Study on the Anti-influenza Virus A type Activity of Citrus junos (유자의 항 Influenza 바이러스 A형 활성에 관한 연구)

  • To evaluate anti-influenza virus activity of 113 specimens of Korean traditional medicine both water and methanol extracts were examined using haemagglutination inhibition test. The water extract from Citrus junos was found to inhibit influenza virus A/Taiwan/l/86(H1N1). The survival rates of virus were determined by in situ cellular enzyme-linked immunosorbent assay. The water extract of Citrus junos was fractionated by chromatographic separating using Amberlite XAD-4, 40% MeOH and 60% MeOH layer had antiviral activity. The half inhibition concentration $(IC_{50})$ of 40% MeOH layer on survival of influenza virus was $MIC>361.5{\mu}g/ml$ and $IC_{50}$ value of fr. 40-4 fractionated from 40% MeOH layer was $677.19{\mu}g/ml$. These results suggested that the fractions of Citus junos have potent anti-influenza A virus activity.

Inhibition of Adrenergic Agonists-induced Metastatic Ability of Liver Cancer Cells by Ethanol Extract of Premature Citrus Unshiu Peel (청피 에탄올 추출물이 스트레스성 카테콜아민으로 유도한 간암세포의 전이를 억제하는 효과 및 기전 연구)

  • Previous studies have highlighted the pivotal role of the β-adrenergic receptor (β-AR) signaling pathway in stimulating cancer metastasis induced by chronic stress. According to the theory of traditional Korean medicine, chronic stress can induce Qi stagnation. Based on the traditional role of premature citrus unshiu peel in moving Qi, we hypothesized that an ethanol extract of premature citrus unshiu peel (EPCU) can attenuate chronic stress-induced cancer progression. In this study, we investigated the potential role of EPCU on modulating the adrenergic agonists-induced metastatic properties of liver cancer cells. Our findings revealed that adrenergic agonists, including norepinephrine (NE), epinephrine (E), and isoproterenol (ISO), augmented the migratory capacity of Hep3B human hepatocellular carcinoma cells, which was completely abrogated by EPCU treatment in a concentration-dependent manner. Consistently, EPCU inhibited the E-induced invasive property of Hep3B cells in a dose-dependent manner. These results suggest that EPCU efficiently attenuates adrenergic agonists-induced metastatic abilities of liver cancer cells. As a molecular mechanism, EPF suppressed the phosphorylation of major components of β-AR signaling pathway, including Src, signal transducer and activator of transcription 3 (STAT3) and ERK, induced by E treatment. Taken together, our results demonstrate that EPCU impedes the adrenergic agonists-driven metastatic potential of cancer cells by inhibiting β-AR signaling pathway. This study provides basic evidence supporting the probable use of premature citrus unshiu peel to prevent metastasis in liver cancer patients under chronic stress.

  • Citrus unshiu Marc., a family of Rutaceae, has been used as antitussives, antiemetics, expectorants, spasmolytics, stomachics and digestants in oriental medicine. But there is few reports on the pharmacological effects of Citrus unshiu pericarpium. Present experiment was performed to observe the effects of the water extract of Citrus unshiu on isolated auricle and intestine in rabbits and blood pressure in rats and also to study on the active component on the cardiovascular system by means of thin layer chlomatography and spectrophotometer. 1. The water extract showed positive inotropic and positive chronotropic effect in the concentration of $1{\times}10^{-4}g/ml$, $5{\times}10^{-4}g/ml$, and $1{\times}10^{-3}g/ml$. 2. The motility of isolated rabbit intestine was significantly depressed by the water extract. 3. The blood pressure was markedly increased by the water extract. 4. The blood vessel of rabbit ear was constricted by perfusion of the ether extract. 5. The purified alkaloid of Citri pericarpium by ion exchange resin showed the same Rf value with l-synephrine on thin layer chromatography. 6. The purified alkaloid of Citri pericarpium by thin layer chromatography showed the same absorption curve with l-synephrine in the ultraviolet scanning.

Current Status of the Occurrence of the Insect Pests in the Citrus Orchard in Cheju Island (감귤해충의 최근 발생 동향)

  • As the pest of the citrus in Cheju, 69 insect species and 5 animal species were investigated during 1996 to 1998. Of these, Panonychus citri, Phyllocnistis citrella, Aphis citricola, Aphis gossypii and Frankliniella occidentalis are major pests. Especially, F. occidentalis and Peridroma saucia are very important species because of increasing damage. The number of pests species injuring leaf, branch, fruit and flower of the citrus are 36, 16, 41 and 2 species respectively. The rate of damaged fruits by pests is 20.5% in 1997 and 18.6% in 1998 and the rate of bad quality fruits is 6.4% and 6.7% respectively.

Effect of Incubation Time, Concentration of Enzyme, and 2,4-D on Isolation and Callus Formation of Protoplast from Callus of Citrus junos (遊離시간 , 酵素處理 및 2,4-D 농도가 재래 유자(Citrus junos)의 캘러스由來 原形質體 遊離 및 培養에 미치는 영향)

  • The factors affecting the isolation and culture of the protoplast of embryogenic callus, derived from immature ovule in Citrus junos, were examined. An incubation time in enzyme solution of 16 hrs was preferable for protoplast isolation. Efficient protoplast yields were obtained from the treatment of equal concentration of 0.7 M $\textrm{BH}_{3}$ to the enzyme solution containing 1.0% cellulase, 1.0% macerozyme and 0.2% pectolyase. Protoplast cultured in MT medium with 0.1 mg/L 2,4-D showed vigorous division and some of them formed callus. Induced callus was subcultured on solid MT medium but the callus showed very slow growth. The above results show the possibility to culture from protoplast fusion in Citrus genera.

Changes in the Constituents of Citrus Juice by Ultrafiltration (한외여과에 의한 온주 밀감주스의 성분 변화)

  • The citrus juice obtained from Jeju mandarines, Citru unshiu, was filtered to remove 20% of its original volume through the hollow fiber ultrafiltration systems equipped with various pore sizes of membranes. As the results of ultrafiltration, the contents of total acids, ascorbic acid, ascorbic acid, free sugars and neohesperidin in retentate showed the tendencies of gradual decreases with the increase of membrane pore sizes from 10K to 100K daltons, but tendencies were inverted when the membrane with 500k was used. The changes of color, soluble solids, total nitrogen, amino-nitrogen, naringin and hesperidin were not consistent with the membrane pore size. Considering all the data obtained using various pen sizes of membranes, the filtration system with NMWC 500K daltons was the most effective to produce citrus juices with hither quality.

Monitoring of Quality Properties with Drying of Citrus (감귤의 건조에 따른 품질특성 모니터링)

  • The response surface methodology was performed by central composite design for moisture content, titratable acidity, hunter's color value and organoleptic properties based on drying temperature and time of Citrus, to apply basic data to preparation of dried Citrus. Moisture content, b value and organoleptic color were affected by drying temperature ; That decreased with increase of drying temperature. Titratable acidity was affected by drying time. Organoleptic flavor and taste were affected by drying temperature and time. The optimum drying conditons for organoleptic taste were 67.56$^{\circ}C$ in drying temperature and 8.06 hr in drying time.

Protective effects of citrus based mixture drinks (CBMDs) on oxidative stress and restraint stress

  • In the current study investigated the protective effects of citrus based mixture drinks (CBMDs) using oxidative stress in human dermal fibroblast (HDF) cells and restraint-stressed rats. The CBMDs contained citrus bioflavonoids including narirutin and hesperidin. The cell viability of HDF cells treated with $H_2O_2$ was observed at 53.9% but treated with CBMD-1 and CBMD-2 ($500{\mu}g/mL$) on $H_2O_2$ exposed HDF cells significantly increased the relative cell viability at 65.0 and 72.2%, respectively. In the treadmill test, the time spent on the electrode plate in the restraint-stressed group was analyzed 24.1 s, but restraint-stressed rats with administered CBMDs (300 mg/kg) had significantly decreased the time at 2.4 (CBMD-1) and 4.7 (CBMD-2) s, respectively. In addition, number of touches the electrode plate in restraint-stressed group was observed at 42.4 ea, but, restraint-stressed rats with administered CBMD-1 and CBMD-2 (300 mg/kg) were significantly decreased at 7.0 and 10.2 ea, respectively.

Detection of citrus-infecting viruses and sequence analysis of Satsuma dwarf virus(SDV) and SDV-CiMV in Jeju island

  • To investigate occurrence and variability of satsuma mandarin ( Citrus unshiu)-infecting viruses in Jeju island, several sets of diagnostic RT-PCR primers were designed and applied to samples collected randomly. Each primers set used in this survey was designed to detect Satsuma dwarf virus (SDV, Sadwavirus) and Citrus mosaic virus (CiMV) which is reclassified as an isolate of SDV (SDV-CiMV, Saduavirus). RT-PCR methods could detect SDV-CiMV and CTV from leaf . samples of unshui citrus. CTV was the prevalent and SDV-CiMV was not common in Jeju island. RT-PCR product of SDV-CiMV-JJl2 were cloned and sequenced. Sequence of the isolate revealed that it was 96.9 % identical to SDV-CiMV-Jp isolate at the nucleotide level. SDV-CiMV-JJl2 was propagated on Physalis floridana and sequencing of entire sequences of genome is in progress. Variability of SDV in Jeju island was confirmed by sequence comparisons and restriction mapping analysis.
