Mixed-type Inhibition of Human Hepatic Cytochrome P450 1-Catalyzed Ethoxyresorufin O-deethylation by Volatile Allyl Sulfides

  • Effects of allyl sulfides on kinetic behavior of cytochrome P450 1 (CYP1)-catalyzed ethoxyresorufin O-deethylase (EROD) activity were studied using microsomes from benzo[a]pyrene-treated human hepatoma cells. Apparent $K_m$ and $V_{max}$ values were calculated as $2.8\;{\mu}M$ and $3.0\;{\mu}mol$ resorufin/min/mg protein based on Lineweaver-Burk plot of microsomal EROD activity, respectively. Diallyl disulfide (DADS) and diallyl trisulfide (DATS) affected $K_m$ and $V_{max}$ values of EROD activity and acted as mixed-type inhibitors for CYP1 isozymes. Apparent Ki values of DADS and DATS were calculated as 1.07 and 0.88 mM, respectively, by re-plotting slopes of Lineweaver-Burk plot and inhibitor concentrations.

Combelen 과량투여(過量投與)가 산양(山羊)에 미치는 영향 (Influence of Overdosed Combelen on Korean Native Goats)

  • In order to detect the influence of overdosed combelen on the Korean native goats, 5mg of combelen per kg of body weight were injected intramusculary. Clinical findings, electrocardiograms and some blood chemical values were determined and the results obtained were as follows: 1. After administration of 5 mg/kg dose of combelen the animals were moderately depressed and often tried to take sitting position but showed moderate response to pinching stimulation of skin. 2. Body temperature and respiratory rate were not suppressed but heart rate were accelerated during sedative stage. 3. During sedation a slight change in the QRS complex and T wave appeared but it was not significant. 4. Hemoglobin concentration showed a tendency to decrease. SGOT activities were markedly increased 24 hours after administration of combelen and BUN values were slightly lowered. Blood sugar values were markedly elevated during sedative stage.

  • In this study, we compared Oxygen Index by ISO 4589-2 with Fire Spread by ISO 5658-2. The result shows that the tset values of Fire Spread such as IT, CFE, Qsb have mutual relations with thoses of Oxygen Index values in some samples. And we found that the tset values of Fire Spread make up for the insufficient points of Oxygen Index has its limits to evaluate the properties of fire for the interior materials of the railroad vehicles.

  • This paper describes a proposal for crack width and deflection in RC flexural members. Because the serviceability provisions of the current codes are mainly based on only empirical relationships developed from test result and effective moment of inertia, crack width and deflections are contrary to the actual values. Based on nonlinear bond characteristics, tension stiffening effect, arch action and effective concrete tensile area. Then an equation is developed for predicting crack width and deflection in flexural members. The predicted results shows that as proposed model employed, crack width and deflections are different from estimated by the current KCI, MC 90 and EC 2 provisons, and the values predicted are in good agreement with experimentally measured values.

Purification and Characterization of a Xylanase from Bacillus sp. KK-1

  • A thermophilic Bacillus sp. strain KK-1 isolated from soil produced an extracellular xylanase. From the culture supernatant of Bacillus sp., the xylanase was purified to homogeneity by ammonium sulfate precipitation and DEAE-Sephadex A-50 chromatography. The molecular weight of the purified xylanase was estimated to be 45 kDa by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and gel permeation chromatography. The apparent $K_m$ values for xylanase, using oat spelt xylan and birchwood xylan as substrates, were 7.1 mg/ml and 3.2 mg/ml, and $V_{max}$ values were $27.0\;{\mu}mol{\cdot}min^{-1}{\cdot}mg^{-1}$ and $29.0\;{\mu}mol{\cdot}min^{-1}{\cdot}mg^{-1}$, respectively. The xylanase hydrolyzed oat spelt xylan to mostly xylobiose, xylotriose, and xylose. The amino acid composition indicated that the xylanase contained high amounts of amino add residues of glutamic acid and glutamine (Glx) and aspartic acid and asparagine (Asx).

  • Proteolytic activity of live Uronema manum was analyzed by fluorescence polarization (FP) technique. Protease activity was measured by a decrease in FP value using fluorescein isothiocynate (FITC)-casein as a protein substrate. The results demonstrated an inverse linear relationship between fluorescence polarization (FP) values and live ciliate concentration over the range $1\times10^4\;to\;2\times10^5$ cells/well. However, the FP values of $10-10^3$ live parasites were not different significantly from that of control. Time-dependent decrease in FP value was shown in the wells containing live U. marinum. In the present study, FP assay had the benefit to provide measurements of substrate hydrolysis by live parasites in real-time, and did not require separations, precipitations, or transfers of reaction mixture.

  • $Cu(In_xGa_{1-x})Se_2$ thin films were prepared and characterized with various Ga contents. As the Ga content increased, the grain size of CIGS film became smaller. The 2 $\theta$ values in XRD patterns were shifted to larger values and the overlapped peaks were splitted. The energy bandgap increased from 1.04 to 1.67 eV and the resistivity decreased. The solar cell fabricated with ZnO/CdS/$Cu(In_{0.7}Ga_{0.3})Se_2/Mo$ structure yielded an efficeincy of 14.48% with an acitive area of 0.18 $cm^2$. The efficiency decreased with further increase of Ga content.

Effect of Fermented Spinach as Sources of Pre-Converted Nitrite on Color Development of Cured Pork Loin

  • The effect of fermented spinach extracts on color development in cured meats was investigated in this study. The pH values of raw cured meats without addition of fermented spinach extract or nitrite (negative control) were higher (p<0.05) than those added with fermented spinach extract. The pH values of raw and cooked cured meats in treatment groups were decreased with increasing addition levels of fermented spinach extract. The lightness and yellowness values of raw cured meats formulated with fermented spinach extract were higher (p<0.05) than those of the control groups (both positive and negative controls). The redness values of cooked cured meats were increased with increasing fermented spinach extract levels, whereas the yellowness values of cooked cured meats were decreased with increasing levels of fermented spinach extract. The lowest volatile basic nitrogen (VBN) and thiobarbituric acid reactive substances (TBARS) values were observed in the positive control group with addition of nitrite. TBARS values of cured meats added with fermented spinach extract were decreased with increasing levels of fermented spinach extract and VBN values of curing meat with 30% fermented spinach extract was lower than the other treatments. Total viable bacterial counts in cured meats added with fermented spinach extract ranged from 0.34-1.01 Log CFU/g. E. coli and coliform bacteria were not observed in any of the cured meats treated with fermented spinach extracts or nitrite. Residual nitrite contents in treatment groups were increased with increasing levels of fermented spinach extract added. These results demonstrated that fermented spinach could be added to meat products to improve own curing characteristics.
