식물체의 플라보노이드 성분 함량 분석에 필요한 적정 가수분해 조건을 구명하기 위하여 은행잎을 이용하여 중심합성법으로 설계한 산 가수분해 시간과 HCl 농도에 따라 myricetin, quercetin, kaempferol 함량을 HPLC로 분석하고, SAS의 반응표면분석법으로 산 가수분해 조건의 최적화를 시도하였다. HCl 0.5~2.5 M, 0.5~2.5시간 범위 내에서 10개의 구간을 잡아서 최적화한 결과 1.5~1.9 M의 HCl 농도와 1.4~2.0시간의 가수분해 조건에서 myricetin, quercetin, kaempferol 성분이 가장 높았다. Superimposing하여 구한 식물체의 플라보노이드의 적정 가수분해 조건은 HCl 1.5 M, 1.5시간이었으며, 이 조건에서 구한 플라보노이드 함량의 예측치와 측정치는 95% 이상 일치하였다. 본 연구에서 구한 적정 가수분해 조건을 이용하여 녹차잎, 무화과잎, 상동나무잎의 플라보노이드 함량을 분석하여 보았다.
Dimeric ${\beta}$-cyclodextrin linked by a thioether bridge was synthesized from a reaction of mono-6-iodo-6-deoxy-${\beta}$-cyclodextrin with sodium sulfide, and the structure was analyzed using nuclear magnetic resonance spectroscopy and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. The effects of thioether-bridged dimeric ${\beta}$-CD on the aqueous solubility of flavonols (myricetin, quercetin, and kaempferol) were investigated by ultraviolet-visible spectroscopy. The aqueous solubility of myricetin, quercetin, and kaempferol were enhanced 33.6-, 12.4-, and 10.5-fold following the addition of 9 mM of thioether-bridged dimeric ${\beta}$-CD. In comparison, the aqueous solubility of myricetin, quercetin, and kaempferol were enhanced 5.4-, 3.3-, and 2.7-fold using the same concentration of monomeric ${\beta}$-cyclodextrin. Furthermore, the formation of flavonol/thioether-bridged dimeric ${\beta}$-CD inclusion complexes was confirmed with nuclear magnetic resonance, Fourier-transform infrared spectroscopy, differential scanning calorimetry, and scanning electron microscopy. The results showed that the nature of the complexes significantly differed from that of free flavonols. Herein, we suggest that the thioether-bridged dimeric ${\beta}$-CD can act as an effective complexing agent for flavonols.

The peel of astringent persimmon (Diospyros kaki Thunb. cv. Cheongdo-Bansi) is a by-product of dried persimmon (gotgam). We investigated if deastringent peel extracts of persimmon cv. Cheongdo-Bansi had antioxidative and neuroprotective properties. Two different extracts were prepared: thermally and nonthermally treated persimmon peel extracts (TPE and NTPE, respectively). Both TPE and NTPE were fractionated sequentially in n-hexane, chloroform, ethyl acetate, n-butanol, and water. The TPE and NTPE ethyl acetate fractions had the highest total phenolic and flavonoid contents as well as antioxidant capacities among all the fractions. Pretreatment of neuronal PC-12 and SH-SY5Y cells with the TPE and NTPE ethyl acetate fractions increased cell viability after exposure to oxidative stress. The ethyl acetate fraction of TPE attenuated oxidative stress inside both PC-12 and SH-SY5Y cells more effectively than that of NTPE. Furthermore, the TPE and NTPE ethyl acetate fractions inhibited acetylcholinesterase and butyrylcholinesterase. Analysis of ultra-high-performance liquid chromatography-electrospray ionization-tandem mass spectrometry results revealed gallic acid, kaempferol, kaempferol-3-O-galactoside, kaempferol-3-O-glucoside, quercetin, quercetin3-O-galactoside, quercetin-3-O-galactoside-2'-O-gallate, and quercetin-3-O-glucoside as the major phenolics of the TPE and NTPE ethyl acetate fractions. Taken together, these results suggest that the ethyl acetate fraction of deastringent persimmon peel is rich in antioxidants and has potential as a functional food to reduce oxidative stress.
In the course of screening for antioxidant compounds by measuring the DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging effect and superoxide quenching activity, methanol extract of Cyperus flaccidus (Cyperaceae) was found to show potent antioxidant activities. Subsequent activity-guided fractionation of the methanolic extract led to the isolation of ten compounds, kaempferol-3,7-O-${\alpha}$-L-dirhamnopyranoside (1), caffeic acid (2), quercetin-3-O-${\alpha}$-L-rhamnopyranosyl($1{\rightarrow}6$)-${\beta}$-D-glucopyranoside (3), kaempferol-3-O-${\alpha}$-L-rhamnopyranoside (4), quercetin-3-O-${\alpha}$-L-rhamnopyranoside (5), luteoiln-7-O-${\beta}$-D-glucopyranoside (6), kaempferol-3-O-${\alpha}$-L-rhamnopyranosyl($1{\rightarrow}6$)-${\beta}$-D-glucopyranoside (7), luteoiln (8), quercetin (9) and quercetin-3-O-${\beta}$-D-glucuronide (10). Their structures were elucidated by spectroscopic studies. Compounds 1 - 10 were isolated for the first time from this plant. Among them, compounds 2, 8 and 9 showed the significant radical scavenging effects on DPPH, and the potent xanthine-originated superoxide quenching activities.
오동나무 열매를 채취하여 건조시킨 후 분말로 제조하여 5.0 kg을 아세톤-물 (7:3, v/v) 혼합용액으로 추출하고 유기용매를 제거한 후 헥산, 메틸렌클로라이드, 에틸아세테이트 및 물 등 네 개의 분획으로 분리하여 에틸아세테이트용성 분획물을 Sephadex LH- 20으로 칼럼 크로마토그래피를 수행하였으며 용리용매는 메탄올 수용액과 에탄올-헥산 혼합액을 사용하였다. 그 결과 phenolic acid 화합물인 caffeic acid와 flavonoid 화합물인 naringenin, kaempferol, apigenin, luteolin 및 quercetin이 단리 되었으며 NMR 및 MS 스펙트럼을 이용하여 구조를 결정하였다. 각 분획물과 단리 된 화합물들 그리고 조추출물은 DPPH radical 소거법을 이용하여 항산화 실험을 실시하였으며, 분획물중에는 에틸아세테이트용성이 그리고 화합물중에는 caffeic acid, kaempferol luteolin 및 quercetin이 우수한 항산화 효능을 나타내었다.
This research was conducted in order to understand the hybridization between Quercus aliena Blume and Q. serrata Murray in Korea which show wide range of morphological variations within species and interspecific variations of diverse overlapping characteristics caused by hybridization. Morphological analysis (principal components analysis; PCA) of 116 individuals representing two species and their intermediates were performed. As a result, two species were clearly distinguished in terms of morphology, and intermediate morpho-types assumed to be hybrids between the two species were mostly located in the middle of each parent species in the plot of the principal components analysis. There was a clear distinction between two species in trichome distribution pattern which is an important diagnostic character in taxonomy of genus Quercus, whereas intermediate morpho-types showed intermediate state between two species' trichome distributions. Forty-two individuals representing two species and their intermediates were examined for leaf flavonoid constituents. Twenty-three flavonoid compounds were isolated and identified: They were glycosylated derivatives of flavonols, kaempferol, quercetin, isorhamnetin and myricetin. The flavonoid constituents of Q. aliena were five glycosylated derivatives: kaempferol 3-O-galactoside, kaempferol 3-O-glucoside, quercetin 3-O-galactoside, quercetin 3-O-glucoside, and Isorhamnetin 3-O-glucoside. The flavonoid constituents of Q. serrata had 20 diverse flavonol compounds including five flavonoid compounds found in Q. aliena. It was found that there is a clear difference in flavonoid constituents of Q. aliena and Q. serrata. Flavonoid chemistry is very useful in recognizing each species and putative hybrids. The flavonoid constituents of intermediates were a mixture of the two species' constituents and they generally showed similar characteristics to morpho-types. The hybrids between Q. aliena and Q. serrata showed morphologically and chemically diverse characteristics and it is assumed that there are frequent interspecific hybridization and introgression.
본 연구는 포도수확 후 버려지는 가지를 이용한 포도전정가지 추출물(Grape Pruning Stem Extracts, GPSE)에 함유된 polyphenol류 중 항산화, 항염증, 항암 등의 효과가 있다고 알려진 rutin, procyanidin B3, quercetin, kaempferol의 함량을 분석하고, UVB로 유도된 HR-1 mice의 손상된 피부에 대한 교원섬유, 탄력섬유 파괴억제 효과를 측정하여 기능성 화장품 소재로서의 응용 가능성을 확인하고자 하였다. 포도전정가지에서 polyphenol 성분을 80% EtOH로 추출하고 여과하여 농축한 후, 동결건조하여 $-20^{\circ}C$에 보관하면서 사용하였다. GPSE의 유효성분 함량은 HPLC를 사용하여 분석하였다. 피부손상을 유도하기 위해 UVB를 HR-1 hairless mice에 조사하였고, 손상된 피부에 교원섬유 파괴억제 효과를 보기 위해 Masson's trichrome stain, 탄력섬유 파괴 억제 효과를 보기 위해 Verhoeff's elastic fiber stain, MMP-9 단백 발현을 보기 위해 면역조직화학적 염색을 실시하였다. 본 연구에 사용된 포도전정가지 시료 53 kg에서 EtOH 분획 추출물 2.34 kg을 추출하여 4.42%의 수율 결과를 얻었다. 유효성분 분석결과, procyanidin B3, 0.28 mg/g, rutin 12.81 mg/g, quercetin 0.51 mg/g 및 kaempferol 8.24 mg/g로 나타났다. GPSE와 serum base를 혼합 도포한 그룹에서 UVB에 의한 교원섬유와 탄력섬유를 분해하는 MMP-9 단백발현 결과, 대조군 대비 평균 약 90% 감소 억제하는 효과를 볼 수 있었다. 교원섬유 파괴억제 효과는 모든 농도에서 평균 24%의 억제율과, GPSE와 serum base의 혼합물 농도 2,000 mg/kg 에서 43%로 가장 우수한 교원섬유 파괴억제효과가 나타났으며, 탄력섬유파괴 억제 효과는 모든 농도에서 평균 12%의 억제율과 GPSE와 serum base의 혼합물 농도 1,000 mg/kg에서 35%로 가장 우수한 탄력섬유 파괴 억제 효과가 있는 것으로 나타났다. 이 결과는 GPSE가 진피층의 세포 외 기질에 UVB 조사에 의해 파괴되는 교원섬유, 탄력섬유의 파괴를 억제하는 작용을 한다는 것을 알 수 있었다. 이상의 결과로 포도전정가지로부터 rutin, kaempferol, quercetin, procyanidin B3 등의 폴리페놀성 화합물이 화장품 기초 원료인 serum base에 함유토록 제조된 GPSE 는 교원섬유 파괴 억제, 탄력섬유파괴 억제효과 및 피부손상 기능개선 효과의 결과를 얻을 수 있었다. 본 연구의 의의는 지역포도농가에서 수확 후 버려지는 포도전정가지에서 폴리페놀류와 같은 유효성분을 확보하여 화장품 원료로서의 활용 가치가 높을 것으로 기대된다.
본 연구에서는 우리나라 설악산 지역을 중심으로 신갈나무와 졸참나무 두 종의 수직분포 양상을 관찰하고, 두 종간의 교잡이입 및 유전자 전달 가능성을 플라보노이드 분석을 통해 추론하고자 하였다. 중부지방인 설악산의 신갈나무와 졸참나무의 수직분포는 남부지방인 지리산에서와는 차이가 난다. 설악산의 경우, 해발 100 m 이상 거의 전 고도에서 신갈나무가 나타나나 졸참나무는 해발 500 m 이상에서는 드물게 나타나며 500 m 이하에서 신갈나무와 혼생한다. 지리산의 경우, 졸참나무의 수직 분포 범위는 해발 0-1,200 m로 설악산에 비해 훨씬 넓으며, 신갈나무는 높은 해발고도에서는 순림을 이루지만 해발 1,000 m 이하에서는 고도가 낮아질수록 빈도가 낮아지고 해발 300 m 이하에서는 거의 분포하지 않는다. 전반적으로 우리나라에서 신갈나무는 높은 고도에서, 졸참나무는 낮은 고도에서 생육하나 상당한 범위의 고도 구간에서 두 종은 혼생한다. 설악산 지역의 신갈나무와 졸참나무 32개체와 지리산 고지대 및 저지대에 분포하는 신갈나무, 졸참나무 각 1개체 등 총 34개체를 대상으로 잎의 플라보노이드 성분을 분석한 결과, 이들로부터 총 24종류의 서로 다른 화합물이 분리, 동정되었다. 이들 플라보노이드 화합물은 모두 flavonol인 kaempferol, quercetin, myricetin 및 isorhamnetin에 당이 결합된 flavonol glycoside이었으며, 이 중 5개는 acylated flavonoid compound이다. 이들 중 kaempferol 3-O-glucoside, quercetin 3-O-glucoside와 quercetin 3-O-galactoside 및 이들의 acylated compounds가 주요 성분으로 두 종의 모든 개체에서 나타났다. 신갈나무의 플라보노이드 조성은 acylated kaempferol 3-O-glucoside, acylated quercetin 3-O-galactoside 및 acylated quercetin 3-O-glucoside가 다량 나타나고, 졸참나무에서는 나타나지 않는 diglycoside인 quercetin 3-O-arabinosylglucoside가 분포하는 특징을 가진다. 졸참나무의 flavonoid 조성은 3개의 rhamnosyl flavonol compounds가 전체 졸참나무 개체에 걸쳐서 나타나고 또한 이들 compound가 신갈나무에 비해 다량으로 나타나는 특징을 갖는다. 신갈나무와 졸참나무 두 종간에는 이러한 정량 정성적인 플라보노이드 조성 차이와 함께 소량으로 분포하는 몇몇 compound들에 있어서 정성적인 차이도 나타나 두 종은 플라보노이드 조성에 있어 뚜렷이 구분된다. 한편, 두 종의 플라보노이드 조성은 고도에 따라 종내 개체 간 변이가 있으며, 동소적으로 분포하는 두 종 개체들의 플라보노이드 조성은 대체로 정량적 또는 정성적으로 상대 종의 플라보노이드 조성을 닮는 경향이 있었다. 이러한 현상은 두 종간에 교잡이입을 통한 유전자 교환이 일어나고 있음을 강하게 암시한다.
잡곡은 대사성 질환의 예방과 치료에 우수한 기능성 식품소재로 인식되고 있다. 국내산 잡곡들(기장, 황금찰수수, 노랑차조, 율무 및 식용피)을 대상으로 항염증 활성을 비교조사하고자, 이들 잡곡의 80% 에탄올 추출물을 이용하여 마우스 대식세포주 RAW264.7을 지질다당류(LPS)로 자극할 때 생성되는 염증 매개인자인 산화질소(NO)의 생성에 미치는 저해능을 조사하였다. 그 결과, 식용피(Echinochloa crus-galli var. frumentacea) 유래의 에탄올 추출물이 LPS에 의해 유도되는 NO의 생성을 저해하는 항염증 활성에 있어서 가장 우수함을 확인하였다. 식용피의 에탄올 추출물로부터 항염증 관련 성분을 규명하기 위해, 80%에탄올 추출물을 물에 녹인 후 n-hexane, methylene chloride (MC), ethyl acetate (EtOAc), n-butanol (BuOH)과 물 분획들로 단계별 분획하고, 각 분획의 항염증 활성을 조사하였다. RAW264.7 세포에 MC 분획($100{\mu}g/ml$)을 전처리하였을 때, LPS처리에 의해 유도되는 iNOS, COX-2, 그리고 친염증성 사이토카인들(IL-$1{\beta}$, IL-6, 및 TNF-${\alpha}$)의 발현이 현저하게 저해되는 것으로 나타났다. 또한, LPS처리에 의해 유도되는 p38MAPK, JNK 및 ERK의 활성화의 경우도 MC 분획에 의해 농도-의존적으로 저해되는 것으로 나타났다. HPLC분석을 통해, 식용피 유래 MC 분획의 항염증 활성은 식용피의 MC 분획 속의 주요성분인 kaempferol과 biochanin A에 기인함을 확인하였다. 이상의 연구 결과는 식용피 및 식용피의 MC 분획이 염증성 질환과 이와 관련된 대사성 질환의 예방과 예후개선에 유리한 기능성 식품소재 개발에 활용될 수 있음을 시사한다

- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
