Biocatalysis and Biotransformation for the Production of Chiral Epoxides (바이오촉매 및 생물전환을 이용한 광학활성 에폭사이드 제조)

  • Chiral epoxides are important chiral synthons in organic synthesis for the production of chiral pharmaceuticals and functional food additives. Chiral epoxides can be synthesized by enantioselective introduction of oxygen to double bond of substrate by monooxygenase. Peroxidase also carry out asymmetric epoxidation of alkene in the presence of hydrogen peroxide. Kinetic resolution of racemic epoxides via enantioselective hydrolysis reaction by epoxide hydrolase (EH) is a very promising method since chiral epoxides with a high optical purity can be obtained from cheap and readily available racemic epoxides. In this review, various biocatalytic approaches for the production of chiral epoxides with several examples are presented and their commercial potential is discussed.

Production of Chiral (S)-styrene Oxide by Rhodosporidium sp. SJ-4 which has an Epoxide Hydrolase Activity (에폭사이드 가수분해효소를 갖는 Rhodosporidium sp. SJ-4를 이용한 광학활성 (S)-styrene Oxide의 생산)

  • A yeast strain utilizing styrene epoxide as a sole carbon and energy source was isolated from soil samples for the production of enantiopure of styrene epoxide by kinetic resolution. The strain, identified as a Rhodosporidium toruloides SJ-4, expressed an epoxide hydrolase which preferentially converted (R)-styrene epoxide into the corresponding diol. A maximum activity of 135 U/L was observed when biomass (dry cell mass) reached 6.7 g/L at 21 h of batch culture. Under the partially optimized reaction conditions ($35^{\circ}C$ and pH 8.0), the optically pure (S)-styrene epoxide was obtained with the yield of 21% when the initial substrate concentration was 100 mM. The reaction was completed at 9 h.

  • Enantiopure epoxides are valuable intermediates for the asymmetric synthesis of enantiopure bioactive compounds. Microbial epoxide hydrolases (EHs) are versatile biocatalysts for the preparation of enantiopure epoxides by enantioselective hydrolysis of cheap and easily available racemic epoxide substrates. EHs are commercially potential biocatalysts due to their characteristics such as high enantioselectivity, cofactor-independent catalysis, and easy-to-prepare catalysts. In this paper, recent progresses In molecular engineering of EHs are reviewed to evaluate the commercial feasibility of EH-catalyzed hydrolytic kinetic resolution for the production of enantiopure epoxides.

Enantioselective Kinetic Resolution of Racemic Styrene Oxide using Recombinant Marine Fish Epoxide Hydrolase of Mugil cephalus (해양 어류 Mugil cephalus 유래의 에폭사이드 가수분해효소를 이용한 라세믹 styrene oxide의 입체선택적 분할 반응)

  • The microsomal epoxide hydrolase gene (referred to as mMCEH) of Mugil cephalus was cloned by PCR, and then inserted to pColdI and pET-21b(+) vector, respectively. The recombinant E. coli possessing the recombinant plasmids exhibited the enantioperference toward (R)-styrene oxide. When enantioselective kinetic resolutions were conducted with 20 mM racemic styrene oxide, enantiopure (S)-styrene oxide was obtained with high enantiopurity more than 99% enantiomeric excess (ee) and 24.50% yield by using the recombinant E. coli harboring pET-21b(+)/mMCEH.

Lipase-catalyzed Remote Kinetic Resolution of Quaternary Carbon-containing Alcohols and Determination of Their Absolute Configuration

  • The quaternary carbon-containing alcohols (1-6) were resolved enantioselectively by various lipases such as PFL (Pseudomonas fluorescens lipase), LAK (Pseudomonas fluorescens lipase), CRL (Candida rugosa lipase) and PCL (Pseudomonas cepacia lipase). The enzymatic resolution of racemic alcohol $({\pm})-2$ gave the excellent enantioselectivity in favor of (S)-2d in 99% ee, while those of the racemic alcohols (1, 3, 4, 5 and 6) gave the resolved alcohols with moderate to good enantioselectivity. Also, their absolute configurations were determined by chemical transformation to the known compounds.

  • A high resolution picosecond absorption kinetic spectrometer utilizing dye emission and a streak camera is presented and compared with other methods of picosecond transient absorption measurements. Typical transient absorption and bleach recovery kinetics measured with this spectrometer are shown. Single wavelength transient absorption or ground state bleach recovery kinetics are determined on the basis of a single laser shot, so that the samples are relatively free frm decomposition by irradiation. Excellent kinetics may be obtained from the near UV to the near IR and are not subject to interference from luminescence of samples. The sensitivity of this spectrometer is very high and it is reasonably easy and convenient to set up and use.

  • Kinetic resolution of various racemic aromatic epoxides by newly isolated Aspergillus niger LK has been investigated, and enantioselectivity of whole-cell biocatalyst was analyzed. The epoxide hydrolase (EHase) of A. niger LK was cloned using RT-PCR. The sequence homology was compared with that of other microbial EHase and the gene for EHase was characterized at molecular level.
