몇종의 선충천적 진균과 화학약제를 이용한 약용작물 뿌리혹선충 방제효과 검토 (Field Application of Egg and Larval Parasitic Fungi and Chemicals for Controlling Root-knot Nematodes on Some Medicinal Herb)

  • Series of studies were conducted to establish biological and chemical control method for Meloidogyne spp to medical herbs by applied of nematophagous fungi, Arthrobotrys spp, Fusarium spp, and egg parastic fungi, Paecilomyces lilacinus were applied for root-knot nematodes on medicinal herbs, Paeonia albiflora, Codonopsis lanceolata, Cnidium officinale. The results are as follow. In pot experiments, The no. of root gall and egg mass and larvae of Cnidium officinale. The results are as follow. In pot experiments. The no. of root gall and egg mass and larvae of Cnidium officinale, Codonopsis lanceolata, Paeonia japonica lowered in P. Lilacinus treated plots compare to untreated control plots. But A. thaumasia F. oxysporum treated plots were less effective. Effect of egg parasitic fungi and chemical treatment at divided root of Paeonia japonica after sterilized in pot were increased in the fresh weight, root weight, control effect in P. lilacinus treated plots as chemical, Carbo G treated plots compare to untreted control plots. I field experiment, the number of root gall, egg mass and nematode density of Paeonia were also suppressed in P. lilacinus treated plots. It was very effective continuous 2 years and transplanting time on Paeonia japonica infested soil with the M. hapla in field in both region, Chillgok and Euisung treated P. lilacinus as chemical treated plots. Soaking effect of insecticide for Paeonia japonica at diving shoot before transplating in pot were effective for 12hours immersion into 1,000 ppm of Benlate T + Mep Ec, Benlate T + Fenthion EC in pot and field Experiment. In the Examination of fungi activily on P.japonica field 1 year after soil treatment, Number of spore of P. lilacinus were 1,000~1,300 in 3 region except Euisung.

뿌리혹 선충기생 정도에 따른 인삼의 생육 및 무기성분 변화 (Effect of Root- Knot Nematode Infection on Ginseng Growth of Inorganic Nutrients in Ginseng Roots.)

  • Effect of root-knot nematode (Meloidigyne sp.) 1 infection on the growth and quantity of the inorganic substances in 6 year-old ginseng (Panax ginseng C.A. Meyer) was investigated. Naturally infected 6 year-old ginseng roots were grouped into 3 such as healthy, moderate, and heavy according to the degree of the nematode infection. The roots were sampled in the middle of October, 1980 for chemical analysis. Growth of roots in terms of fresh weight, dry weight, length and diameter was greatly reduced by the nematode infection. The content of inorganic substances was different among the various parts of ginseng roots and according to gall index. Phosphorous and nitrogen contents of the healthy ginseng roots were tend to higher than those of the infected ones, whereas calcium was high in infected roots in the order of heavy, moderate, and healthy. Content of the other inorganic substances was highest in the moderately infected roots, and lowest in the heavy one.

뿌리혹선충 유전자의 RNA 간섭 억제에 의한 선충저항성 식물 개발 및 선충방제의 최근 연구 동향 (Recent Studies on Development of Transgenic Plants Induced Root-Knot Nematode Resistance by RNA Interference Suppression of Nematode Genes and Nematode Prevention)

  • Root-knot nematodes cause billions of dollars in crop losses annually have a broad range of host over 2,000 species of plants. These nematodes are known as obligate, sedentary endo-parasites in a plant host to feed upon to complete their life cycle. To prevent the plant parasitic nematode, methyl bromide was widely applied as a soil fumigant. Other strategies to prevent or control nematodes involve RNAi-mediated suppression, R gene transformation, natural products or chemical treatments, the expression of peptide or proteins in susceptible plants, and others. Over the last decade, the entry in GenBank for Meloidogyne reveals 73,340 ESTs and recently two complete Meloidogyne spp. genomes sequences have simultaneously been presented by two groups. Recent works have demonstrated the effect of RNAi suppression to nematode target genes. These results will provide novel members of genes as a foundation for studies focused on understanding the function of M. incognita nematode genes as well as for the development of novel target genes for parasite control. Thus the successful development of biotechnology-derived plants with nematode resistance will result in large yield benefits for producers as well as environmental benefits and will accelerate the research related to pathogensresistant crops.

Use of Dactylaria brochopaga, a Predacious Fungus, for Managing Root-Knot Disease of Wheat (Triticum aestivum) Caused by Meloidogyne graminicola

  • A laboratory experiment was conducted to study the induction of constricting rings and test predation of Dactylaria brochopaga isolates against second stage juveniles (J2s) of Meloidogyne graminicola. Among the five fungal isolates, isolate D showed the greatest number of predatory rings and, consequently, trapped the maximum number of M. graminicola J2s in dual cultures. Another pot experiment was conducted to study the effect of D. brochopaga (isolate D) on the management of wheat root-knot disease. Applying a mass culture (10 g/pot) and a spore suspension of the fungus with and without cow dung manure to soil infested with 2,000 M. graminicola juveniles significantly improved plant height, root length, weights of shoots, roots, panicles and grains per hill compared to those in the control. Moreover, the fungus significantly reduced the number of root-knots, the number of egg masses, juveniles, and females per hill compared to those in the control. Bio-efficacy of the fungus was heightened when the mass culture and a spore suspensions were used in combination with cow dung manure to improve the plant growth parameters and reduce the number of root-knot and reproductive factors. Further investigations should be conducted to identify the impact of this fungus in the field.

밀기울 토양 혐기발효 처리가 멜론의 뿌리혹선충 방제 및 품질에 미치는 영향 (The Effect of Anaerobic Fermentation Treatment of Wheat bran on the Root-Knot Nematodes and the Quality of Melons in Plastic Film House Soil)

  • 본 연구는 10년 이상 과채류를 재배하여 뿌리혹선충 피해가 많이 발생되었던 시설재배 토양에서 친환경적인 뿌리혹선충방제를 위하여 밀기울 토양환원처리 효과를 구명하였다. 토양소독처리후 뿌리혹선충의 밀도를 조사한 결과 시험전과 비교하여 모든 처리구에서 뿌리혹선충밀도가 감소하는 경향을 보였지만 특히 밀기울처리구에서는 높은 방제효과를 보였다. 처리후 90일경에도 밀기울처리구가 매우 낮게 유지되었으며 멜론의 생육도 가장 양호하였다. 수확기 멜론의 고사율을 조사한 결과 무처리구 65%인데 비해 밀기울처리구, 3%로 현저히 낮았으며 과실의 크기와 품질도 밀기울처리구가 우수하였다. 따라서 밀기울 토양환원처리는 뿌리혹선충 발생포장에 농약을 사용하지 않고 멜론을 1작기 동안 안전하게 재배할 수 있는 유망한 소독방법일 뿐만 아니라 멜론의 품질을 개선하는데 매우 효과적인 방법으로 생각되었다.

지중가온시설을 이용한 고구마뿌리흑선충 방제 (Control of Root-Knot Nematode(Meloidogyne incognita Chitwood) by Root Zone Warming System)

  • 지중가온시설을 이용한 고구마뿌리혹선충(Meloidogyne incognita Chitwood) 방제를 위하여 지하 20cm 깊이의 최저지온을 각각 30. 40, $50^{\circ}C$ 가온구 및 관행의 무지중가온구로 설정하고 담수구와 무담수구로 나누어 8월 1일부터 8월 5일까지 5일간 가온한 결과, 담수구의 경우 지온 4$0^{\circ}C$에서 5일간, 무담수구의경우 지온 $50^{\circ}C$에 5일간 지중가온처리한 구에서 생존한 유충이 발견되지 않아 선충밀도를 경제적 피해수준 이하로 유지할 수 있을 것으로 사료되었다. 지중가온으로 토양내 OM. P$_2$ $O_{5}$, Ca, Mg함량과 EC는 시험전 토양에 비하여 감소하였는데, 특히 담수처리에서 EC가 현저히 저하하였다.

시설과채류(오이, 참외)의 뿌리혹선충 종합방제에 관한 연구 (Study on Integrated Control against Root-knot Nematode of Fruit Vegetables (Oriental Melon and Cucumber) in Vinyl House)

  • 시설 과채류 재배지의 뿌리혹선충의 피해경감을 위해서 경북도내 시설 오이 재배단지인 칠곡과 참외재배 주산지인 성주에서 1992년부터 1994년까지 3년간 방제시험한 결과 뿌리혹선충의 피해를 경감시키기 위한 토양개량 적기는 모래산흙인 마사토의 경우에 3년 붉은산흙인 경우 4년마다 하는 것이 효과적이고 농약과 조합한 오이의 방재시험에서 빗사마드 분제가 78.2%, 카보입제+토양반전+담수가 71.2%, 카보입제+담수가 66.3%이었으며 참외에서 살충제처리후 20일에는 77.7~80.6%의 방제효과를 보였으나 60일 후에는 33.7~49.5%로 방제가 낮아 후기에는 효과가 낮았다. 참외재배 논에서의 작부체계에 따른 뿌리혹선충 밀도 억제효과는 참외 재배후 담수하여 벼를 재배할 때에 벼 수확시 방제효과가 91.1%로 대부분의 선충이 사멸되었다. 각 처리구 공히 처리당시에는 뿌리혹선충 증식을 억제하였으나 생육추기로 갈수록 선충밀도가 증가되었다. 그러나 토양개량은 3년간 효과적이였으며 가장 경제적으로 선충의 증식을 억제하 수 있는 처리는 3년마다 한번씩 벼를 재배하는 것이었다.

Identification of 2-methylbutyric Acid as a Nematicidal Metabolite, and Biocontrol and Biofertilization Potentials of Bacillus pumilus L1

  • The present study described the isolation of 2-methylbutyric acid (2-MBA) produced from Bacillus pumilus L1, to subsequently investigate its nematicidal activity for the control of the root-knot nematode. The results showed that 2-MBA could be purified by chromatographic techniques and was identified using nuclear magnetic resonance and liquid chromatography-mass spectrometry. Crude extract and partially purified compounds had a significant effect on the inhibition of egg hatchability and second-stage juvenile (J2) mortality. A dose-dependent effect of 2-MBA was observed for J2 mortality and egg hatchability. Egg hatchability was 69.2%, 59.9%, 32.7%, and 0.0% at 125, 250, 500, and $1000{\mu}g\;mL^{-1}$ of 2-MBA after 4 d of incubation, respectively. Meanwhile, J2 mortality was in the range of 24.4%-100.0% after 2 d of incubation, depending on the concentrations of 2-MBA used. A pot experiment also demonstrated that treatment of B. pumilus L1 culture caused a significant reduction in the number of galls, egg masses, and J2 population than that of the tap water (TW) control. However, as the B. pumilus L1 culture concentration was decreased, the efficacy of nematode control by treatment of B. pumilus L1 culture was reduced compared to that of TW. B. pumilus L1 inoculation at different concentrations also promoted cucumber plant growth. Therefore, our study demonstrated the potential of 2-MBA from B. pumilus L1 as a biocontrol agent against the root-knot nematode and a plant growth promoter for cucumber plants.

  • 당근뿌리혹선충(Meloidogyne hapla)이 더덕(Codonopsis lanceolata)ml 생장에 미치는 영향을 구명하기 위하여 pot와 포장에서 실험을 수행하였다. 그 결과 당근뿌리혹선충의 밀도가 증가함에 따라 pot 실험에서는 더덕의 덩굴생장이 60%, 뿌리생장이 45% 억제되었다. 포장 실험에서는 밀도 수준별 생육차이가 뚜렷하지 않았다. 이 결과는 더덕 구당 선충 접종 밀도가 낮아 피해수준에 도달하지 않은 것으로 사료된다.

  • 뽕나무 근유선충의 방제법을 구명하고자 1985년부터 1986년까지 2개년에 걸쳐 상묘포에서 실시한 결과를 요약하면 다음과 같다. 1. 공시약제를 처리하였을 때 묘목의 활착율에는 영향을 미치지 않았고 생육은 무처리보다 우수하였다. 2. 근근유선 충방제효과는 Fenamiphos가 가장 좋았고 D-D(55%), Ethoprop, Carbofuran순으로 좋았다.
