홍삼 추출물에 의한 유전독성 감소효과(II) -배양 NIH3T3 세포에서 MMS에 의한 유전독성의 감소에 미치는 홍삼추출물 처리효과 (Decrease of Genotoxicity by Red Ginseng Root Extract (II) -Decrease of MMS- induced Genotoxicity by Red Ginseng Root Extract in Cul tared NIH3T3 Cells)

  • 알킬화제인 MMS에 의한 유전독성의 감소에 미치는 홍삼추출물의 영향을 배양 NIH3T3 세포계에서 분석하였다. 1mM의 MMS를 30분간 처리한 후 정상 배지에서 배양한 시간간격에 따라 세포의 생존률은 증가하였는데 홍삼추출물이 함유된 배지에서 배양한 경우는 약 17%정도 증가한 생존률을 보였다. MMS 처리 후 감소된 DNA 복제가 정상배지 배양시간에 따라 증가하는 정도도 홍삼추출물을 후처리할 경우 현저한 증가를 보였다. MMS 상해를 회복하기 위한 절제회복능은 홍삼추출물을 처리할 경우 유의미한 증가를 보였다. 이러한 절제회복과정 중 효소에 의한 절제단계가 홍삼추출물 처리에 의해 활성화됨을 단사절단 분석을 통하여 규명하였다. 자외선 상해의 경우와 비해서 MMS 상해의 절제단계를 활성화하는 홍삼의 효과는 약간 떨어지는 것으로 보이나, MMS 상해회복의 전과정에 대한 홍삼의 효과는 자외선의 경우와 유사하였으며, MMS 상해에 의한 DNA 복제억제의 완화나 세포생존률은 자외선의 그것들보다 홍삼에 의해 더 증가된 측면을 보였다. 이상의 결과는 홍삼추출물이 MMS 상해의 절제회복에 유의미한 증가를 보이며 따라서 유전독성을 감소시키는 항노화제로써 사용할 수 있음을 시사한다.

  • 본 연구는 근치적 위절제 및 림프절 절제와 혹은 절제불가능 환자에서 항암화학요법 치료를 받는 위암환자에서 홍삼액기스 투여군에서 비록 각군의 대상 개체의 표본수가 적음에도 불구하고 홍삼엑기스 투여군에서 항암 cytokine으로 알려진 IL-2가 위암 대조군에 비하여 높게 나타나고 숙주의 항암 면역기능을 저해하는 cytokine인 IL-10은 수술 후 1개월에 홍삼엑기스 투여군에서 위암 대조군에 보다 그 감소비가 높게 나타났으며, 수술 후 3개월에는 홍삼엑기스 투여군에서만 건강 대조군 값에 접근하는 결과를 보여 보조 항암 화학요법 기간에서 홍삼엑기스의 투여는 위암 환자에서의 숙주의 항암 면역 억제의 현상을 빠른 시간 내에 회복시킬 수 있는 효과가 있는 것으로 보여진다. 대단위 개체를 포함하는 지속적인 추가 연구의 필요성이 절실하며 이러한 추가 연구가 진행 된다면 홍삼엑기스의 위암환자에서의 항암 면역기능의 역할을 임상적으로 증명 할 수 있으리라고 기대 된다.

  • This study was conducted to elucidate the effects of light intensity using polyethylene (p.E) net shading on the specific gravity, red ginseng quality, sugar and saponin contents of ginseng root. The specific gravity significantly increased in the ginseng roots grown under the P.E net shading as compared with that of common straw shading. The red ginseng quality under the P.E net shading was improved in order of 10, 5, 15, 20, 30% of light intensity and the inside cavity and inside white part decreased remarkably as compared with those of common straw shading. The ginseng roots grown under the P.E net shading at 10% and 15% light intensity showed a significant increase in the total sugar content but a significant decrease in the reducing sugar content at 15% light intensity as compared with those of common straw shading. The amount of total saponin of ginseng roots was increased under the P.E net shading at high light intensity as compared with that of common straw shading and the ginseng roots grown under the P.E net shading at 10% light intensity showed an increase in the diol group saponin but the ratio of PT/PD was decreased. Extract contents of ginseng root under the P.E net shading was higher than those of common straw shading and the roots grown under the P.E net shading at 15% and 20% light intensity resulted in a remarkable increase in extract contents.

Protective Effect of Fermented Red Ginseng on a Transient Focal Ischemic Rats

  • Red ginseng and fermented red ginseng were prepared, and their composition of ginsenosides and antiischemic effect were investigated. When ginseng was steamed at 98-$100{\circ}C$ for 4h and dried for 5h at $60{\circ}C$, and extracted with alcohol, its main components were ginsenoside $Rg_3$ > ginsenoside $Rg_1$> ginsenoside $Rg_2$. When the ginseng was suspended in water and fermented for 5 days by previously cultured Bifidobacterium H-1 and freeze-dried (fermented red ginseng), its main components were compound K > ginsenoside $Rg_3{\geq}$ ginsenoside $Rg_2$. Orally administered red ginseng extract did not protect ischemia-reperfusion brain injury. However, fermented red ginseng significantly protected ischemica-reperfusion brain injury. These results suggest that ginsenoside Rh2 and compound K, which was found to be at a higher content in fermented red ginseng than red ginseng, may improve ischemic brain injury.

Safety of red ginseng and herb extract complex (RHC) in menopausal women: A randomized, double-blind, placebo-controlled trial

  • Background: Various treatments are used to relieve menopausal symptoms for women. However, herbal substances are frequently used as complementary and alternative therapies as other treatments can increase ovarian and breast cancer risk. While the herbal substances' therapeutic effect is essential, the safety of their use is considered more important. This study aims to confirm the safety of red ginseng and herb extract complex (RHC), which are used to relieve menopausal symptoms. Methods: This randomized, double-blind, placebo-controlled clinical study recruited and divided 120 women experiencing menopausal symptoms into the RHC and placebo groups (60 women per group). Subjects were administered with 2 g RHC or placebo daily for 12 wk. Adverse reactions, female hormonal changes, and uterine thickness were observed and recorded on wk 0, 6, and 12. Hematologic and blood chemistry tests were also conducted. Results: The reactions of the subjects who received RHC or placebo at least once were analyzed. A total of six adverse reactions occurred in the RHC group, while nine occurred in the placebo group; common reactions observed in both groups were genital, subcutaneous tissue, and vascular disorders. However, there was no statistically significant difference between the administration groups (p = 0.5695), and no severe adverse reactions occurred in both groups.

홍삼농축액에서 Saccharomyces cerevisiae와 Saccharomyces carlsbergensis에 의한 Ginsenosides의 bioconversion (Bioconversion of Ginsenosides in Red Ginseng Extract by Saccharomycescerevisiae and Saccharomyces carlsbergensis)

  • Ginseng(Panax ginseng C.A. Meyer) is reported to have many pharmaceutical activities. The minor ginsenosides(Rd, Rg3, Rh2 and compound K) display pharmaceutical properties superior to those of the major ginsenosides. These minor ginsenosides, which contribute a very small percentage, are produced by hydrolysis of the sugar moieties of the major ginsenosides. The pH of red ginseng extracts fermented with S. cerevisiae and S. carlsbergensis decreased rapidly during 3 days of fermentation, with no further significant change thereafter. After 20 days of fermentation, a relatively small difference remained in the acidity of extracts fermented with S. cerevisiae (0.54%) and S. carlsbergensis (0.58%). Reducing sugar in the S. cerevisiae and S. carlsbergensis extracts decreased from 25.86 to 4.54 mg/ml and 4.32 mg/ml glucose equivalents, respectively; and ethanol contents increased from 1.5% at day 0 to 16.0 and 15.0%, respectively, at 20 days. Ginsenosides Rb1, Rb2, Rc, Re, Rf, and Rg1 decreased during the fermentation with S. cerevisiae, but Rd and Rg3 increased by 12 days. Ginsenosides Rb1, Rb2, Rc, Re and Rg1 decreased gradually in the extract with S. carlsbergensis, but Rd and Rg3 were increased at 6 days and 9 days.

Biological Activities and Metabolite Analysis of Various Extracts and Fractions from Red Ginseng Marc

  • Red ginseng marc (RGM) has been used on primary industries using fertilizer or forage, and it mostly has been dumped. To improve utilization of RGM, the biological activities of RGM were examined. RGM was extracted and fractionated using various solvents and their biological activities were compared. The hexane fraction from the methanol extract of RGM (RGMMH) showed strong anti-cancer activity (58.56 ± 6.04% at 100 ㎍/mL) and anti-inflammatory effect (65.72 ± 1.33% at 100 ㎍/mL). But, oil extract of RGM extracted with hexane (RGMH) showed low activities (anti-cancer: 16.42 ± 3.33%, at 100 ㎍/mL, anti-inflammatory activity: 29.46 ± 2.10%, at 100 ㎍/mL). Their metabolites were analyzed using HPLC. Panaxydol known as anti-cancer compound of RGM was one of major compounds in RGMMH. Meanwhile, panaxydol was detected in trace amount in red ginseng marc oil (RGMH). In addition, RGMMH and RGMH showed big differences in HPLC profiling. This research suggests optimal extraction method of RGM oil.

치자의 스트레스 관련 생리 활성: 홍삼과의 비교 연구 (Stress Related Activities of Gardenia Jasminoides: Comparative Study with the Effects of Red Ginseng)

  • Gardenia Jasminoides(GJ) is traditionally used for treatment of hepatic disease, insomnia, anxiety, and inflammatory disease. The aim of this study is to examine effects of GJ extract in response to stress. Animals of the normal group were not exposed to any stress and the control group were exposed to stress. The rats of the Ginseng and GJ supplementary group were orally administered once a day with 100mg of red ginseng extract, 100mg of GJ extract/kg body weight. The mice were given water containing 200mg of red ginseng extract, 200mg of GJ extract/100ml potable water. Animals were given supplements for 7 days without stress, and then were given supplements for 5 days with restraint and electroshock stress. After loading final stress, we examined stress related behavioral changes of experimental animals and measured the levels of blood corticosterone. GJ-supplementation partially blocked the stress effect on locomotion and elevated plus maze test in rats, and also partially blocked stress-induced behavioral changes such as freezing, burrowing, face-washing, smelling and rearing behavior in rats. The effect was almost equipotent to Ginseng's effect. GJ-supplementation didn't influence on fatigue related behavior or physical stress resistance. GJ-supplementation decreased the levels of blood corticosterone which is increased by stress in rats. These results suggest that GJ protects partially the living organism from stress attack and it has the potential to be used as a functional material to alleviate stress response.

Red Ginseng Marc and Steamed Extraction Powder Enhance Proliferation and Inflammatory Cytokine Modulation in Canine PBMCs Stimulated by IL-2

  • The growing market for companion animals, combined with their increasing lifespan, has generated an increased interest in companion animal immunity enhancers. Ginsenoside, a saponin component of ginseng and an essential ingredient of red ginseng marc (produced during red ginseng production), is effective in improving immunity. In this experiment, a powder mixture of red ginseng marc and steamed red ginseng extract powder (RGME) was orally administered to dogs for eight weeks. Subsequently, blood samples were collected and tested every four weeks. In addition, canine peripheral blood mononuclear cells (cPBMCs) were stimulated with or without interleukin-2 (IL-2) to evaluate their proliferation and cytokine secretion abilities. Proliferation assay suggests that the administration of RGME effectively enhanced numbers of cPBMCs under IL-2 stimulation. Furthermore, in the RGME group, a significant increase in the concentration of interferon gamma released from cPBMCs under IL-2 stimulation was observed. In conclusion, RGME might be an effective health supplement for improving immunity in dogs.

Changes in Chemical Composition of Korean Red Ginseng (Panax ginseng C.A. Meyer) Extract With Alcohol Extraction

  • We extracted red ginseng with various alcohol concentrations and evaluated total carbohydrate, uronic acid, polyphenols compounds and ginsenoside contents, and yields of alcohol extract. The water extraction (0% alcohol extraction) showed a high level of total carbohydrate content. 10% and 20% alcohol extraction showed the highest uronic acid contents (7,978.8 and $7,872.7\;{\mu}g/mL$ of extract, respectively). The efficiency order of the red ginseng extract (RGE) preparations in liberating polyphenols was: $0{\sim}50%$ alcohol${\geq}\;60%$ alcohol> $70{\sim}90%$ alcohol. Solid contents in RGE were decreased with increased alcohol concentration; the same tendency as with the results of total carbohydrate content. Total ginsenoside contents in $20{\sim}50%$ alcohol extracts showed similar levels ($442,962.9{\sim}47,930.8\;{\mu}g/mL$ of extract). Water extraction showed the lowest ginsenoside content ($14,509.4\;{\mu}g/mL$ of extract). The ginsenoside contents at above 60% alcohol were decreased with increased alcohol concentration. Generally, ginsenoside (Rg2, Rg1, Rf, Re, Rd, Rb2, Rc and Rb1) contents were increased with increased alcohol concentrations. However, Rg3 content was decreased with increases in alcohol concentration.
