Breeding of a New Yellow Gerbera hybrida "Golden Time" with High Yield for Cut Flower (황색 대륜 절화용 다수성 거베라 신품종 "골든타임" 육성)

  • A new yellow gerbera (Gerbera hybrida Hort.) cultivar "Golden Time" was developed from a cross between '99-ga-2' and '99-na-7-6', followed by seedling and line selections at the Flower Research Institute, Gyeongnam Agricultural Research and Extension Services (ARES) in 2007. Detailed characteristics of the new cultivar were evaluated from 2004 to 2007. "Golden Time" has semi-double type large flowers in a good harmony with yellow (RHS, 17-B) ray floret and brown center. It has good, stable flower shape and strong peduncles, and its vase life was 12.3 days. The average flower yield of "Golden Time" was about 49.5 stems per plant/year in greenhouse trails during 2004 and 2007. This cultivar was registered to the Korea Seed and Variety Service (KSVS) for commercialization in 2009. Year-round production of this cultivar is possible in the greenhouse condition throughout the Korean climate.

  • A new spray chrysanthemum (Dendranthema grandiflorum) cultivar, 'Magic Yellow' was developed from a cross between 'Artist pink' and 'S03-47' by selection of seedlings and lines at the Flower Research Institute, Gyeongnam Agricultural Research and Extension Services (ARES) from 2003 to 2008. Its characteristics were investigated three times from 2006 to 2008 under condition of forcing culture in spring and retarding culture in autumn. The natural flowering time of 'Magic Yellow' was October 18th, and year-round production was possible by day length treatment. This cultivar has single-type flowers in good harmony with bi-color ray floret and green center. Its flower was 6.0 cm in diameter, and it had 11.4 flowers per stem in autumn. The days to flowering under the short day treatment is about 45 days in spring season, and 'Magic Yellow' showed the vase life of 22.9 days in autumn. This cultivar was registered for the protection in 2008.

  • A new spray chrysanthemum (Dendranthema grandiflorum) cultivar, 'Dimple' was developed from a cross between 'champy' and 'SP05-135', by selection of seedlings and lines at the Flower Research Institute, Gyeongnam Agricultural Research and Extension Services (ARES) from 2005 to 2009. Its characteristics were investigated three times from 2007 to 2009 under condition of forcing culture in spring and retarding culture in autumn. The natural flowering time of 'Dimple' was October 24th, and year-round production was possible by day-length control. This cultivar has single type flowers with bright yellow ray floret and green center. The plant is very vigorous and very stable in the color of petals when it is cultivated even under high temperature condition in summer. Its flower was 4.7 cm in diameter, and it had 20.3 flowers per stem in autumn. The days to flowering under the short day treatment is about 50 in spring season, and 'Dimple' showed the vase life of 21.3 days in autumn. This cultivar was registered for the protection in 2009.

Effects of Ethephon Treatments on Accelerating of Maturity of Sweet Persimmon 'Nishimurawase' Cultivar (서촌조생 단감의 숙기촉진에 관한 에세폰 효과)

  • This study was carried out to investigate the effects of treatments and adequate concentration of ethephon on coloring promotion of peel of sweet persimmon 'Nishimurawase' cultivar. High concentration(30 and 40㎎/l) of ethephon treatments increased the coloring of peel. but they were unfavorable to marketability due to the decrease of firmness, the increase of softening ratio, and the short of marketing critical period of fruits. Low concentration(20㎎/l) of ethephon treatment maintained higher marketability in terms of coloring of peel, firmness, softening ratio, and marketing critical period of fruit.

Pi5 and Pii Paired NLRs Are Functionally Exchangeable and Confer Similar Disease Resistance Specificity

  • Effector-triggered immunity (ETI) is an effective layer of plant defense initiated upon recognition of avirulence (Avr) effectors from pathogens by cognate plant disease resistance (R) proteins. In rice, a large number of R genes have been characterized from various cultivars and have greatly contributed to breeding programs to improve resistance against the rice blast pathogen Magnaporthe oryzae. The extreme diversity of R gene repertoires is thought to be a result of co-evolutionary history between rice and its pathogens including M. oryzae. Here we show that Pii is an allele of Pi5 by DNA sequence characterization and complementation analysis. Pii-1 and Pii-2 cDNAs were cloned by reverse transcription polymerase chain reaction from the Pii-carrying cultivar Fujisaka5. The complementation test in susceptible rice cultivar Dongjin demonstrated that the rice blast resistance mediated by Pii, similar to Pi5, requires the presence of two nucleotide-binding leucine-rich repeat genes, Pii-1 and Pii-2. Consistent with our hypothesis that Pi5 and Pii are functionally indistinguishable, the replacement of Pii-1 by Pi5-1 and Pii-2 by Pi5-2, respectively, does not change the level of disease resistance to M. oryzae carrying AVR-Pii. Surprisingly, Exo70F3, required for Pii-mediated resistance, is dispensable for Pi5-mediated resistance. Based on our results, despite similarities observed between Pi5 and Pii, we hypothesize that Pi5 and Pii pairs require partially distinct mechanisms to function.

A New Gerbera Cultivar, 'Red Auction' with Red Color and Semi-double for Cut Flower (적색 반겹꽃 절화용 거베라 '레드옥션' 육성)

  • A new standard red color gerbera (Gerbera hybrida Hort.) 'Red Auction' was developed from a cross of '01-120-195' and 'Misty red' followed by seedling and line selections at the Flower Research Institute, Gyeongsangnam-do Agricultural Research & Extension Services (ARES) in 2008. Characteristics trials were conducted three times from 2006 to 2008. 'Red Auction' has semi-double type in good harmony with red color (RHS 45-A) ray floret and a good stable flower shape and strong peduncle. Its vase life was 11.3 days. The average yield of 'Red Auction' was 48.8 flowers per plant a year in greenhouse yield trials carried out from 2006 to 2008. This cultivar registered for commercialization in 2010.

Inheritance of P34 Allergen Protein in Mature Soybean Seed

  • Soybean proteins are widely used for human and animal feeds worldwide. The use of soybean protein has been expanded in the food industry due to their excellent nutritional benefits. But, antinutritional and allergenic factors are present in the raw mature soybean. P34 protein, referred as Gly m Bd 30K, has been identified as a predominant immunodominant allergen. The objective of this research is to identify the genetic mode of P34 protein for the improvement of soybean cultivar with a very low level of P34 protein. Two $F_2$ populations were developed from the cross of "Pungsannamulkong" ${\times}$ PI567476 and "Gaechuck2ho" ${\times}$ PI567476 (very low level of P34 protein). Relative amount of P34 protein was observed by Western blot analysis. The observed data for the progeny of "Pungsannamulkong" and PI567476 were 133 seeds with normal content of P34 protein and 35 seeds with very low level of P34 protein (${\chi}^2=1.157$, P=0.20-0.30). For the progeny of "Gaechuck#1" and PI567476, the observed data were 177 seeds with normal content of P34 protein and 73 seeds with very low level of P34 protein (${\chi}^2=2.353$, P=0.10-0.20). From pooled data, observed data were 310 seeds with normal content of P34 protein and 108 seeds with very low level of P34 protein (${\chi}^2=0.156$, P=0.50-0.70). The segregation ratio (3:1) and the Chi-square value obtained from the two populations suggested that P34 protein in mature soybean seed is controlled by a single major gene. Single gene inheritance of P34 protein was confirmed in 32 $F_2$ derived lines in $F_3$ seeds, which were germinated from the low level of P34 protein obtained from the cross of "Pungsannamulkong" and PI567476. These results may provide valuable information to breed for new soybean line with low level of P34 protein and identification of molecular markers linked to P34 locus.

  • Twelve URP primers were used to assess genetic characteristics of oyster mushroom including 59 Pleurotsu ostreatus cultivars, two of P. florida cultivars, one P. sajor-caju cultivars, one P. abalonus cultivar and two P. eryngii cultivars registered in Korea. Six URP primers produced PCR polymorphic bands within and between the Pleurotus species. Primer URP2F produced distinct cultivar specific PCR polymorphic bands that profiled to 15 cultivar types. PCR polymorphic bands amplified by URP2F, URP6R, URP4R and URP2R were used for UPGMA cluster analysis. Fifty nine cultivars of Pleurotus ostreatus are genetically clustered into 5 groups, showing genetic similarity over 70% among them and P. abalonus. P. eryngii and P. sajor-caju, were involved in outside groups.

Correlation of saponarin content with biosynthesis-related gene expression in hulled and hulless barley (Hordeum vulgare L.) cultivars

  • Saponarin found in young barley sprouts has a variety of beneficial biological and pharmacological properties, including antioxidant, hypoglycemic, antimicrobial, and hepatoprotective activities. Our previous work demonstrated that saponarin content was correlated with the expression levels of three biosynthetic pathway genes [chalcone synthase (HvCHS1), chalcone isomerase (HvCHI), and UDP-Glc:isovitexin 7-O-glucosyltransferase (HvOGT1)] in young barley seedlings under various abiotic stress conditions. In this study, we investigated the saponarin content and expression levels of three saponarin biosynthetic pathway genes in hulled and hulless domestic barley cultivars. In the early developmental stages, some hulled barley cultivars (Kunalbori1 and Heukdahyang) had much higher saponarin contents than did the hulless barley cultivars. An RNA expression analysis showed that in most barley cultivars, decreased saponarin content correlated with reduced expression of HvCHS1 and HvCHI, but not HvOGT1. Heat map analysis revealed both specific increases in HvCHS1 expression in certain hulled and hulless barley cultivars, as well as general changes that occurred during the different developmental stages of each barley cultivar. In summary, our results provide a molecular genetic basis for the metabolic engineering of barley plants to enhance their saponarin content.

Characterization of Rice Mutants with Enhanced Susceptibility to Rice Blast

  • As a first step towards identifying genes involving in the signal transduction pathways mediating rice blast resistance, we isolated 3 mutants lines that showed enhanced susceptibility to rice blast KJ105 (91-033) from a T-DNA insertion library of the japonica rice cultivar, Hwayeong. Since none of the susceptible phenotypes co-segregated with the T-DNA insertion we adapted a map-based cloning strategy to isolate the gene(s) responsible for the enhanced susceptibility of the Hwayeong mutants. A genetic mapping population was produced by crossing the resistant wild type Hwayeong with the susceptible cultivar, Nagdong. Chi-square analysis of the $F_2$ segregating population indicated that resistance in Hwayeong was controlled by a single major gene that we tentatively named Pi-hy. Randomly selected susceptible plants in the $F_2$ population were used to build an initial map of Pi-hy. The SSLP marker RM2265 on chromosome 2 was closely linked to resistance. High resolution mapping using 105 $F_2$ plants revealed that the resistance gene was tightly linked, or identical, to Pib, a resistance gene with a nucleotide binding sequence and leucine-rich repeats (NB-LRR) previously isolated. Sequence analysis of the Pib locus amplified from three susceptible mutants revealed lesions within this gene, demonstrating that the Pi-hy gene is Pib. The Pib mutations in 1D-22-10-13, 1D-54-16-8, and 1C-143-16-1 were, respectively, a missense mutation in the conserved NB domain 3, a nonsense mutation in the 5th LRR, and a nonsense mutation in the C terminus following the LRRs that causes a small deletion of the C terminus. These findings provide evidence that NB domain 3 and the C terminus are required for full activity of the plant R gene. They also suggest that alterations of the resistance gene can cause major differences in pathogen specificity by affecting interactions with an avirulence factor.
