Objectives : The purpose of this study was to investigate antiaging and antioxidant effects on cultured human skin fibroblast with 80% ethanol extracts of plants including of stem of Dendropanax morbifera, Corni fructus and Lycii Fructus. Methods : An ethanol extract of three medicinal plants including stem of Dendropanax morbifera, Corni fructus and Lycii Fructus. Extracts were assessed to determine the mechanism of antioxidant and antiaging activities. Antioxidant activity of extract was evaluated by two different assays as 2,2-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and super oxide dismutase (SOD) like activities. These extracts were tested for cell viability on HS68 skin fibroblast by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide (MTT) assay. We investigated the effects of Ultraviolet-B irradiation on cytotoxicity, type 1 collagen, elastin level and oxidative damage in cultured human skin fibroblast (HS68). Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. Results : The extracts obtained dose-dependently increased the scavenging activity on DPPH radical scavenging activity and SOD like activity. The extracts of complex herbal medicine showed low cytotoxicity as more than 100% cell viability in 100ppm/ml concentration. HS68 fibroblasts were survived 70% at 120 $mJ/cm^2$ UVB irradiation and treated tumor necrosis factor (TNF)-alpha. The levels of aging factors and cytotoxicity were decreased by ethanol extract of complex herbal medicine. Conclusions : These results suggest that ethanol extracts of complex medicinal plants of including of stem of Dendropanax morbifera, Corni fructus and Lycii Fructus may have value as the potential antioxidant and antiaging medicinal plant.
산수유를 항암 식품 소재로 활용하기 위하여 sarcoma-180 암세포에 대한 산수유 에탄올 추출물의 증식 억제 및 마우스에서의 수명 연장효과를 검토하였다. 산수유 에탄올 추출물을 10, 100, 300 및 $500\;{\mu}g/mL$ 농도로 24, 48 및 72시간 별로 sarcoma-180 암세포에 처리하여 암세포 증식억제 효과를 측정한 결과, 농도 및 시간 의존적으로 억제하였으며, $500\;{\mu}g/mL$ 농도에서 48시간 배양 시 60% 이상의 높은 암세포 증식 억제 효과를 나타내었다. 산수유 에탄올 추출물을 $100\;{\mu}g/mL$ 농도 이상으로 처리한 암세포에서는 대조군의 증식이 감소되었으며, 형태학적 변화도 관찰되었다. 또한 산수유 에탄올 추출물은 농도 의존적으로 핵을 응축하고 apoptotic bodies를 생성하였으며, 300 및 $500\;{\mu}g/mL$의 농도에서는 DNA fragmentation을 유도하였고 이러한 apoptosis 유도는 caspase-3 활성과 관련이 있었다. 산수유 에탄올 추출물을 sarcoma-180 암세포가 접종된 마우스 복강에 7일간 100 및 300 mg/kg/day의 농도로 연속적으로 투여하였을 때, 산수유 투여군의 수명이 대조군에 비하여 증가되었다. 따라서 이 결과를 종합하여 볼 때 산수유는 암 예방 기능성 소재로서 활용이 가능하리라 생각된다.
산수유를 기능성식품 소재로서 이용하기 위하여 산수유 에탄올추출물의 생리활성을 조사하였다. 산수유 에탄올추출물의 수소공여능은 농도 의존적으로 증가되었으며, 300 및 $500{\mu}g/mL$ 농도에서 각각 64, 73%의 활성을 나타내었다. 대식세포 RAW264.7에 산수유 에탄올추출물을 100, 300 및 $500{\mu}g/mL$의 농도로 처리하였을 때 산수유추출물은 농도 의존적으로 NO(nitric oxide)의 생성을 유도하였다. 또한 산수유 에탄올추출물을 유방암세포에 다양한 농도로 처리하여 24, 48, 72시간 반응시킨 결과 농도 및 시간에 의존적으로 유방암세포의 증식을 억제하였으며, $500{\mu}g/mL$ 농도로 72 시간 처리 시 60% 이상의 높은 증식억제율을 보여주었다. 또한, MCF-7 유방암 세포에 bisphenol과 $17{\beta}$-estradiol과 같은 환경호르몬을 $0.1{\mu}M$의 농도로 처리하였을 때 세포의 증식이 유도되었으며, 이 세포에 산수유 에탄올추출물을 농도별로 72시간 처리하였을 때 유방암 세포의 증식이 억제되었다. 따라서 본 연구결과는 산수유의 기능성식품 소재로서의 활용 가능성을 시사한다.
본 연구에서는 신생혈관형성 제어에 바탕을 둔 비만세포제어 정도를 확인하기 위하여 4가지 지역 천연산물인 전호(Anthrisci radix), 파고지 (Psoraleae semen), 희첨 (Siegesbeckiae herba) 및 산수유 (Corni fructus)를 이용한 지방 축적물 변화 및 기전을 확인하기 위해 3T3-L1 adipocyte를 이용한 Oil Red O 염색 및 western blot을 실시하였다. 그 결과 전호, 파고지, 희첨, 산수유의 세포 독성 이내의 농도 증가에 따라 지방 축적물이 감소됨을 보였다. 또한 western blot을 위해 lipogenesis와 관련된 SREBP-1 및 adipogenesis와 관련된 $PPAR\gamma$와 C/$EBP\alpha$의 신호전달 정도를 확인한 결과 4가지 지역 천연산물의 농도 증가에 따라 단백질의 발현양이 감소됨을 확인하였다. 이는 4가지 지역 천연산물 추출물이 지방분화와 관련된 신호분자를 차단함으로써 지방형성이 억제되었음을 보였다. 따라서 4가지 지역 천연산물인 전호, 파고지, 희첨 및 산수유는 신생혈관형성 억제에 따른 항비만제제로서의 이용 가능함을 시사하였다.
산수유에 함유된 유효성분을 극성이 다른 용매로 계통분획하여 얻은 추출물(water 분획물, CWF; EtOAc 분획물, CEF; butanol 분획물, CBF)의 항산화활성및 항염증 효과를 확인하였다. 그 결과, 모든 분획물은 농도가 증가함에 따라 자유 라디칼 소거능이 증가하였으며, 특히 가장 높은 폴리페놀 함량(179.3 mg TAE/g)이 확인된 CEF의 경우 대조군으로 사용된 BHA와 ${\alpha}-tocopherol$보다 뛰어난 자유 라디칼 소거활성(DPPH: $RH_{50}$; $25.1{\mu}g/mL$, ABTS: $RC_{50}$; $36.1{\mu}g/mL$)이 확인되다. Mouse macrophage RAW 264.7 cell을 사용하여 NO, $PGE_2$ 및 염증성 사이토카인($TNF-{\alpha}$, $IL-1{\beta}$, IL-6)의 생성억제 측정을 통한 항염증 효과 분석 결과, 산수유 분획물(CWF, CEF, CBF)은 용매에 따른 유의적 차이 없이 처리농도 범위($1{\sim}100{\mu}g/mL$)에서 NO, $PGE_2$ 및 염증성 cytokine 생성을 유의적으로 억제하였다. CWF와 CEF는 농도 의존적으로 NO 생성을 억제하였으며, $100{\mu}g/mL$ 농도에서 CWF($0.5{\mu}M$)와 CEF($1.2{\mu}M$)는 LPS 비처리군과 유사하게 NO 생성을 억제하였다. $100{\mu}g/mL$ 농도에서 가장 높은 염증성 cytokine 생성 억제 효과는 CWF가 $TNF-{\alpha}$의 생성을 51.2%, CEF가 $IL-1{\beta}$와 IL-6의 생성을 각각 67%와 58% 억제하여, 산수유 분획물의 뛰어난 항염증 효과가 확인되었다.
Objectives: This study was designed to assess neuroprotective effects of herb medicine against Alzhheimer's disease related brain damage in organotypic hippocampal slice culture. Methods: We induced dementia related brain damage in organotypic hippocampal slices by $\beta$-amyloid. Those slices were treated with herb medicines - Hwangryeonhaedoktang, Sopungsoongiwon. Using by PI staining, the extents of cell death were assessed. After that, we selected the best effective one among those herb medicines and the major components of that medicine were studied to reveal neuroprotective effects and related proteins by using PI stating. Results: In PI staining, Sopungsoongiwon is the best effective herb medicine between Hwangryeonhaedoktang and Sopungsoongiwon. Notopterygii Rhizoma, Corni Fructus, Areca Catechu, Aurantii Fructus Immaturus, Plantaginis Semen is the best effective one among the components of Sopungsoongiwon. Conclusions: We suggested that purgative effect would be the best effetive medicine on dementia related brain damage between clearing heat and toxic materials.

To investigate the effect of Corni Fructus(CF) water extracts on the relaxation of the corpus cavernosum, organ bath studies, histochemical and immunohistochemical methods were used and obtained the following results. In the organ bath study, the maximal contraction of the corpus cavernosum tissue by PE showed a significant relaxation effect at $3.0mg/m{\ell}$ of CF water extract. When ${\text\tiny{L}}$-NNA was pretreated corpuscular relaxation effect of CF water extract was significantly inhibited compared without ${\text\tiny{L}}$-NNA pretreatment. In $Ca^{2+}$-free solution, the increase of contraction due to $Ca^{2+}$ influx significantly inhibited in the pretreatment compared with no pretreatment of CF water extract. Histochemical and immunohistochemical studies showed that the ratio of smooth muscle to collagen fiber was increased in the CF group compared to the PE group in the corpus cavernosum, and the eNOS positive reaction increased and the PDE5 positive reaction decreased. These results suggest that CF extract has increased NO production through activation of eNOS and inhibited the action of PDE5 to block the extracellular $Ca^{2+}$ influx, thereby relaxing the smooth muscle of the corpus cavernosum.
This study is conducted to investigate vasorelaxant effect of Corni Fructus(CF) on rabbit carotid artery. To determine vasorelaxant effect of CF on rabbit carotid artery, arterial sections with intact or removed endothelium were used in this organ bath study. After being contracted by phenylephrine(PE), arterial sections were treated with CF extract in a dose-dependent manner. To identity its mechanism, the contracted arterial sections by PE were pretreated with indomethacin(IM), tetraethylammonium chloride(TEA), Nω-nitro-L-arginine(L-NNA) or methylene blue(MB) and 1.0 ㎎/㎖ CF extract. We also studied to confirm the effect on influx of extracellular calcium chloride(Ca2+) of the CF extract in rabbit carotid artery. To measure the cytotoxicity of the CF extract, cell viability of human umbilical vein endothelial cell(HUVEC) was measured by MTT assay. Generation of nitric oxide(NO) was also measured by Griess reagent. The arterial sections with intact endothelium were relaxed significantly by CF extract, but this effect was inhibited in the arterial sections with damaged endothelium. The vasorelaxant effect was inhibited significantly when arterial sections were pretreated with IM, TEA, L-NNA, MB. In Ca2+-free krebs solution, increasing of arterial contraction by Ca2+ was also inhibited by CF significantly. The treatment of CF extract increased NO concentration in HUVEC. This study suggested that the vasorelaxant effect of CF extract would be related with endothelium derived relaxing factor(EDRF) such as NO, prostacyclin(PGI2), endothelium derived hyperpolarization factor(EDHF).
Carbon tetrachloride ($CCl_4$)-induced liver injury depends on a toxic agent that has to be metabolized by the liver NAPDH-cytochrome P450 enzyme system to a highly reactive intermediate. Alternations in the activity of cytochrome P450 enzymes affect the susceptibility to hepatic injury from $CCl_4$. In this study, we evaluated the potential protective activity of the traditional Korean medicinal herb, Corni fructus (CF), against an experimental model of hepatotoxicity induced by $CCl_4$. The CF exhibited a hepatoprotective activity against $CCl_4-induced$ liver damage in Sprague-Dawley (SD) rats, as measured by GOT, GPT, ALP and histological observation. The CF also showed significant decrease of malodialdehyde (MDA) and increase of glutathion (GSH), catalase activity in rat liver homogenate. In addition, the expression of CYP2E1, as measured by reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blot analysis, was significantly decreased in the liver of CF treated SD rats. But $CCl_4$ and CF has no significant effect on 1A1 and 3A1 isoform of cytochrome P450. Based on these findings, it is suggested that hepatoprotective effects of CF possibly related to antioxidative effects and regulation of CYP2E1 expression.
