Construction of Ovine Customer cDNA Chip and Analysis of Gene Expression Patterns in the Muscle and Fat Tissues of Native Korean Cattle (cDNA microarray를 이용하여 한우의 근육과 지방조직의 유전자 발현 패턴 분석 및 bovine customer cDNA chip 구성 연구)

  • To investigate the molecular events of controlling intramuscular fat (or marbling), which is an important factor in the evaluation of beef quality, we performed cDNA microarray analyses using the longissimus dorsi muscle and back fat tissues. For this study, we constructed normalized cDNA libraries: fat tissues in native Korean cattle (displaying 1,211 specific genes), and muscle tissues in native Korean cattle (displaying 1,346 specific genes). A bovine cDNA chip was constructed with 1,680 specific genes, consisting of 760 genes from muscle tissues and 920 genes from fat tissues. The microarray analysis in this experiment showed a number of differentially expressed genes, which compared the longissimus dorsi muscle (Cy5) with back fat tissue (Cy3). Among many specific differentially expressed genes, 12-lipoxygenase (oxidizing esterified fatty acids) and prostaglandin D synthase (differentiation of fibroblasts to adipocytes) are the key candidate enzymes that should be involved in controlling the accumulation of intramuscular fat. In this study, differentially and commonly expressed genes in the muscle and fat tissues of native Korean cattle were found in large numbers, using the hybridization assay. The expression levels of the selected genes were confirmed by semi-quantitative RT-PCR, and the results were similar to those of the cDNA microarray.

Seroprevalence of Paratuberculosis in Pure-bred Breeding Cattle in Korea (국가 종축우에 대한 요네병 항체 양성률 조사)

  • Paratuberculosis (Johne's disease), a chronic wasting disease caused by Mycobacterium avium subspecies paratuberculosis (MAP), is a major cause of economic loss in the cattle industry. In Korea, national monitoring of breeding stock for MAP has been implemented. In this study, we report the results of serological testing to determine the prevalence of MAP in breeding stock of Korean native and dairy cattle during 2008 and 2009. A total of 3,927 serum samples were submitted (3,692 Korean native cattle and 235 dairy cattle) to Animal Disease Diagnostic Division, Animal, Plant and Fisheries Quarantine and Inspection Agency. The samples were classified into four different age groups for MAP; group 1 (${\leq}2$ year, n = 1,509), group 2 (>2 years to ${\leq}3$ years, n = 486), group 3 (>3 years to ${\leq}4$ years, n = 441), and group 4 (>4 years, n = 1,491). Overall seroprevalence of MAP in this study was 0.5% (21/3,927), which was much lower than that of conventional cattle (1.2-16.4%) in Korea. Also, the seroprevalence was determined by age groups: three of group 1 (0.2%), two of group 2 (0.4%), three of group 3 (0.7%), and 13 of group 4 (0.9%) were seropositive for MAP, respectively. Although seropositive samples were found in all age groups, the seroprevalence tended to increase with age. Our study showed that the seroprevalence of MAP in pure-bred breeding dairy cattle (0%) was lower than that in pure-bred breeding Korean native cattle (0.6%).

Effect of $\textrm{Ca}^{2+}$ Concentration on Electric Activation of In Vitro Matured Oocytes of Korean Native Cattle ($\textrm{Ca}^{2+}$ 씨오투 농도가 체외성숙 유래 한우 난자의 전기적 활성화에 미치는 영향)

  • The present study was undertaken to examine the critical effect of $Ca^2$+ concentration on electrostimulation and post-electrostimulation media for electric activation of in vitro matured oocytes of Korean Native Cattle. Oocytes collected from slaughterhouse ovaries were matured in TCM 199 containing FSH, estradiol-17$\beta$ and FBS with granulosa cell monolayer for 24 hours and denuded with hyaluronidase. And then cumulus-free oocytes were submitted to a DC field of 1.0 kV/cm for 60 $\mu$sec in electroporation media(0.28 M mannito' and PBS) with different $Ca^2$+ concentations (0.00, 0.05, 0.10 and 0.15 mM). Stimulated oocytes were stained and examined for pronuclear formation after incuhation in SOF for 12 hours. The rates of pronuclear formation in hovine oocytes electrically stimulated in 0.28 M mannitol with 0.05, 0.10 and 0.15 mM $Ca^2$+(60.3, 82.2 and 75.0%) were significantly higher than without $Ca^2$+(6.3%) at 12 hours after an electric pulse(p<0.005). The activation rates of Korean Native Cattle oocytes stimulated in PBS supplemented with 0.05, 0.10 and 0.15 mM $Ca^2$+(71.0, 75.8 and 75.4%) were significantly higher than without $Ca^2$+(23.5%) after post-stimulation incubation(p<0.005). After incubation of oocytes in SOF with and without $Ca^2$+ following electric stimulation in 0.28 M mannitol with 0.10 mM $Ca^2$+, the rates of pronuclear formation of bovine oocytes in $Ca^2$+-free SOF(85.7%) was significantly higher than in SOF with 1.71 mM $Ca^2$+(62.5%, p<0.05). When oocytes were stimulated in two electrostimulation media supplemented with $Ca^2$+ and incubated in $Ca^2$+-free SOF, there were no significant differences in the rates of pronuclear formation hetween 0.28 M mannitol and PBS. These results indicate that a single electric pulse could induce activation of Korea Native Cattle oocytes in 0.28 M mannitol and PBS supplemented with $Ca^2$+. Furthermore, to improve the activation rates, it was hetter that stimulated oocytes were incubated in $Ca^2$+-free SOF after electric stimulation than in SOF with $Ca^2$+.

An Outbreak of Lnugworm Disease in a Flock of Korean Native Cattle (한우사육단지(韓牛飼育團地)에 발생(發生)한 우폐충증(牛肺蟲症)에 대하여)

  • In a pasture located at Kyong Puk area, a group of 200 Korean native casttle showed severe coughing, dyspnea and emaciation, of which 30 cattle died from the respiratory disease. To the alived cattle, clinical, pathologicoparasitological observations and treatment with terramycin, Mintic and tetramisole (Bonasol) were carried out and the following results were obtained. 1) Paroxymal coughighings, increased respiratory rate and heart rate were observed in almost all cases. But, larval output in feces was recognized in 32 percent of the cases. 2) Morderate eosinophilia was observed in a half of the cases. 3) At autopsy, lungs were enlarged, and areas of dark consolidations were scattered in all lobes. In the bronchi, numerous adult lungworms were found. 4) The adult worms detected from the bronchi were morphologically identified as Dictyocaulus viviparus. 5) The clinical signs and larval output were disappeared after the administration of Mintic or Bonasol. 6) From these results, it was concluded that the respiratory disease of the cattle was caused by the infestation of lungworm, Dictyocaulus viviparus.

A survey for tick-borne pathogens in Korean native cattle from northern area of Gyeongbuk (경북 북부지역의 한우에 대한 진드기매개성 병원체의 감염실태 조사)

  • This study was carried out to investigate the tick-borne pathogens from one hundred nineteen cattle farms (38 farms of Andong, 41 of Yeongju, 12 of Uiseong, 5 of Cheongsong, 5 of Yoengyang, 18 of Bonghwa) in northern areas of Gyeongbuk province by polymerase chain reaction (PCR). Among 119 cattle farms, the positive ratios against Babesia, Theileria, Anaplasma, Ehrlichia and Rickettsia were 3.4% (4/119), 10.1% (12/119), 6.7% (8/119), 1.7% (2/119) and 16.8% (20/119), respectively. Also, the PCR results revealed that 8 farms were positive for T. sergenti in positive of Theileria and 2 farms were positive for A. phagocytophilum in positive of Anaplasma. Therefore, further studies regarding vectors, environmental condition, interaction between domestic and wild animals and development of control program are needed to reduce the numbers of bovine tick-borne disease in northern areas of Gyeongbuk province.

Development Strategies of The Hanwoo [Korean Native Cattle] Industry (한우산업의 발전전략)

  • The structure of the Hanwoo (Korean Native Cattle) Industry remains very weak and vulnerable to the WTO/IMF system. Considering that the majority of cattle farmers are small sized, and that marketing systems are outdated and inefficient, rapid expansion of lower priced beef imports by WTO system and rapid increasing of production costs by IMF system would lead to the deprivation of a regular source of farm income and threaten the stability of rural life. Accordingly, the Hanwoo industry should be expanded in accordance with progress in the implementation of (1) programs for the structural adjustment and (2) measures to compensate for the loss. Efforts for lowering major production factor costs needs to continue, In order to increase the supply of calves at low cost, the programs of collective cow-calf farms should be expanded, thereby reducing the cost of calf purchase, which constitutes the largest share of Hanwoo production cost. Also, feedlot operations should be encouraged for small herd farms in order to achieve a substantial saving in beef production costs by integrated operations from calf production to cattle fattening. A substantial saving would also be made by collective purchase and distribution of various inputs through the cooperatives' channels. Extension services should be strengthened for cattle farm management, cattle care and feeding, prevention of cattle disease, etc. In order to minimize cash outlays for commercial mixed feeds, utilization of far by-products as feeds should be enhanced and production of forage crops productive of resources, such as land and rural labor, during the farm o(f-season, needs to be encouraged. Also, technological development for enhancing the nutritional value of farm by-products should be encouraged. Measures for successful segregation of the Hanwoo beef market should be implemented, thereby enhancing incentive for quality beef producers and protecting consumers willing to pay higher price for quality beef. For development of the Hanwoo industry, a considerable time frame would be required in order for (1) small livestock farmers to make a successful adjustment by staying in the enterprise and achieving increased price efficiency (2) livestock farmers to acquire know-how for producing quality Hanwoo beef, (3) the ongoing Government policy of enhancing price-quality competitiveness, and for improving the quality Hanwoo beef marketing to take root. (4) consumers to increase their ability to distinguish meat quality, and others.

  • The livers of 300 cases of Korean native cattle which were collected at slaughted house in Pusan were examined by gross and histopathological means to study patterns of pathological changes of the organ. 1. The most dominated lesion of the liver was those of fascioliasis showing incidence rate as 907 per cent of inspected cattle. 2. On age distribution of the fascioliasis lesion in the liver, higher incidence was seen in animals that were from 6 to 9 years of age. 3. Significant changes of the hepaticparenchyma caused by immature flukes included thrombosis, migrational tracks such as hemorrhagic foci with massive eosinophilia and hepatic necrosis. The changes of chronically infected livers of cattle were recognized as proliferation of periductal eosinophilic granuloma of various degrees in each portal triad. And this easential cholangiohepatitis might develop into multiple focal cirrhosis or diffuse perilobular cirrhosis. 4. Most cases with gross lesion of swelled bile ducts on visceral surface of the liver showed histological evidences of fascioliasis lesion throughout hepatic lobes.

Survey on Housing Types of Korean Native Cattle and Beef Cattle by Farm Scale and Region (한우농가의 사육규모별, 지역별 축사시설 현황 분석)

  • This survey was conducted to investigate housing types of Korean native cattle and beef cattle farms in Korea. The farm sized over 50 heads of cattle, 7,433 farms were surveyed. Regarding housing types for Korean native cattle and beef cattle, litter barn, freestall, mooring+litter ground and others accounted for 87.1, 9.8, 3.0, and 2.9 %, respectively. Most of Korean native cattle and beef cattle farms (94.7%) used litter floor rather than scraper. As for roof types, slate, panel, iron plate, galvanized plate, colored iron plate and sunlight represented 32.2, 13.7, 12.2, 10.2, 9.7, and 8.9 %, respectively. Open side wall type and winch curtain were 55.6, and 47.6 %, respectively. Bigger farms seemed to used more winch curtain than open style. Utilization period of automatic feeder, waterer, electric facility, and cooling facility was 6.7, 8.0, 8.5, and 6.0 years, respectively.

Research on the Working Environment and Personal Protective Equipment of Korean Native Cattle Raising Farmers (한우 사육자 작업환경 및 작업복과 보호구 착용 실태 분석)

  • This study examined the working site environment of Korean native cattle raising farmers and their actual condition of wearing personal protective equipment as part of preventing disasters during agricultural work. To this end, 160 Korean cattle raising farmers were surveyed and a site visit was performed on 10 farms. According to an examination of the cattle breeding environment, the major harmful factors were in the following order: fine dust (12.8%), organic feed dust (10.3%), contact with cattle (9.7%), manure (8.2%), germs and viruses (8.1%), harmful gases (7.4), contact with obstacles (6.7), and temperature (6.6%). The current status of the rate of wearing protection was in the order of gloves (20.9%), working hats (19.7%), boots (19.6%), masks (10.9%), protective clothing (8.9%), and specialized working clothing (6.6%). Nevertheless, most Korean cattle raising farmers recognized the risks when they did not wear protective equipment and the need for wearing protectors, but they mostly did not wear personal protective equipment due to a lack of knowledge on the selection of appropriate personal protective equipment and the discomfort they experience when they put on protectors. Even when they put on protective equipment, 38% was inappropriate for the farming work environment. Given the research results, improvements on and the development of specialized working clothing and personal protective equipment to protect farmers from harmful and dangerous materials from the cattle nurturing environment is necessary. Overall, based on the study data, objectified data collection, a determination of the necessary performance elements of personal protective equipment, and R&D will be needed through an on-site current status investigation.
