In this study, to determine the importance of lactic acid bacteria (LAB) in Kombucha fermentation, biological functions, such as organic acid production and anti-inflammatory and antibacterial activities, of Kombucha, with or without LAB inoculation, were evaluated. Lactobacillus paracasei DK215, Saccharomyces cerevisiae C3, and Acetobacter pasteurianus P2 were selected as the inoculants. Organic acids were measured every 3 days from the end of fermentation using HPLC; the organic acid content of LAB-inoculated Kombucha was relatively high. Samples with or without LAB inoculation showed high antibacterial activity against Escherichia coli. The MTT assay results indicated no significant difference in concentration difference and cell death. In the NO production test, compared with the uninoculated Kombucha sample, the LAB-inoculated Kombucha sample exhibited a value similar to that of the group without LPS treatment. The levels of cytokine (IL-1α, IL-6, TNF-α) production were significantly lower than those of the LPS(+) group, indicating the anti-inflammatory activity potential of the Kombucha sample. This improvement in the biological function of the LAB-inoculated Kombucha further verifies the value of LAB in the fermented food and beverage industry.
Factors affecting the production of bacterial cellulose and organic acids in Kombucha fermentation were investigated. Kombucha was obtained by the fermentation (for 12 days at 3$0^{\circ}C$) of the green/black tea extract, supplemented with 10% white sugar, using an Oriental tea fungus as starter. Hitgher initial pH increased acid production with decreased cellulose production. With a cellulose pellicle or tea fungus broth as a starter, a 1~3 mm thick cellulose layer developed as a top layer every four days, and was removed subsequently while continuing fermentation. Addition of 30 mL tea fungus broth (13%, v/v) in Kombucha fermentation resulted in maximum production of a cellulose pellicle, indicating weak acid production. Yield of cellulose production at an early stage of fermentation was also higher when Kombucha was inoculated with a cellulose pellicle. In fact, addition of 1% (v/v) alcoholic beverage in the Kombucha fermentation activated the cellulose production, coupled with four times higher acid production.
Kombucha fermentation broth has been used as a popular health beverage and an alternative therapy with prophylactic and therapeutic benefit. We tried to establish optimal culture conditions for Kombucha fermentation in milk and to investigate cytotoxicity and antioxidative enzyme activity of Kombucha broth against normal human fibroblasts. The optimal conditions of Kombuch culture were established to 3$0^{\circ}C$, 20∼23 hours by DPPH radical scavenging test. There were positive effects on cell growth while no cytotoxicity against primary normal human diploid fibroblasts was found. The activites of glutathione peroxide and catalase in the cells treated by hydrogen peroxide (1 mM) alone and by hydrogen peroxide with Kombucha broth (1 mg/mι) were significantly different (p<0.05). These results suggest that Kombucha broth could be developed as an antioxidant agent for a new cosmetic material.

Kombucha, a fermented beverage, is gaining popularity due to its numerous beneficial health effects. Various substrates such as herbs, fruits, flowers, and vegetables, have been used for kombucha fermentation in order to enhance the flavor, aroma, and nutritional composition. This study aims to investigate the potential suitability of cascara as a novel ingredient for kombucha production. Our findings suggested that cascara is a suitable substrate for kombucha production. Fermentation elevated the total phenolic and flavonoid content in cascara, which enhanced the antioxidant, antibacterial, and prebiotic characteristics of the product. Furthermore, the accumulation of acetic acid-induced the pH lowering reached 2.7 after 14 days of fermentation, which achieved the microbiological safety of the product. Moreover, 14 days of fermentation resulted in a balanced amalgamation of acidity, sweetness, and fragrance according to sensory evaluation. Our findings not only highlight the potential of cascara kombucha as a novel substrate for kombucha production but also contribute to repurposing coffee by-products, promoting environmentally friendly and sustainable agricultural development.
Kombucha beverages were made from sweetened tea by Oriental, European and Tibetan tea fungus starters. The hot water extracts o green tea, black tea, Gugija and Omija were mixed with white and/or brown sugar, and were fermented under a static culture at 3$0^{\circ}C$. Titrable acidity, pH, color and cellulose production in kombucha beverages were evaluated. All tea fungus starters showed a higher acid production in green/black tea extracts rather than Gugija and Omija extracts. In green/black tea extracts Oriental tea fungus produced a kombucha beverage with a higher titrable acidity and lower pH than those of European and Tibetan tea fungus starters. By the static fermentation of green/black tea extract for 18 days, Oriental, Tibetan and European tea fungus starters produced cellulose pellicles of 0.43g, 0.16g, and 0.19 g (dry weight) on the top in the culture, respectively. As a mother starter, the cellulose pellicle was more efficient in acid production compared with tea fungus broth. Oriental/Tibetan mixed tea fungus showed the best acid production in the green/black tea extract supplemented with brown sugar.
본 연구는 황칠나무(Dendropanax morbiferus, DM)의 잎과 뿌리 추출물을 함유한 발효음료인 콤부차를 제조하고, 소재의 항산화 및 세포내 활성을 분석하였다. 홍차만으로 발효한 콤부차와 발효 과정에서 DM 잎이나 뿌리 추출물을 첨가한 홍차를 사용하여 발효한 콤부차의 pH 변화, 전체 산도, 라디칼 소거능을 비교하였다. 또한 RAW 264.7 세포주를 활용하여 DM의 잎이나 뿌리 추출물을 함유한 콤부차가 세포 내 산화질소(NO) 생성 및 염증 관련 사이토카인 함량에 미치는 영향을 평가하였다. DM 잎(BTKE-DML)과 뿌리(BTK-E-DMR)의 에탄올 추출물을 함유한 콤부차는 홍차만으로 제조한 콤부차(BTK-Ori)보다 발효 시작 3일 후 더 높은 라디칼 소거능을 나타내었다. RAW264.7 세포주를 이용한 in vitro 실험에서 세포독성을 고려하여 샘플을 8 mg/mL 콤부차로 처리한 결과, 지질다당류(LPS)로 유발된 NO 함량이 BTK-Ori 처리와 비교하였을 때 BTK-E-DML 및 BTK-E-DMR 처리에서 유의하게 감소하였다. 또한 LPS에 의해 자극되는 염증성 사이토카인인 인터루킨-6와 종양괴사인자-알파의 수준은 발효 15일 후 BTK-E-DML과 BTK-E-DMR을 처리한 세포에서 대조군에 비해 유의하게 감소하였다. 종합하면, 이러한 결과들은 DM의 잎 및 뿌리와 함께 발효된 콤부차는 항산화 활성이 증가되고, 세포 수준에서 염증 반응을 유의하게 조절할 수 있음을 입증하였다.
본 연구에서는 쌀의 소비를 증가시키기 위해 쌀을 당화시킨 쌀당화액을 이용해 콤부차를 제조하였다. 쌀 품종별 당화액 및 콤부차 제조 특성으로는 멥쌀은 참드림, 보람찬, 추청을 사용했으며 중간찰벼로 가와지1호, 찹쌀로 동진찰벼를 이용했다. 쌀 당화에 있어 가장 중요한 전분 함량은 가장 높은 것이 추청으로 80.40±1.33%를 나타내었으며 당화액 분석결과 90분 당화시 가장 높은 10.42°Brix를 나타내었다. 산도는 참드림이 0.38%로 가장 높았으며 가와지1호가 0.24%로 가장 낮았다. 쌀 품종별 콤부차의 발효 15일 유기산 분석결과 acetic acid는 참드림이 111.70±1.09 ppm이었으며 동진찰벼는 46.86±1.00 ppm이었다. 효소 종류에 따른 쌀당화액 콤부차 특성으로는 α-amylase를 처리한 쌀당화액이 1.06%로 가장 높았으며 β-amylase가 0.58%로 가장 낮은 산도를 나타내었다. 차 종류별 콤부차 특성에서는 녹차 첨가 콤부차의 산도가 1.09%로 가장 높았으며 루이보스 첨가 콤부차가 0.37%로 가장 낮았다. Polyphenol 분석결과에서 녹차를 이용한 발효 0일에 1,623.75 ppm에서 3,989.00 ppm으로 가장 많이 증가하였다. 유기산 분석에서 녹차첨가 콤부차가 acetic acid가 발효 0일에는 172.89 ppm에서 발효 15일에는 2,649.11 ppm으로 증가하였다. 알코올(주정) 첨가 콤부차 특성에서는 알코올 2.0% 첨가 콤부차의 산도가 1.32%로 가장 높았으며 알코올 0.5% 첨가 콤부차가 0.97%로 가장 낮았다. 이러한 결과를 종합해볼 때 새로운 쌀 음료로써 쌀당화액을 이용해서 콤부차를 만드는 것이 가능하다는 것을 확인할 수 있었다.
본 연구에서는 홍차버섯이라고 알려진 콤부차(Kombucha, K)에 플라보노이드 성분 및 각종 기능성 물질이 풍부한 감귤액을 첨가하여 감귤의 생리활성 물질들이 콤부차로 이행되는 효과를 기대하여 감귤 콤부차(citrus Kombucha, CK)를 배양한 후 항산화 능력 및 인체 방광암세포(T-24와 5637)를 이용한 항암 효과를 확인하고 더 나아가 암의 증식을 억제시킬 수 있는 천연소재 탐색을 목적으로 연구를 진행하였다. 항산화 및 총페놀 함량 결과는 K보다 CK의 항산화 능력과 페놀 함량이 높게 확인되었으며 방광암세포 T-24와 5637에 K 혹은 CK를 24시간 처리한 후 MTT assay를 통해 세포독성을 확인한 결과 농도 의존적으로 생존율이 감소하였다. 특히 T-24 세포에서는 CK를 처리하였을 때 현저한 세포의 형태적 변화를 확인하였다. Western immunoblot을 통해 apoptosis 관련 단백질들의 발현을 확인하였는데 T-24에 CK 처리하였을 때 Bcl-2의 발현은 크게 감소하였으며, pro-caspase-9, pro-caspase-8, pro-caspase-3는 농도가 높아질수록 감소하였으며, cleaved caspase-9, cleaved caspase-8, cleaved caspase-3는 농도가 높아질수록 증가하는 경향을 보였다. 또한, cleaved PARP가 증가함을 확인할 수 있었다. 이상의 결과에서 일반 콤부차보다 감귤액을 첨가한 감귤 콤부차가 인체 방광암세포 T-24에 caspase에 의한 apoptosis가 유도됐음을 확인할 수 있었다.
배, 포도, 자두, 오렌지, 사과와 골든키위 껍질을 첨가한 콤부차의 pH, 산도, 가용성 고형분, 총당, 환원당, 폴리페놀 함량, 플라보노이드 함량, 안토시아닌 함량, DPPH 라디칼 소거능과 색도를 조사하였다. 모든 실험구와 대조구는 발효 진행에 따라 pH는 감소하고 산도는 증가하였다. 가용성 고형분도 증가하였고, 대부분의 실험구가 대조구보다 높았다. 총당은 시간이 지날수록 감소했지만 환원당은 증가하였다. Day 0에서 총당과 환원당은 껍질 첨가군이 대조군보다 높았다. 폴리페놀, 플라보노이드, 안토시아닌 및 DPPH 라디칼 소거능은 발효 진행에 따라 증가하였고, 배를 제외한 모든 첨가군은 비첨가군보다 높게 나타났다. 시간이 지남에 따라 Anthocyanins이 높은 자두와 포도를 제외하고 L값은 증가하였고, 자두와 포도의 a값은 증가하였다(p<0.05).
본 연구는 감귤 콤부차의 산업화를 위한 발효 균주 표준화를 위하여 콤부차에서 분리된 3가지 균주(Gluconacetobacter xylinus, Gluconacetobacter medellinensis, Gluconobacter oxydans)를 이용한 감귤 발효액(CK-MOX)의 기능적 특성을 탐색하고자 하였다. CK-MOX 제조 후 15일간 3일 마다 샘플링을 하였으며, 발효 기간에 따른 pH, 산도, 항산화 능력을 평가하였다. 발효에 따라 pH는 감소하였고, 산도는 증가하였다. DPPH, ABTS 라디칼 소거능, ORAC assay를 통한 항산화 능력 측정 결과 발효에 따라 항산화 능력이 향상하는 것으로 나타났으며, 방광암 세포주(EJ 세포)의 생존 억제 및 이동 억제 효과가 있는 것으로 나타났다. 특히 CK-MOX로 유도된 EJ 세포의 사멸에 MAPK pathway의 중추적인 역할을 하는 것으로 알려진 ERK의 발현이 깊이 관여하는 것으로 나타났다.
