Apoptosis inhibitor 5 increases metastasis via Erk-mediated MMP expression

  • Apoptosis inhibitor 5 (API5) has recently been identified as a tumor metastasis-regulating gene in cervical cancer cells.However, the precise mechanism of action for API5 is poorly understood. Here, we show that API5 increases the metastatic capacity of cervical cancer cells in vitro and in vivo via up-regulation of MMP-9. Interestingly, API5-mediated metastasis was strongly dependent on the Erk signaling pathway. Conversely, knock-down of API5 via siRNA technology decreased the level of phospho-Erk, the activity of the MMPs, in vitro invasion, and in vivo pulmonary metastasis. Moreover, the Erk-mediated metastatic action was abolished by the mutation of leucine into arginine within the heptad leucine repeat region, which affects protein-protein interactions. Thus, API5 increases the metastatic capacity of tumor cells by up-regulating MMP levels via activation of the Erk signaling pathway. [BMB Reports 2015; 48(6): 330-335]

  • 경쟁 시스템에서 최종 우승자를 결정하기 위해 다양한 방법을 사용한다. 많은 스포츠 경기에서는 1-in-2 토너먼트 방식으로 우승자를 가리거나 이의 변형된 방법으로 우승자를 가린다. 본 연구에서는 일반적인 경기보다 훨씬 많은 경쟁 참여자가 존재하고, 이를 많은 임의의 심사자가 심사하는 상황에서 최종 우승자를 가리는 상황에서 활용할 수 있는 경쟁 시스템의 한 방법을 제안한다. 이와 같이 다수의 경쟁자와 다수의 심사자가 존재하는 상황은 가상공간에서 흔히 보는 각종 앱의 선호도 조사 등에서 발생한다. 본 연구에서는 knock-out 토너먼트를 발전시켜 각 라운드마다 경쟁자를 임의배정하는 다단 랜덤 토너먼트를 제안하고 제안된 방식의 공정성에 대해서 알아본다. 또한 일정한 조건 하에서 특정 심사자가 미치는 심사 영향력을 수치화하여 각 심사자의 영향력을 비교할 수 있게 하였다.

레스베라트롤에 의한 인간 암세포주, A549와 SKOV3의 p53의존적 Apoptosis 유발 (Induction of p53-dependent Apoptosis by Resveratrol in Human Cancer Cells, A549 and SKOV3)

  • Resveratrol은 포도, 오디, 땅콩과 같은 많은 과일과 채소에 존재하는 폴리페놀 화합물로써 다양한 생물학적 효과를 가진다고 보고되어있다. 그러나, resveratrol이 A549 폐암세포에서 유도하는 apoptosis에 관여하는 분자적 기전 및 anoikis에 관해서는 명백하게 밝혀지지 않았다. 본 연구에서, 우리는 정상적인 p53 유전자를 갖는 A549 세포에서 resveratrol의 효과를 조사하고, p53 유전자가 결실된 SKOV3 난소암 세포와 그 효과를 비교했다. Resveratrol은 확실하게 SKOV3 세포에 비해 농도, 시간의존적으로 A549 세포의 생존과 증식을 억제했다. 또한 resveratrol은 A549 세포의 apoptosis를 유도했지만 세포의 anoikis 저항성에는 영향을 미치지 않았다. 더불어, p53 유전자 기능이 불완전 소실된(knock-down) A549 세포의 생존과 증식은 resveratrol에 의해 변하지 않았다. 그러므로, 본 연구의 결과는 resveratrol의 항암 효과가 기능을 하는 p53 유전자의 존재에 의존한다는 것을 보여준다. 결론적으로, 우리는 resveratrol이 p53 유전자에 의존적으로 A549 세포에 대해 항암효과를 가진다는 것을 입증했다.

The Role of Stem Cells and Gap Junctional Intercellular Communication in Carcinogenesis

  • Understanding the process of carcinogenesis will involve both the accumulation of many scientific facts derived from molecular, biochemical, cellular, physiological, whole animal experiments and epidemiological studies, as well as from conceptual understanding as to how to order and integrate those facts. From decades of cancer research, a number of the "hallmarks of cancer" have been identified, as well as their attendant concepts, including oncogenes, tumor suppressor genes, cell cycle biochemistry, hypotheses of metastasis, angiogenesis, etc. While all these "hallmarks" are well known, two important concepts, with their associated scientific observations, have been generally ignored by many in the cancer research field. The objective of the short review is to highlight the concept of the role of human adult pluri-potent stem cells as "target cells" for the carcinogenic process and the concept of the role of gap junctional intercellular communication in the multi-stage, multi-mechanism process of carcinogenesis. With these two concepts, an attempt has been made to integrate the other well-known concepts, such as the multi-stage, multi-mechanisn or the "initiation/promotion/progression" hypothesis; the stem cell theory of carcinogenesis; the oncogene/tumor suppression theory and the mutation/epigenetic theories of carcinogenesis. This new "integrative" theory tries to explain the well-known "hallmarks" of cancers, including the observation that cancer cells lack either heterologous or homologous gap junctional intercellular communication whereas normal human adult stem cells do not have expressed or functional gap junctional intercellular communication. On the other hand, their normal differentiated, non-stem cell derivatives do express connexins and express gap junctional intercellular communication during their differentiation. Examination of the roles of chemical tumor promoters, oncogenes, connexin knock-out mice and roles of genetically-engineered tumor and normal cells with connexin and anti-sense connexin genes, respectively, seems to provide evidence which is consistent with the roles of both stem cells and gap junctional communication playing a major role in carcinogenesis. The integrative hypothesis provides new strategies for chemoprevention and chemotherapy which focuses on modulating connexin gene expression or gap junctional intercellular communication in the premalignant and malignant cells, respectively.

A Study on Furniture Design for Disassembly

  • Modernity which is superficial phenomenon set off the mass scale for mass consumption and provide uniformly artificial environment. But natural destruction, environment pollution, resources exhaustion and so on has been caused by this and now ecology is threatened by destruction and damage beyond the limitation and human beings survival is even threatened. Accordingly furniture development for environment preservation considered environment problem is the urgent real situation. Recent paradigm is the concept of Eco-design which is the green design possible to live together in symbiosis, and new types of alternative furniture are needed in Korea as well. 'Furniture for disassembly' is presented as new method for alternative furniture. Furniture for disassembly can be presented by mainly two directions. The first main characteristic is what is assembled by the use of woodworking joints technique as an assembly structure system without any hardware. The second is what is presented as the structure possible to be assembled by simple manual tools with hardware without any glue. The advantages of furniture for disassembly are environment preservation, space application, transportation efficiency and shapeliness. In manufacture method which is different from present furniture, the application of traditional truss technique which uses various types of custom-made and connection technique in case of assemble structure system without hardware is the typical differences. This assembly method expects not only interest induction about assembly and disassembly of diagram per sub materials but also the development of emotion, the improvement of collaboration, space perception ability and shape sense, the improvement of solid body structure insight and so on, when it use in the furniture for children with the application to many kinds of structure with BANGDOOSANJ (Wedged), JUMUGJANGBU (Dovetail) or NABIEUNJANG (Dovetail Keys) and so on.

Multi-zone 모델링을 통한 온도성층화와 농도성층화가 존재하는 DME HCCI 엔진의 운전영역에 관한 수치해석연구 (Comparison of DME HCCI Operating Ranges for the Thermal Stratification and Fuel Stratification based on a Multi-zone Modeling)

  • This work investigates the potential of in-cylinder thermal stratification and fuel stratification for extending the operating ranges in HCCI engines, and the coupling between thermal stratification and fuel stratification. Computational results areemployed. The computations were conducted using both a custom multi-zone version and the standard single-zone version of the Senkin application of the CHEMKINII kinetics rate code, and kinetic mechanism for di-methyl ether (DME). This study shows that the potential of thermal stratification and fuels stratification for extending the high-load operating limit by a staged combustion event with reduced pressure-rise rates is very large. It was also found that those stratification offers good potential to extend low-load limit by a same mechanism in high-load. However, a combination of thermal stratification and fuel stratification is not more effective than above stratification techniques for extending the operating ranges showing similar results of fuel stratification. Sufficient condition for combustion (enough temperature for) turns misfire in low-load limit to operate engines, which also leads to knock in high-load limit abruptly due to the too high temperature with high. DME shows a potential for maximizing effect of stratification to lower pressure-rise rate due to the characteristics of low-temperature heat release.

Memantine Induces NMDAR1-Mediated Autophagic Cell Death in Malignant Glioma Cells

  • Objective : Autophagy is one of the key responses of cells to programmed cell death. Memantine, an approved anti-dementia drug, has an antiproliferative effect on cancer cells but the mechanism is poorly understood. The aim of the present study was to test the possibility of induction of autophagic cell death by memantine in glioma cell lines. Methods : Glioma cell lines (T-98 G and U-251 MG) were used for this study. Results : The antiproliferative effect of memantine was shown on T-98 G cells, which expressed N-methyl-D-aspartate 1 receptor (NMDAR1). Memantine increased the autophagic-related proteins as the conversion ratio of light chain protein 3-II (LC3-II)-/LC3-I and the expression of beclin-1. Memantine also increased formation of autophagic vacuoles observed under a transmission electron microscope. Transfection of small interfering RNA (siRNA) to knock down NMDAR1 in the glioma cells induced resistance to memantine and decreased the LC3-II/LC3-I ratio in T-98 G cells. Conclusion : Our study demonstrates that in glioma cells, memantine inhibits proliferation and induces autophagy mediated by NMDAR1.

DOBI is cleaved by caspases during TRAIL-induced apoptotic cell death

  • Downstream of Bid (DOBI) known as Pus10, has been identified as a modulator of TRAIL-induced cell death using RNAi library screening. The crystal structure of DOBI has revealed that it is a crescent-shaped protein containing the pseudouridine synthase catalytic domain and a THUMP-containing domain. Here, we demonstrated that DOBI is expressed in various tissues such as heart and lung, and is also expressed in various tumor cells such as HeLa and A549. Although ectopic expression of DOBI does not promote TRAIL death signaling in HeLa cells, knock-down of DOBI expression using shRNA inhibited TRAIL death signaling. DOBI is cleaved into a 54 kD cleaved DOBI during cell death, and the recombinant DOBI protein can be directly cleaved by caspases-3, or -8 in vitro. Together, these data suggest that the cleaved DOBI may acquire a new function, possibly by cooperating with tBid in the mitochondrial event of cell death caused by TRAIL.

A Research on Industrial Trend Analysis of Materials & Components in Jeollanam-do

  • Domestic materials & components industry is making a shift from quantitative growth to qualitative growth, while the paradigm of a future society is changing rapidly to energy, mobility, smartification and etc. The main features of materials & components industry are analyzed broadly based on three aspects that are: ① industry Status, ② the value chain structure, ③ the competitiveness. With regard to the national aspect, it is shown that the number of an export and a trade surplus increased consistently after the government advanced the policy on materials & components and Korea's trade deficit with Japan has been decreased. Moreover, the state's export ranking increased from tenth place in 2001 into sixth place in 2017, dedicating to the growth of a national economy. The analysis of the chain value indicates that specifications of an end product are set when the materials & components are combined with manpower, infrastructure and technologies. In terms of the competitiveness, it requires a long time and high costs to develop technologies on materials and it is important for components to have a process innovation and credibility wired. Following the industry structure with priority given on materials, Jeonnam Province accounts for 14.8% of national material production. As a result of analyzing Jeonnam's export competitiveness, it is measured that a serious polarization exists, revealing there are 6 major technology industries by 11 categories for materials & components, Jeonnam has presented 6 major materials & components considering the key industry and the new industry in future to remedy the mentioned problem so far, and plans to knock on the industry development through 10 different strategies.

딥러닝 기반 컨테이너 적재 정렬 상태 및 사고 위험도 검출 기법 (Shipping Container Load State and Accident Risk Detection Techniques Based Deep Learning)

  • 최근 항만에서는 부정확한 컨테이너 적재로 인해 컨테이너가 강풍에 쉽게 쓰러지는 컨테이너 붕괴 사고가 빈번이 발생하고 있으며 이는 물적 피해와 항만 시스템 마비로 이어지고 있다. 본 논문에서는 이런 사고를 미연에 방지하기 위해 딥러닝 기반 컨테이너 적재 상태 및 사고 위험도 검출 시스템을 제안한다. 제안된 시스템은 darknet 기반 YOLO 모델을 활용하여 컨테이너 상하의 코너캐스팅을 통해 컨테이너 정렬 상태를 실시간으로 파악하고 관리자에게 사고 위험도를 알리는 시스템이다. 제안된 시스템은 추론 속도, 분류 정확도, 검출 정확도 등을 성능 지표와 실제 구현 환경에서 최적의 성능을 보인 YOLOv4 모델을 객체 인식 알고리즘 모델로 선택하였다. 제안된 알고리즘인 YOLOv4가 YOLOv3보다 추론속도와 FPS의 성능 측면에서 낮은 성능을 보이기는 했지만, 분류 정확도와 검출 정확도에서 강력한 성능을 보임을 증명하였다.
