금강밀과 dark northern spring밀의 기울과 배아에서 추출한 기름의 자동산화 안정성 및 미량성분 비교 (Comparison of Autoxidative Stability and Minor Compounds in Oils Extracted from Bran and Germ of Keumkang Wheat and Dark Northern Spring Wheat)

  • 금강밀 또는 Dark Northern Spring(DNS)밀의 기울과 배아로부터 추출한 유지(WBG유)를 빛을 차단하고 $50^{\circ}C$에서 12일 동안 저장하면서 과산화물값, 지방산조성 변화로 자동산화안정성을 평가하였고 토코페롤과 토코트리엔올, 카로티노이드, 인지질 함량 변화도 함께 측정하였다. 금강밀과 DNS밀에서 추출한 WBG유는 초기 과산화물값에 뚜렷한 차이를 보이지 않았으나 금강밀 WBG유는 DNS밀 WBG유에 비해 높은 유리지방산값을 보였다. 또한 금강밀 WBG유는 DNS밀 WBG유보다 리놀레산이, DNS밀 WBG유는 금강밀 WBG유보다 올레산이 유의하게 더 많았다. 인지질 함량은 DNS밀 WBG유에서 많았으며 토코페롤과 카로티노이드 함량은 반대 경향을 보였다. 금강밀과 DNS밀에서 추출한 WBG유는 산화기간이 증가함에 따라 과산화물값이 유의하게 증가하였으며, 산화속도는 두 기름 사이에 유의한 차이가 없었다. 금강밀과 DNS밀 WBG유의 자동산화 중 카로티노이드와 인지질은 모두 유의한 차이를 보이며 분해되었고, 인지질함량이 WBG유의 자동산화정도와 높은 상관관계를 보여주었다.

Genomics Approach to Identify the Cause of the Missing Omega-5 Gliadin Protein in O-Free Wheat

  • A previous work developed and identified a new omega-5 gliadin deficient wheat line named O-free by crossing Keumkang and Olgeuru, which is nutritionally quite meaningful in that omega-5 gliadin is one of the known wheat allergens. To verify the characteristics of the O-free, we performed RNA sequencing (RNAseq) analysis of the O-free and the two parent lines (Keumkang and Olgeuru). The results of the similarity analysis with the ESTs for gliadins and glutenins showed that the O-free ESTs had no similarity with the omega-5 gliadin sequences but had similarity to other gliadins and glutenins. Furthermore, mapping results between the raw RNAseq data from the O-free and the omega-5 gliadin sequence showed a clear deletion of the N-terminal sequences which are an important signature of omega-5 gliadin. We also designed specific PCR primers that could identify omega-5 gliadin in the genomic DNA. The results showed that no omega-5 gliadin fragments were detected in the O-free. According to these results, we confirmed that the deficiency of omega-5 gliadin in the O-free is not caused by post-transcriptional or post-translational regulations such as epigenetic phenomena but by a simple deletion in the chromosome. Furthermore, we showed that the low-molecular weight glutenin subunit (LMW-GS) gene in the O-free had a single nucleotide polymorphism (SNP) causing a premature stop codon, resulting in a truncated polypeptide. We expect that the O-free line may serve as an excellent source of wheat that could prevail in the hypo-allergen wheat market, which has recently gained interest world-wide.

호남지역 논 재배에 적합한 곡실 사료용 밀 품종 선발 및 사료가치 평가 (Selection and Feed Value Evaluation of Wheat for Grain Feed at the paddy field in Honam Region)

  • 본 연구는 호남지역 유휴 논토양에서 곡실 사료용 밀 생산에 적합한 품종 선발을 위해 국내산 총 8개(금강, 조경, 백중, 수안, 청우, 고소, 호중, 조중)의 품종을 선택하여 전북 완주군에서 2015년부터 2016년까지 파종 및 수확하였으며, 사료가치 평가를 위해 일반성분 및 반추위 소화율 분석을 실시하였다. 시험품종 중 출수기는 "조중"이 4월 15일로 가장 빨랐으며, 성숙기 또한 5월 24일로 가장 빨랐다. "백중"의 출수기는 4월 18일로 "금강"과 같았고, 성숙기는 5월 29일로 하루 늦었지만 전 시험품종 간의 유의성은 없었다. 경수는 "고소"가 513개/㎡로 가장 적었으며, "청우"가 974개/㎡로 가장 많았다. "백중"의 경수는 901개/㎡로 시험품종 중 경수가 많은 품종에 속하며, 수확량은 542kg/10a으로 가장 많았다(p<0.05). 일반성분 분석, 반추위내 건물 및 조단백질 소화율 평가에서 시험품종 간 유의적인 차이는 보이지 않았으며, 옥수수보다 높은 영양 가치와 소화율을 확인하였다. 따라서 상기 8개 밀 품종은 공히 수입곡물을 대체 할 수 있는 훌륭한 사료자원이며, 수확량이 높은 품종을 선택하여 곡실 사료로의 이용성을 확대하면, 논경지이용효율 증대와 더불어 축산농가 사료비 절감에 도움이 될 것으로 사료된다.

밀 품종 혼파와 밀가루 혼합에 따른 품질 및 가공적성 분석 (Analysis of Quality and Processing Suitability of Mixed Seeding and Flour Blending between Wheat Varieties)

  • 본 연구는 국산밀의 생산량 증대와 밀가루 품질 균일성 증진을 위해 용도가 다른 금강밀과 백중밀 두 가지 밀 품종의 혼합파종 재배와 밀가루 혼합에 따른 품질과 가공특성을 분석하였고 결과를 요약하면 다음과 같다. 밀 혼파 재배에 따른 수량은 품종이 나타내는 고유 수량에 따라서만 종실 수량이 결정 되었는데, 이는 금강밀과 백중밀의 간장과 수장의 차이가 거의 없어 입지 공간의 증대에 따른 수량 증가는 없고 단순히 단위면적당 수수의 차이로만 수량이 결정된 것으로 보인다. 품질특성은 단백질 함량, 침전가, 손상전분 함량은 혼파 및 밀가루 혼합 둘 다 비율에 따라 품질의 변이의 경향이 같게 나타났고, 특히 밀가루 혼합이 더욱 예측 가능하고 비례적인 품질 변이 결과를 보였다. 밀가루 용매 보유 능력(SRC) 분석 결과 금강밀이 백중밀에 비해 물 흡수도와 손상전분이 적고 글루텐 강도와 arabinoxylan 기여도는 큰 것으로 관찰되었고, 밀가루 혼합의 시료는 비율에 따라 유의성 있게 차이가 났지만 혼파에서 얻은 밀가루는 유의적인 차이를 나타내지 못하였다. Mixograph 반죽 패턴은 금강밀이 피크 높이가 크고 밴드 넓이가 커서 단백질, 침전가, 젖산SRC 값과 같은 경향을 나타내었고, 혼파에 비해 밀가루 혼합에서 더욱 비례적으로 밴드가 변화되는 것을 관찰 할 수 있었다. 이러한 결과를 보아 생산성을 제외하고 품질 균일성을 위해서는 혼파보다는 밀가루 혼합이 더욱 유리하였고, 생산된 밀가루의 여러 가지 품질 특성을 고려 한 후 밀가루 혼합 비율을 조절한다면 원하는 가공적성을 지닌 균일한 품질의 밀가루를 안정적으로 생산 할 수 있을 것으로 판단된다.

Screening Wheat Germplasm for high Amylose Content

  • Wheat (Triticum spp.) is an important source of food worldwide and the focus of considerable efforts to identify new combinations of genetic diversity for crop improvement. In particular, wheat starch composition is a major target for changes that could benefit human health. Starches with increased levels of amylose are of interest because of the correlation between high amylose content and elevated levels of resistant starch, which has been shown to have beneficial effects on health for combating obesity and diabetes. In this study, high amylose wheat germplasms from other countries were collected and cultivated in Korea, and then the content of amylose was evaluated, we examined amylose content in 614 wheat germplasm. Furthermore, amylose content was validated using several milling processes such as roller, hammer, and grinding mill. As a result, the amylose content distribution was divided into five groups. The range of the amylose levels in whole wheat flour was 18.3% to 29.6%. In addition, the mutant lines were screened for high amylose, and two mutant lines (WX-1046 and WX-1074) exhibited a comparable amylose content to Keumkang whole wheat (19.6%). It has been established that high amylose indicated SS IIa null and necessitate GBSS. Based on these findings, it may be helpful to develop high amylose wheat germplasm and production techniques, particularly in Korea.

Substituting Normal and Waxy-Type Whole Wheat Flour on Dough and Baking Properties

  • Normal (cv. Keumkang, KK) and waxy-type (cv. Shinmichal, SMC) whole wheat flour was substituted at 20 and 40% for white wheat flour (WF) during bread dough formulation. The flour blends were subjected to dough and baking property measurement in terms of particle size distribution, dough mixing, bread loaf volume and crumb firmness. The particle size of white wheat flour was the finest, with increasing coarseness as the level of whole wheat flour increased. Substitution of whole wheat flour decreased pasting viscosity, showing all RVA parameters were the lowest in SMC40 composite flour. Water absorption was slightly higher with 40% whole wheat flour regardless of whether the wheat was normal or waxy. An increased mixing time was observed when higher levels of KK flour were substituted, but the opposite reaction occurred when SMC flour was substituted at the same levels. Bread loaf volume was lower in breads containing a whole wheat flour substitution compared to bread containing only white wheat flour. No significant difference in bread loaf volume was observed between normal and waxy whole flour, but the bread crumb firmness was significantly lower in breads containing waxy flour. The results of these studies indicate that up to 40% whole wheat flour substitution could be considered a practical option with respect to functional qualities. Also, replacing waxy whole flour has a positive effect on bread formulation over normal whole wheat flour in terms of improving softness and glutinous texture.

Evaluation of Stomatal Characteristics of Adaxial and Abaxial Side of Flag Leaves of Korean Wheat Cultivars

  • Stomatal traits such as stomata density (SD), aperture length (APL) and width (APW), guard cell length (GCL) and width (GCW), and distance between stomata (DIS) were investigated to identify correlation with agronomic traits for 35 Korean wheat cultivars. Flag leaf width (FLW) of Korean wheat cultivars was the widest in Ol-mil, and the narrowest in Keumkang. SD tended to be higher on the adaxial side than on the abaxial side in Korean wheat cultivars. SD of adaxial and abaxial sides was classified into a cultivar with a significantly different or not. In APL, 18 wheat cultivars showed significant differences according to leaf side, and APL of adaxial was longer than APL of abaxial in 13 wheat cultivars. In APW, 15 wheat cultivars showed a significant difference, and APW of abaxial was wider than APW of adaxial among them. In GCL, 14 wheat cultivars showed a significant difference, and the GCL of abaxial was longer than the GCL of adaxial in 10 wheat cultivars. In GCW, 10 wheat cultivars showed a significant difference, GCW of adaxial was wider than GCW of abaxial and in 6 wheat cultivars. FLW of adaxial and abaxial showed a negative correlation with GCL and a positive correlation with grain number per panicle. FLW of only abaxial showed a positive correlation with DIS. The SD of the adaxial showed a negative correlation with GCL, while the SD of the abaxial showed a negative correlation with APL. APL of both sides of the leaf showed a positive correlation with GCL, and APW of only abaxial showed a negative correlation with GLC. DIS of adaxial showed a negative correlation with tiller number (TN), while DIS of abaxial showed a positive correlation with GNP.

Analysis of Differentially Expressed Genes Between Leaves and Grain Tissues of Three Wheat Cultivars

  • Wheat is a very important crop as a food source worldwide, but gluten in wheat causes a variety of allergic reactions. Previous studies have developed ${\omega}-5$ gliadin deleted O-free, known as the central antigen of WDEIA (wheat-dependent exercise-induced anaphylaxis). In this study, we performed RNA sequencing on the grains and leaves of the allergic-reduced species O-free and their cultivars, Keumkang and Olgeuru, to analyze differentially expressed genes (DEG) based on different cultivars and tissues. Tissues of all species were biologically repeated three times. We used bowtie2 version 2.3.5.1 to get sequence data from RNAseq and used cufflinks and Tophat programs to find DEG. When comparing leaf and grain tissues, a total of 1,244 DEGs were found in the leaf tissues while only 563 DEGs were found in the grain tissues. As a result of gene ontology analysis of differentially expressed genes, the leaf tissues were mostly included in the "catalytic activity" part of molecular function, "metabolic process" part of biological process, and "membrane" part of cell component. The grain tissues were mostly included in the "metabolic process" part of biological process, "binding" and "catalytic activity" part of molecular function, and "membrane, cell, cell part" parts of cell component. Based on these results, we present information on the differentially expressed genes of the three cultivars of leaves and grains. This study could be an important basis for studying the characteriztion of O-free.

Comparison of Milling and Flour Quality Characteristics of Foreign Wheat and Korean Wheat

  • This study was investigated to compare the milling and physicochemical characteristics of six Korean wheat cultivars (Keumkang, KK; Jokyung, JK; Goso, GS; Joongmo2008, JM; Baekkang, BK; Saekeumkang, SKK) and five foreign wheat classes (Australian standard white wheat, ASW; Australian hard, AH; US northern spring, NS; US hard red winter, HRW; Soft wheat, SW). Korea and foreign wheat grains were milled using a Buhler MLU-202. Flour moisture, ash, protein, gluten, sedimentation, particle size, solvent retention capacity (SRC) and dough properties of flour were analyzed. Results showed that the hard wheats had a greater total flour yield and reduction fraction yield than the soft wheats regardless of the country. However, there were in the milling characteristics between the US and Korean soft wheats. GS, a soft wheat in Korea, had the lowest flour yield (59.6%) and the highest bran fraction yield (21.4%). The particle sizes of flour by milling fraction were B1>B2>B3 for the largest, and the R1〈R2〈R3 for the smallest. Particle size, ash, protein contents and the values of lactic acid SRC showed highly correlated with flour yield. The gluten-performance-index (GPI) is the ratio of the lactic acid SRC value to the sum of sodium carbonate and sucrose SRC values, and it has been used as a quality indicator for overall performance potential of flour. GPI values differed depending on the wheat variety or class, JM (0.82) was the highest value, and SKK (0.56) and SW (0.59) were low. The curve pattern of the Mixolab result also gives a quality indication of the flour sample. JM and NS flour had similar pattern at water absorption and gluten strength parameters and BK and HRW had similar viscosity patterns. These results will enable further study for blending Korean wheat cultivar to improve the flour quality.

Analysis of agricultural characteristics and qualities of CIMMYT resources in seeding time

  • We received 73 wheat resources from CIMMYT for analysis of agricultural characteristics and qualities of them. We sowed 73 lines and 4 Korean varieties in fall and spring to compare differences of wheat traits between fall and spring seeding. The heading and maturity date of fall seeding lines has coming faster average 18 days than spring seeding lines. Fall seeding lines have more number of spike per $m^2$ and grain number of spike. Spike and awn length of spring seeding lines were shorter than fall seeding lines and stem length was variable to varieties in both condition. Protein contents of five of 73 lines, Jokyoung and Jopoom varieties decreased but the others increased. The sedimentation values of 34 lines include Jokyoung and falling number (F/N) of 44 lines include two varieties, Keumkang and Jokyoung, decreased but the others increased, respectively. Particle size was presented that most spring seeding lines were bigger. However, when it sowed in fall, the lightness (L) of wheat flour was lighter. Consequently, we confirmed when it was sowed in spring, agricultural traits and qualities of wheat got weak.
