Reproductive Ecology of the Dusky Mud Hopper, Periophthalmus modestus in Western Korea (한국 서해산 말뚝망둥어, Periophthalmus modestus의 번식생태)

  • Reproductive cycle with the gonad developmental phases, first sexual maturity, spawning frequency, sex ratio of the dusky mud hopper, Periophthalmus modestus were investigated by histological observations. Monthly variations of the gonadosomatic index (GSI) began to increase in May and reached a maximum in June when the gonad was getting mature during the period of higher ground (water) temperature-long day length. Changes in the GSI showed a negative correlation to the HSI, but coincided with the fatness index. The reproductive cycle can be classified into five successive stages: in female, early growing stage (April to May), late growing stage (April to May), mature stage (May to June), ripe and spent stage (June to August), and recovery and resting stage (August to March); in males, growing stage (April to May), mature stage (May to June), ripe and spent stage (June to August), and recovery and resting stage (August to March); According to the frequency distributions of egg diameters during the breeding season, Periophthalmus modestus is presumed to be a summer breeder, asynchronous group and polycyclic species to spawn 2 times or more during the spawning season. Total eggs and mature eggs in absolute fecundity and relative fecundity (per cm) increased with the increase of body length. Total eggs and mature eggs in absolute fecundity and relative fecundity (per g) did not increase with the increase of body weight. Percentages of first sexual maturity of females and males ranging from 5.1 to 5.5 cm in body length are over 50%, and 100% for fish over 7.1 cm in body length. The sex ratios of females to males over 5.1 cm in body length were not significantly different from a 1 : 1 sex ratio.

Comparison of Plant Growth Characteristics and Biological Activities of Four Asparagus Cultivars by Cultural Method (재배방법에 따른 아스파라거스 4 품종의 생장과 생리활성 비교)

  • In the present study, we investigated the plant growth characteristics and biological activity of four asparagus cultivars grown using two cultural methods and tested the possibility of domestic open field. The number of shoots, buds, roots, shoot and root fresh and dry weights, and total dry weight of the four asparagus cultivars grown in a plastic house were higher than those of the same cultivars grown in an open field. Of the cultivars grown in the open field, Jersey Giant had greater shoot number than the other cultivars. In plastic house cultivation, the number of buds in Jersey Supreme was greater than the other cultivars. The total flavonoid content of the Jersey Giant was greater than the other cultivars, but cultural method was unaffected. The total polyphenol contents in asparagus cultivars grown in the plastic house were higher than those of cultivars grown in the open field. The total polyphenol content of the Jersey Giant grown the plastic house was significantly higher than those of other cultivars. Antioxidant activity such as catalase (CAT) and peroxidase (POX) did not differ significantly with cultural methods and among the cultivars. Ascorbate peroxidase (APX) activity of asparagus cultivars grown in the open field was higher than that of cultivars grown in the greenhouse; the highest APX activity was detected in UC157. Thus, greenhouse cultivation is expected to improve plant growth characteristics and biological activities of asparagus cultivars; each cultural method should be considered when selecting a suitable cultivar for high yield and high bioactive compound content.

Comparison of growth, condition index and mortality of manila clam (Ruditapes philippinarum) between originated from China (Liaoning Dandong) and Chungnam (Taean) in Gochang tidal flats (고창 갯벌에서 중국산 및 충남산 바지락의 성장, 비만도 및 폐사 특성 비교)

  • In this study, we transplanted Chinese (Liaoning Dandong) and Chungnam (Taean) manila clam seeds to Gochang tidal flats in Jeonbuk province, and compared growth, mortality, and condition indices from May 2015 to August 2016. Within 2 months after transplantation, clam mortality of Chungnam and Chinese origin were $6.9{\pm}4.3%$, $16.9{\pm}7.9{\sim}21.0{\pm}6.3%$, respectively. We supposed that higher mortality of Chinese manila clams might be due to higher stress and weakening of physiological functions by air-exposure and unavailability of food intake for 5 days harvesting, transportation, quarantine procedures. Unlike the local clam farmer's opinion based on their field experience, growth of Chungnam clams in shell length and total weight were a little better than Chinese without statistical significant difference (P > 0.05). Condition indices of Chinese clams showed declining tendency after reaching a peak in May 2014, and in June 2015, respectively. Meanwhile, Chungnam clams has reached its peak of condition indices in May 2014 and in April 2015, respectively. From this result, we supposed that there might be at least one month difference in reproduction between the Chinese and Chungnam manila clams during the first-transplanted year. Because the mortality of adult clam have sharply increased from June 2015 (due to high temperature, few rainfall and spawning) in Gochang tidal flat, it would be recommendable that farmers harvest the clams before June when its shell length are larger than 40 mm. In conclusion, we could directly compare the productivity of Chinese and Chungnam manila clam at Gochang tidal flats by field experiment, and these results are also meaningful to manila clam farmers.

Optimum Culture Density for the Intensive Mass Production in Cyclopoid Copepod, Paracyclopina nana (Cyclopoid 요각류, Paracyclopina nana의 대량배양을 위한 최적밀도)

  • This study was performed to investigate the optimum density for the intensive mass production of cyclopoid copepod, Paracyclopina nana in terms of nauplii and adults production. Effect of three development stages on the fecundity of adult female for nauplii production, survival rate of P. nana nauplii with different initial nauplii culture densities for adults production and cannibalistic feeding behavior of P. nana was examined, respectively. The fecundity of adult female by different female, copepodite ana nauplii density in 2 ml water volume decreased with the density of adult female, but was not affected by the density of either copepodite or nauplii. The average daily nauplii production for a adult female in 8 L water volume was $2.3{\times}10^5$ individuals with the incubation density of 7 adult females/ml, and this average value was significantly higher than those values of 0.6 to $1.7\times10^5$ individuals with the incubation density of 1,3,5, 10 adult females/ml (P<0.05). Survival rate of P. nana nauplil with different initial nauplii culture densities in 5 L vessels for 15 days were 32.7, 30.7, 28.9 and $23.0\%$ with the culture density of 50, 100, 150 and 200 inds./ml, respectively, but these were significantly higher than those of values 19.7 and $18.4\%$ with the culture density of 250 and 300 inds./ml (P<0.05). Cannibalistic behavior of P. nana adults on their offspring was observed, but the behavior decreased when phytoplankton was supplemented though there was no statistical difference (P>0.05). These results may indicate that P. nana is adaptable to the hatchery conditions and this species is cultured at the high densities. Optimum culture density for nauplii and adults production of P. nana were 7 adult females/ml and 200 nauplii/ml, respectively.

Long-tenn Evaluation of Extruded Pellet Diets Compared to Raw Fish Moist Pellet Diet for Growing Flounder, Paralichthys olivaceus (넙치 미성어 건조 배합사료 및 습사료의 장기사육 평가)

  • This experiment was conducted to compare the effects of extruded pellets and raw fish-based pellet on olive flounder Paralichthys. olivaceus. Six diets were prepared for this study: two formulated extruded pellets (FEP1 & FEP2), three commercially available extruded pellets (CEP1, CEP2 & CEP3) and moist pellet (MP). Weight gain offish fed FEP1 and CEP3 were significantly higher (P<0.05) than those of fish fed FEP2, CEP1, CEP2 and MP, while that of fish fed MP was not significantly different (f<0.05) from those of fish fed the FEP2, CEP1 and CEP2. Feed efficiency of fish fed CEP2 was significantly lower (P<0.05) than those of fish fed FEP1, FEP2, CEP1, CEP3 and MP. There was no significant difference in protein efficiency ratio and hepatosomatic index between fish fed FEP1 and CEP3, and among fish fed FEP2, CEP1 and CEP2. There was no significant difference in condition factor among fish fed the FEP1 and CEP3, and between fish fed FEP2, CTP1 and MP. However, fish fed MP had a lower survival rate than fish fed the other five EP These results suggest that diet FEPl could be developed to replace MP for the owing stage of flounder without adverse effects on growth performance.

  • In the rapidly to change global market in recent years, as the era of merging and integrating industries and the evolution of technology have come to an era in which everything can not be solved as a single company, it is evolving into competition for the enterprise network rather than the competition for the enterprise unit. In a competitive business environment, it is necessary to provide not only for the efforts as an individual companies but also the mutual development efforts to enhance output through the innovation activities based on the interrelationship with the business partners. In spite of the recent efforts and research through core competencies and innovation activities, some of business activities were unable to achieve enough progress in business performance and this study mainly focused to improve business performance for those companies. This study targeted CEOs and Directors who participates in "manufacturing performance innovation partnership project" carried by The foundation of Large, SMEs, Agriculture, Fisheries cooperation Korea and studied the influences of innovation activities to the core competencies and business performance. Detailed variables in this study were extracted from the previous research and used for verification. The study is designed to determine the influence of individual innovation activities to the core competencies and business performance. Innovation activities as a parameter, the relationship between core competencies and business performance was examined. In the examination of the innovation activities as a meditated effect, those activities carried by SMEs (Collaboration in Technology, Manufacturing, and Management innovations with Large Scale Business) through partnership in manufacturing innovation is significantly related business performance. Therefore, the result reveals that the individual SMEs are having own limitation in the achievement of significant progress in business performance with their own capabilities, and using the innovation activities act as catalyst through the collaboration with large scale businesses would result significant progress in business performance. Mutual effort in collaborative innovation activities between large scale businesses and SMEs is one of the most critical issues in recent years in Korea and the main focus of this study is to provide analysis which demonstrates where the SMEs are required to focus in their innovation activities.

Mathematical Models to Predict Staphylococcus aureus Growth on Processed Cheeses

  • This study developed predictive models for the kinetic behavior of Staphylococcus aureus on processed cheeses. Mozzarella slice cheese and cheddar slice cheese were inoculated with 0.1 ml of a S. aureus strain mixture (ATCC13565, ATCC14458, ATCC23235, ATCC27664, and NCCP10826). The inoculated samples were then stored at $4^{\circ}C$ (1440 h), $15^{\circ}C$ (288 h), $25^{\circ}C$ (72 h), and $30^{\circ}C$ (48 h), and the growth of all bacteria and of S. aureus were enumerated on tryptic soy agar and mannitol salt agar, respectively. The Baranyi model was fitted to the growth data of S. aureus to calculate growth rate (${\mu}_{max}$; ${\log}CFU{\cdot}g^{-1}{\cdot}h^{-1}$), lag phase duration (LPD; h), lower asymptote (log CFU/g), and upper asymptote (log CFU/g). The growth parameters were further analyzed using the square root model as a function of temperature. The model performance was validated with observed data, and the root mean square error (RMSE) was calculated. At $4^{\circ}C$, S. aureus cell growth was not observed on either processed cheese, but S. aureus growth on the mozzarella and cheddar cheeses was observed at $15^{\circ}C$, $25^{\circ}C$, and $30^{\circ}C$. The ${\mu}_{max}$ values increased, but LPD values decreased as storage temperature increased. In addition, the developed models showed acceptable performance (RMSE = 0.3500-0.5344). This result indicates that the developed kinetic model should be useful in describing the growth pattern of S. aureus in processed cheeses.

Cryptocaryoniasis of cultured flounder, Paralichthys olivaceus in low temperatures (저수온 양식 넙치 Paralichthys olivaceus의 Cryptocaryoniasis)

  • In the winter of 1995, mass mortality occurred in cultured flounder, Paralichthys olivaceus in Gurongpo, Kyoungbuk, Korea. From the observations of moribund and dead fish, parasitic ciliates, which were shown as white spots to the naked eye, were considered to be involved in the mass mortality. From heavily infected flounders, histopathological, morphological and biological characterization of these ciliates were carried out. In the histological observation, many ciliates were found under the epithelia of gill filaments and skin, and caused hyperplasia of epithelial and mucus cells at the infected areas. The ciliates found on the body surface, fins and gills were very similar to Cryptocaryon irritans. However the ciliates showed two different patterns of reproductian, i.e., typical form(palintomy)and atypical form(budding plus multiple fission) at $16^{\circ}C$ of water temperature. The occurrence ratio between typical and atypical form was about 3:2. Tomitogenesis takes 8-14 days in the typical and 13-15 days in the atypical form. In the viability test at different temperatures and salinities, the typical form died below 30‰ at $12^{\circ}C$, below 20‰ at $16^{\circ}C$, below 15‰ at $20^{\circ}C$, and below 25‰ at $24^{\circ}C$, respectively. On the other hand, the atypical form died below 20‰ at $12^{\circ}C$, below 15‰ at 16-$20^{\circ}C$, and below 25‰ at $24^{\circ}C$, respectively. The results suggested that the atypical has better viability at low salinity than that of the typical at low temperatures. In the excystment time and success rates of excystment according to temperatures, the typical form showed 8 days, 30% at $12^{\circ}C$ : 6.5 days, 50%, at $16^{\circ}C$ : 5.5 days, 75% at $20^{\circ}C$ : and 7 days, 10% at $24^{\circ}C$, respectively. On the other hand, the atypical form showed 15.5 days at $12^{\circ}C$ : 14 days, 76.6% at $16^{\circ}C$ : 12 days, 72.2% at $20^{\circ}C$ : 10 days 31.6% at $24^{\circ}C$, respectively. The results suggested that the atypical form had longer excystment time than that of the typical form at any temperature and showed better stability at low temperatures.

Effect of Selenium Supplementation on Beef Color Stability (셀레늄 급여가 쇠고기 육색 안정성에 미치는 영향)

  • The objective of this study was to investigate the supplementation effect of selenium on beef color stability. A total of 15 Hanwoo steers were divided into 3 groups and 2 groups were administered with 0.9 ppm of one of two organic-selenium products, Organic-Se and Se-SMC (Se-spent mushroom compost) for 4 mon. The third group was the control group, which was not with fed selenium during the same period. The result of this study showed that there was no significant difference in meat color between the control and treatments when Hunter $L^*$, $a^*$, $b^*$, chroma, hue and total color difference (${\Delta}E$) were measured after 30 min of blooming. When the oxymyoglobin (OxyMb) contents were measured after beef samples were ground and stored for 48 h at $20^{\circ}C$ in an incubator, they were 26.04%, 28.52% and 33.78% for the control, Organic-Se and Se-SMC after 14 d of storage and 12.65, 18.98 and 18.72 after 21 d of storage at $4^{\circ}C$, respectively (p<0.05). The control had a significantly higher metmyoglobin (MetMb) content than Organic-Se and Se-SMC (p<0.05). This result indicated that selenium supplementation was effective in preventing the oxidation of myoglobin(Mb) and production of MetMb and thus was able to maintain the purplish fresh red color of the meat.

Isolation and Identification of Oceanisphaera sp. JJM57 from Marine Red Algae Laurencia sp. (Ceramiales: Rhodomelaceae) (해양 홍조류 Laurencia sp. (Ceramiales: Rhodomelaceae)에서 분리한 Oceanisphaera sp. JJM57의 분리 및 동정)

  • A taxonomic study was carried out to assess the phylogenetic characteristics of isolate JJM57 from marine red algae Laurencia sp. collected from intertidal zone in Jeju Island, South Korea. Comparative analysis of 16S rRNA gene sequence shows that this isolate belongs to the genus Oceanisphaera. It shows 98.02% and 97.7% sequence similarity with Oceanisphera litoralis DSM $15406^T$ and Oceanisphera donghaensis KCTC $12522^T$, respectively. Strain JJM57 is a Gram-negative, aerobic, moderately halophilic bacterium able to grow in different NaCl concentration ranges from 0.5 to 8.0% and at varying temperatures from 4 to $37^{\circ}C$. Sharing some of the physiological and biochemical properties with O. litoralis and O. donghaensis, JJM57 strain differs in the utilization of ethanol, proline, and alanine. The G+C contents of the strain JJM57 is 61.94 mol% and it is rich in $C_{16:1}$ ${\omega}7c$ and/or iso-$C_{15:0}$ 2-OH, $C_{16:0}$, and $C_{18:1}$ ${\omega}7c$ fatty acids. The DNA-DNA relatedness data separates the strain JJM57 from other species such as O. litoralis and O. donghaensis. On the basis of these polyphasic evidences, present study proposed that strain JJM57 (=KCTC 22371 =AM983543 =CCUG 60764) represents a novel bacterial species of Oceanisphaera.
