The CC chemokine receptor 5 (CCR5) is a G protein-coupled receptor that regulates chemotaxis and effector functions of immune cells. It also serves as the major co-receptor for the entry of human immunodeficiency virus (HIV). Recently, CCR5 inhibitors have been developed and used for the treatment or prevention of HIV infections. Additionally, it has been identified that CCR5 controls bone homeostasis by regulating osteoclastogenesis and the communication between osteoblasts and osteoclasts. However, the effects of CCR5 inhibition on bone tissue in elderly patients are unknown. This study aimed to examine the bone phenotype of aged CCR5 knockout (KO) mice. Femoral and tibial bones were isolated from 12-month and 18-month old wild-type (WT) and CCR5 KO mice, and microcomputed tomography and histology analyses were performed. Twelve-month-old CCR5 KO mice exhibited a decreased trabecular bone mass and cortical bone thickness in both femoral and tibial bones compared with age-matched WT mice. Eighteen-month-old mice also showed a decreased trabecular bone mass in femurs compared with control WT mice, but not in tibial bones. Unlike in 12-month-old mice, the cortical margin of femurs and tibias in 18-month-old mice were rough, likely because they were aggravated by the deficiency of CCR5. Overall, our data suggest that the deficiency of CCR5 with aging can cause severe bone loss. When CCR5 inhibitors or CCR5 inactivating technologies are used in elderly patients, a preventive strategy for bone loss should be considered.

Paf1 복합체는 H2BK123 monoubiquitination에 기여하고, histone crosstalk에 의해 간접적으로 H3K4의 di-, tri-methylation에 기여하는 것이 알려져 있다. 하지지만, Paf1 복합체 구성요소들의 개별적인 기능에 대해서는 연구가 되어있지 않다. 이 연구에서는, Paf1 복합체 구성요소들의 단일 결핍 돌연변이 균주를 만든 후, 이들의 H2BK123 monoubiquitination 및 H3K4 mono-, di-, tri-methylation에 미치는 영향을 관찰했다. 놀랍게도, ${\Delta}paf1$, ${\Delta}rtf1$, ${\Delta}ctr9$ 돌연변이 균주에서는 H2Bub에 영향을 받는 H3K4me2와 H3K4me3뿐 아니라, H2B monoubiquitination에 영향을 받지 않는 H3K4 monomethylation의 심각한 감소를 관찰했다. 그러나, methyl기 전달 효소인 Set1의 발현 정도는 이 돌연변이 균주들에서 변하지 않았다. 이러한 결과로부터, Paf1 복합체가 Set1의 활성이나 Set1 복합체의 안정성을 직접 조절함으로써 H3K4 methylation을 조절할 수 있음을 제시한다.
Purpose: The trefoil factor family 1 (TFF1) has a protective effect against gastric mucosal damage induced by nonsteroidal anti-inflammatory drugs or ethanol. In addition, a TFF1 knockout mouse model has exhibited circumferential adenomas with high-grade dysplasia, of which $30\%$ progressed into frankly invasive carcinomas. We tried to determine whether the expression pattern of the TFF1 could be involved in the development of sporadic gastric carcinomas. Materials and Methods: We examined TFF1 expression in a series of 43 sporadic gastric carcinomas and 18 gastric adenomas by immunohistochemistry. Results: Strong positive TFF1 staining was identified primarily in the normal gastric mucosa, mainly in the cytoplasm of the superficial and foveolar epithelium. We found TFF1 expression in $55.8\%$ (24 out of 43) of the gastric carcinomas and in $16.7\%$ (3 out of 18) of the gastric adenomas. Statistically, TFF1 immunoreactivity was significantly higher in diffuse-type ($82.4\%$) than in intestinal-type ($38.5\%$) carcinomas(p=0.0058, Fisher's exact test). Conclusion: Our findings provide sufficient evidence that the expression of TFF1 in gastric cancer may simply disclose gastric-type differentiation of neoplastic cells and provide further support for the existence of at least two pathways of malignant transformation of the gastric mucosa: one via intestinal metaplasia and adenomatous dysplasia, leading to glandular carcinomas with intestinal-type differentiation, and the other via hyperplastic changes or de novo changes, leading to diffuse carcinomas and to a subset of glandular carcinomas displaying gastric-type differentiation.
Bmi1은 PcG 단백의 일종으로 PRC를 구성하는 성분이다. Bmi1에 관한 초기의 연구는 주로 암세포에서의 역할에 집중되었고, 그 결과 Bmi1은 암세포의 증식과 생존에 중요한 역할을 하는 것으로 받아들여지고 있다. 그러나, 최근의 연구 결과 Bmi1은 퇴행성 신경계 질환이 있는 뇌에서 발현이 감소되어 있고 미토콘드리아의 기능과 활성 산소 수준을 조절하는 것으로 알려져 있다. 본 연구에서는 Bmi1 발현 저하 동물을 이용하여 간질에서 Bmi1의 약물 치료 표적 가능성을 밝히고자 하였다. Bmi1의 발현은 pilocarpine에 의한 경련중첩증 이후 해마의 CA1, CA3 그리고 치상회에서 뚜렷하게 증가하였다. 경련 양상을 볼 때 경련중첩증이 유도되는 비율은 Bmi1+/+와 Bmi1+/- 생쥐에서 각각 43.14%와 53.57%로 나타났다. 그러나, 치사율과 해마의 신경세포 손상에는 두 군 간에 통계적으로 유의한 차이를 보이지 않았다. 경련중첩증 유도 2개월 후에 나타난 간질 경련의 빈도수는 통계적 유의성은 없었으나 Bmi1+/- 생쥐에서 약 50% 낮게 나타났다. 반면, 이끼섬유발아는 Bmi1+/- 생쥐에서 통계적으로 유의하게 증가하였다. 이러한 결과를 종합하면, Bmi1의 발현이 감소할 때 pilocarpine에 의한 경련 유도와 이끼섬유발아가 증가함을 보여준다.
목적: 아급성경화범뇌염은 퇴행성 신경질환으로 홍역바이러스의 지속적 중추신경계 감염으로 나타난다. 저자들은 마우스 모델을 통해 아급성경화범뇌염 발병에서 만성 바이러스 감염에 관여하는 PD-1 유전자의 역할을 알아보고자 하였다. 방법: 3주령의 동형 PD-1 유전자 제거 마우스, 이형 PD-1 유전자 제거 마우스, 야생 BALB/c 마우스를 대상으로 측뇌실내 홍역바이러스를 주입하여 동물 모델로 하였다. 바이러스 주입 3개월 후, 마우스의 뇌 조직학적 소견을 관찰하고 혈청을 분리하여 IL-21의 혈청 농도를 ELISA kit을 통해 측정하였다. 결과: 야생 BALB/c 마우스에서 가장 많은 국소적 뇌백질의 괴사 및 성상세포의 증가가 관찰되었다. 이형 마우스에서 뇌실질의 병변은 적었으며 동형 마우스는 거의 보이지 않았다. 세 그룹에서 모두 혈청 IL-21의 증가는 보이지 않았다. 결론: 이 결과는 PD-1 유전자가 만성 바이러스 감염에 중요한 역할을 함을 시사한다.
Rodobacter sphaeroides에서 orf282 유전자는 cbb3 terminal oxidase를 암호화하는 ccoNOQP 오페론과 혐기적 활성자인 FnrL을 암호화하는 fnrL 유전자 사이에 있으며, 아직은 기능이 잘 알려지지 않았다. orf282 유전자의 기능을 알기 위해 우리는 orf282의 일부를 삭제함으로써 유전자를 붕괴시켜 orf282-minus mutant를 제조하였다. 두개의 FnrL 결합 부위가 orf282의 upstream에 존재한다는 것이 밝혀져 있으며, orf282 유전자가 FnrL에 의해 양성적으로 조절된다는 것이 증명되었다. orf282 유전자는 B875와 B800-850 spectral complexes의 형성과 관련이 없다. orf282 mutant에서의 cbb3 oxidase 활성을 wild type와 비교해보면 orf282 유전자가 ccoNOQP 오페론의 조절과 cbb3 cytochrome c oxidase의 생합성과 무관하다는 것을 알 수 있다. orf282 mutant의 구조 유전자인 nifH와 조절유전자인 nifA의 프로모터 활성이 증가한 것은 orf282 유전자 산물이 nifH와 nifA의 발현에서 음성적 effector로 작용한다는 것을 시사한다.
The p73 gene contains an extrinsic P1 promoter and an intrinsic P2 promoter, controlling the transcription of the pro-apoptotic TAp73 isoform and the anti-apoptotic ${\Delta}Np73$ isoform, respectively. The DNA methylation status of both promoters act equally in the epigenetic transcriptional regulation of their relevant isoforms. The aim of this study was to analyze the different effects of these p73 isoforms in 5-aza-2'-deoxycytidine (5-aza-dC)-induced apoptosis in breast cancer cells. We investigated the effects of the DNA demethylation agent, 5-aza-dC, on the T-47D breast cancer cell line, and evaluated the methylation status of the p73 promoters and expression of TAp73 and ${\Delta}Np73$. Furthermore, we assessed the expression of p53 and p73 isoforms in 5-aza-dC-treated T-47D cells and p53 knockout cells. 5-aza-dC induced significant anti-tumor effects in T-47D cells, including inhibition of cell viability, G1 phase arrest and apoptosis. This was associated with p73 promoter demethylation and a concomitant increase in TAp73 mRNA and protein expression. In contrast, the methylation status of promoter P2 was not associated with ${\Delta}Np73$ mRNA or protein levels. Furthermore, demethylation of P2 failed to inhibit the expression of ${\Delta}Np73$ with 5-aza-dC in the p53 knockdown cell model. Our study suggests that demethylation of the P1 and P2 promoters has opposite effects on the expression of p73 isoforms, namely up-regulation of TAp73 and down-regulation of ${\Delta}Np73$. We also demonstrate that p53 likely contributes to 5-aza-dC-induced ${\Delta}Np73$ transcriptional inactivation in breast cancer cells.

Serum complement proteins comprise an important system that is responsible for several innate and adaptive immune defence mechanisms. There were three well described pathways known to lead to the generation of a C3 convertase, which catalyses the proteolysis of complement component C3, and leads to the formation of C3 opsonins (C3b, iC3b and C3d) that fix to bacteria. A pivotal step in the complement pathway is the assembly of a C3 convertase, which digests the C3 complement component to form microbial-binding C3 fragments recognized by leukocytes. The spleen clears microorganisms from the blood. Individuals lacking this organ are more susceptible to Streptococcus pneumoniae. Innate resistance to S. pneumoniae has previously been shown to involve complement components C3 and C4, however this resistance has only a partial requirement for mediators of these three pathways, such as immunoglobulin, factor B and mannose-binding lectin. Therefore it was likely that spleen and complement system provide resistance against blood-borne S. pneumoniae infection through unknown mechanism. To better understand the mechanisms involved, we studied Specific intracellular adhesion molecule-grabbing nonintegrin (SIGN)-R1. SIGN-R1, is a C-type lectin that is expressed at high levels by spleen marginal-zone macrophages and lymph-node macrophages. SIGN-R1 has previously been shown to be the main receptor for bacterial dextrans, as well as for the capsular pneumococcal polysaccharide (CPS) of S. pneumoniae. We examined the specific role of this receptor in the activation of complement. Using a monoclonal antibody that selectively downregulates SIGN-R1 expression in vivo, we show that in response to S. pneumoniae or CPS, SIGN-R1 mediates the immediate proteolysis of C3 and fixation of C3 opsonins to S. pneumoniae or to marginal-zone macrophages that had taken up CPS. These data indicate that SIGN-R1 is largely responsible for the rapid C3 convertase formation induced by S. pneumoniae in the spleen of mice. Also, we found that SIGN-R1 directly binds C1q and that C3 fixation by SIGN-R1 requires C1q and C4 but not factor B or immunoglobulin. Traditionally C3 convertase can be formed by the classical C1q- and immunoglobulin-dependent pathway, the alternative factor-B-dependent pathway and the soluble mannose-binding lectin pathway. Furthermore Conditional SIGN-R1 knockout mice developed deficits in C3 catabolism when given S. pneumoniae or its capsular polysaccharide intravenously. There were marked reductions in proteolysis of serum C3, deposition of C3 on organisms within SIGN-$R1^+$ spleen macrophages, and formation of C3 ligands. The transmembrane lectin SIGN-R1 therefore contributes to innate resistance by an unusual C3 activation pathway. We propose that in the SIGN-R1 mediated complement activation pathway, after binding to polysaccharide, SIGN-R1 captures C1q. SIGN-R1 can then, in association with several other complement proteins including C4, lead to the formation of a C3 convertase and fixation of C3. Therefore, this new pathway for C3 fixation by SIGN-R1, which is unusual as it is a classical C1q-dependent pathway that does not require immuno globulin, contributes to innate immune resistance to certain encapsulated microorganisms.
해양생물을 포함한 천연물질은 신약개발의 원천 소재로서 매력적이며, 특히 무수한 미지의 해양생물들의 연구가 관심을 받고 있다. 기존의 연구에서 미크로네시아에서 채취한 해면동물 40여종에 대하여 항증식 효과를 다양한 암세포주에서 검색한 바 있다. 본 연구에서는 그 중 Coscinoderma sp.의 작용 및 그 기전을 살펴보았다. 특히, 암 억제유전자 p53의 발현을 억제시킨 세포주(HCT116 p53KO과 RKO-E6)에서의 차이점을 비교하였다. 세포생존률 시험에서 Coscinoderma sp. 추출물은 p53의 유무와 상관없이 암세포의 증식을 억제하였음을 확인하였다. 이 암세포 증식 억제 효과가 p53 존재에 따라 다르게 나타나는지 알아보기 위하여 세포사멸 관련 단백질 발현양을 Coscinoderma sp. 처리한 각 세포주에서 비교하였다. 그 결과, Coscinoderma sp.를 HCT16 세포주에 처리하였을 때, p53과 Noxa의 발현이 증가하는 것을 관찰하였고, caspase-9이 분절되면서 감소하는 것으로부터 apoptosis를 일으킨다고 여겨진다. 반면, p53이 결핍된 HCT116세포주에서는 Coscinoderma sp.에 의하여 p21과 mTOR의 발현이 증가되는 것을 확인하였고, 이는 senescence를 야기할 수 있다고 여겨진다. 본 연구로부터 Coscinoderma sp.는 p53의 존재여부에 따라 상이한 작용기전을 매개하여 대장암 세포주의 증식을 억제한다는 것을 알 수 있었다. 이는 새로운 항암제의 개발 가능성을 제시하는 것으로, Coscinoderma sp.의 활성 성분에 대한 지속적인 연구가 이루어 져야 할 것으로 보인다.
알츠하이머 질환은 인지능력과 행동능력 두 가지에 모두 영향을 미치는 진행적 노인성 치매증의 일종으로 정확한 발병기전은 아직 알려져 있지 않으나 환경적 요인 및 유전적 요인이 모두 중요한 위험인자로 알려져 있다. 본 연구에서는 8주간 에탄올에 노출된 Aldh2 knockout mouse 뇌조직을 분리하여 알츠하이머 질환의 지표로 잘 알려진 아밀로이드 베타와 NF-kB 발현을 평가하고 이러한 변화가 ALDH2 효소의 활성에 따라 어떻게 달라지는 지 비교하였다. 그 결과, 8주간 에탄올을 경구 투여한 마우스에서 ALDH2 효소의 활성에 따른 NF-kB 농도의 차이는 확인할 수 없었으나, ALDH2 효소 활성은 알코올 노출에 의한 해마조직의 아밀로이드 베타 축적에서 중요한 영향을 미치는 것으로 나타났다(p<0.05). 이러한 결과는 ALDH2 효소가 결핍된 사람이 결핍되지 않은 사람에 비해 알코올에 의한 알츠하이머 질환의 발생에 보다 민감할 수 있음을 시사한다.

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
