한우와 Holstein종의 κ -casein 유전자의 발현조절부위의 염기서열 분석 (Analysis of Sequence on promoter of κ -casein Genes in Korean Native Cattle and Holstein)

  • 본 연구는 한우와 Holstein종의 $\kappa$ -CN 유전자 발현조절부위, 즉 $\kappa$ -CN gene 5'-flanking region에 대한 특성을 구명하기 위하여 specific primer를 이용하여 PCR(polymerase chain reaction) 방법으로 분석하였다. PCR 증폭 결과, 349bp 크기의 증폭산물을 얻었으며 한우와 Holstein종의 $\kappa$ -CN 유전자의 발현조절부위의 염기서열 분석결과에 있어서는 -82bp에서 -431bp까지 분석되었으며, 종간의 염기서열 비교에서 -386bp에서는 Holstein종에서는 T 염기가 한우에서는 C 염기로 치환되었으며, -241bp와 -192bp에서는 Holstein종에서는 T 및 C 염가가 존재하였으나, 한우에서는 결실되었다. 또한, -183bp에서는 한우에서는 T 염기가 존재하였으나, Holstein종에서는 염기가 결실되어 있음을 알 수 있었다. 또한 한우와 Holstein 종간의 상동성 분석에 있어서는 한우 $\kappa$ -CN 유전자 발현조절부위의 염기서열이 Holstein종과 98.9% 의 상동성을 보였다.

Physicochemical properties of crust derived from dry-aged Holstein and Hanwoo loin

  • This study evaluated the quality characteristics of crust derived from dry-aged Holstein and Hanwoo loins and their effects on food as additives. With respect to physicochemical properties, we examined the proximate composition, pH value, salinity, color, water and fat absorption, emulsifying capacity, and swelling yield. The protein and ash contents in the Holstein crust were significantly higher than those in the Hanwoo crust (p < 0.0001). The fat content in the Hanwoo crust was significantly higher than that in the Holstein crust (p < 0.01). The salinity, lightness, and yellowness of the Hanwoo crust were significantly lower than those of the Holstein crust (p < 0.001). Furthermore, the pH value and emulsifying capacity of the Hanwoo crust were significantly higher than those of the Holstein crust (p < 0.001). The fat absorption of the Holstein crust was significantly higher than that of the Hanwoo crust (p < 0.001). The swelling yield of the Holstein crust was significantly higher than that of the Hanwoo crust at pH 3 and 4 (p < 0.001), whereas the swelling yield of the Hanwoo crust was significantly higher than that of the Holstein crust at pH 7 (p < 0.001). Principal component analysis of dry-aged Hanwoo, Holstein, and non-aged Holstein showed different flavor patterns for each sample. Finally, the results showed that the crusts derived from dry-aged Hanwoo and Holstein loins were suitable flavor enhancers.

충남 남부지역의 소 유행열, 아까바네병 및 이바라끼병의 항체 상황 조사 (A Sero-epizootiological Study on Bovine Ephemeral Fever, Akabane Disease and Ibaraki Disease in Holstein of Southern Area of ChungNam Province)

  • To investigate serum neutralizing antibodies against Bovine ephemeral fever (BEF) virus, Akabane virus and Ibaraki virus in southern area of Chungnam province, Holstein sera were collected from April-May(269 heads /37 farms) and October-November (226 heads /35 farms), 1993. The results were summarized as follows ; 1. Bovine ephemeral fever.-antibody positive rates to BEF virus were 46.1% (124 heads /269 Holstein) in April-May and 53.9%(122 heads /226 Holstein) in October-November. 2. Akabane disease.-antibody positive rates to Akabne virus were 34.2%(92 heads /269 holstein) in April-May and 51.3%(116 heads /226 Holstein) in October-Novermber. 3. Ibaraki disease.-antibody positive rates to Ibaraki virus were 57.6%(155 heads /269 Holstein) in April -May and 38.5%(87 heads /226 Holstein) in October-November.

Meat Quality of Loin and Top Round Muscles from the Hanwoo and Holstein Veal Calves

  • This study was conducted to compare the meat qualities of loin (m. longissimus dorsi) and top round (m. semimembranosus) from Hanwoo and Holstein veal. Ten Hanwoo and Holstein calves were randomly selected from a local cattle farm and raised. They were slaughtered when they were 8 mon old and weighed. Weight and percentage in primal cuts and slaughter performance of Hanwoo and Holstein veal calves are obtained. Immediately after weighting, slices of loin and top round muscles were sampled. After vacuum packaging, the samples were subjected to proximate composition, physicochemical and microbiological analyses. Dressing weight and percentage were heavier and greater (p<0.05) in the Holstein than in the Hanwoo. Water contents of the top round muscle was higher in the Holstein than in the Hanwoo (p<0.05). Water-holding capacity, protein content and CIE L* (lightness) of both muscles were higher in the Holstein than in the Hanwoo veal, whereas fat content, pH, cooking loss, a* (redness), and b* (yellowness) were higher in the Hanwoo than in the Holstein veal (p<0.05). Thiobarbituric acid and volatile basic nitrogen values of both the muscles were lower in the Hanwoo than in the Holstein veal during the first 10 d of storage (p<0.05).

한우와 젖소의 발정 행동 반복수와 지속 시간에 관한 연구 (Studies on the Relationship between Number of Repeat and Duration of Estrous Behavior in Hanwoo and Holstein Cattle)

  • The objective of this study was to investigate the relationship between behavioral signs and duration of estrous in Hanwoo and Holstein cattle for the prediction of optical insemination. Fifty two of 58 and 71 of 89 in Hanwoo and Holstein cows or heifers showed estrous exhibition within 72 h after 2 days following $PGF_{2}\alpha$ administration, respectively. The number of mounting and standing heat were 54.2 and 57.6, 59.4 and 53.5, 42.0 and 30.8 and 16.2 and 10.7 times in Hanwoo cows, Hanwoo heifers, Holstein cows and Holstein heifers, respectively. Duration of mounting were no significantly difference for Hanwoo cows ($21.2\pm11.3$), Hanwoo heifers ($19.9\pm11.4$), Holstein cows ($8.7\pm4.4$) and Holstein heifers ($16.9\pm8.0$). Duration of standing heat were significantly (p<0.05) shorter for Holstein cows ($5.4\pm3.4$) than for Hanwoo cows ($17.1\pm9.6$), Hanwoo heifers ($16.5\pm6.3$) and Holstein heifers ($15.0\pm7.2$). Time until mounting after injection of $PGF_{2}\alpha$ were significantly (p<0.05) longer for Holstein cows ($56.3\pm11.45$) than for Hanwoo cows ($42.71\pm10.44$), Hanwoo heifers ($36.6\pm8.21$) and Holstein heifers ($40.70\pm6.15$). Time until standing heat after injection of $PGF_{2}\alpha$ were significantly (p<0.05) longer for Holstein cows ($61.6\pm8.92$) than for Hanwoo cows ($46.2\pm11.49$), Hanwoo heifers ($42.7\pm6.06$) and Holstein heifers ($44.1\pm6.72$). In the results of this study, duration of standing heat was the shortest in Holstein cows ($5.4\pm3.4$). The estimation of estrous with Holstein cows has more difficulty because to significantly shorter duration of standing heat than for Hanwoo cows, Hanwoo heifers and Holstein heifers. The standing heat can be a good predictor for time of ovulation but it is concluded that mounting behavior could be the best predictor for time of ovulation.

Holstein 유우의 안검암종 증레 (A Case of a Holstein Cow with Eyelid Carcinomatosis)

  • A ten-year-old holstein cow was presented because of prolapse of the third eyelid and apparent hyperplasia of the right lower eyelid. Historical findings included increased appetite as well as polyuria and polydipsia for about two weeks. The most remarkable fadings on physical examination were a large periocular proliferative tissue and bleeding. Surgical incision was used both as a biopsy and therapeutic tool in holstein cow with mass. Histopathological examination of the mass revealed squamous cell carcinoma. Blood and milk tests of patient with squamous cell carcinomatosis were peformed, but normal values.

한우와 홀스타인 암송아지의 성장, 유선발달, 혈청 단백질과 혈청 중 Growth Hormone, Prolactin 및 Estradiol의 농도 비교 (Comparisons of Growth, Mammary Development, Serum Proteins and Serum Concentrations of Estradiol, Growth Hormone and Prolactin between Korean Native and Holstein Heifers)

  • Growth, mammary development, serum concentrations of growth hormone, prolactin and estradiol which affect the growth and mammary development, were investigated in Korean Native heifers and Holstein heifers from 5 months old to 15 months old. From 5 months old to 15 months old, body weights in Korean Native heifers averaged 37%, lower than those in Holstein heifers of the same age (P<.01). Teat volumes in Korean Native heifers are average 67% smaller than those in Holstein heifers. Analysis of serum protein by secondary electrophoresis showed differences in protein pattern between the breeds and between the individuals. From 5 months old to 15 months old, serum concentration of growth hormone in Korean Native heifers was averge 47% lower than that in Holstein (P<.01), yet prolactin concentrations in serum were about the same between the two breeds. However serum concentration of estradiol in Korean Native heifers was average 20% higher than that in Holstein of same age (P<.01). This result showed evidently that the decrease of the growth hormone concentration in serum, while growing up, causes to retard the growth and mammary development in Korean Native heifers compared to Holstein heifers.

모색 발현 유전자의 DNA Marker를 이용한 쇠고기 품종 판별 (Identification of Beef Breed using DNA Marker of Coat Color Genes)

  • 본 연구는 축우의 모색발현에 관여하는 MC1R, MGF 및 TYRP1 3종류의 모색 유전자의 PCR-RFLP marker를 이용하여 쇠고기 품종 판별기술을 개발하고자 수행하였다. MC1R 유전자의 104번째 아미노산을 지정하는 codon에 GGT 염기를 갖고 있는 Holstein 젖소와 Angus 육우는 제한효소 인지부위가 존재하여 537 bp증폭산물이 절단되어 329와 208bp 두개의 band가 검출되었으나 한우에서는 GTG로 G 염기가 T염기로 치환됨으로써 제한효소 인식부위가 소실되어 537 bp의 단일 bind 만이 검출되었다. 따라서, 이처럼 MC1R 모색유전자의 품종 간 특정 염기서열의 차이가 곧 특정 제한효소의 염기 서열상의 인지 부위 차이를 가져와 한우와 Holstein 젖소 및 Angus 육우 품종간의 RFLP 유전자형 출현에 확실한 차이가 인정되어 한우 품종에 특이적인 MC1R 유전자의 RFLP marker를 이용한 한우육 판별이 가능하였다. 또한, MGF 유전자의 RFLP 유전자형 출현빈도에서 한우는 r/r형이 75%로 출현율이 매우 높은 유전자형으로 분석된 반면 Hereford종은 R/R 형이 80%로 출현율이 매우 높았고 Holstein종과 Angus종은 R/r형이 100% 출현함으로써 한우와 Holstein 및 수입육우 품종간의 MGF 유전자형 출현빈도에 뚜렷한 차이가 인정되었다. 한편, TYRP1 유전자의 RFLP유전자형을 분석한 결과 모든 품종에서 동일한 RFLP type이 검출되어 TYRP1 모색 유전자를 이용한 쇠고기 품종 구별은 불가능한 것으로 나타났다. 따라서, 소 모색 관련 MC1R과 MGF 두 유전자의 품종 특이적 PCR-RFLP 유전자형은 한우육과 국내산 Holstein젖소고기 및 Angus 수입육간의 품종을 식별하는데 매우 유용한 DNA marker로 이용될 수 있음이 확인되었다.

한우 및 홀스타인육의 품종간 특이성분의 검색에 관한 연구 (Identification of Species-Specific Components between Hanwoo and Holstein Meat)

  • Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) of muscles extracted with distilled water, saline solution, SDS or Trition X-100 showed simular protein patterns between Hanwoo and Holstein meat, indicating that SDS-PAGE technique may not be useful for the identification between Hanwoo and Holstein meat. Lectine blot analysis of muscle extracted with distilled water demonstrated that Hanwoo and Holstein meat had similar affinities for concanavalin A (Con A), ricinus communis agglutinin (RCA-120), ulex europaeus agglutinin (UEA-1) or peanut agglutinin (PNA) lectins. However, approximately 32.1 kDa component of Hanwoo meat showed high affinity for dolichos biflorus agglutinin (DBA) lectin. On the contrary, high molecular weight components of Holstein meat had the specific affinity for wheat germ agglutinin (WGA) lectin. Hanwoo meat-specific components were observed by lectin staining of heat-denatured meat at 100$^{\circ}C$ for 30 sec. Also, the component of heat-denatured meat at 100$^{\circ}C$ for 30 sec, which was slightly smaller than Hanwoo meat-specific component, was concentrated specifically in Holstein meat.

Holstein과 교잡종 거세우 및 처녀우의 성장발육, 사료이용성 및 도체특성 비교 연구 (Study on Comparison of Growth Performance, Feed Efficiency and Carcass Characteristics for Holstein and F1(Holstein ♀ x Hanwoo ♂) Steers and Heifers)

  • Holstein 및 교잡종(Holstein♀×한우♂)의 거세우 및 처녀우 각 8두씩 총 32두(평균체중, Holstein 거세우:196.9±25.2kg, Holstein 처녀우:163.4±11.3kg, 교잡종 거세우:169.6±24.9kg, 교잡종 처녀우 156.9±15.6kg)를 대상으로 7개월령부터 24개월령까지 비육시험을 실시한 결과를 요약하면 다음과 같다. 성장단계별 일당증체량은 육성기, 비육전기, 비육중기 및 비육후기에 각각 0.733~1.018(평균 0.869), 0.994~1.255(평균 1.094), 0.947~1.259(평균 1.122) 및 0.736~0.824kg(평균 0.790kg)으로 비육중기>비육전기>육성기>비육후기의 순으로 높았다. 그러나 전 비육기간의 일당증체량은 0.882~1.061kg(평균 0.957kg)으로 순종이 교잡종보다 0.072kg(7.3%), 그리고 성별로는 거세우가 처녀우에 비해 0.106kg(10.5%)이 더 증체되었다. 전 비육기간의 농후사료 급여수준은 체중의 1.9% 내외이고, 볏짚은 농후사료 급여량의 25% 수준이 바람직한 것으로 나타났다. 그리고 1kg 증체에 소요된 농후사료 및 TDN량은 순종이 교잡종보다 4.6%가 많았으며, 처녀우가 거세우보다 7.3%가 많아 순종보다는 교잡종이, 그리고 처녀우보다 거세우의 사료이용성이 높았다. 도체조사 결과, 등지방두께는 순종이 교잡종보다 엷었고, 등심단면적은 순종이 교잡종보다 적었다. 도체중 1kg에 대한 등심단면적은 순종보다 교잡종이, 그리고 거세우보다 처녀우가 넓었다. 그리고 육색은 교잡종보다 순종이 우수하였으나 성별에 따른 뚜렷한 차이는 없었다. 쇠고기의 물리화학적 특성에서 전단력, 가열감량 및 보수력 등과 관능검사 결과의 다즙성, 연도 및 향미 등은 순종보다 교잡종이, 그리고 거세우보다 처녀우가 우수하였다. 이상과 같은 결과들을 종합해 볼 때, 고급육생산은 순종보다 교잡종이, 그리고 거세우보다 처녀우가 유리하고, Holstein 및 교잡종의 거세우 및 처녀우 비육시 농후사료는 체중의 1.9%, 볏짚은 농후사료 량의 25% 수준에서 결정하는 것이 바람직 할 것으로 판단된다.
