개에서 페노바비탈 장기 투여로 유발된 표재성 괴사성 피부염 1례 (Superficial Necrolytic Dermatitis (Hepatocutaneous Syndrome) in a Dog with History of Long-Term Phenobarbital Administration)

  • 10세령의 암컷 말티즈가 족부 괴양 및 안면부 괴양성 피부병으로 서울대학교 수의과대학 동물병원에 내원하였다. 환자는 장기간의 페노바르비탈 투여 경력을 가지고 있었으며, 안면부 및 족부의 피부병변의 분포 양상과 간효소 수치의 상승 및 초음파 영상에서의 간의 저에코성병변들을 참고하여, 최종적으로 피부 생검을 통해 표재성 괴사성 피부염으로 진단하였다. 8개월간의 간보조제 경구 투여 및 아미노산제제의 말초 혈액 투여로 피부 증상은 약간 호전되었으나, 비가역적인 간의 손상은 회복이 힘들었으며, 8개월간의 치료 후 사망하였다.

Oral Administration of Lactobacillus plantarum HY7714 Protects Hairless Mouse Against Ultraviolet B-Induced Photoaging

  • Ultraviolet (UV) irradiation alters multiple molecular pathways in the skin, thereby inducing skin damage, including photoaging. In recent years, probiotics have gained interest due to their beneficial effects on skin health, such as inhibiting atopic dermatitis and improving skin immunity or inflammation. However, little is known about the effects of probiotics on UVB-induced photoaging. In this study, we evaluated the effect of Lactobacillus plantarum HY7714 against UVB-induced photoaging in human dermal fibroblasts and hairless mice. The results showed that L. plantarum HY7714 treatment effectively rescued UVB-reduced procollagen expression through the inhibition of UVB-induced matrix metalloproteinase (MMP)-1 expression in human dermal fibroblasts. Data from a western blot showed that L. plantarum HY7714 inhibited the phosphorylation of Jun N-terminal kinase, thereby suppressing the UVB-induced phosphorylation and expression of c-Jun. Oral administration of L. plantarum HY7714 clearly inhibited the number, depth, and area of wrinkles in hairless mouse skin. Histological data showed that L. plantarum HY7714 significantly inhibited UVB-induced epidermal thickness in mice. Western blot and zymography data also revealed that L. plantarum HY7714 effectively inhibited MMP-13 expression as well as MMP-2 and -9 activities in dermal tissue. Collectively, these results provide further insight regarding the skin biological actions of L. plantarum HY7714, a potential skin anti-photoaging agent.

국내 유통 축·수산물 중 페니실린계 동물용의약품에 대한 잔류실태조사 (Residue analysis of penicillines in livestock and marine products)

  • 페니실린계 항생물질은 ${\beta}$-lactam 계열의 항생물질로서 인간과 동물에게 널리 사용되어지는 항생물질이다. 이러한 항생항균물질들의 오남용은 소비자가 항생제에 노출될 위험성을 증가시킨다. 본 연구에서는 국내 유통 중인 축 수산물을 대상으로 LC-MS/MS를 이용하여 페니실린계 동물용의약품 7종인 아목시실린, 암피실린, 벤질페니실린, 클록사실린, 디클록사실린, 나프실린, 옥사실린에 대한 잔류량을 조사하였다. 특이성, 정밀성, 회수율, 직선성을 통해 시험법의 유효성을 확인하였으며 아목시실린을 제외한 나머지 6종은 모두 76% 이상의 회수율을 보였고, RSD는 10% 이하로 나타내었다. 시료는 80% 아세토니트릴로 추출한 후 고체상 추출 카트리지를 사용하여 정제 한 후 분석하였다. 축 수산물 225건을 대상으로 잔류량을 조사한 결과 벤질페니실린이 축산물에서 12건, 수산물에서 7건으로 총 19건이 검출되었고 나머지 6개 항목인 아목시실린, 암피실린, 클록사실린, 디클록사실린, 나프실린, 옥사실린은 검출되지 않았다. 벤질페니실린이 검출된 축산물은 우유와 알을 제외한 소고기, 돼지고기, 닭고기이었으며 검출 수준은 0.001~0.009 mg/kg으로 잔류허용기준(MRL)인 0.05 mg/kg 이하로 검출되었다. 수산물에서 검출된 7건은 잔류허용기준인 0.03 mg/kg 이하로 검출되었다. 본 연구사업에서 수행한 모니터링 결과 해당항목의 경우 대부분 기준치 이하로 검출되어 비교적 안전한 수준인 것으로 나타났으나, 향후 지속적인 잔류실태조사와 제도적인 관리지도를 통하여 동물용의약품의 오남용에 대한 안전관리가 이루어져야 할 것으로 사료된다.

우분으로부터 Bacillus subtilis CH-10의 분리 및 균주가 분비하는 Cellulase의 특성에 관한 연구 (Isolation and Characterization of Bacillus subtilis CH-10 Secreting Cellulase from Cattle Manure)

  • 우분으로부터 cellulase를 생산하는 미생물을 congo red 염색과 활성측정을 통해 선발하여 cellulase 활성이 우수한 균을 분리하였다. 분리균은 생화학적, 형태학적, 균체 지방산 조성을 근거로 Bacillus subtilis CH-10으로 동정하였다. 분리균이 분비하는 효소학적 특성 중 효소생산 조건은 초기 pH7.5 및 배양돈도 50${\circ}C$ 그리고 48시간 배양이 가장 적합하였다. 조효소액에서 CMCase 최적 온도는 75${\circ}C$이었고, 온도안정성은 50${\circ}C$까지 70%의 효소활성을 유지하였다. 조효소액에서 CMCase 최적 pH는 7.5이었고, pH 안정성은 pH7.5~9.0 영역에서 70%의 효소활성을 유지하였다. 분리균을 CMC 배지에서 37${\circ}C$, 24시간 배양시 나타난 CMCase와 Fpase 활성은 각각 1.13 U/㎖와 0.16U/㎖ 였으나 avicelase와 ${\beta}$-glucosidase의 활성은 검출되지 않았다. CMC-SDS-PAGE 방법으로 3개의 효소활성 band를 확인하였고, Cel 1 및 2 그리고 Cel 3의 분자량은 각각 약 39 및 41 그리고 57kDa이었다.

  • 세균포자는 살균소독제에 대해 영양세포보다 수백~수천배이상 저항성을 가지고 있기 때문에 과산화수소 및 과산화초산을 원료로 하는 아포살균용 살균소독제는 Escherichia coli 및 Staphylococcus aureus의 감소율을 측정하는 현행 살균소독제 유효성 평가방법을 적용하기 어렵다. 따라서, 아포살균용 살균속독제의 유효성 평가를 위해 Bacillus subtilis ATCC 6633 포자를 표준균주로 하여 아포살균용 살균소독제를 처리했을 때 초기포자수를 $10^3$이상 감소시키는지를 측정하는 정량적 표준시험방법을 확립하였다. 확립된 시험법은 아포살균력을 측정하는 국가표준시험법으로서 아포살균용 살균소독제의 유효성 평가에 적합하였다.

Characterization of Rice Mutants with Enhanced Susceptibility to Rice Blast

  • As a first step towards identifying genes involving in the signal transduction pathways mediating rice blast resistance, we isolated 3 mutants lines that showed enhanced susceptibility to rice blast KJ105 (91-033) from a T-DNA insertion library of the japonica rice cultivar, Hwayeong. Since none of the susceptible phenotypes co-segregated with the T-DNA insertion we adapted a map-based cloning strategy to isolate the gene(s) responsible for the enhanced susceptibility of the Hwayeong mutants. A genetic mapping population was produced by crossing the resistant wild type Hwayeong with the susceptible cultivar, Nagdong. Chi-square analysis of the $F_2$ segregating population indicated that resistance in Hwayeong was controlled by a single major gene that we tentatively named Pi-hy. Randomly selected susceptible plants in the $F_2$ population were used to build an initial map of Pi-hy. The SSLP marker RM2265 on chromosome 2 was closely linked to resistance. High resolution mapping using 105 $F_2$ plants revealed that the resistance gene was tightly linked, or identical, to Pib, a resistance gene with a nucleotide binding sequence and leucine-rich repeats (NB-LRR) previously isolated. Sequence analysis of the Pib locus amplified from three susceptible mutants revealed lesions within this gene, demonstrating that the Pi-hy gene is Pib. The Pib mutations in 1D-22-10-13, 1D-54-16-8, and 1C-143-16-1 were, respectively, a missense mutation in the conserved NB domain 3, a nonsense mutation in the 5th LRR, and a nonsense mutation in the C terminus following the LRRs that causes a small deletion of the C terminus. These findings provide evidence that NB domain 3 and the C terminus are required for full activity of the plant R gene. They also suggest that alterations of the resistance gene can cause major differences in pathogen specificity by affecting interactions with an avirulence factor.

국내 유통 농산물 중 EBI계 농약 모니터링과 위해도 평가 (Monitoring of Ergosterol Biosynthesis Inhibitor (EBI) Pesticide Residues in Commercial Agricultural Products and Risk Assessment)

  • 우리 국민의 주식이 되는 쌀(백미)과 콩류, 감귤류, 열대과 일류, 엽채류, 엽경채류, 근채류, 과채류, 버섯류에 속하는 농산물(989건)에 대해 8종의 EBI계 농약의 잔류 여부를 분석하였다. 그 결과 8건의 검체에서 농약이 검출되었고 농약 수는 5종이 검출되었다. 기준 미설정 농약은 2종이었으며, 곡류 134건 중 4건 검출로 3.0%, 채소류 471건 중 4건 검출로 0.8%의 검출율을 나타내었다. 그 외 콩류(48건), 견과종실류(72건), 과실류(197건), 버섯류(67건)에서는 전혀 검출되지 않았으며 부적합율은 0.1%로 나타났다. 국내 미등록농약인 paclobutrazole이 근대에서 잠정허용기준을 초과하는 것으로 확인되었다. 그리고 EBI계의 검출율은 0.8%로 매우 낮았으며 검출된 농약의 경우도 기준규격 이하로 안전한 수준이었다.

당귀사역탕(當歸四逆湯)이 백서의 갑상선기능저하증에 미치는 영향 (The Effects of Danggwisaeyeoktang(當歸四逆湯) on the Hypothyroidism of Rat)

  • Objectives : The aim was to study experimentally the effects of Danggwisaeyeoktang(當歸四逆湯) on the hypothyroidism of rats induced by thiouracil. Methods : Spargue-Dawley rat were separated into three groups. Except normal group, the other two groups were treated with thiouracil 50mg/kg/days for 30 days by oral administration. Among the two group, one group was treated DGSYT extract 0.54g/kg/days for 20 days separately. T3-uptake;, T3, T4, TSH, hematological values, enzyme activity, total cholesterol and Na+ content charge in the Serum of rats were measured after medication of solid extract of DGSYT. Result : As a result of this study, in the serum T3-Uptake and T4 level, hematological values and sodium were increased in DGSYT group compared to controls. The serum TSH, total cholesterol and body weight were decreased in DGSYT group compared to controls. Conclusion : According to the above results, it is suggested that DGSYT is effective to cure hypothyroidism.

마그네슘합금내 개재물 함유량에 따른 파단면의 명도변화 (The Brightness Change of Fractured Surface in Accordance with Inclusion Contents of Magnesium Alloy)

  • Pure magnesium and magnesium alloys have been applied to various kinds of industrial fields, especially automotive and electronic parts. These parts are manufactured mainly through a diecasting process. These days, magnesium ingots are used as raw material, and recycled ingots are often used for commercial purposes. But the quality of virgin magnesium and recycled ingots is not secure. Therefore, massive casting defects can occur, and some things manufactured can be damaged by these defects. This study evaluated the inclusions of virgin magnesium and recycled ingot. It also included composition analysis by spectrometer, measuring inclusion contents by SEM & EDS, and performing a brightness test on fractured surfaces. The brightness test is generally very easy and obtains results quickly, so its results have been compared with the results obtained from various test methods. From the test results, we obtained a satisfactory result in evaluating inclusion and oxide. The brightness values are lower as the inclusion contents are higher. When the brightness value is over 47 in AM50A and 44 in AZ91D, the mechanical properties are expected to be good.

Comparative Analysis of Historical National Image Advertising in Korea - Video Advertisements Produced between 1998-2017

  • Since national image video advertisements are a means of public relations with wide delivery and major repercussions at home and abroad, this study performs a comparative analysis of video advertisements aired by previous governments as reported by Daejung Kim, Moohyun Roh, Myungbak Lee, and Geunhye Park. The characteristics of the previous governments, whether or not social trends were reflected, and the importance of traditional and modern elements were examined. As we describe, there are clear differences in video advertisements by government regime, and while messages expressing traditional values of Eastern culture have gradually decreased, reflecting the trends of the times, the messages emphasizing the modern values of the West have gradually increased. Our research confirmed that traditional elements such as 'Samulnori' and 'Taekwondo' are gradually disappearing. In addition, it was confirmed that the collective values, traditional values, and humanism represented by the 'large crowd' and 'traditional elements' in the Kim Dae-jung and Roh Moo-hyun administration changed in the Lee Myung-bak and Park Geun-hye administrations to more individualistic, materialistic, Western values. This study is meaningful in that it analyzed the components and characteristics of national image advertisements by governments in the past, how social trends were reflected, and the weight difference between traditional and modern elements. Based on this research, the significance of the current Moon Jae-in government's groundwork for follow-up research should not be understated.
