Mountain Ginseng Pharmacopuncture Treatment on Three Amyotrophic Lateral Sclerosis Patients (산삼약침 치료를 시행한 근위축성 측삭 경화증 3례에 대한 임상보고)

  • Objectives : The objective of this study is to report the change of progress in symptoms and various scales after treated with Mountain Ginseng Pharmacopuncture(MGP) on the patients of Amyotrophic Lateral Sclerosis(ALS). Methods : The three ALS patients who treated with MGP, were checked the change of progress by ALS Functional Rating Scale(ALSFRS), ALS Severity Score(ALSSS), grasping power on both arms and circumference of both thighs and calves. Results : After MGP treatment on three ALS patients, first case did not grow worse during MGP treated whereas she got worse rapidly during none treated period. Second case did not grow worse during treated period and third case got worse in progress by slow degrees despite of MGP treatment. Conclusions : Although MGP could not control the progress of ALS completely, MGP may help the improving of quality of life(QOL) in ALS patients and have the effect of delayed ALS progression.

Research on the Amount of Stimulus Differences According to Pharmacopuncture Injected dose and Characters Method (약침 주입 용량 및 자입 방법에 따른 자극량 차이 연구)

  • Objectives : This study was designed to discover the difference in stimulation effect when the volume and insertion method of mountain ginseng pharmacopuncture was varied. Methods : 48 healthy people were randomly injected with 0.1, 0.3, 0.5, and 0.7 cc of mountain ginseng pharmacopuncture, perpendicularly or transversely at acupoints $LI_9$ and $LI_{10}$. The subjects noted their level of stimulation on a 100 mm visual analog scale(VAS) immediately after, 3 minutes after, and 5 minutes after the treatment. Results : There was no significant statistical difference in the stimulation strength between perpendicular injection and transverse injection pharmacopuncture. There was significant statistical difference in the stimulation strength when the volume of pharmacopuncture was varied. As the volume of pharmacopuncture increased from 0.1 cc to 0.3 cc, and to 0.5 cc, the stimulation strength increased as well. Conclusions : We found that volume of pharmacopuncture may be a controlling element for mountain ginseng pharmacopuncture. Additional study is needed for pharmacopuncture's stimulation adjusting factors.

Change of Ripened Persimmon Vinegar with Mountain Ginseng Ingestion on Energy Metabolism in Rats (산양삼 혼입 숙성 감식초 섭취에 의한 흰쥐의 에너지 대사 변화 연구)

  • In this study, a 4-year-old mountain ginseng was mixed and ripened with 4-year-matured persimmon vinegar, and then it was diluted 5 times and orally administerd to rats. Afterwards, by analyzing the protein expression rate which affects both the carbohydrate metabolism and the lipid metabolism, this study examined the anti-obesity effect of the fusion material. The rats were divided into a control group (CON), a persimmon vinegar group (PV) and a mountain ginseng+persimmon vinegar fusion material group (MPV). The weight gain rate was found to be low in PV and MPV, and the concentration of glucose was also low in PV and MPV. However, GLUT-2 was found to be significantly high in these two groups on the contrary. Both the concentration of free fatty acid and CPT-1 protein expression rate were high in PV and MVP, but MVP was higher than PV. Cytochrome C oxidase was found to be higher in MPV than in CON. AMPK, $PPAR-{\gamma}$ and $PGC1-{\alpha}$ were all high in PV and MPV, but MPV was higher than PV. All the results above verified the thermogenesis effect of the fusion material, leading to an increase of energy metabolism, and it was thought that the fusion material could be effectively used for anti-obesity. However, it seems necessary to verify the anti-obesity effect through various further studies.

Mountain-cultivated ginseng protects against cognitive impairments in aged GPx-1 knockout mice via activation of Nrf2/ChAT/ERK signaling pathway

  • Background: Escalating evidence shows that ginseng possesses an antiaging potential with cognitive enhancing activity. As mountain cultivated ginseng (MCG) is cultivated without agricultural chemicals, MCG has emerged as a popular herb medicine. However, little is known about the MCG-mediated pharmacological mechanism on brain aging. Methods: As we demonstrated that glutathione peroxidase (GPx) is important for enhancing memory function in the animal model of aging, we investigated the role of MCG as a GPx inducer using GPx-1 (a major type of GPx) knockout (KO) mice. We assessed whether MCG modulates redox and cholinergic parameters, and memory function in aged GPx-1 knockout KOmice. Results: Redox burden of aged GPx-1 KO mice was more evident than that of aged wild-type (WT) mice. Alteration of Nrf2 DNA binding activity appeared to be more evident than that of NFκB DNA binding activity in aged GPx-1 KO mice. Alteration in choline acetyltransferase (ChAT) activity was more evident than that in acetylcholine esterase activity. MCG significantly attenuated reductions in Nrf2 system and ChAT level. MCG significantly enhanced the co-localization of Nrf2-immunoreactivity and ChAT-immunoreactivity in the same cell population. Nrf2 inhibitor brusatol significantly counteracted MCG-mediated up-regulation in ChAT level and ChAT inhibition (by k252a) significantly reduced ERK phosphorylation by MCG, suggesting that MCG might require signal cascade of Nrf2/ChAT/ERK to enhance cognition. Conclusion: GPx-1 depletion might be a prerequisite for cognitive impairment in aged animals. MCG-mediated cognition enhancement might be associated with the activations of Nrf2, ChAT, and ERK signaling cascade.

Comparison of Physicochemical Properties of Extruded Ginseng Samples

  • This study compared the physicochemical properties of root hair of white ginseng (WG), root hair of tissue cultured mountain ginseng (MG), root hair of red ginseng (RG) and extruded ginseng samples. The comparison of crude ash and total sugar resulted insignificant differences between extruded and raw samples. MG had a higher content of crude ash, crude protein, amino acids and polyphenolic compound than WG and RG; the total sugar and reducing sugar were highest in RG. Crude fat and acidic polysaccharide in RG and WG were similar to and higher than MG. Crude saponin of treated samples WG1 (moisture content 25%, barrel temperature $110^{\circ}C$) and WG3 (moisture content 35%, barrel temperature $110^{\circ}C$) were 9.80% and 9.73%, respectively, which were the highest among ginseng samples. In conclusion, the extrusion process can be applied to red ginseng manufacturing, and some characteristics of MG were higher than in RG and WG.

Study on Sleeping Behaviors of The Combined-Preparation of Crude Drugs -on The $Well^{TM}$ Preparation- (복합한약제제의 수면에 대한 연구 -더웰 제제에 대하여-)

  • This experiment was performed to investigate whether the combined-preparation of crude drugs (The $Well^{TM}$ Preparation, TW), has hypnotic effects and/or enhances pentobarbital-induced sleep behaviors. TW was mixed with water extracts of Ginseng Radix red, Germinated brown rice, cultured mountain ginseng, and 50% ethanol extracts of Longanae Arillus, Nelumbinis Folium and Chrysanthemi Flos. TW (100 mg/kg, p.o.) reduced sleep onset and prolonged sleep time induced by pentobarbital similar to muscimol (0.2 ${\mu}M$), a $GABA_A$ receptor agonist. Also, TW (2 ${\mu}g$/ml) and pentobarbital (2.5 ${\mu}M$) did not affect the chloride influx in primary cultured cerebellar granule cells, respectively, but the combined-treatment of TW (2 ${\mu}g$/ml) and pentobarbital (2.5 ${\mu}M$) increased the chloride influx onto the cells. In conclusion, TW augments pentobarbital-induced sleep behaviors; these effects may result from chloride channel activation.

Pancreatic Lipase Inhibitors Isolated from the Leaves of Cultivated Mountain Ginseng (Panax ginseng) (산양삼 잎으로부터 Pancreatic lipase 저해 활성물질의 분리)

  • Activity-guided fractionation of an ethyl acetate (EtOAc)-soluble portion of an ethanolic extract from the leaves of cultivated mountain ginseng, using pancreatic lipase inhibition assay, led to the isolation and identification of three flavonoids of a previously described structure, kaempferol-3-O-sophoroside (I), kaempferol-3-O-${\beta}$-Dglucopyranoside (astragalin, II) and kaempferol (III). All compounds (I.III) showed pancreatic lipase inhibitory activities, with $IC_{50}$ values ranging from $20.3{\pm}2.2$ to $9.1{\pm}1.5$ ${\mu}M$, kaempferol (III) showed the most potent inhibitory activity with an $IC_{50}$ of $9.1{\pm}1.5$ ${\mu}M$. The level of activity may depend on the number of C-3 glucosyl group(s) linked to the kaempferol backbone, and the isolated compounds may have promise as pancreatic lipase inhibitors.
