The Effect of Cultivation Period on Growth Characteristics , Palatability and Forage Yield of Soybean Cultivars (대두 품종별 재배기간이 생육특성 , 기호성 및 수량에 미치는 영향)

  • This experiment was canied out to select the soybean of high productivity suited for soiling. and growth characterisitcs, palatability, dry matter and crude protein yield of 10 cultivars of soybean have been evaluated at the experimental forage field. College of Natural Science of Kon-Kuk University in Chungju over three years (1 989-1 09 1 ). The results obtained are summarized as follows: JangBaek showed the tallest plant length from the 70th day to 80th day during cultivation period. and BaekUn showed the tallest at 90th day. PalDal among all cultivars showed the fastest growth rate and the strongest resistant lodging during cultivation period. The stem diameter of 70th day showed highly in order of PalDal and HwangKeum > BaekUn > KwangKyo and JangYeob, the 80th day showed highly in the order of TogYu > BaekCheon > JangBaek and 90th day showed highly in order of BangSa > BaekCheon > JangYeob. Stem hardness was highly increased with increasing of cultivation period, but no significant difference were found among the soybean cultivan. Especially, BaekUn and TogYu of 90th day showed very high as 17.9 and 17.6 kg/cm2, respectively. In the number of branch, PalDal was the lowest during cultivation period(70, 80 and 90th day), and 90th day showed lower two and three times than other soybeans(P<0.05). In the leaf rate, BaekCheon showed the highest according to cultivation period and the lowest in JangBaek. The fresh yield was tended to increase by increasing cultivation period at all cultivars except KwangKyo, but no significant differences were found among cultivars. Dry matter yield of JangYeob was the highest as 5, 861 kg/ ha at 70th day, and that of KwangKyo and TogYu was the highest as 7, 471 and 10, 603 kgha at 80th and 90th day, respectively. Relative palatability of 70th days showed very high at all soybean cultivars(81-100%), and that of 80th and 90th day showed the highest at TogYu, while showed the lowest at PalDal. In crude protein content of the 70th, 80th and 90th day, DanYeob, JangYeob and HwangKeum were the highest as 20.1, 17.9 and 20.4 percentage, respectively. Crude protein yield during cultivation period showed the highest at JangYeob(7Oth : 1, 068, 80th : 1, 211, 90th : 1, 882 kglha), but showed the lowest at BangSa(70th : 61 1, 80th : 814, 90th : 1, 078 kg/ha).

  • In this study, we observed the effects of repeated perms on the morphology of virgin hair of a healthy thirteen year-old girl with scanning electron microscopy. After the first treatment, the outer parts of cuticle cell were broken unevenly and roughly. Cuticle cells were lifted upward making a space. After the third treatment, cuticle cells were lifted off one another and the folded scales showed irregular surface areas. Broken pieces of cells were stuck on the surface and an empty hole was present in the endocuticle of the cytoplasm. We observed that cortex separated from cuticle layer more easilywith repeated treatments.

Probing the Critical Residues for Intramolecular Fructosyl Transfer Reaction of a Levan Fructotransferase

  • Levan fructotransferase (LFTase) preferentially catalyzes the transfructosylation reaction in addition to levan hydrolysis, whereas other levan-degrading enzymes hydrolyze levan into a levan-oligosaccharide and fructose. Based on sequence comparisons and enzymatic properties, the fructosyl transfer activity of LFTase is proposed to have evolved from levanase. In order to probe the residues that are critical to the intramolecular fructosyl transfer reaction of the Microbacterium sp. AL-210 LFTase, an error-prone PCR mutagenesis process was carried out, and the mutants that led to a shift in activity from transfructosylation towards hydrolysis of levan were screened by the DNS method. After two rounds of mutagenesis, TLC and HPLC analyses of the reaction products by the selected mutants revealed two major products; one is a di-D-fructose-2,6':6,2'-dianhydride (DFAIV) and the other is a levanbiose. The newly detected levanbiose corresponds to the reaction product from LFTase lacking transferring activity. Two mutants (2-F8 and 2-G9) showed a high yield of levanbiose (38-40%) compared with the wild-type enzyme, and thus behaved as levanases. Sequence analysis of the individual mutants responsible for the enhanced hydrolytic activity indicated that Asn-85 was highly involved in the transfructosylation activity of LFTase.

If spleen and stomach cause lack of Youngwi(營衛) harmorny losing function, used Gyejitang (桂技揚) Because spleen course stomach is weak, so, because there are been a lot of cold liquids, if vomiting catches continuously, used Geonganginsambanhahwan(乾薑附子半夏九). When treat pregnancy stomachache because Yang(陽) is weak, it is cold and make to be warm by Bujatang(附子楊) in case have a stomachache. Because Chungimmaek(衝任脈) weak and cold, use Gyoaetang(膠艾揚) in case there are stomachache and bleeding. Because liver and spleen do not harmonize, use Dangguijakyaksan(當歸芍藥散) if stomachache and vertigo occur. ect. There is no contents that foster embryo, but I am thought by something affected greatly at future generations.

Glycosylation Enhances the Physicochemical Properties of Caffeic Acid Phenethyl Ester

  • In this study, we synthesized a glycosylated derivative of caffeic acid phenethyl ester (CAPE) using the amylosucrase from Deinococcus geothermalis with sucrose as a substrate and examined its solubility, chemical stability, and anti-inflammatory activity. Nuclear magnetic resonance spectroscopy showed that the resulting glycosylated CAPE (G-CAPE) was the new compound caffeic acid phenethyl ester-4-O-${\alpha}-{\small{D}}$-glucopyranoside. G-CAPE was 770 times more soluble than CAPE and highly stable in Dulbecco's modified Eagle's medium and buffered solutions, as estimated by its half-life. The glycosylation of CAPE did not significantly affect its anti-inflammatory activity, which was assessed by examining lipopolysaccharide-induced nitric oxide production and using a nuclear factor erythroid 2-related factor 2 reporter assay. Furthermore, a cellular uptake experiment using high-performance liquid chromatography analysis of the cell-free extracts of RAW 264.7 cells demonstrated that G-CAPE was gradually converted to CAPE within the cells. These results demonstrate that the glycosylation of CAPE increases its bioavailability by helping to protect this vital molecule from chemical or enzymatic oxidation, indicating that G-CAPE is a promising candidate for prodrug therapy.

Interpretation of Agronomic Traits Variation of Sesame Cultivar Using Principal Component Analysis

  • This study was conducted to evaluate the growth characters and yield components of 18 collected sesame cultivars to get basic information on the variation for the sesame breeding using principal component analysis. All characters except days to flowering, days to maturity and 1,000 seed weight showed significantly different. Seed weight per 10 are showed higher coefficient of variance. Capsule bearing stem length and liter weight showed positive correlation with seed yield per 10 are. The principal components analysis grouped the estimated sesame cultivars into four main components which accounted for 83.7% of the total variation at the eigenvalue and its contribution to total variation obtained from principal component analysis. The first principal component ($Z_1$) was applicable to increase plant height, capsule bearing stem length and 1,000-seed weight. The second principal component ($Z_2$) negatively correlated with days to flowering and maturity by which it was applicable to shorten flowering and maturity date of sesame. At the scatter diagram, Yangbaek, Ansan, M1, M2, M4, M7 and M9 were classified as same group, but M10, Yanghuk, Kanghuk, M5, M6, M12 and M13 were classified as different group. This results would be helpful for sesame breeder to understand genetic relationship of some agronomic characters and select promising cross lines for the development of new sesame variety.

Genotype $\times$ Environment Interaction for Yield in Sesame (Sesamum indicum L.)

  • Application of genotype by environment ($G\;{\times}\;E$) interaction would be used for identifying optimum test condition of the varietal adaptation in the establishment of breeding purpose. Yield and yield components were used to perform additive main effect and multiplicative interaction (AMMI) analysis. Significant difference for $G\;{\times}\;E$ interaction were observed for all variable examined. For yield, 0.18 of total sum of squares corresponded to $G\;{\times}\;E$ interaction. Correlation analysis was carried out between genotypic scores of the first interaction principal component axis (IPCA 1) for agronomic characters. Significant correlations were observed between IPCA 1 for yield and capsule bearing stem length (CBSL), number of capsule per plant (NOC). The biplot of grain yield means for IPCA1 which accounted for 34% of the variation in total treatment sums of squares showed different reaction according to $G\;{\times}\;E$ interaction, genotypes and environments. Taegu showed relatively lower positive IPCA1 scores, and it also showed smaller coefficient variation of yield mean where it is recommendable as a optimal site for the sesame cultivar adaptation and evaluation trial. In case of variables, Yangbaek and M1 showed relatively lower IPCA1 scores, but the score direction showed opposite each other on the graph. Ansan, Miryang1, Miryang4, and Miryang6 seemed to be similar group in view of yield response against IPCA1 scores. These results will be helpful to select experimental site for sesame in Korea to minimize $G\;{\times}\;E$ interaction for the selection of promising genotype with higher stability.

Temporal and Spatial Characteristics of Water Quality in a River-Reservoir (Paldang) (하천형 호수인 팔당호 수질의 시공간적 특성)

  • This study is to investigate the allochthonous load and water quality of a typical river-reservoir, Paldang during spring (March ~ May) of 17 years (2001 ~ 2017). Phosphorus loading from point sources seems to have been reduced by 74 % in the 2010s. As a result, trophic state of the Paldang reservoir, eutrophic during the 2000s, has returned to the lmesotrophic state. Along with decrease in phosphorus concentration, standing crops of algae (Chl.a) decreased, and concentration of biodegradable organic material decreased to the past level. Concentration of total suspended solids has decreased, and it is due to the decrease of phytoplankton standing crops since the mid-2000s. As transparency increased, it is estimated that euphotic area increased by 22 % and euphotic capacity expanded by 27 %. In the river/transition zone of Paldang, concentration of organic matter increases slightly due to algal growth, but concentration of all water quality items decreases in the lacustrine zone. Although algal growth rate revealed positive correlation with concentration of phosphorus, it was insignificant. Algal growth appeared to be dependent on renewal of phosphorus by flow, than either flow rate or phosphorus concentration. The empirical model including inflow phytoplankton concentration fit well with observed values, and indicates the Paldang reservoir is greatly influenced by allochthonous loads.

Analysis of the Cytotoxicity of Green Pigment Produced on the Surface of Roasted and Retorted Chestnuts Using Immune Cells and Gastrointestinal Cancer Cells

  • Roasted and retorted (RR) chestnuts develop green pigment spots on their surface during storage. The purpose of this study was to evaluate the cytotoxicity of the green pigment using RAW 264.7, MOLT-4, KATOIII and HT-29 cells. The pigment scraped from RR chestnuts (GP), whole RR chestnuts with green pigment spots (GC), whole RR chestnuts without green pigment (WC) and roasted and frozen stored chestnuts (FC) were extracted in 10% DMSO. MOLT-4 cells were less resistant to the cytotoxicity of the chestnut extracts than the RAW 264.7 cells. The GP extracts did not show different responses against $H_2O_2$-induced oxidative stress and LPS-induced NO production compared to the other extracts. The chestnut extracts did not have proliferative activity on either of the KATOIII or HT-29 cells (p>0.05). Our results from the comparison of the green pigment produced on the surface of the RR chestnuts to chestnuts that do not develop the green pigment suggest that the pigment may not be harmful in terms of either cytotoxicity towards immune cells or proliferation of gastrointestinal cancer cells.
