Hypoglycemic Activity of the Hexane Extract of Oriental Melon (Cucumis melo L. var. makuwa Makino) Seeds and Its Active Compounds

  • The aim of this work is to evaluate the potential of oriental melon (Cucumis melo L. var. makuwa Makino) seeds for the management of type 2 diabetes by controlling glucose absorption. The ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibitory effects of the hexane extracts from oriental melon seeds were investigated. A bioassay-guided fractionation technique was used to elucidate the principal active components. The results show that the hexane extract from oriental melon seeds exhibited high inhibitory activities against ${\alpha}$-glucosidase and ${\alpha}$-amylase. The hexane extract was further fractionated into four sub-fractions. Among them, the sub-fraction F-1 exhibited the most potent anti-diabetic effect. The active components were isolated and identified by gas chromatography/mass spectroscopy (GC-MS). Free fatty acids showed significant hypoglycemic activity (p < 0.001) and fatty acid composition influenced enzyme inhibitory activities. These results suggest that oriental melon seeds could be used to prevent type 2 diabetes.

Key-point detection of fruit for automatic harvesting of oriental melon (참외 자동 수확을 위한 과일 주요 지점 검출)

  • In this study, we suggested a key-point detection method for robot harvesting of oriental melon. Our suggested method could be used to detect the detachment part and major composition of oriental melon. We defined four points (harvesting point, calyx, center, bottom) based on tomato with characteristics similar to those of oriental melon. The evaluation of estimated key-points was conducted by pixel error and PDK (percentage of detected key-point) index. Results showed that the average pixel error was 18.26 ± 16.62 for the x coordinate and 17.74 ± 18.07 for the y coordinate. Considering the resolution of raw images, these pixel errors were not expected to have a serious impact. The PDK score was found to be 89.5% PDK@0.5 on average. It was possible to estimate oriental melon specific key-point. As a result of this research, we believe that the proposed method can contribute to the application of harvesting robot system.

De Novo Transcriptome Analysis of Cucumis melo L. var. makuwa

  • Oriental melon (Cucumis melo L. var. makuwa) is one of six subspecies of melon and is cultivated widely in East Asia, including China, Japan, and Korea. Although oriental melon is economically valuable in Asia and is genetically distinct from other subspecies, few reports of genome-scale research on oriental melon have been published. We generated 30.5 and 36.8 Gb of raw RNA sequence data from the female and male flowers, leaves, roots, and fruit of two oriental melon varieties, Korean landrace (KM) and Breeding line of NongWoo Bio Co. (NW), respectively. From the raw reads, 64,998 transcripts from KM and 100,234 transcripts from NW were de novo assembled. The assembled transcripts were used to identify molecular markers (e.g., single-nucleotide polymorphisms and simple sequence repeats), detect tissue-specific expressed genes, and construct a genetic linkage map. In total, 234 single-nucleotide polymorphisms and 25 simple sequence repeats were screened from 7,871 and 8,052 candidates, respectively, between the KM and NW varieties and used for construction of a genetic map with 94 F2 population specimens. The genetic linkage map consisted of 12 linkage groups, and 248 markers were assigned. These transcriptome and molecular marker data provide information useful for molecular breeding of oriental melon and further comparative studies of the Cucurbitaceae family.

Effect of Silicate Fertilizer on Oriental Melon in Plastic Film House (시설재배 참외에 대한 규산 비료 시용 효과)

  • Although the requirement and optimum soil level of Si for oriental melon are still not well understood, silicate fertilizer is commonly applied to the oriental melon in plastic film houses where soil silicate level is relatively high. In this research the effects of silicate fertilizer on growth, fruit yield and fruit quality of oriental melon, and soil properties were investigated in plastic film house where the soil available silicate was $212mg\;SiO_2\;kg^{-1}$. Silicate fertilizer was applied in the rates of 100, 200, and $300kg\;10a^{-1}$. The application of silicate fertilizer could not increase the early growth of oriental melon, and also the fruit yield and quality were not different among the treatments. Available Si and P contents in soils and also Si and P contents in leaf of oriental melon of the different treatments were not significantly different. In the relationship between total Si in oriental melon leaf and soil silicate extracted by 1 N sodium acetate, optimum soil available silicate level for oriental melon was found to be around $100mg\;SiO_2\;kg^{-1}$. These results indicate that the additional silicate fertilization in soils of available silicate higher than $100mg\;SiO_2\;kg^{-1}$ is unnecessary, and such application of silicate can not have any beneficial effect on the growth and fruit yield of oriental melon.

Residues of New Fungicide, Isopyrazam on Cucumber and Oriental Melon (Pyrazolecarboxamide계 살균제 Isopyrazam의 오이 및 참외 중 잔류특성)

  • Isopyrazam, a new pyrazolecarboxamide fungicide developed by Syngenta, was highly active against foliar powdery mildew pathogens on cucumber, oriental melon and various vegetables. Following foliar applications on cucumber and oriental melon, crop residues were determined using high performance liquid chromatography. For all studies, limit of quantification was 0.02 mg/kg and minimum detection level was 2.0 ng and recoveries were 83.0-88.0% on cucumber, 92.4-104.5% on oriental melon. Isopyrazam was detected 0.07-0.72 mg/kg on cucumber and < 0.02-0.68 mg/kg on oriental melon, respectively. The TMDI (Theoretical Maximum Dailly Intake) of isopyrazam on cucumber and oriental melon was estimated to less than 1.765% of ADI.

Screening of Resistance Melon Germplasm to Phytotpthora Rot caused by Phytophthora Capsici

  • Melon (Cucumis melo) is an annual herbaceous plant of the family Cucurbitaceae. Phytophthora rot, caused by Phytophthora capsici is a serious threat to cucurbits crops production as it directly infects the host plant, and it is difficult to control because of variable pathogenicity. This study investigated the resistance of 450 accessions of melon germplasm against Phytophthora rot by inoculating the seedlings with sporangial suspension ($10^{5\;or\;6}$ zoosporangia/ml) of P. capsici. Disease incidence of Phytophthora rot was observed on the melon germplasm at 7-day intervals for 35 days after inoculation. Susceptible melon germplasm showed either severe symptoms of stem and root rot or death of the whole plant. Twenty out of 450 tested accessions showed less than 20% disease incidence, of which five accessions showed a high level of resistance against Phytopthtora rot. Five resistant accessions, namely IT119813, IT138016, IT174911, IT174927, and IT906998, scored 0% disease incidence under high inoculum density of P. capsici ($10^6$ zoosporangia/mL). We recommend that these candidate melon germplasm may be used as genetic resources in the breeding of melon varieties resistant to Phytophthora rot.

Desalinization Effect of Off-season Crop Cultivation in Long-term Oriental Melon Cultivated Plastic Film House Soils (휴경기 후작물 재배에 의한 참외 장기연작 비닐하우스 토양의 제염 효과)

  • BACKGROUND: During the off-season, the cultivation of Chinese cabbage and water dropwort is often used to desalinize plastic film house soils. The objective of this study was to verify the effect of double-cropping systems on the salt removal in oriental melon cultivated plastic film house soils.METHODS AND RESULTS: Electrical conductivity (EC) and soluble salt contents were measured in soils collected from plastic film houses of oriental melon cultivation before and after the off-season crop cultivation. Also the same measurements were performed in the next oriental melon season to estimate the desalinization effect of double-cropping systems. During the cultivation of Chinese cabbage under open-field condition, ECeof surface soil was reduced from 6.0 to 0.8 dS/m. Double-cropping of water dropwort in flooded soil was also efficient in removing the salts accumulated during oriental melon cultivation. In the house soils where salts were removed during the off-season crop cultivation, soil ECewas maintained below 3 dS/m during the next oriental melon cultivation season.CONCLUSION: The off-season cropping under open-field or flooded condition was effective in desalinization of plastic film house soils. Since the salt removal effect is not expected to last for several years, the double-cropping system should be introduced every season to maintain soil EC below the critical level.

가공전 열처리가 Fresh-cut Muskmelon의 품질변화에 미치는 영향

  • Muskmelon(Cucumis melo L.)은 독특한 향기와 높은 당도로 각광받고 있는 과일로 주로 지중해, 아시아, 북유럽과 미주지역에서 광범위하게 재배되고 있으며 우리나라에서는 주로 시설 내에서 재배되고 있다. 점차 그 수요가 증가하고 있는 fresh-cut제품은 유통, 저장 기간 동안 제품의 신선함 유지가 요구된다. 본 연구에서는 가공 전 열처리가 fresh-cut muskmelon의 품질에 미치는 영향과 최적 열처리 조건을 조사하였다. Half-cut melon을 대상으로 blanching 온도 설정을 위한 예비 실험을 한 결과 5$0^{\circ}C$ 열처리한 구가 향, 연화 저해 정도, 그리고 전체적인 기호도에 대한 관능평가 가장 우수하였다. 이를 바탕으로 5$0^{\circ}C$의 water bath상에서 whole melon을 10, 20, 30, 40분 blanching하여 5$^{\circ}C$에서 24시간 저장한 후 cork borer를 이용하여 cylinder 형의 fresh-cut melon을 가공하였다. 가공한 melon은 5$^{\circ}C$에서 6일 저장하면서 L-value, chroma-value, soluble solid, pH, 경도, 호흡 변화 등을 통해 품질변화를 측정하였다. 저장 6일후 L-value의 변화는 대조구에서 가장 크게 나타났으며, chroma-value는 저장 시간이 경과함에 따라 감소하였으며 10분 열처리 한 구에서 변화정도가 가장 낮게 나타났다. 수용성 고형분 함량은 저장기간의 경과에 따른 큰 변화는 없었으나 40분 열처리한 구에서 수용성 고형분 함량이 가장 낮았으며 20, 30분 열처리한 구에서 높게 나타났다. 저장 중 fresh-cut melon의 pH는 증가하는 경향을 보였으며 10분 열처리한 구에서 가장 낮게 나타났다. 호흡변화는 저장 기간이 경과함에 따라 $O_2$는 감소하고 $CO_2$는 증가했으며 특히 20분 열처리한 구에서 $CO_2$가 크게 증가한 것으로 나타났다. 증가 정도가 가장 크게 나타났다. 열처리한 fresh-cut melon의 hardness는 저장 3일 후 대조구와 40분 열처리 구에서 가장 낮아 연화현상을 확인했으며 20분 열처리구에서 초기 hardness가 유지되는 것으로 확인했다. 따라서 fresh-cut melon을 위한 가공 전 열처리를 위해서는 5$0^{\circ}C$에서 10분 열처리가 품질 변화를 유지하는데 적당하다고 판단된다.

Monitoring on Heating Condition and Enzyme Treatment Condition for Manufacture of Oriental Melon Concentrate (참외 농축액 제조를 위한 가열 및 효소 처리조건 모니터링)

  • This study was carried out to observe the change of oriental melon juice quality during manufacture by heating condition and enzyme treatment. To use over-production oriental melon effectively, oriental melon juice was prepared with change of heating temperature and heating time. The heating conditions for minimum filtration time were 98.57$^{\circ}C$ and 11.29 min. The optimum conditions Predicted for separation ratio of clear solution, filtration time and browning color intensity of oriental melon juice were 98$^{\circ}C$, 13 min. In the pectin-degrading enzyme treatment test, turbidity showed minimum value in 4.40 mg%(pectin-degrading enzyme content), 39.72 mg% (gelatin content) and 24.09 min (treatment time).Browning color intensity showed minimum value in 9.33 mg% (pectin-degrading enzyme content),44.70 mg% (gelatin content) and 115.56 min (treatment time). The optimum conditions predicted for turbidity and browning color intensity of oriental melon concentration Juice were 6.6 mg% (pectin-degrading enzyme content), 33.6 mg% (gelatin content) and 70 min (treatment time).

Study about the Determinant Factors of Consumers about Environment-friendly vegetables : Focusing on Carrot and Oriental Melon (친환경 채소의 소비자 선택 속성에 관한 연구: 당근과 참외를 대상으로)

  • Even though Korean consumers have shown the strong preference for environment-friendly agricultural products, the real market of environment-friendly agricultural products may reach a plateau. In order to overcome this problem, researchers should find the more specific consumers' needs, including taste, flavor, shape, and others. and offer some implications to farmers and marketers. The object of this paper is to analyze the determinant factors of Korean consumers about environment-friendly carrot and oriental melon, through survey and fuzzy multi-criteria decision making approach. The results of analyses present that Korean consumers have the unique determinant factors about environment-friendly carrot and oriental melon, which are different from conventional carrot and oriental melon. Korean consumers' experience of the buying of environment-friendly vegetable is also influential in the determinant factors. Findings and implications from this research may be useful to Korean producers and other researchers.
