone materials can be applied as a reliable bone substitute in the clinical treatment.

The Effects of Bone Morphogenetic Protein-4 and Resorbable Membrane on the Regeneration of Periodontal Tissues (골형태형성단백질과 흡수성차폐막이 치주조직 재생에 미치는 영향)

  • The aim of our study is to achieve complete periodontal tissue regeneration by the application of BMP and resorbable membrane. Three beagle dogs aged over one and half years and weighed 14 to 16 kg were used in this study. Mandibular 1st, 2nd premolars were extracted bilaterally. Horizontal furcation defects were induced around 3rd, 4th premolars bilaterally. BMP-4 were applied in the right side with resorbable membranes and only resorbable membranes were applied in the left side respectively. Each animal was sacrificed at 2, 4, and 8weeks, after regenerative surgery. Specimens were prepared with Hematoxylin-Eosin stain and Goldner's modified Masson Trichrome stain for light microscopic evaluation. The results were as follows: 1. At 2 weeks after regenerative surgery, downgrowth of junctional epithelium was observed both in the membraneapplied site and BMP-4-and-membrane-applied site. 2. At 4 weeks after regenerative surgery, resorbable membranes were completely resolved, therefore would not prevent downgrowth of junctional epithelium. New bone formation, new cementum formation and Sharpey's fiber were observed in BMP-4-andmembrane-applied site. 3. At 8 weeks after regenerative surgery, downgrowth of junctional epithelium was observed in the membrane-applied site. But, new cementum formation was observed in the same site. The extensive regeneration of new bone, new cementum and remarkable formation of Shapey's fiber were showed in BMP-4-and-membrane-applied site. 4. Resorbable membranes were resolved via the cell-mediated processes. 5. Periodontal tissue regeneration were better achieved in the BMP-4-andmembrane-applied site than in the membrane-applied site. Within the above results, BMP-4 may have the strong capability to form the new bone and resorbable membrane may be able to prevent the bony ankylosis. However, resolution rate of resorbable membrane may not be enough to protect rapid epithelial downgrowth for ideal periodontal regeneration. Bone reaction to bovine hydroxyapatite grafted in the mandibular defects of beagle dogs. (성견의 하악 골 결손부에 이식한 생체 유래 골 이식재 (OCS-B)에 대한 치조골의 반응)

  • 1. 목 적 이 연구의 목적은 성견의 하악 골 결손부에 이식한 생체 유래 골 이식재에 대한 치조골의 반응을 알아보는 것이다. 2. 연구방법 및 재료 생후 1년 이상 된 성견 4마리의 하악 제2소구치 및 제 4 소구치를 발거하고 발치와에 금원심 폭경 8mm, 협설 폭경 5mm, 치조정에서의 깊이 6mm인 결손부를 형성하였다. 4주간의 자연 치유 후 판막을 형성하여 결손부의 크기를 확인하였다. 각각의 결손부 크기가 일정하도록 수정한 후 '이식재+차폐막'군에는 OCS-B을 이식하고 Bio-gide을 차단막으로 사용한 후 봉합하고 '이식재군'은 OCS-B 이식 후 차폐막 없이 봉합하였으며 '비이식'군은 아무런 처치없이 일차봉합하였다. 수술 4, 6주에 실험동물을 각각 희생시켜 실험부위를 적출하고 비탈회 연마 표본을 제작하여 골 치유 양성을 조직학적 및 조직계측학적으로 관찰하였다. 3. 연구결과 이식재 비이식군 및 이식군 모돼서 별다른 부작용없이 잘 치유되었다. 세 실험군 모두에서 술후 4주에 비교하여 술 후 6주에서의 결손부 산생골 형성량이 증가하였다. 술후 4주 소견에서 비이식군은 결손부 주변부위에서 골이 생성되어 나오는 양상을 보였으며 이식군은 이식재 주변으로 골침착 시작되는 것을 관찰할 수 있었다. 술후 6주 소견에서 비이식군은 결손부 경계부로부터의 지속적인 골 생성을 관찰할 수 있었으며 이식군은 이식재 주변으로 침착된 골의 양이 많아지고 신생골이 가교를 형성하는 것을 관찰할 수 있었다. 4. 결 론 차폐막 유무와 상관없이 OCS-B는 염증반응을 전혀 일으키지 않았으며 우수한 골 전도성을 보였다. 또한 결손부의 형태를 잘 유지하여 골재생을 위한 공간을 확보할 수 있었다. 이는 OCS-B가 골이식재로서의 필요조건을 갖추었음을 확인한 결과이며 보다 장기적인 관찰에서 OCS-B의 흡수 가능성을 확인하는 것이 필요할 것으로 보인다.

The effect of Ca-P coated bovine bone mineral on bone regeneration around dental implant in dogs (개 모델에서의 임플란트 주위 골결손시 Ca-P 표면 처리된 이종골의 효과)

  • 목적 : 최근 발치 후 즉시 임플란트 식립은 널리 사용되는 수술 방식이다. 이 연구의 목적은 임플란트 주위 골결손시 Ca-P으로 표면 처리된 이종골을 사용하여 골재생을 평가하기 위함이다. 재료와 방법 : 두 마리의 개 모델에서 하악 소구치와 제일 대구치를 발치하였다. 발치 6주 후 trephine bur를 이용하여 7.5 mm 지름과 5 mm 깊이를 가진 결손부를 형성하였다. 이 후 이 결손부의 중앙에 3.5 mm 지름과 15mm 길이의 fixture(GS II)를 식립하였다. 결과적으로 임플란트와 주변을 둘러싸고 있는 골 사이에는 2.0 mm정도의 gap이 만들어진다. 준비된 결손부 내로 자기골 또는 $Biocera^{(R)}$를 채웠다. 각각 4주, 8주 후 조직 절편을 제작하였다. 조직학적 평가를 위해 Block biopsy를 시행하였다. 결과 : 두 집단 모두 임상적으로 골이 완전히 채워졌다. 자가골이 이식된 부위(control)의 평균 골-임플란트 접촉(BIC)은 각각 4주째 $28.2{\pm}19%$였고, 8주째 $44.9{\pm}9%$였다. $Biocera^{(R)}$가 이식된 부위(test)의 평균 BIC는 각각 4주째 $34.6{\pm}27%$였고, 8주째 $27.6{\pm}23%$였다. 자가골이 이식된 부위(control)의 평균 골밀도는 각각 4주째 $32.7{\pm}25%$, 8주째 $37.4{\pm}17%$였다. 골-임플란트 접촉(BIC)과 골밀도의 평균 비율(%)은 비슷하였다. 조직학적으로 자가골과 이종골 이식 부위 모두 주변골과 잘 조화를 이루었고 유사한 치유 양상이 관찰되었다. 자가골과 이종골 이식 부위간 유의한 차이는 없었다.(P>0.05) 결론 : 임플란트 주위 2 mm의 골 결손부위에 자가골 또는 이종골로 채운 경우 유사한 결과를 얻었다. 이 결과 임플란트 fixture 주위의 골 결손부 해소를 위해 자가골을 대체할 수 있는 재료로 $Biocera^{(R)}$를 사용할 수 있음을 보여준다.

Periodontal Regeneration Using the Mixture of Human Tooth-ash and Plaster of Paris in Dogs (성견에서 치아회분말과 연석고를 이용한 치주조직재생술)

  • 흡수성 차페막을 이용한 조직 유도 재생술시 차폐막의 견고성으로 미루어 보아 재생을 위한 공간의 유지가 어려울 수 있다. 조직 유도 재생술과 함께 골이식술을 시행함으로써 공간 확보와 함께 적절한 혈병의 유지를 도모할 수 있고 이식된 골은 선생골 형성을 위한 핵으로 작용할 수도 있다. 최근에 사람의 치아회분말과 연석고를 혼합한 골이식재가 여러 연구를 통해 좋은 골이식재로 평가되었다. 본 연구에서는 성견 하악 소구치 2급 치근이개부위에 외과적으로 형성하여 흡수성 차폐막과 치아회분말-연석고 혼합 이식재를 이용한 조직유도재생 술을 시행하여 치주 조직 재생의 양상을 조직학적으로 관찰하고자 한다. 생후 12개월에서 16개윌 된 체중 15 Kg 내외의 성견 4마리를 이용하였다. 실험 재료로 생체흡수성 차폐막 (Biogide(R), Swiss) 를 사용하였고, 골이식재로 치아회분말-연석고를 혼합매식 하였다. 양측 상악 소구치 부위에 변연 치조골하방에 4 mm ${\times}$ 4 mm ${\times}$ 4 mm, (깊이 ${\times}$ 근원심 ${\times}$ 협설폭경) 깊이로 골내낭을 형성하였다. 형성된 골내낭의 기저부위 치근 표면에 1/4 round bur로 notch를 형성하여 참고점으로 하였다. 무작위로 선택된 한 쪽의 결손부를 대조군으로 오직 생체 흡수성 차폐막을 사용하였고, 실험군으로 치아회분말-연석고와 생체 흡수성 차폐막을 결손부로부터 2 mm 이상 덮을 수 있도록 다듬어 결손부 위에 위치시킨 후 협측 판막을 덮고 봉합하였다. 4주 후 2마리 ,8주 후 2마리를 희생시키고 통상의 방법으로 고정, 탈회, 포매의 과정을 거쳐 광학 현미경으로 검경하였다. 그 결과, 1. 4주 대조군에서 Bio-gide(R)는 완전한 흡수를 보였고, 치근이개부내에는 큰 공간이 존재하였다. 2. 4주 실험군에서 역시 Bio-gide(R)는 완전한 흡수를 보였고, 골 결손부내에 더 많은 신생골 관찰되었다. 그러나 아직까진 기존골과 신생골간에 명확한 차이가 있어서 쉽게 구분할 수 있었다. 또한 골이식재 주변으로 파골세포가 다수 관찰되며 이로 미루어 보아 활발한 골흡수가 일어남을 알 수 있었다. 3. 8주 대조군에서 결손부내에서는 기존골에 인접하여 신생골 형성이 부분적으로 일어났으나 연조직 침입이 관찰되었다. 4. 8주 실험군은 신생골이 기존골과 매우 유사한 형태로 관찰되었고, 신생골 형성 부위에 신생 혈관 증식이 관찰되었다. 또한 골내낭 기저부위에서는 백악질과, 치주인대가 재생됨이 관찰되었다. 이상의 결과에서 치아회분말-연석고 혼합매식은 골재생을 위한 골전도성이 있는 재료로 사료되며, 이를 이용히여 치주조직재생술시 흡수성 차폐막과 병행하여 사용한다면 더 많은 골재생이 있을 것으로 기대된다.
