Nicotinic acid 결핍이 메추리 혈청의 여러 대사물질 수준에 미치는 효과 (Effects of Nicotinic Acid Deficiency on the Levels of Various Metabolites in the Serum of Quail)

  • Nicotinic acid 결핍시 메추리는 심각한 체중의 감소를 보였으며, 심장 및 간의 무게도 약간 감소하였다. 포도당의 농도는 현저하게 증가하였으나 콜레스테롤, 알부민 그리고 총 단백질 양의 변화는 없었다. Glutamic oxaloacetate iransaminase 와 glutamic pyruvate transaminase의 활성은 증가하였으나 alkaline phosphatase와 LDH의 활성은 변화가 없었다. 혈청속의 아미노산중 tryptophan, tyrosine, aspartic acid, glutamic acid 등의 농도는 감소하였으나, arginine, histidine, lysine 등은 변화가 없었다.

QT35 세포주에서 제조합 에리스로포이에틴 생산을 위한 무혈청 배지의 개발 (Serum Free Medium Development for Recombinant Erythropoietin Production using Novel Cell Line (QT35))

  • Human Erythropoietin (EPO) gene is cloned in quail fibrosarcoma cell, QT35. Because molecular weight of EPO is similar to that of serum albumin, cell culture with serum containing medium makes purification of EPO very difficult. Using fractional factorial study, we have developed serum free medium for the recombinant QT35 cell lines, QT N4D4 and QT SY-IMP, which have cloned EPO with glutamine synthetase (GS) gene amplification system and with puromycin selective marker, respectively. Among the seven frequently used medium components, fibronectin, BSA, and EGF were the most important for EPO production. However, sufficient fibronectin supplement to the medium did not make any good attachment of QT35 to culture plate over 3 days. Therefore, to maximize EPO production, we attempted a medium-shift at confluence from serum containing medium to serum free medium(QT SFM6). Using the medium-shift protocol with QT SFM6, nearly the same productivity of EPO was achieved comparing with that without medium-shift. This result was true in both QT35 cell lines in three types of culture, i.e. T flask, microcarrier and roller bottle cultures.

항 대사물질 6-Aminonicotinamide가 메추리 조직 내 유리 아미노산에 미치는 영향 (Effects of Antimetabolite 6-Aminonicotinamide on Levels of Free Amino Acids in Various Tissues of Quail)

  • 항 대사물질 6-aminonicotinamide가 메추리 조직의 수용성 단백질, 유리 아미노산 및 단백질 가수분해효소에 미치는 영향을 조사하였다. 간(P<0.05), 심장(P<0.01), 흉부근육 조직(P<0.05)의 수용성 단백질의 함량은 대조군에 비해 현저히 감소하였다. 신장과 흉부근육 조직의 단백질 가수분해효소의 비활성도는 대조군에 비하여 현저히 감소하였다. 신장과 흉부근육 조직의 단백질 가수분해효소의 비활성도는 대조군에 비해 현저히 증가하였다. (P<0.05). 간(P<0.05)의 aspartic acid / asparagine, alanine, valine, methionine, isoleucine, leucine, lysine의 농도는 대조군에 비해 증가하였다. 신장(P<0.05)의 acid / asparagine, alanine, threonine, alanine, proline과 lysine은 증가하였으나 glutamic acid / glutamine의 농도는 감소하였다. 심장에서는 glycine과 methionine 농도는 증가하였으나 glutamic acid / glutamine의 농도는 감소하였다. 흉부근육 조직에서는 arginine의 농도는 감소하였으나 (P<0.05) alanine과 threonine의 농도는 증가하였다 (P<0.05). 본 연구 결과는 항 대사물질인6-aminonicotinamide가 유리 아미노산의 농도를 증가시키므로써 기포 물질 대사에 필요한 에너지를 유지하는 것으로 사료된다.

메추리알 장조림에서 녹차 Epigallocatechin gallate (EGCG)의 Salmonella Enteritidis에 대한 저장 온도에 따른 항미생물 활성 (Antimicrobial activity of epigallocatechin gallate from green tea (Camellia sinensis) on pathogenic Salmonella Enteritidis in braised quail eggs)

  • 천연 녹차추출물인 epigallocatechin gallate (EGCG)의 메추리알 장조림에서 천연식품첨가제로서의 적용 가능성을 탐색하고자 S. Enteritidis에 대한 항미생물 활성을 냉장온도($4^{\circ}C$)와 일반실내온도($25^{\circ}C$)에서 평가하였다. 순수배양액에서의 실험결과 첨가된 EGCG의 농도($200-800{\mu}g/mL$)에 따라 $4^{\circ}C$에서 5일에서 16일 사이에서 그리고 $25^{\circ}C$에서 1일부터 6일 사이에서 S. Enteritidis균이 검출한계 이하로 억제되는 것이 관찰되었다. 이와 대조적으로 간장소스에서의 실험에서는 $25^{\circ}C$에서 첨가된 EGCG의 농도와 관계없이 S. Enteritidis균이 억제되지 않았으며, $4^{\circ}C$에서는 $400{\mu}g/mL$ 이상의 농도일 때 16일에 S. Enteritidis균이 검출한계 이하로 억제되는 것이 관찰되었다. 메추리알 장조림의 경우, $25^{\circ}C$에서 간장소스의 결과와 동일하게 어떠한 EGCG의 항미생물 활성이 나타나지 않았지만, $4^{\circ}C$에서 $400{\mu}g/mL$의 EGCG 농도 첨가 시 16일에 S. Enteritidis 균이 검출한계 이하로 억제되는 것이 확인되었다. 따라서, 메추리알 장조림 완제품에서 EGCG는 $4^{\circ}C$ 이하의 저온유통체계(cold chain system)하에서 S. Enteritidis과 같은 식중독세균을 제어할 수 있는 적합한 천연 식품첨가제로서 활용될 수 있을 것으로 기대된다.

가임기 여성의 주요 엽산급원식품 및 동 식품의 엽산함량 분석 (Major Foods for folate and Their Folate Contents of Korean Child-bearing Women)

  • In this study, we determined major foods and food groups which contributed to the intake of folate in korean child-bearing women. Folate content of the major foods was also analyzed. A total of 91 healthy women with childbearing potential participated. They were divided into one of three groups by age; A (15∼24 yrs), B(25∼34 yrs) and C (35∼49 yrs). Food consumption data of the subjects were obtained by the 24-hr recall method. The rate of contribution of each food to the total intake of dietary folate was determined. Seventy major foods for folate were selected from the 178 foods they consumed. Two sets of the 70 foods were collected from different markets. After extracting folates from the foods by heating with a HEPES-CHES buffer, the samples were treated with α-amylase, protease and folate intakes was Korean cabbage kimchi followed by laver, soybean sprout, rice, yolmu kimchi, eggs, lettuce, perilla leaves, sea mustard and radish root. Food items that contributed to folate intake were slightly different among the age groups. Although the folate content of Korean cabbage kimchi was 51.6 ㎍/100g, but due to the high amount of intake, it was ranked the first major food prviding folate. High folate foods containing over 100㎍/100g were laver, corn flakes, spinach, mungbean, sesame, quail's eggs, small radish kimchi, kidney beans, leeks and peanuts. However, based on the serving size, spinach, corn flakes, leeks, sweet potato, pepper leaves, quail's eggs, crown daisy, small radish kimchi and perilla leaves contributed above 50㎍ of folate per serving size.

종합병원 직원의 기호도 조사 (A Survey of Food Preferences of Employees in Hospital)

  • This study was carried out to obtain the menu preferences of frequently served set menu of the staff working at Samsung Medical Center. The frequently served set menus were 72 menus including 23 Korean meals, 12 bowel of rices, 18 noodles, 10 soups, 6 fried rices, 3 western meals. The subjects of this study were 624 staffs who works at Samsung Medical Center. The results are summarized as followers : The subject group for this study was composed of 23% males and 58% females, 80% of subjects are twenties and thirties. The best preferred menu was fried food 4(boiled rice with assorted mixtures, boiled quail egg). And noodles 8(cold buckwheat noodles, fried mung-beans), noodles 2(buckwheat noodles, fried vegetable), Western meals 3(hamburg steak, steamed dumpling), noodles 18(rice-cake dumping soup, fried sweet potato with sugar syrup) were preferred by the subjects. The worst preferred menu was soups 4(loach soup, fried sea food and welsh onion). In the preference each kinds of set menus, Korean meals 3(soup to relive the hangover with pollack, grilled meat, lettuce, fruit salad, Kimchi, ice tea) was the most preferred of the Koreans meals, bowel of rice 8(omelet containing fried rice, chicken salad) of the bowel of rices, noodles 8(cold buckwheat noodles, fried mung-beans) of the noodles, soups 3(spicy beef soup, chicken salad) of the soups, fried rice 5(boiled rice with assorted mixtures, boiled quail egg). There was no significant sexual difference in the menu preferences except 7 menus. There was significant age difference in the 17 menu preference, the younger age subjects were the better like noodles.

Cook-Chill System과 Sous vide Cook-Chill System으로 생산된 메추리알 어묵조림의 저장기간에 따른 미생물적 품질 및 관능특성의 변화(2) (Changes in the Microbial Qualities and Sensory Characteristics of Boiled Quail Egg and Fish Paste in Soy Sauce as Prepared with the Cook-Chill System and Sous Vide Cook-Chill System)

  • This study was performed to provide basic data for the sous vide cook-chill(SVCC) System by comparing and evaluating food quality, according to preparation method and days of storage, of foods that were prepared by SVCC to those prepared using the cook-chill(CC) System. Boiled quail eggs and fish paste in soy sauce were prepared using CC and SVCC and their quality was evaluated at the time of preparation and by days of storage. Viable cell counts were increased to 5.65(CC) and 3.40(SVCC) LogCFU/g by 15 days(p<.0001) and increased more over time in foods prepared with CC than with SVCC. For the CC method, Coliform counts increased to 4.58 LogCFU/g by 15 days(p<.0001). With SVCC, colirorms were not detected at 0 days, but counts reached 3.70 LogCFU/g by 15 days(p<.0001). After reheating, no coliforms were detected for CC or SVCC at 0 days, however, by 15 days, coliform counts reached 3.61 and 2.52 LogCFU/g(p<.0001) for CC and SVCC, respectively. Finally, the sensory scores of SVCC were higher than those of CC.

Purification and Characterization of [Ala2]-Neuromedin N from the Visceral Tissue of the African Lungfish, Protopterus dolloi

  • A new biologically active peptide with structural similarity to neuromedin N (NMN) has been isolated from extracts of visceral tissue of the African lungfish, Protopterus dolloi, using the rectum of the quail as the bioassay system. The primary structure of NMN-related peptide was established as Lys-Ala-Pro-Tyr-Ile-Leu-OH ([$Ala_2$]-NMN) and contained one substitution ($Ala_2\rightarrow$Ile) compared with the porcine NMN. [$Ala_2$]-NMN was found to have an excitatory effect on rectal muscle tissues of quail (Coturnix japonica), newt (Cynops pyrrhogaster) and black bass (Micropterus sulmoides). The threshold concentration of [Ala2]-NMN for contraction of C. japonica muscle was found to be approximately $10^-11$M. [$Ala_2$]-NMN showed contractile activities in the following order: C. japonica > C. pyrrhogaster > M. sulmoides. The identification of [Ala2]-NMN provides evidence that NMN family, hitherto confined to mammals, has a widespread occurrence in lungfish.

DNA methyltransferase 3a is Correlated with Transgene Expression in Transgenic Quails

  • DNA methyltransferases (DNMTs) are closely associated with the epigenetic change and the gene silencing through the regulation of methylation status in animal genome. But, the role of DNMTs in transgene silencing has remained unclear. So, we examined whether the knockdown of DNMT influences the reactivation of transgene expression in the transgenic quails. In this study, we investigated the expression of DNMT3a, and DNMT3b in blastoderm, quail embryonic fibroblasts (QEFs) and limited embryonic tissues such as gonad, kidney, heart and liver of E6 transgenic quails (TQ2) by RT-PCR. We further analyzed the expression of DNMT3a at different stages of whole embryos during early embryonic development by qRT-PCR. DNMT3a expression was detected in all test samples; however, it showed the highest expression in E6 whole embryo. Embryonic fibroblasts collected from TQ2 quails were treated with two DNMT3a-targeted siRNAs (siDNMT3a-51 and siDNMT3a-88) for RNA interference assay, and changes in expression were then analyzed by qRT-PCR. The siDNMT3a-51 and siDNMT3a-88 reduced 53.34% and 64.64% of DNMT3a expression in TQ2 QEFs, respectively. Subsequently the treatment of each siRNA reactivated enhanced green fluorescent protein (EGFP) expression in TQ2 (224% and 114%). Our results might provide a clue for understanding the DNA methylation mechanism responsible for transgenic animal production and stable transgene expression.

메추리와 오골계 초생추의 생체중과 두 및 장기중량에 관한 연구 (Study on the Visceral Organs, Head and Body Weight in Day-Old Chicks of Quail and Silky Fowl)

  • 본 시험에 공시된 오골계 50수는 본 대학교 가금학교실에서 부화한 것이고. 메추리 50수는 경남금해에서 부화한 것이었다. 본 시험은 부화후 24시간경에 실시하였으며, 조사항목은 12개로서 생체중, 두, 안구, 뇌, 심장, 간장, 신장, 위, 소장, 대장 및 란황낭의 중량을 측정한 종과는 다음과 같았다. 1. 메추리의 조사항목중, 생체중량은 6.54$\pm$0.082g, 두의 중량은 1.38$\pm$0.013g, 안구의 중량은 0.29$\pm$0.004g, 뇌의 중량은 0.24$\pm$0.006g, 심장의 중량은 0.08$\pm$0.002g, 간장의 중량은 0.21$\pm$0.005g, 폐의 중량은 0.06$\pm$0.002g, 신장의 중량은 0.05$\pm$0.002g, 위의 중량은 0.46$\pm$0.010g, 소장의 중량은 0.24$\pm$0.008g, 대장의 중량은 0.10$\pm$0.004g 그리고 란황낭의 중량은 0.28$\pm$0.016이었다. 2. 오골계의 조사항목중, 생체중량은 26.60$\pm$0.491g, 두의 중량은 4.28$\pm$0.071g, 안구의 중량은 0.64$\pm$0.039g, 뇌의 중량은 0.55$\pm$0.025g, 심장의 중량은 0.21$\pm$0.012g, 간장의 중량은 0.77$\pm$0.030g, 폐의 중량은 0.31$\pm$0.006g, 신장의 중량은 0.08$\pm$0.006g, 위의 중량은 1.53$\pm$0.076g, 소장의 중량은 0.73$\pm$0.036g, 대장의 중량은 0.42$\pm$0.031g 그리고 란황낭의 중량은 2.65$\pm$0.297g 이었다. 3. 메추리와 오골계의 중란중량에 대한 생체중량 및 각 장기중량의 비율을 타 실용계와 비교한 결과, 메추리의 안구중량과 뇌중량은 타 실용계와 오골계에 비하여 월등히 큰 것이었고, 오골계의 뇌의 중량, 심장의 중량, 간장의 중량, 폐장의 중량, 신장의 중량, 위의 중량 및 소장의 중량은, 타 실용계와 메추리에 비하여 적은 경향이었다는 것이 특이하였으며, 그 외의 항목간에는 대동소이하였다.
